4.Effects of relaxed music therapy on pain of patients undergoing colonoscopy

Objective To study the effect of listening to music with headphone or background music on pain reduction of patients undergoing colonoscopy. Methods 180 patients undergoing colonoscopy were randondy selected as the headphone group,background music group and the control group.The headphone group listened to relaxed music with headphone after measurement of vital signs.The background music group listened to relaxed music played by CD after measurement of vital signs.The control group did not listen to music.Pain degree was measured by pain intensity number scale during colonoscopy.Heart rate,blood pressure,saturation of blood oxygen and appraisal to the music by the former two groups were observed.Results Pain degree of the headphone group and the background music group was lower than that of the control group(P<0.05).Pain degree of the headphone group was lower than that of the background music group with no significant difference(P>0.05).Heart rate of the control group after colonoscopy was faster than before colonoscopy(P<0.05).Conclusions Relaxed music can alleviate pain of patients undergoing colonoscopy.Listening to background music is a convenient and economical method for alleviation of pain without any side effects for patients undergoing colonoscopy.
5.Investigation and analysis of the current human resources in clinical laboratories of hospitals in Ningxia province

Objective To investigate the current human resources and continuing education of clinical laboratories in Ningxia's hospitals and identify problems to resolve.Methods A questionnaire survey was made to study the current human resources in clinical laboratories of hospitals in Ningxia,and the data obtained were analyzed.Results There were 676 laboratory staff employed in 75 clinical 5.1.In terms of education,the ratio their titles ranging from postgraduate,graduate,college Differences in their continuing education are significant statistically.Conclusion Imbalance is found in the distribution of academic titles,education and age groups in such clinical laboratories,with a shortage of those of high academic title and education,and great gaps in continuing education.
6.The clinical significance of DJ-1 over expression in pancreatic neuroendocrine neoplasms

Background and purpose:The symptoms of pancreatic neuroendocrine neoplasms (PNENs) are complicated, that might lead to misdiagnosis. This study aimed to detect the expression level of DJ-1 in PNENs and explore the clinical significance of DJ-1 in PNENs.Methods:DJ-1 protien levels in the serum of 16 cases of PNENs patients and 25 cases of healthy persons were detected by ELISA analysis; The expressions of DJ-1 in 78 cases of PNENs tissues were detected by immunohistochemical staining.Results:The DJ-1 level in the serum of PNENs patients was signiifcantly higher than healthy persons (36.19±6.71vs 24.68±5.94 ng/mL;P<0.001);53.8% (42/78) cases of PNENs tissues showed DJ-1 positive staining; DJ-1 expression level in PNENs tissues had signiifcantly positive correlation with lymph node metastasis (P=0.033), distant metastasis (P=0.017), TNM stage (P=0.012), and pathology grade (P=0.049); As a dependent risk factor, DJ-1 expression was signiifcantly associated with shorter OS (P=0.003) and DFS (P=0.018) of PNENs patients.Conclusion:High expression of DJ-1 protein correlated with invasion and metastasis, disease progression, and poor prognosis in PNEN.
7.Study on the isoniazid induced cellular damage and the expressions of Fas/Fas ligand of HepG2

Objective To establish a model of isoniazid induced necrosis and apoptosis in HepG2 cell and to observe the expressions of Fas/Fas ligand (FasL) in this model.Methods HepG2 cells were treated with different dosages of isoniazid (0,1,2,4,6 and 8 mg/mL) or blank control for 24 hours.Flow cytometer was used to observe the cellular morphology of the HepG2 cell.Annexin V/propidium iodide double staining and flow cytometry were employed to detect the necrosis and apoptosis of HepG2 cells.The expressions of Fas/FasL on the cells were also determined by flow cytometry.The data were analyzed by one-way ANOVA.The comparisons between the drug groups and the control group were performed by using Dunnett t test. Results The higher the dose of isoniazid (4,6,8 mg/mL) was,the more necrosis and apoptosis were observed.In the 4,6 and 8mg/mL isoniazid arms,the total mortality rates were all higher than the control group [(32.1 ±7.5)%,(34.9±8.1)%,(38.2±9.4)% vs (7.2±1.5)% respectively](t=4.62,5.14 and 5.75,respectively; all P<0.01 ).The expression levels of Fas increased along with the dose of isoniazid increasing [(8.7±2.2)%,(11.5±2.8)%,(12.3±3.0)% and (10.6±2.9)% in isoniazid 2,4,6and 8 mg/mL arms,respectively],which were all higher than that in control arm [(3.1 ±0.8) %](t=2.97,P<0.05; t=4.46,P<0.01; t=4.88,P<0.01; t=3.98,P<0.05).Furthermore,the expressions of FasL increased as well when the dose of isoniazid increased.The expression levels of FasL were (16.2±3.5)%,(21.7±4.8)% and (18.7±4.9)%,respectively in isoniazid 4,6 and 8 mg/mL arms,which were all higher than that in the control group [(7.4±1.4)%](t=3.11,P<0.01; t=5.06,P<0.01; t=3.99,P<0.05).HepG2 cell necrosis increased with isoniazid of 8 mg/mL.However,the increase of apoptosis was not observed.Conclusion Isoniazid can induce HepG2 cell necrosis and apoptosis,and the apoptosis may be related with the increased expressions of the Fas/FasL on the cells.
8.Gastrointestinal stromal tumors: A pathomorphologic and immunohistochemical study of 77 cases

Purpose To investigate the pathomorphological and immunohistochemical features of gastrointestinal stromal tumors (GIST). Methods Light microscopy was used to study the morphologic characteristics of 77 cases of GIST. The expression of c-kit(CD117), CD34, vimentin, SMA and S-100 protein were detected in all cases with S-P immunohistochemical method. Results GIST tended to have a higher degree of cellularity and lesser degree of cytoplasmic eosinophilia than observed in classic leiomyomas. The neoplastic cells of GIST were spindle, epithelioid or both in different proportion. Focal to prominent cytoplasmic vacuolization was often seen. The histological spectrum included interlacing fascicles, diffuse sheets, storiform and palisading pattern. The formation of cell clusters was rather special. Stromal and vascular hyalinization was common. GIST primary in the mesentery tended to have a higher potentiality of malignancy. The positivity of CD117 and CD34 were 90%, 92%, respectively. Conclusion The characteristic histological features exist in GIST. The detection of CD117 and CD34 is helpful to differential diagnosis.
9.PLAGL1 gene expression and methylation level and correlation with SDHB mutations in benign and malignant pheochromocytoma

Background and purpose:PLAGL1 gene is a newly discovered gene, which could induce tumor cells’ apoptosis and cell cycle arrest. This study aimed to investigate the expression and the promoter methylation level ofPLAGL1 gene and its correlation withSDHB gene mutations in pheochromocytoma.Methods:The expressions of PLAGL1 mRNA was detected by real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) method in 13 cases of normal adrenal medulla, 32 cases of benign pheochromocytoma group and 54 cases of malignant pheochromocytoma. We examined the promoter methylation status by methylation-speciifc polymerase chain reaction (MSP) andSDHB gene mutation by qRT-PCR ampliifcation and direct sequencing.Results:The relative expression volume ofPLAGL1 gene in malignant tumor tissue was (0.527±0.201), which was signiifcant lower than those in benigntumor tissue and normal adrenal medulla[(1.517±0.662) and (1.734±0.756)]. There was no remarkable difference between benign tumor tissue and normal tissue forPLAGL1 gene expression and promoter methylation.PLAGL1 gene methylation rate in malignant tumor tissue, benign tumor tissue and normal adrenal medulla were 68.5%, 18.8% and 15.4% respectively.SDHB gene mutation was detected in 7 malignant tumor tissues, while no mutation was found in benign and normal group.Conclusion:Lower expression status ofPLAGL1 gene showed signiifcant correlation with promoter methylation which may be associated withSDHB gene mutation.
10.Correlation of Traditional Chinese Medical Syndrome Patterns with Renal Pathological Changes and Clinical Laboratory Parameters in Patients with Lupus Nephritis

Objective To explore the relationship of traditional Chinese medical syndrome patterns with renal pathological changes and clinical laboratory parameters in patients with lupus nephritis.Methods Syndrome patterns were differentiated according the clinical manifestations.The data of laboratory examination and renal pathology were collected and analyzed in 40 patients with lupus nephritis.Results(1) Of the fundamental syndrome patterns,pattern of spleen and kidney Qi deficiency had higher activated partial thromboplastin time(APTT),erythrocyte sedimentation rate(ESR) and 24-hour urine protein amount than the pattern without spleen and kidney Qi deficiency.(2) Of the incidental syndrome patterns,water-damp pattern had higher APTT than damp-turbidity pattern;hemoglobin(HGB) level in water-damp pattern and damp-turbidity pattern was lower than the normal,and HGB level in water-damp pattern differed from that in blood-staiss pattern and damp-heat pattern(P
