1.Evaluation of the Combination of APC and DCC Gene Methylation in the Early Diagnosis of Lung Cancer

Objective To investigate the significanceof both the adenomatous polyposis coli (APC)and deleted in colorectal carcinoma (DCC)gene methylation in the early diagnosis of lung cancer.Methods 245 patients with lung cancer and 150 patients with non-malignant lung disease patients and 40 healthy volunteers were drawn for the experiment.A methylation specific PCR (MSP)was used to detect the methylation status of APC and DCC in the peripheral blood.Results The posi-tive rates of APC and DCC genes promoter methylation Peripheral blood of patients with lung cancer were 26.53% (65/245)and 36.33% (89/245),respectively.The positive rates of APC and DCC genes promoter methylation Peripheral blood of patients with benign lung diseases were 2.67% (4/150),8.00%(12/150),respectively.The positive rates of APC and DCC genes promoter methylation Peripheral blood of healthy volunteers were 0.There was a significant difference between patients with the lung cancer,those with the benign lung diseases and Healthy volunteers (P <0.01).The sensitivity and specificity of lung cancer was respectively 52.65% and 89.33% by the combination of APC and DCC genes methylation. Comparing with the separate APC detection,sensitivity and specificity statistical differences were significant in joint meas-urement (P <0.01).Comparing with the separate DCC detection,sensitivity statistical differences were significant,but spe-cificity statistical differences was not obvious (P >0.05).There was no correlation between methylation test results and pa-tient gender,age,pathological type,pathological grade,TNM stage and so on (P >0.05).Conclusion APC and DCC gene methylation were closely related to the development of non-small cell lung cancer.They can be used as an early diagnostic marker of lung cancer.

Five organic compounds were isolated from the marine sponge Axinyssa aplysi- noides collected from South China Sea. Their chemical structures were identified by UV, IR, NMR, and MS as 4-hydroxybenzaldehyde (I); (22E)-24,26,26-trimethylcholesta-6,22,25 (30)-trien-3?-ol(Ⅱ); 3- (17Z-tetracose-nyloxy)-1, 2-propanediol (Ⅲ); 24-isobutylcholesta-3, 25(31)-dien-3?-ol(Ⅳ); Uracil (V). It's the first report about the marine sponge Axinyssa aplysinoides from South China Sea.
3.Clinical Study on Bushen Huoxue Decoction for Elderly Patients with Chronic Cerebral Circulation Insufficiency

Objective To observe the effects of Bushen Huoxue Decoction on clinical symptom, vertigo degree and cognitive function for elderly patients with chronic cerebral circulation insufficiency (CCCI); To explore its possible mechanism. Methods Totally 70 cases were randomly divided into observation group and control group according to the random digits table method, with 35 cases in each group. The two groups were given Western medicine consisting of blood pressure control, glucose control and blood lipid control. Based on this, the control group was given Flunarizine Hydrochloride capsules, 5 mg once a day orally before sleeping. The observation group was given Bushen Huoxue Decoction, one dose a day and taken twice a day. The treatment lasted for 30 days. The TCM symptom score, Chinese version of dizziness handicap inventory (DHI) and Montreal cognitive assessment (MoCA) score were observed. The mean blood flow velocity (Vm) of basilar artery (BA), bilateral vertebral artery (VA), bilateral middle cerebral artery (MCA) and the content of plasma acid phospholipid (AP) were detected, and the clinical efficacy was evaluated. Results The total effective rate of the observation group was 94.3% (33/35), which was significantly higher than that of 82.9% (29/35) in the control group, with statistical significance (P<0.05); after treatment, the TCM symptom score, DHI score, and the content of plasma AP were significantly lower than those of pre-treatment in the same group (P<0.05); the MoCA score and the Vm of VA, LBA, RBA, LMCA, RMCV were significantly higher than those of pre-treatment in the same group (P<0.05); the TCM symptom score, DHI score, MoCA score, the Vm of LVA, LMCA, RMCA and the content of plasma AP in the observation group were improved better than those in the control group, with statistical significance (P<0.05). Conclusion Bushen Huoxue Decoction has an obvious therapeutic effects for elderly patients with CCCI, which can improve clinical symptom, vertigo degree and cognitive level, which possible mechanism is related to increasing the cerebral blood flow velocity and reducing the content of plasma AP.

5.Correlation between depressive symptoms, chronic diseases and daily activities in the elderly

Objective To analyze the correlation between chronic diseases,depression and activities of daily living (ADL) in the elderly,and to explore the risk factors of depressive symptoms and ADL.Methods Randomized multi-stage sampling method was used to select 504 elderly (aged ≥65 years old) in Jining City.The participants were then investigated and assessed by using Geriatric Depression Scale15,Katz-Activity of Daily Living (Katz-ADL) and Lawton instrumental ADL (Lawton-IADL).Results The prevalence of chronic diseases was 74.01% among the elderly who lived in Jining City.The positive rate of depressive symptom was 7.14% and the rate of ADL or IADL limitation was 19.05% and 66.67%,respectively.The positive rate of depressive symptom and the rate of ADL or IADL limitation of the male were lower than those of female (x2 values were 18.15,12.31 and 30.18,respectively; all P< 0.05).The number of chronic diseases was a risk factor of depressive symptom and ADL limitation (odds ratio (OR) =1.71 or 1.53,respectively).ADL and IADL limitations were also the risk factors of depressive symptom (OR =3.15 or 9.36,respectively).Over adjustment of gender and age,the impact of the number of chronic diseases on depressive symptom,ADL and IADL and the influence of ADL and IADL limitations on depressive symptom showed no statistical significance.Conclusions Gender and age may be fundamental risk factors of depressive symptom and ADL and IADL limitations in the elderly.Family members and society workers should improve ADL and positive emotion and reduce the likelihood of depressive symptom among aging population.
6.The analysis and countermeasures of postoperative wound infection in patients who undergo the aseptic surgery of urinary system

Objective To investigate the influencing factors and preventive measures of postoperative wound infection in patients who undergo the aseptic surgery of urinary system.Methods Clinical data of 302 cases of patients underwent the aseptic surgery of the urinary system between January 2011 to May 2013 were reviewed and studied by Logistic regression analysis.Results Wound infection occurred in 17 cases,the incidence of wound infection was 5.6% among 302 patients in this study.Postoperative wound infection risk factors were open surgery,operative time ≥ 2 h,to replace the drainage bag every one or two days once,and with some other underlying diseases.Conclusions To strengthen the basic disease treatment,select endoscopic operation if the surgery and treatment of disease allowed,shorten the operation time,reduce the number of drainage tube and reduce the frequency of replacement of drainage bag may effectively prevent postoperative wound infection in patients who undergo the aseptic surgery of the urinary system.
7.Investigation of risk factors for mild cognitive impairment in elderly people

Objective To investigate the risk factors for mild cognitive impairment (MCI) in elderly people in Beijing.Methods Using multistage cluster random sampling,129 elderly people aged 60-80 years living around Dongzhimen communities were interviewed by mini-mental state examination (MMSE) and National Institute of Neurological and Communicative Diseases and Stroke/ Alzheimer's Disease and Related Disorders Association (NINCDS-ADRDA) criteria for screening MCI and Alzheimer's disease (AD).Results Thirty-seven cases (28.7%) had amnestic MCI (aMCI),thirty-six cases (27.9%) had AD,and fifty-six cases (43.4%) were with normal cognitive state (NCS).The age was older in aMCI patients or AD patients than in NCS[(67.6±7.5)y vs.(62.5±7.9)y,(67.6±7.5)y vs.(62.5±7.9)y,both P<0.01],Andthe aMCI or AD patients had low level of education (P<0.05).The blood pressure was higher in the patients suffered from aMCI or AD than in people with NCS more or less (P<0.05).The prevalence of aMCI was related to the body mass index (BMI) (P<0.05),while that of AD had no significant relation with BMI (P>0.05).The prevalence of aMCI or AD was not significantly different between male and female or between different birth months.(all P>0.05).Conclusions The prevalence of aMCI increases with age,lower level of education,higher level of blood pressure and BMI,while it has no significant relations with gender or birth month.

AIM: To study the relationship between the activity of phospholipase A 2 (PLA 2) and pulmonary structural remodeling with the model of chronic isobaric hypoxic pulmonary hypertension. METHODS: 29 healthy Sprague-Dawley rats were randomly divided into normal control group, chronic hypoxic group and hypoxia plus Polidatin (PD) group. By diameter, the arteries were divided into two groups: arteries of group I (30 ?m-100 ?m) and group II (101 ?m-200 ?m). The mean pulmonary arterial pressure (mPAP) was measured by inserting a microcatheter into the pulmonary artery. The PLA 2 activity was measured with modified microtitrimetic method. The pulmonary tissue and arterioles morphology changes were examined under light microscope. AIM:To establish the methods of determining 2,3,5,4′-tetrahydroxystilbene-2-O-?-D-glycoside,puerain;ginsengnoside Rg_1,ginsengnoside Rb_1,notoginsengnoside R_1,and salvianolic B in Jiangzhi Jianfei Tablets(Radix Polygoni multiflori,Radix Puerariae lobatae,Radix et Rhizoma Notoginseng,Radix et Rhizoma Salviae miltiorrhizae,etc) with HPLC. METHODS: All chromatogram columns used were ODS columns.The mobile phase of acetonitrile-wate(25(∶)75) was chosen for 2,3,5,4′-tetrahydroxystilbene-2-?-D-glycoside.The flow rate was 1.0 mL/min.The detection wavelength was at 320 nm.The mobile phase of methanol-water(25(∶)75) was chosen for puerain.The flow rate was 1.0 mL/min.The detection wavelength was at 250 nm.The mobile phase of acetonitrile-water was chosen for ginsengnoside Rg_1,ginsengnoside Rb_1 and notoginsengnoside R_1.The flow rate was 1.0 mL/min.The detection wavelength was at 203 nm.The mobile phase of methanol-acetonitrile-methane acid-water(30(∶)10(∶)1(∶)59) was chosen for salvianolic B.The flow rate was 1.0 mL/min.The detection wavelength was 286 nm. RESULTS: The linear rangers were 0.476 ?g-4.760 ?g(r=0.999 6) for 2,3,5,4′-tetrahydroxystilbene-2-?-D-glycoside,0.168 96 ?g-1.689 6 ?g(r=0.999 8) for puerain,0.724 ?g-7.240 ?g(r=0.999 8) for ginsengnoside Rg_1,0.728 ?g-7.280 ?g(r=0.999 9) for ginsengnoside Rb_1,0.23 ?g-2.30 ?g(r=0.999 8) for notoginsengnoside R_1,0.306 ?g-3.060 ?g(r=0.999 9) for salvianolic B.Their average recoveries were 99.43%(RSD=1.08%,n=6),99.43%(RSD=1.66%,n=6),101.17%(RSD=2.08%,n=6),and 102.39%(RSD=2.60%,n=6),respectively. CONCLUSION: These methods of determining components in four kinds of herbs are simple,accurate,reproducible and can be used for quality control of Jiangzhi Jianfei Tablets.

AIM: To investigate the effects of polidatin on phospholipase A_2(PLA_2),nitric oxide(NO),and endothelin-1(ET-1) levels in serum and lung homogenate of hypoxic pulmonary hypertension rats.METHODS: 29 healthy Spraugue-Dawley rats were randomly divided into normal control group,chronic hypoxic group and hypoxic plus polidatin group.The model of chronic hypoxic pulmonary hypertension was made by method of intermittent isobaric hypoxia for 21 days.The mean pulmonary arterial pressure(mPAP) was measured by inserting a microcatheter into the pulmonary artery.The ratio of right ventricular wall and that of left ventricular wall and ventricular septum weight(R/L+S) was measured as well.RESULTS: After exposed to hypoxia for 21 days,the mPAP,R/L+S,PLA_2 and ET-1 activities in blood and lung homogenate increased significantly. The NO concentration decreased significantly.Pretreatment with PD attenuated these changes mentioned above.CONCLUSION: Pretreatment with PD effectively prevents the formation of chronic hypoxic pulmonary hypertension.Its mechanism may be related to the inhibition of PLA_2 and ET-1 and promotion of NO production.
