Ductular reaction (DR) is a phenomenon that refers to impaired hepatocellular proliferation and activated hepatic stem cells at times of demand such as hepatic regeneration.The components of ductular reactions are thought to be derived from activated hepatic progenitor cells,intermediate hepatobiliary cells,and reactive ductular cells.DRs are thought to play two roles in hepatocarcinogenesis:their hepatic progenitor cells may undergo malignant transformation and migration,and they may contribute to the development of the tumor's microenvironment.Exploring DR phenotype,cellular diversity,and relationship to hepatocarcinogenesis will further enhance our understanding of the pathogenesis of human hepatocellular carcinoma with an ultimate goal of developing new therapies.
3.Immunology study of chronic corneal allograft dysfunction after penetrating keratoplasty

Objective To study the expression of immune and non-immune related cytokine in corneal grafts of chronic graft dysfunction after penetrating keratoplasty and reveal the possible mechanism of it.Design Clinical experimental study.Participants Including two groups.In chronic corneal allograft dysfunction(CCAD) group,corneas of 8 patients fulfilling the diagnostic criteria for CCAD who underwent repeated PK operation.Three normal corneas supplied by Shandong provincial eye bank of China were used as control group.Methods The conreal graft were analyzed by immunohistochemical studies.Moreover,the comprehensive analysis was carried out combining the patient's clinical data.Main Outcome Measures CD4~+,CD8~+,F4/80,TGF-?,bFGF and?-SMA in cornea. Results The results of immnunohistochemical studies showed that compared with normal corneas,full-thickness corneal grafts of eight corneas which underwent repeated PK operation had no CD4~+ and CD8~+ T lymphocyte infiltration.F4/80 positive inflammatory cells were more abundant near the stroma of five corneal grafts which underwent once PK operation.Full-thickness corneal grafts of other three corneas which underwent no immune rejection had no F4/80 positive inflammatory cells.The TGF-?positive inflammatory cells were more abundant near the epithelium and stroma of all corneal grafts.The moderate bFGF positive inflammatory cells were present in stroma of all corneal grafts.The?-SMA positive inflammatory cells were more abundant near stroma and the elastic layer of all corneal grafts.Conclusions After PK operation,the results of immnunohistochemical studies of the corneal graft which underwent CCAD showed that there is no evidence of clinical acute rejection.The abnormal expression of antigen presenting cells and non-immune cell specific factors suggest that immune or non-immune factors maybe involve in the occurrence and development of CCAD.
4.Study on Quality Control of Rupining Strapping

OBJECTIVE:To establish quality control method for rupining strapping.METHODS:The qualitation of Herba Epimedii,Rhizoma Bolbostemmae and fructus toosendan in rupining strapping was conducted by TLC.The principal drug-epimedium herb was determined by column chromatography-HPLC.RESULTS:Herba Epimedii,Rhizoma Bolbostemmae and fructus toosendan could all be identified in TLC.Good linear correlation of icariin was observed when the sample size was0.428?g~2.568?g(r=0.9998).The average recovery was94.57%(RSD=1.43%).CONCLUSION:The method is sensi-tive,accurate,and reproducible.It can be used for the quality control of rupining stripping.

Objective To compare the clinical outcomes of pre-emptive renal transplantation and transplantation after dialysis, and to evaluate the safety and advantages of pre-emptive renal transplantation. Methods The data of 50 cases of pre-emptive renal transplantation between January 1999 and January 2003 in our hospital were analyzed.Another 50 cases of renal transplantation after dialysis were selected as control group.The 2 groups were matched in the following variables:age,gender,blood type,cold (warm) ischemic time of the grafts,human leukocyte antigen (HLA),primary diseases, and use of immunosuppressants. The patient/allograft survival,incidence of rejection and delayed graft function were compared. 7.Detection of HBV DNA with full automatic system and HBV genotyping in blood donor screening

Objective To establish fully automated sample pooling, nucleic acid extraction, amplification and detection method for HBV DNA testing, and investigate the seroconversion and genotype in HBV DNA positive donors. Methods Individual donor plasma samples serologically negative for HBV were pooled by STAR2000 sampling processor with a size of 24. Nucleic acid were automatically extracted by MPLC simultaneously, amplified and detected by Roche COBAS AMPLICOR system. The sensitivity of detection was determined by international standard. HBV DNA positive donors were genotyped and followed up by serological tests. Results The 95% detection limit for this automated HBV DNA testing system was 38.9IU/ml,with 95% confidence interval (21323),eight out of 16512 specimens were PCR positive for HBV DNA,with a positive rate of 0.049%. Three of the 8 DNA positive donors were genotype C,2 genotype B, 1 genotype D,and the other 2 uncertain。Six of the eight HBV DNA positive donors were followed up, and three of them seroconverted。 Conclusion Fully automated HBV DNA detection method can be applied in blood screening,and will further increase the safety of blood supply.
8.Assessment of glomerular filtration rate in renal transplant recipients using serum cystatin C during follow-up

Objective To evaluate whether serum cystatin C (SCys C) could be used as an ideal index to assess renal function of renal transplant recipients during posttransplant follow-up.Methods Seventy patients who were followed up for at least 6 months after renal transplantation in our centre were recruited in the study. SCys C and serum creatinine (SCr) were determined during the follow-up period, and glomerular filtration rate (GFR) was measured using an isotope Tc99m DTPA.The correlation between SCys C, SCr and GFR was analyzed. The performance of SCys C and SCr in diagnosing the mild impairment of renal allgraft function (GFR < 1 ml/s) was evaluated using ROC curve. Results Both SCys C and SCr had a linear negative correlation with GFR (r = -0. 82 and -0. 66 respectively, P<0. 01 ). The sensitivity, specificity and positive predictive values (PPV) of SCys C for diagnosing the mild impairment of renal allgraft function were higher than those of SCr,but the AUC of SCys C did not differ from that of SCr significantly (0. 935 vs. 0. 877, P>0. 05).Conclusion SCys C could be used an ideal index to evaluate the allograft renal function for its better correlation with actual GFR.
9.Effects of comprehensive schistosomiasis control in marshlands of Pukou District,Nanjing City

Objective To evaluate the effects of comprehensive schistosomiasis control in marshlands of Pukou District Nanjing City so as to provide the evidence for developing the strategy of the disease control. Methods The information of the prevention and control of schistosomiasis was collected in Pukou District from 2004 to 2013. The variations of the infection rates of human livestock and Oncomelania hupensis snails were analyzed. Results In 2013 the area with snails in the entire dis-trict was 384.09 hm2 which was 66.24%decrease compared to 1 137.61 hm2 in 2004. Moreover after 2008 no infected-snails and schistosomiasis patients were found and both the density of live snails and serum positive rate of schistosomiasis declined with years. Conclusions After the comprehensive prevention and control is conducted the schistosomiasis situation in Pukou District is stable. However in order to consolidate the results it is still essential to continue monitoring the previous infected snail environments and implementing the comprehensive prevention and control measures.
10.Optimization of the Extraction Process for Xiaozheng Plaster by Orthogonal Test

OBJECTIVE: To optimize the extraction process of Xiaozheng plaster, a traditional Chinese medicine. METHODS: Xiaozheng plaster was prepared by water extraction and alcohol precipitation method with stachydrine content in water and the yield of powdered extract as evaluation indexes. The orthogonal test was carried out to optimize the reasonable parameters of the extraction process. RESULTS: The optimal extraction process for the prescription was adding 12-fold of water and decocting for 90 minutes at each time for three times altogether with alcohol precipitation concentration at 45%. CONCLUSION: The extractive condition can be optimized through visual analysis and the preparation technology is feasible.
