1.Extraction of anti-thrombotic contituent from Earthworm (Pheretima)

AIM: To investigate the extraction of anti-thrombotic contituent from Earthworm ( Pheretima). METHODS: The prothrombin time (PT)of rabbit blood and antithrombin of 14 samples that were extracted by different methods were determined in vitro. RESULTS: The sample that was percolated with 70% alcohol and the sample that was extracted by semi-bionic method were better than ones that were extracted by other methods. CONCLUSION: It is a simple and practicable method to percolate Earthworm with 70% alcohol.
2.Invasion ability and collagenolytic activity of human glioma cells in vitro

Objective To investigate the invasion ability and collagenolytic activity of human glioma cells in vitro.Methods Boyden chamber invasion assay was employed to evaluate the cell migration ability of human malignant glioma cell line U87MG in vitro and the effect of conditioned medium of U87MG cells on matrix collagenolytic activity was tested by agar-gelatin gel.Results The number of U87MG glioma cells migrating through the Matrigel-coated membrane was more than that of addition of EDTA or anti-MMP-2 antibody in U87MG glioma cells(P0.05).EDTA or anti-MMP-2 antibody markedly inhibited the number of the migrated cells,the inhibitory rates were 79.2% and 77.1%,respectively. The conditioned medium collected from the U87MG cells showed an increase in the transparent ring area on agar-gelatin gel.Inhibition of MMP-2 enzymatic activity by EDTA or anti-MMP-2 antibody reduced the transparent ring area on agar-gelatin gel.Conclusion Both invasion ability and collagenolytic activity of U87MG cells are depended on MMP-2,suggesting that MMP-2 plays an important role in glioma invasiveness.
3.Position nursing for neonates with type three congenital esophageal atresia after operation

Objective To discuss the position nursing for neonates with type three congenital esophageal atresia after operation. Methods We selected 10 patients with type three congenital esophageal atresia from September 2004 to May 2007.The respirator, trachea catheter was removed after operation and the SpO2 was lower than 95% through head mask.Then the SpO2 was compared in different positions of patients,that is face forward,face lateral, face forward with hypsokinesis,face lateral with hypsokinesis. Results All the ten patients survived the operation.The Sp02 in the four positions under oxygen inhalation were (90.30±0.95)%, (89.90±0.74)%, (96.80±1.48)%, (95.90±0.88)%.The former two results were statistically different from the latter two results (P < 0.01). Conclusions The position of face forward with hypsokinesis and face lateral with hypsokinesis facilitated the transition to natural respiration for patients with type three congenital esophageal atresia after remove of respirator and trachea catheter.This could prevent the incidence of complications and the probability of reuse of respirator.
4.Differentiation of bone marrow mesenchymal stem cells into chondrocytes induced by transforming growth factor beta 1

BACKGROUND: Transforming growth factor beta 1 (TGF-β1) is a first selected growth factor in study of articular cartilage tissue engineering. Suitable concentration can stimulate proliferation, division and differentiation of articular chondrocytes. OBJECTIVE: To establish an special inducing system for differentiation of bone marrow mesenchymal stem cells (BMSCs) into chondrocytes in contained TGF-β1 culture in vivo,and to observe the changes in cell form and phenotype.METHODS: BMSCs were separated and purified from rabbit tibial tubercle using attachment culture method. Surface antigen of the third-generated BMSCs was determined by using flow cytometry. Subsequently,the third-generated BMSCs were induced with TGF-β1-contained special inducing system in 21-day culture, which was compared with human septal cartilage cells of nose following induction. Collagen type Ⅱ was qualitatively determined using immunohistochemical method.RESULTS AND CONCLUSION: Rabbit BMSCs were separated and purified using attachment culture method. The third-generated BMSCs were positive for surface antigen CD44,but negative for surface antigen CD34 and CD45. Cell form was irregular 21 days after induction. Immunohistochemical staining for type II collagen demonstrated positive cells. Results suggested that under culture system containing TGF-β1 BMSCs may directionally differentiate into chondrocytes,and there were not significant differences with normal chondrocytes.
5.Feasibility of differentiating human umbilical cord blood stem cells into chondrocytes induced by rabbit chondrocytes

BACKGROUND: Embryonic stem cells or bone marrow mesenchymal stem cells possess potential to treat cartilage defects, but the practicability is restricted by many factors. Therefore, it is significant to explore a new source of cells.OBJECTIVE: To investigate the feasibility of differentiating human umbilical cord blood stem cells into chondrocytes. METHODS: Human mesenchymal stem cells was obtained from term delivered umbilical vein, labeled by BrdU, and co-cultured with rabbit chondrocytes using Transwell method. The harvested cells were identified by immunocytochemistrical analysis. RESULTS AND CONCLUSION: The human umbilical cord blood stem cells were differentiated into chondrocytes under induction of rabbit chondrocytes, cytoplasm of which were stained into purple, and cellular nucleus was yellow stained with BrdU. The results revealed that human umbilical cord blood stem cells can in vitro differentiate into chondrocytes.
6.Evaluation of the primary outcomes of minimally invasive lumbar microdiscectomy

Objective To evaluate the primary outcomes of minimally invasive lumbar microdiscectomy. Methods There were 34 cases with single lumbar disc herniation treatment of tubular retractor access with METRx system and microdiscectomy(MMD group)and same cases with treatment of conventional discectomy(CD group).Operating time,blood loss,time of leaving the bed and length of hospital stay were compared in two groups.The postoperative wound pain was measured using a visual analog scale(VAS).The modified MacNab criteria were used to evaluate the outcomes. ResuIts The mean operating time was 65 min in MMD group,63 min in CD group.The mean blood loss was 34 ml in MMD group,85 ml in CD group.To compare in two groups,there was significant difference(P<0.05).The mean time of leaving the bed after operation was 3.2 d in MMD group.5.8 d in CD group.The mean length of hospital stay was 5.8 d in MMD group.9.6 d in CD group.The mean VAS of the postoperative wound pain was 3.2 scores in MMD group,5.6 scores in CD group.To compare in two groups,there was significant difference(P<0.05).There was one complication in MMD group,4 in CD group.Thirty-three cases got follow-up mean 8.5 months in MMD group.The success rate was 93.9%. Conclusion The minimally invasive lumbar microdiscectomy has the advantages on less trauma,safety,quickly postoperative recovery,and satisfactory outcomes.

8.Research on the relationship between the expression of estrogen receptor, progesterone receptor and epidermal growth factor receptor in uterine leiomyoma and the recurrence after myomectomy

Objective To investigate the relationship between the expression of estrogen receptor (ER),progesterone receptor(PR)and epidermal growth factor receptor(EGFR)in uterine leiomyoma and the recurrence after myomectomy.Methods Eighty cases underwent myomectomy were enrolled in this study.ER,PR and EGFR expression in uterine leiomyoma tissue and normal myometrium tissue were examined by immunohistochemical technique.All the patients were followed up regularly after myomectomy.Results The positive expression rate of ER,PR and EGFR in uterine leiomyoma tissue were significantly higher than those in normal myometrium tissue[88.8%(71/80)vs.48.8%(39/80),92.5%(74/80)vs.30.0% (24/80),86.2%(69/80)vs.57.5%(46/80),P < 0.05].The recurrence rates in ER,PR and EGFR positive expression patients were significantly higher than those in negative expression patients[42.3%(30/71)vs.22.2%(2/9),41.9%(31/74)vs.16.7%(1/6),42.0%(29/69)vs.27.3%(3/11),P < 0.05].The recurrence times in ER,PR and EGFR positive expression patients were significantly shorter than those in negative expression patients(P <0.05).The expression of EGFR had positive correlation with ER and PR(r =0.837,0.702,P < 0.05).Conclusions The recurrence of uterine leiomyoma may have correlation with high expression of ER,PR and EGFR in uterine leiomyoma.
9.Effects of Astragalus Granules on the Cav-3 and Smad3 Expression in the Myocardial Cells of Rats with Viral Myocarditis

OBJECTIVE:To study the effects of Astragalus granules on the expression of Caveolin-3(Cav-3)and Smad family member 3 (Smad3) in the myocardial cells of rats with viral myocarditis. METHODS:90 rats were randomly divided into a nor-mal group,a model group,a Shenmai injection group [positive drug,0.2 g/(kg·d)] and the groups of low,medium and high-dose Astragalus granules [0.42,0.84,1.68 g/(kg·d)],with 15 rats in each group. The rats in all groups except for the normal group were given CVB3 ip for the establishment of viral myocarditis model. Meanwhile,the rats in the drug administration groups were given corresponding drugs ig,while those in the normal group and the model group were given normal saline ig,for 15 consecu-tive days. 5 rats were selected from each group respectively on the 3rd,9th and 15th days of drug use to take an experiment. For the rats,the pathological change of the cardiac muscle tissue was observed and scored,and the mRNA and protein expression of Cav-3 and Smad3 in the myocardial cells were detected. RESULTS:After 15 days of drug use,compared to the normal group,the rats of the model group had hyperplasia of a large number of cardiac muscle fibers,obvious lesions at cardiac muscle fibers, and significantly higher pathological score and levels of the mRNA and protein expression of Cav-3 and Smad3 in the myocardial cells (P<0.05). Compared to the model group,the rats in the drug administration groups had cardiac muscle tissue lesions improved and had obviously lower pathological score and levels of the mRNA and protein expression of Cav-3 and Smad3 in the myocardial cells(P<0.05). CONCLUSIONS:Astragalus granules can markedly downregulate the gene expression of Cav-3 and Smad3 in the myocardial cells of rats with viral myocarditis,which is inferred as a prevention and treatment mechanism of viral myocarditis.
10.Significance of SF-36 Scale as Main Content of Life Quality Assessment in Efficacy Evaluation System of TCM Treatment for Rheumatoid Arthritis

SF-36 scale has been widely accepted because of its objectivity and scientificity. However, until now its application in TCM treatment for rheumatoid arthritis is not common. This article discussed the significance of SF-36 scale as the main content of life quality assessment in the efficacy evaluation system of TCM treatment for rheumatoid arthritis, which can provide certain references for the promotion of the application in this field.
