With the understanding of the function mechanism of RNAi,it has been widely applied in various fields.RNAi is a powerful tool to study gene functions and provides a new technical method for the specific gene treatment as well.The further development of RNAi technique opens up a new way for tumor gene treatment.

Objective To observe the expression and relevance of human papillomavirus L1 (HPVL1)capsid protein and tumor suppressor gene p16~4A in cervical lesions.Methods The expression of HPVL1 capsid protein and p16INK4A in liquid-based cytology specimens and organization from 210 cases infected with HPV virus were detected by Max-vision immunohistochemistry method.Results In different grade of liquidbased cytology specimens,the positive rates of HPVL1 capsid protein had statistic difference (x 2 =70.50,P ＜ 0.005).The rate in LSIL couples was 68 ％ (34/50),which was the highest in all couples.The positive rates of tumor suppressor gene p16INK4A were increased gradually.In SCC couples,the positive rates was 100 ％(30/30),which was the hishest.In LISL couples,the rate of HPVL1+ p16INK4A-was 32 ％ (16/50),which was the hishest.In SCC couples,the rate of HPVL1-p16INK4A+ was 100 ％ (30/30),which was the highest.In orgnizational specimens,the positive rates of HPVL1 capsid protein in cervical intraepithelial neoplasia had statistic difference (x2 =54.37,P ＜ 0.005).The rate in CIN Ⅰ group was 60.4 ％ (32/53),which was the highest.The positive rates of tumor suppressor gene p16 were increased gradually.In cervical cancer group,the positive rate was 100 ％ (28/28),which was the highest.In CIN Ⅰ group,the rate of HPVL1+ p16INK4A-was 45.3 ％ (24/53),which was the highest.In cervical cancer couples,the rate of HPVL1 (-) p16INK4A(+) was 100 ％ (28/28),which was the highest.Conclusion Detection on the expression of HPVL1 capsid protein and tumor suppressor gene p16INK4A can diverse different levels of cervical lesions and separate cases from aggravated or self-healing,to avoid over-treatment or misdiagnose.

OBJECTIVETo evaluate the short effect and safety of the acupoint application of dog days for allergic rhinitis.

METHODSTwo hundred and forty-nine patients were randomly divided into an application group (166 cases) and a placebo group (83 cases). On the first day of the first dog days, the first day of the second of the three ten-day periods of the hot season and the first day of the last of the three ten-day periods of the hot season, acupoint application was adopted mainly at Dazhui (GV 14), Feishu (BL 13), Pishu (BL 20) and Shenshu (BL 23) in the application group. The plaster was made of Chinese herbs, including Baijiezi (semen brassicae), Xixin (asarum), Yanhusuo (corydalis tuber) and Gansui (euphorbia kansui). Buckwheat plaster without medical ingredient was used in the placebo group. The application was pasted for 2-6 h every time. On the first three days of all the three periods of dog days, treatment was used continuously. The whole period of dog days was made into one course. The changes of symptom and syndrome scores, visual analogue scale (VAS) for symptom before and after treatment and the adverse reaction with the condition disposed were observed in the two groups.

RESULTSAfter treatment, the symptom and syndrome scores and VAS for symptom were improved compared with those before treatment in the two groups (all P < 0.05), and the improvements of the application group were more obvious than those of the placebo group (all P < 0.05). The short effective rate of the application group was 68.1% (113/166), which was better than 48.2% (40/83) of the placebo group (P < 0.05). The adverse reaction rates were 4.2% (7/166), 2.4% (2/83) respectively, with no statistical significance between the two groups (P > 0.05).

CONCLUSIONThe dog days plaster achieves obvious effect with good safety for allergic rhinitis.

ObjectiveTo screen for proteins related to invasive and metastatic potential of ovarian cancer.MethodsThe nude mice were intraperitoneally injected with the human ovarian cancer 3AO cell line.The metastatic tumors were isolated and cultured in vitro.The cell lines were established from tumors and transplanted intraperitoneally for several passages.Invasive potential of cell lines was evaluated by cell adhesion and matrigel invasion assays.Lysates from different cell lines with high or low invasive potential were analyzed by surface enhanced laser desorption/ionization-time of flight-mass spectrometry(SELDI-TOF-MS)and protein-chip array technology.The proteomic fingerprint patterns were compared. ResultsThe subclones 3AOF3 and 3AOF4 were isolated from repeated passaging in vivo and vitro,and showed higher invasive capacity than 3AO cells.By comparing their proteomic fingerprints,9 distinct proteins (3494,3831,8121,8293,9123,9332,10 184,15 436 and 17 134 at m/z value) were found,which were expressed only in 3AO cells.On the contrary,1 distinct protein (4360 at m/z value) was expressed only in 3AO F3-4 cells,and 2 distinct proteins (1842 and 3145 at m/z value) were expressed only in 3AO F2-4 cells.Additionally,6 distinct proteins (1236,5476,5649,6575,6897 and 7016 at m/z value) and a mass cluster (13 794 and 14 027 at m/z value)were expressed in all 3AO and 3AOF1-4 cells,while the amount of protein was different.ConclusionBy comparing the proteomic fingerprints of isogenic ovarian cancer cells with different invasive potential,a group of proteins are identified which are correlated with high invasive and metastatic potential.

Objective To analyze the clinicopathologic features,prognosis and influencing factors of invasive cervical adenocarcinoma.Methods Clinical and pathologic data of 96 cases of invasive cervical adenocarcinoma and 100 cases of squamous carcinoma of the cervix treated in Shanxi Cancer Hospital from years of 1998 to 2007 were retrospectively analyzed.Results The morbidity of cervical adenocarcinoma was showing the arise and low-age trend.The overall 5-year survival rate was 60.4 ％,The overall survival rates of patients with stage Ⅰ,Ⅱ,Ⅲ,and Ⅳ were 82.9 ％,51.5 ％,13.3 ％ and 0,respectively.Ocervical adenocarcinoma had many prognosis-related factors:clinical stage,lymmphatic spread,vascular invasion,volume of tumor,histologic grade.Multivariate analysis showed that clinical stage,lymph nodal metastasis and vascular invasion were independent prognosis-related factors.However,whether ovary was reserved,it was irrelative to prognosis.Conclusion The morbidity of cervical adenocarcinoma is showing the arise and low-age trend.Earlier diagnosis is extremely important for improving the survival rate of cervical adenocarcinoma,the reserved ovary doesn't influence the overall 5-year survival rate for young women with early cervical adenocarcinoma.

Objective To compare therapeutic effect of Losartan and hydrochlorothiazed compound prepara- tion and Losartan in patients with mild to moderate hypertension and to conduct economical evaluation. Methods The trial began with 2 weeks lead-in period and followed by 8 weeks therapy. 99 cases of mild to moderate hyperten- sion were randomly divided into two groups: compound preparation group ( n = 50) and Losartan (n = 49). The pa- tients were given Losartan 50 mg/h ydrochlorothiazed 12.5 mg compound preparation and Losartan 50 mg ,once a day. If the SeDBP≥85 mm Hg at 4th week, Losartan was added to 50 rag. Results After two, four and eight weeks treatment, systolic blood pressure and diastolic decreased obviously (P < 0.01 for each) ;The overall effective rate of of antihypertension was the same in both groups, but the different ratio of availability and validity in two groups was significant ( P < 0.05 for each). Conclusion Losartan/hydrochlorothiazed compound preparation has significant advantage in improving both systolic and diastolic pressure compared with monotherapy of Losartan. In ad- dition, the patients receiving compound preparation are well tolerated, with higher compliance.

Objective To investigate the expression of Skp2 and Cx43 in cervical carcinoma and to study their relationships and clinical significance. Methods The expression of Skp2 and Cx43 was examined by immunohistochemical method in 48 cases of cervical carcinoma tissue, 84 cases of cervical intraepithelial neoplasia (CINⅠ - Ⅱ 45 cases, CINⅢ 39 cases), 28 cases of chronic cervicitis. Results Skp2 expression in cervical carcinoma was higher than that in the CINⅠ - Ⅱ and chronic cervicitis (P <0.05), Cx43 expression was lower in cervical carcinoma than that in the CINⅢ, CINⅠ - Ⅱ and chronic cervicitis (P <0.05). The expression of Skp2 was correlated with histological differentiation and lymph node metastasis (P <0.05). The expression of Cx43 was correlated with the lymph node metastasis (P <0.05). Besides, the expression of Skp2had a negative correlation with that of Cx43. Conclusion Skp2 and Cx43 may play an important role in the genesis and development of cervical carcinoma.

Objective:To explore the application of three-level storage pattern(on-line,near-line,off-line)to picture archiving and communication system(PACS),and to find a solution to the long-term storage of medical imaging data in PACS of the basic unites.Methods:To realize the on-line storage of medical imaging data by the redundant array of independent disk(RAID)of host-server;To realize the near-line storage of medical imaging data by the network attached storage(NAS);To realize the off-line storage of medical imaging data by the tape-drive.Results:The long-term storage of medical imaging data in PACS has been available in our hospital for one year since the three-level storage pattern used in PACS.It not only realizes the on-line access of enormous amount of medical imaging data,but also protects the imaging data's integrity when the on-line data damaged by human's activities or fire,etc.Conclusion:The application of three-level storage pattern in PACS,which is characterized as low expense,dependable performance,simple maintenance,can become perfect pattern in the basic Level hospital.

[Objective] To observe the effect of Qi-strengthening and blood-activating method for chronic nephritis (CN). [Methods] One hundred and ten patients with CN were randomized into 2 groups. Group A (n = 60) was treated with symptomatic treatment combined with Qi-strengthening and blood-activating herbs including Radix Astragali, Radix Codonopsis, Poria, Rhizoma Dioscorease, Fructus Corni, Rhizoma Curculiginis, Herba Lycopi, Radix Salviae Miltiorrhizae, Semen Persicae and Flos Carthami. Group B (n = 50) was treated with symptomatic treatment combined with oral use of Bailing Capsule, the principal ingredient of which is fermented Cordyceps powder with the actions of tonifying lung and kidney and invigorating essence and Qi. The treatment course lasted 8 weeks. Total therapeutic effect, and the changes of symptoms, immune function, hemorrheology indexes and urine routine indexes before and after treatment were observed. Objective To discuss the imaging manifestations and clinical characteristics of Askin tumor.Methods 12 cases of Askin tumor confirmed by surgically and histopathologically were included in this study.The MSCT signs and clinical data were summarized.Results In 12 cases of Askin tumor,7 cases located in the chest wall;7 case were hospitalised with rapid enlargementd painful lump(58.3%,7/12).9 cases were performed surgery and postoperative radio chemotherapy,6 patients experienced metastatic or recurrent tumors during the following year(66.7%,6/9).Askin tumor often occurred in the chest-lung junction region,had the diameters of 2.0-13.0 cm,11 cases showed inhomogeneous density (91.7%,11/12), 1 case showed calcification (8.3%,1/12), ill-defined margin(100%,12/12),12 cases showed intense but inhomogeneous enhancement following contrast administrationin (100%,12/12),11 cases had the feature of pleura invasion (91.7%,11/12), 9 cases with pleural effusion(75.0%,9/12), 1 case with swollen lymph nodes in mediastinum(8.3%,1/12), and 4 cases with lytic lesion of adjacent bone(33.3%,4/12).Conclusion Askin tumor often occurrs in the chest-lung junction region with rapid enlargementd painful lump as the primary symptom;the tumor show the infiltration diffuse growth;calcification and adjacent lymph node metastasis is not very common.
