1.Influence of prophylactic bilateral salpingectomy in ovarian function and its preventive effect on pelvic disease

Objective:To study the influence of prophylactic bilateral salpingectomy during hysterectomy in ovarian function of the premenopausal women with benign uterine disease and its preventive effect on the pelvic diseases(malignant /benign pelvic lesions or pelvic inflammatory disease).Methods:A total of 100 patients with benign uterine disease underwent hysterectomy were collected and divided into two groups,including 50 patients underwent simultaneous hysterectomy and prophylactic bilateral salpingectomy (prevention group) and 50 patients underwent only hysterectomy (control group).The operative time,blood loss,evacuation time,and hospitalization time of the patients in two groups were detected;the antral follicle count,levels of serum estradiol(E2),follicle stimulating hormone(FSH) and luteinizing hormone(LH)) and the incidence of perimenopausal symptoms of the patients in two groups before operation and six months,1 year after operation were compared;the incidence of pelvic disease of the patients after operation were compared.Results:The operative time,blood loss,evacuation time,and hospitalization time of the patients between two groups were not significantly different(P>0.05).Compared with before operation,the antral follicle count and the E2 levels of the patients 6 months and 1 year after operation in two groups were decreased,and the FSH and LH levels of the patients were inc reased (P<0.01).There were no significant differences in the levels of FSH,LH,E2 and the antral follicle count of the patients between two groups 6 months and 1 year after operation(P>0.05).The incidence of perimenopausal symptoms of the patients in two groups 6 months and 1 year after operation showed no significant difference(P>0.05).The incidence of ovarian malignant tumor and ovarian benign tumor of the patients in two groups 6 months and 1 year after operation had no significantly differences (P>0.05).The incidence of pelvic inflammatory disease of the patients in control group was higher than that in prevention group(P<0.05).There were 2 patients diagnosed as tubal carcinoma in control group 1 year after operation,and the pathological findings showed the atypical cells in 2 patients in prevention group.Conclusion:Prophylactic bilateral salpingectomy during hysterectomy does not damage the ovarian function and can reduce the incidence of pelvic malignant/ benign tumor and pelvic inflammatory disease.
2.A Clinical Comparison of the Therapeutic Effect and Safety of Naloxone, Flumazenil and Naloxone Combining Flumazenil in Treatment of Acute Poisoning by Diazepam

Objective To compare the therapeutic effect and safety of naloxone ,flumazenil and naloxone combined wiht flumazenil in treating acute diazepam poisoning and to find out ideal therapeutic treatment.Methods 45 patients were divided into three groups at random: naloxone group (n=15), flumazenil group (n=15) and naloxone combined with flumazenil group (n=15). Besides given the same routine therapy to each group, naloxone group received 0.4mg naloxone through intravenation and 1.2mg naloxone through intravenous drip; flumazenil group received 0.1mg flumazenil through intravenation and 0.9mg flumazenil through intravenous drip; naloxone combined with flumazenil group received 0.4mg naloxone and 0.1mg flumazenil through intravenation and 1.2mg naloxone and 0.9mg flumazenil through intravenous drip. Each patient's consciousness status was graded after given drug treatment 0, 5,15,60,90,180 minute. Testing the BR, SR, CK, CK-MB of every patient and recording to the Bp, HR, RR, SpO 2 and the rhythm of the heart continuously. 3.Endoscopic and endoscopic ultrasonography features of esophageal tuberculosis

Objective To identify endoscopic and the endoscopic ultrasonography (EUS) features of esophageal tuberculosis.Methods We retrospectively analyzed the data of 39 cases (mean age 50.7) of esophageal tuberculosis diagnosed by endoscopy and EUS in past 6 years.Results A total of 29 lesions were found in the middle part of esophagus,and 5 in upper and lower part,respectively.The lesions under endoscope demonstrated as protrusion in 30 and ulceration in 9.EUS found esophageal wall thickness in 9 cases,intra-wall occupying lesion in 17,mediastinum occupying lesions involving esophagus in 13,and calcified lymph nodes in mediastinum which was integrated with esophageal outer wall in 28 cases.Conclusion The esophageal tuberculosis occurs mainly in the middle part of the esophagus,and appears as protrusion and ulceration under endoscopy.EUS can find occupying lesions intra-or out of the esophageal wall,and full layer thickness,which can accompany calcified lymph nodes in meidastinum,and can be the basis of diagnosis.
4.Effect of morphine preconditioning on mitochondrial permeability transition pore after myocardial ischemia-reperfusion injury in rats

Objective To investigate the effect of morphine preconditioning on mitochondrial permeability transition pore (MPTP) and its protective mechanism after myocardial ischemia-reperfusion injury. Methods A rat model of ischemia-reperfusion injury was established. Forty rats were injected with 2-3[H] DOG and then divided into 4 groups randomly: a sham operation (S) group, an ischemia-reperfusion injury (IR) group, a morphine preconditioning (Mp+IR) group, and a cyclosporine A preconditioning (CsA+IR) group. We monitored the concentrations of serum creatine kinase-Mb (CK-Mb) and cardiac troponin I (cTnI), and measured myocardial mitochondrial 2-3[H] DOG, cytochrome c content, Ca2+ concentration ([Ca2+]m), the velocity of Ca2+ intake and reaction half time of mitochondrial permeability transition pore (MPTP t1/2) in the 4 groups. Results The concentrations of serum CK-Mb and cTnI decreased more in the Mp+IR group and the CsA+IR group than those of the IR group. The concentrations of 2-3[H]DOG and [Ca2+]m in the IR group were evidently higher but the level of cytochrome c was lower than those of the sham operation group. The concentrations of 2-3[H] DOG and [Ca2+]m in the Mp+IR group decreased whereas the concentration of cytochrome c increased compared with those in the IR group. Mitochondrial 2-3[H]DOG content was positively correlated with the concentration of calcium (r=0.797, P<0.01). The 2-3[H]DOG and [Ca2+]m content were negatively correlated with cytochrome c in the IR group (r=-0.805 and r=-0.648, respectively, P<0.01). MPTP t1/2 in the IR group was shortened evidently, and that in the Mp+IR and CsA+IR group was significantly lengthened. Conclusion Morphine preconditioning may have myocardial protective effect through unburdening the calcium overload and lengthening the MPTP t1/2.
5.Treatment of axillary hyperhidrosis and bromidrosis with botulinum toxin (BTX)-A injection

Objective To probe the advantages of BTX-A for the treatment of axillary hyper-hidrosis. Methods 42 cases were treated by BTX-A injection, 50 units per axillae, 25 sites with 2 u-nits each, 1.5 cm apart. They consisted of 24 cases of axillary hyperhidrosis, 10 cases of axillary hy-perhidrosis combined bromidrosis, and 8 cases of bromidrosis. Results All hyperhidrosis patients a-chieved good effects. Among patients with axillary hyperhidrosis combined bromidrosis, it had effects on hyperhidrosis, but only one case effected on bromidrosis. Among patients with bromidrosis, only one case had effect. Conclusions In the treatment of hyperhidrosis, BTX-A is very useful, and side effect is temporary and slight. But for bromidrosis, BTX-A is not very useful.

7.THE EXPRESSION OF PROTHYMOSIN ?_1-mRNA IN PREOPTIC AREA OF RAT HYPOTHALAMUS

Objective In order to explore the mechanizms of thymosin action on hypothalamus. Methods RT-PCR and in situ hybridization histochemistry (ISHH)were used. Results The expression of prothymosin ?1-mRNA was detected in preoptic area of hy- pothalamus by using RT-PCR technique. The results of ISHH showed that prothymosin ?1-mRNA was expressed in the preoptic mag- nocellular nucleus, suprachiasmatic nuclei and paraventricular nuclei of hypothalamus. In addition, the positive signal of prothymosin ?1- mRNA was also observed both in the microglicyte near the third ventricle and in medium to small sized pyramidal cells in cerebral cor- tex. Conclusion Prothymosin ?1 is produced in preoptic area of the hypothalamus by means of paracrine, which indicates that prothy- mosin ?1 participates in the regulation of hypothalamic function.
8.Distribution and Drug Susceptibility Analysis of Pathogenic Bacteria in Patients with Cholesteatoma Otitis Media

Objective To explore the distribution and drug resistance of pathogens isolated from patients with cholesteatoma otitis media so as to provide guidance for clinical use of antibiotics.Methods This survey analyzed the spectrum of organisms causing cholesteatoma otitis media and their sensitivities to commonly antimicrobial agents from Hebei province eye hospital in 2014.Results There was 86 positive speciments were cultured from 89 samples,the positive rate was 96.6%.A total of 90 strains of pathogens have been isolated,including 52 strains of gram-positive coccus (57.8%),35 strains of gram-negative bacilli (38.9%),3 strains of gram-positive bacilli (3.3 %),and 0 strain of fungi.Staphylococcus epidermidis,Staphylococcus aureus and Staphylococcus chromogenes ranked the top three species of pathogens,accounting for 20.0%,16.7%,and 12.2%,respectively.The gram-positive cocci were susceptible to vancomycin,rifampicin and amikacin,and showed higher drug-resistancerate to penicillin,amoxicillin and azithromycin.When applied to gram-negative bacilli,the drugs with best resistance were penicillin and cefazolin,and the drugs with the highest sensitivity were levofloxacin and pipercillin/ sulbactam.Conclusion Staphylococcus is the predominant pathogens of cholesteatoma otitis media in hospital,and the bacteria have a high antibiotic resistance.Enhanced monitoring on pathogenic bacteria distribution and drug resistance analysis of cholesteatoma otitis media could benefitthe guide of clinical rational use of antimicrobial agents.
9.Influence of metformin and paclitaxel in proliferation and apoptosis of ovarian cancer SKOV3 cells in vitro

Objective:To study the influence of metformin and paclitaxel in the proliferation and apoptosis of human ovarian cancer SKOV3 cells in vitro, and to clarify the synergistic effect of metformin and paclitaxel. Methods:The ovarian cancer SKOV3 cells were divided into blank control group, different concentrations (0.01, 0.50, 1.00, 5.00, 10.00mmol·L-1) of metformin groups and combined treatment groups(metformin combined with paclitaxel with different concentrations).The inhibitory rates of proliferation of SKOV3 cells after treated with different concentrations of metformin were detected by MTT method.The apoptotic rates of SKOV3 cells after treated with different concentrations of metformin were measured by flow cytometry.The inhibitory rates of proliferation of SKOV3 cells after treated with metformin and paclitaxel were determined by MTT method.Results:The MTT results showed that the inhibitory rates of proliferation of SKOV3 cells in different concentrations of metformin groups were increased in concentration-and timedependent manner;there were significant differences compared with blank control group (P<0.05).The flow cytometry results showed that the apoptotic rates of SKOV3 cells in different concentrations of metformin groups were increased;compared with control group, with the increasing of concentrations of metformin, the apoptotic rates of SKOV3 cells in experimental groups 48 h after treatment were increased significantly (P<0.05);the percentages of SKOV3 cells in G0/G1 phase were decreased with the increasing of metformin concentrations(P<0.05) and the percentages of SKOV3 cells in G2/M phase were increased(P<0.05).The MTT results showed that the inhibitory rate of proliferation of SKOV3 cells in 1.00 mmol·L-1 metformin+paclitaxel group was higher than that in 0.50 mmol·L-1 metformin+paclitaxel group was higher than that in 0.50 mmol·L-1 metformin+paclitaxel group(P<0.05),and the inhibitory rates of proliferation of SKOV3 cells in combined treatment groups were higher than those in paclitaxel alone groups(P<0.05).Conclusion:Metformin inhibits the proliferation of ovarian cancer SKOV3 cells through the induction of apoptosis.Metformin can enhance the cell proliferation inhibition of paclitaxel on ovarian cancer SKOV3 cells.The combination of metformin and paclitaxel has a synergistic reaction on SKOV3 cells.
10.Proteinase activated receptor-2 promotes IL-4 release from mast cells

Objective:To investigate the effect of activation of proteinase activated receptor(PAR)2 on mediator release from mast cells.Methods:P815 cells(a mast cell line) were challenged with various concentrations of PAR-2 agonist peptide,trypsin,tryptase or elasetase with or without PAR-2 antagonist peptide.The supernatants were collected and analyzed by enzyme-linked immunosorbent assay(ELISA) to detect the quantity of released IL-4,IL-6 and histamine.Results:PAR-2 agonist peptide,trypsin or tryptase induced a concentration-dependent IL-4,but not histamine release from P815 mast cells.Trypsin and tryptase induced IL-4 release from the mast cells was blocked by addition of PAR-2 antagonist peptide.No IL-4,IL-6 and histamine release was observed when P815 cells were incubated with elasetase.Conclusion:Induction of IL-4 release from mast cells by trypsin and tryptase through activation of PAR-2 added some novel information on the hypothesis of self-amplification mechanism of mast cell activation.
