1.Effects of different doses of fluvastatin on serum hs -CRP,NT -proBNP in patients with coronary heart disease and chronic heart failure and its clinical efficacy

Objective To explore effects of different doses of fluvastatin on serum hs -CRP,NT -proBNP in patients with coronary heart disease (CHD)and chronic heart failure (CHF)and its clinical efficacy.Methods 120 CHD patients with CHF were randomly divided into three groups,each group included 50 cases.Each group received the therapy of 20mg/d(20mg group),40mg/d(40mg group),and 80mg/d(80mg group),respectively,three groups were treated for 12 weeks.The related indicators were compared among the three groups before and after treat-ment,and the incidence of major cardiovascular events (MACE )and adverse reactions were recorded.Results There was no statistically significant difference between groups before treatment (P >0.05).The serum hs -CRP and BNP of three groups were significantly different after treatment(F =185.956,16.824,all P <0.05).The hs -CRP and NT -proBNP of 80mg group were lower than those of group 20mg and 40mg group (hs -CRP:LSD -t value =3.143,4.113,all P <0.05;NT -proBNP:LSD -t value =2.986,3.765,all P <0.05).The △hs -CRP,△NT -proBNP of C group were higher than those of 20mg group and 40mg group(LSD -t value =4.783,5.819,all P <0.05).The TC,TG,LDL -C of three groups were significantly different after treatment (F =14.402,10.929, 121.968,all P <0.05).The TC,TG,LDL -C of 80mg group were lower than those of 20mg group and 40mg group (LSD -t value =3.538,4.765,4.387,all P <0.05).The ALT,CPK of three groups had no significant differences before and after treatment (P >0.05).The incidence of MACE in 80mg group was lower than that in 20mg group and 40mg group.However,the incidence of three groups had no significant difference after treatment (P >0.05 ). Conclusion The treatment of high -dose fluvastatin can significantly decrease inflammatory of CHD patients with CHF,and also decreases the NT -proBNP level,effectively control lipid levels,the treatment is safe and worthy of clinical recommendations.

3.Differentiation of bone marrow mesenchymal stem cells into chondrocytes induced by transforming growth factor beta 1

BACKGROUND: Transforming growth factor beta 1 (TGF-β1) is a first selected growth factor in study of articular cartilage tissue engineering. Suitable concentration can stimulate proliferation, division and differentiation of articular chondrocytes. OBJECTIVE: To establish an special inducing system for differentiation of bone marrow mesenchymal stem cells (BMSCs) into chondrocytes in contained TGF-β1 culture in vivo,and to observe the changes in cell form and phenotype.METHODS: BMSCs were separated and purified from rabbit tibial tubercle using attachment culture method. Surface antigen of the third-generated BMSCs was determined by using flow cytometry. Subsequently,the third-generated BMSCs were induced with TGF-β1-contained special inducing system in 21-day culture, which was compared with human septal cartilage cells of nose following induction. Collagen type Ⅱ was qualitatively determined using immunohistochemical method.RESULTS AND CONCLUSION: Rabbit BMSCs were separated and purified using attachment culture method. The third-generated BMSCs were positive for surface antigen CD44,but negative for surface antigen CD34 and CD45. Cell form was irregular 21 days after induction. Immunohistochemical staining for type II collagen demonstrated positive cells. Results suggested that under culture system containing TGF-β1 BMSCs may directionally differentiate into chondrocytes,and there were not significant differences with normal chondrocytes.
4.Feasibility of differentiating human umbilical cord blood stem cells into chondrocytes induced by rabbit chondrocytes

BACKGROUND: Embryonic stem cells or bone marrow mesenchymal stem cells possess potential to treat cartilage defects, but the practicability is restricted by many factors. Therefore, it is significant to explore a new source of cells.OBJECTIVE: To investigate the feasibility of differentiating human umbilical cord blood stem cells into chondrocytes. METHODS: Human mesenchymal stem cells was obtained from term delivered umbilical vein, labeled by BrdU, and co-cultured with rabbit chondrocytes using Transwell method. The harvested cells were identified by immunocytochemistrical analysis. RESULTS AND CONCLUSION: The human umbilical cord blood stem cells were differentiated into chondrocytes under induction of rabbit chondrocytes, cytoplasm of which were stained into purple, and cellular nucleus was yellow stained with BrdU. The results revealed that human umbilical cord blood stem cells can in vitro differentiate into chondrocytes.
5.Expression of BDNF and its receptors in polycystic ovary syndrome in rats

Objective To investigate the expression of Brain-derived neurotrophic factor (BDNF)and its receptors mRNAs in ovary tissues of letrozole-induced polycystic ovary syndrome (PCOS)in rats.Methods The SD rats were divided into two groups (moder group and control group),20 rats in each group.The rat models of PCOS were established by letrozole.Serum sex hormone levels were determined by radioimmunoassay.The histologic changes in ovaries were observed by Hematoxylin-eosin stai-ning,and the expression of BDNF and its receptors gene in ovary tissues was detected by Real-time PCR.Results Although the se-rum testosterone,follicle-stimulating hormone and luteinizing hormone levels in model group were markedly increased more than those in the control group (P < 0.01 ),estradiol and progesterone in model group showed a considerable reduction (P < 0.05 ). When compared with the control group,model group rats showed increased ovarian volume and high incidence of subcapsular ovari-an cysts together with decreased number of corpora lutea.The expression of BDNF and p75 mRNAs was significantly higher in model group than that in the control group(P <0.05),but the expression of TrkB mRNA reduced(P <0.05).Conclusion BDNF/TrkB/p75 expressed in ovarian tissues may play a specific role on follicular developmental disorders in letrozole-induced PCOS rats.
6.Study on the risk factors of injuries after Typhoon disaster

Objective To explore the impact on survivors's health and to determine the risk factors involved in the typhoon in order to investigate the characteristics of traumatic symptoms of casualties in the aftermath of the Saomai Typhoon sweeping over the rural area of Zhejiang province in 2006. Method A cluster sampling study was carried out and a total of 389 survivors were assessed. Data were analyzed by univariate and multivariate Logistic analysis to clarify the characteristics and the risk factors of getting traumatize. Results Of 389, there were 113 (29.1% ) victims suffered from a variety of injuries mainly crush trauma (24.4 % ) and wounded by hard object (23.1% ). Moreover, the combined injuries accounted for 9.2%. The risk factors causing injuries in the residential area were the building collapse, glass fragments from broken windows, flying debris as well as fall from high place. Condclusions Outdoor activity and the crumbled building are the leading causes for injuries during the sweeping of typhoon. Meanwhile, we should make more effort to publicize the precautions against typhoon to the local residents.
7.Effect of Xiaozheng Yiliu Decoction on the cytotoxic activity of cisplatin against human ovarian cancerOVCAR/DDP cells and its action mechanism

Objective To study the cytotoxic activity of Chinese compound prescription (Xiaozheng Yiliu Decoction) combined with cisplatin on human ovarian cancer OVCAR/DDP cells and its molecular mechanism. Methods The xenograft model was established by injection of OVCAR/DDP cells. Forty nude mice were randomly allocated into four groupsincluding control group, cisplatin treatment group, low-dose Xiaozheng Yiliu plus cisplatin treatment group,and high-dose Xiaozheng Yiliu plus cisplatin treatment group.The apoptosis was determined by TUNEL analysis.The survivin expression was detected by RT-PCR analysis.Results The inhibit rates in cisplatin treatment group, low-dose Xiaozheng Yiliu plus cisplatin treatment group,and high-dose Xiaozheng Yiliu plus cisplatin treatment group,were(5.56±1.14) %,(18.42±7.73) %,and (31.59±12.58) %,respectively,and there were significant differences among the groups(F=8.64,P<0.01 ).The TUNEL analysis showed that the percentages of cell apoptosis were(2.35±1.04) %,(4.56±1.61) %,(12.42±3.68) %,(21.59±5.17) % in four different groups,respectively.After treated by Xiaozheng Yiliu Decoction,the mRNA expression of survivin was found to be down-regulated by RT-PCR analysis. Conclusion Xiaozheng Yiliu Decoction could iucrease the cytotoxic activity of cisplatin on ovarian cancer OVCAR/DDP cells, which may be related to the down-regulation of survivin gene expression.
8.Expression and prognostic value of TTF-1 of stage Ⅰ non-small cell lung cancer

Objective To study the expression and clinical significance of thyroid transcription factor-1 (TTF-1) in stage Ⅰ non-small cell lung cancer (NSCLC) after operation and to 1 investigate the values in identification of the prognosis of stage Ⅰ NSCLC.Methods The expression of TTF-1 in 129 specimens of stage Ⅰ NSCLC was detected by immunohistochemistry.Results The positive rate of TTF-1 in stage Ⅰ NSCLC was 64.3%.There were significant differences in TTF-1 expression rate among pathological subtypes (x2 =25.231,P < 0.001),different ages (x2 =4.581,P =0.032),sex (x2 =4.900,P =0.027) and differentiation degrees(x2 =11.519,P =0.019).Univariate analysis suggested that in the stage Ⅰ NSCLC patients,the median disease-free survival and overall survival of those positive for TTF-1 expression were superior to those negative:38.9 months versus 27.8 months (P =0.023) and 64.10 months versus 50.68months (P =0.013).Cox regression analysis suggested that TTF-1 expression,tumor differentiation degrees were independent factors affecting the prognosis of stage Ⅰ NSCLC.Conclusion Patients with TTF-1 positive expression often had better survival.Positive expression of TTF-1 and differentiation degree of tumor were independent prognostic factors to affect the prognosis of NSCLC.
9.Factors affecting donation-associated adverse reactions among volunteer blood donors

Objective :
To identify the factors affecting donation-associated adverse reactions among volunteer blood donors, so as to provide the evidence for improving the quality of blood donation services.
Methods :
The volunteer blood donors in Quzhou Blood Center in 2019 were recruited. The Self-rating Anxiety Scale ( SAS ), Social Support Rating Scale ( SSRS ) and Pittsburgh Sleep Quality Index ( PSQI ) scores were measured prior to blood donation, and the presence of donation-associated adverse reactions was observed in the field of blood donation and within one week through short-message-service follow-up. The factors affecting donation-associated adverse reactions were identified using multivariable logistic regression analysis.
Results :
A total of 8 240 volunteer blood donors were enrolled, and adverse reactions occurred in 236 subjects, with an incidence rate of 2.86%. The SSRS, PSQI and SAS scores were 37.68±7.14, 5.97±1.25 and 52.82±3.25 among the donors with adverse reactions, respectively. Multivariable logistic regression analysis identified the educational level ( high school, OR=0.508, 95%CI: 0.289-0.891; diploma and above, OR=0.306, 95%CI: 0.253-0.734 ), SSRS score ( OR=0.399, 95%CI: 0.175-0.913 ), PSQI score ( OR=1.962, 95%CI: 1.116-3.450 ) and SAS score ( OR=2.013, 95%CI: 1.279-3.903 ) as factors affecting donation-associated adverse reactions.
Conclusion
The development of donation-associated adverse reactions correlates with educational level, social support, sleep quality and anxiety among volunteer blood donors.
10.Analysis of prognosis factors of idiopathic sudden hearing loss

Objective:To study the prognosis factors of idiopathic sudden hearing loss. Method:The clinical data of 165 patients with idiopathic sudden hearing loss were retrospectively studied and the computer software was used to investigate the relevant effects on prognosis. Result:The prognosis of patients who had been treated within 14 days under the age of 55 were better. The prognosis of the elderly and the patients with vertigo and high hearing loss were not satisfied. Sex, tinnitus, cardiac or cerebral vascular diseases, diabetic and the medicine or the duration of treatment have no statistical points on prognosis. Conclusion:The prognosis of idiopathic sudden hearing loss related with the treatment time and coverage and extent of the damaged inner ear.
