1.Effects of hypercholesterolemia on hemorrheology and cTn-I in rats

Objective To observe the characteristics of hemorheologic changes of hyperlipidemia rats at an early stage and to explore the mechanism of myocardial ischemic injury.Methods Seventy-two SD rats which were fed with high cholesterol diet(cholesterol+lard+pig bile salt) to result in hyperlipidemia model fell into HC group.Thirty-six SD rats which were fed with common diet fell into control group.The rats were fed for 10 to 90 d.At an interval of 10 d,9 time points were set.Therefore the 2 groups were randomly divided into 9 subgroups.At each time point,the changes of plasma lipids,cTnI,hemorheologic parameters,and pathological change of the rats’ myocardium and coronary vessel were assayed.Results The main characteristic of our models was hypercholesterolemia and high low-density-lipoproteins(LDL).With the extension of feeding time,plasma viscosity of HC group increased significantly,and erythrocyte deformability decreased.After fed with cholesterol for 50 d,myocardial ischemic injury occurred in the HC group.Correlation analysis revealed that the incidence of myocardial ischemic injury in hyperlipidemia rats was closely related with hypercholesterolemia and high low-density-lipoproteins.Conclusion The abnormal change of hemorheologic characteristics of hypercholesterolemia is a cause of myocardial ischemic injury.
2.The elementary discussion of volumetric modulated arc therapy using the orthogonal plane dose verification

ObjectiveThis study was to explore the feasibility of using the orthogonal plane dose formed by the coronal and sagittal plane to verify the volumetric modulated arc therapy (VMAT) plan.MethodsThe VMAT plans of 12 patients were included in this study.The orthogonal plane dose formed by the coronal and sagittal plane were measured based on the combination of 2D ionization chamber array and multicube phantom,and the point dose were measured based on a multiple hole cylindrical phantom attached with two 0.125 cm3 ionization chamber probes.ResultsIn the measurement of the point dose,the average error was 1.5% in high dose area ( more than 80% of maximum),and 1.7% in low dose area ( less than 80% of maximum),respectively.The discrepancy of point dose measurement was 1.3% between the 2D ionization chamber array and the VMAT planning system.In the measurement of the orthogonal plane dose,the pass rate of γ were 93.7% for 2%/2 mm and 97.2% for 3%/3 mm.ConclusionIt is reliable for using the orthogonal plane dose formed by the coronal and sagittal plane to verify the VMAT plan.
3.Inhibitory effect of soybean isoflavone genistein on renal TGF-?_1 in rats with unilateral ureteral obstruction

Objective To investigate the preventive effect of soybean isoflacone genistein on rats with unilateral ureteral obstruction (UUO) and the related mechanisms. Methods Rats were randomly divided into the following groups: sham-operation (group S, n=18), UUO group (group C, n=30), and geinstein treatment group (group G, n=42, at the dose of 200 mg?kg -1 ?d -1 ). At 5, 10, 15, 20, 25, and 30 d after operation, the pathological changes of the ligated kidneys were observed and the expression of mRNA of TGF-? 1 in renal tissues was examined by RT-PCR. 4.Progress in treating diabetes mellitus with adult stem cells.

Diabetes mellitus is a metabolic diseases, mainly including type 1 and type 2 diabetes. Treatment for type 1 and part of type 2 often involves regular insulin injection. However, this treatment neither precisely controls the blood sugar levels, nor prevents the diabetes complications. Transplantation of islets of Langerhans offers an attractive strategy for diabetes therapies, but its wide application has been limited by donor shortage and immunological rejection after transplantation. Stem cells with strong proliferation capacity and multipotential may be potential cell sources in diabetes therapies. For this, adult stem cells are interesting because of absence of teratoma formation and ethnical problems. Adult pancreatic stem cells (PSCs) really exist and could produce insulin-secreting cells both under the condition of pancreatic injury and in vitro culture, but lack of effective markers to enrich PSCs hampers the studies of exploring the expanding and differentiating conditions in vitro. Some other adult stem cells, such as hepatic stem cells, marrow stem cells or intestine stem cells, were also suggested to transdifferentiate into insulin-producing cells under special culture conditions in vitro or by genetic modifications. Moreover, transplanting these adult stem cells-derived insulin-secreting cells into the diabetic mouse could cure diabetes. Thus, adult stem cells would supply the abundant beta-cell sources for cell replacement therapy of diabetes.

5.Prevalence of depression and the association between depressive symptoms and dietary behaviors among students in Beijing

Objective:
To investigate the relationship between depressive symptoms and dietary behaviors among middle school and college students in Beijing,so as to provide the basis for carrying out related research and intervention.
Methods:
A total of 18 130 middle school and college students from 16 districts in Beijing were collected through stratified cluster sampling from October to December, 2020. The relationship between middle school and college students depressive symptoms and dietary behaviors was analyzed by conducting a questionnaire.
Results:
The detection rate of depressive symptoms was 18.2%. The detection rate of depressive symptoms in girls was 19.0%, which was higher than 17.4% in boys( P <0.05), and the differences were of statistical significance( χ 2=8.47, P <0.01). The detection rate of depressive symptoms in city was 17.9%, and in 10 urban areas was 18.5 %( P > 0.05 ), and the differences were of statistical significance( χ 2=1.15, P =0.28). The detection rates of depressive symptoms in vocational high school (21.8%) and high school students(20.6%) were and, higher than that in college students (18.2%) and in junior high school students(15.2%), and the differences were of statistical significance( χ 2=100.38, P <0.01). The rates of students with depressive symptoms drinking sugary drinks(14.4%), eating sweets (22.1%) and eating fried foods every day (12.4%) were higher than that of students without depressive symptoms(7.6%, 12.0%, 5.9%), and the differences were of statistical significance( χ 2=154.62, 215.67, 164.49, P <0.01). The rates of eating fresh fruits(63.3%), vegetables (81.7%) and breakfast every day (53.4%) of the students with depressive symptoms were lower than those without depressive symptoms (72.0%, 88.1%, 74.1%), and the differences were of statistical significance( χ 2=90.77, 90.59, 518.69, P <0.01) Logistic regression analysis showed that the detection rate of depression was the important factor of influencing dietary behaviors( P <0.05).
Conclusion
The detection rates of depressive symptoms of vocational high school students and senior high school students in Beijing are higher than those of junior high school students and college students. Depression is closely related to dietary behaviors. Promoting students mental health may help promote students healthy eating.
6.Cell lineage tracing of regenerating cells after partial pancreatectomy using pseudo-type retrovirus.

Pancreas is an important mixed gland having both endocrine and exocrine functions, and has been proven regeneration after injury. To explore the cell lineage tracing methods in pancreas in vivo and the regenerate cells source, we used pseudo-type retrovirus to transfect adult mouse pancreas which had been partially pancreatectomized by rubbing the kerf using a cotton stick saturated with retrovirus suspension then injecting 100 microL retrovirus suspension into pancreas, injecting 100 microL retrovirus by caudal vein, or interperitoneally injecting retrovirus respectively. The results showed that the method of rubbing the kerf then injection of retrovirus suspension into pancreas could more effectively mark the pancreatic cells than the caudal vein injection and the intraperitoneal injection did in vivo. Furthermore, this study also found that some acinus cells could accept injury stimulus signals to regenerate through resuming mitosis after pancreatic injury. This study establishes a cell lineage tracing method in pancreas in vivo using retrovirus and offers a clue for gene therapy of pancreatic diseases using retrovirus vectors.

7.Monitoring of teaching environmental health in primary and secondary schools in 2019 in Beijing

Objective:
To understand the sanitation status of teaching environment of schools in Beijing, and to provide basis for school environmental improvement through monitoring it.
Methods:
In 2019, 661 primary and secondary schools were selected according to the coverage rate of 50% of all schools. Two classrooms were randomly selected from each school for testing, 1 322 classrooms were monitored. The testing contents included per capita area of the classroom, rate of correct desks-chairs adjusting for school height, blackboard size, reflection ratio of blackboard,illumination, carbon dioxide and noise.
Results:
The qualified rate of noise(92.3%), per capita area(86.5%), blackboard size (85.6%) and illumination uniformity of table top (83.4%) were higher than 80%, while the qualified rate of other indexes were lower than 80%. In 2019, the qualified rate of blackboard size, blackboard average illuminance and blackboard illuminance uniformity were higher than 2018 year(85.6%, 52.7%, 36.2%; 81.9%, 42.7%, 31.7%)(χ2=10.97, 7.80, 5.90, P<0.05), while the noise (92.3%) was lower than 2018 year(98.3%)(χ2=70.82, P<0.05). Both classroom and blackboard, the average illuminance and evenness of the desk and blackboard installed with LED light were higher than those installed with fluorescent lights.
Conclusion
Teaching environment monitoring in Beijing is not optimistic. It is necessary to strengthen the promotion and evaluation of standards,and the classroom environment needs to be further improved.
8.Multi-locus sequence typing of human Brucella isolated in Sichuan Province from 2014 to 2020

Objective:To learn about the genotyping of human Brucella isolated from Sichuan Province. Methods:BCSP31-PCR and AMOS-PCR were used to identify the genus and biotype of the 66 strains isolated from confirmed human brucellosis cases in Sichuan Province from 2014 to 2020, respectively. The isolated strains were genotyped by multi-locus sequence typing (MLST)-9. The sequence type (ST) was compared trough the online MLST database. A minimum spanning tree (MST) was constructed to cluster the newly discovered and known ST using the BioNumerics software version 7.6.Results:The 66 strains isolated from human cases of brucellosis in Sichuan Province from 2014 to 2020 were Brucella, and 65 of them were Brucella melitensis while one strain was Brucella abortus. The MLST method identified three known STs (ST-8, ST-39 and ST-2) and one newly type (ST-101). Among them, ST-8 was the main ST in Sichuan Province (90.91%, 60/66), another 4 strains of Brucella melitensis were ST-39, and 1 strain of Brucella abortus was ST-2. The newly type ST-101 was isolated from Leshan City in 2019, belonging to the Brucella melitensis and closely related to the evolution of ST-8. Conclusion:Brucella melitensis is the main epidemic Brucella strain in Sichuan Province, ST-8 is predominant genotype, with a small amount of ST-39, ST-101 and ST-2.
9.Arterial partial pressure of carbon dioxide combined with Wells score helps predict acute pulmonary embolism

Objective:To explore the clinical value of arterial partial pressure of carbon dioxide (PaCO 2) combined with Wells score in predicting acute pulmonary embolism (PE). Methods:Patients with suspected acute PE admitted to Emergency Department of Beijing Chaoyang Hospital, Capital Medical University from January 1, 2016 to August 31, 2021 were screened. Patients with positive computed tomography pulmonary angiography (CTPA) results were classified as the PE group, and those with negative CTPA results were classified as the non-PE group. Demographic characteristics, symptoms, vital signs, underlying diseases, risk factors for venous thrombosis, arterial blood gas analysis and Wells scores were statistically analyzed and compared between the two groups, and the clinical efficacy of PaCO 2 combined with Wells score in predicting acute PE was evaluated. Results:A total of 1 869 patients with suspected acute PE were screened, and 1 492 patients were finally selected. There were 537 cases in the PE group and 955 cases in the non-PE group. The frequency of chest pain, dyspnea, unilateral lower limb edema, history of PE or deep venous thrombosis, history of surgery or immobilization within 3 months, history of fracture within 3 months, active malignant tumor, elevated Wells score and reduced PaCO 2 in the PE group was significantly higher than that in the non-PE group (all P< 0.05). The area under receiver operating characteristic (ROC) curve (AUC) of Wells score was 0.784 (95% CI: 0.758-0.810), and the sensitivity and specificity of predicting acute pulmonary embolism were 61.64% and 88.48%, respectively. The AUC of reduced PaCO 2 was 0.679 (95% CI: 0.651-0.707), and the sensitivity and specificity of predicting acute pulmonary embolism were 79.89% and 55.92%, respectively. The AUC of reduced PaCO 2 combined with Wells score was 0.837 (95% CI: 0.816-0.858), and the sensitivity and specificity of predicting acute pulmonary embolism were 74.12% and 77.07%, respectively. The AUC of reduced PaCO 2 combined with Wells score was significantly greater than the AUC of Wells score ( P<0.001) and the AUC of reduced PaCO 2 ( P<0.001). Conclusions:The efficacy of PaCO 2 reduction combined with Wells score in predicting acute PE was superior to that of either of them alone. This was a beneficial supplement to the screening of patients with acute PE, and would also help reduce the abuse of CTPA in the emergency department.
10.Application of micro-teaching in life sciences courses based on the "National Universities Micro-teaching Competition of Life Sciences" analysis.

With improvements in information technology and expansion in education reforms, more innovative teaching reform programs have also been launched. Information technology has increased interest in the use of the flipped classroom innovative teaching model. In order to explore new ideas for the improvement of teaching, this paper focuses on the flipped classroom teaching approach with the integration of information technology. Micro-teaching is an important innovative flipped classroom teaching approach with a number of advantages as it is short, concise, and interesting, which therefore helps improve students' self-learning ability. Designing and preparing micro-teaching would become a prerequisite skill for college teachers. Based on the analysis of the entries in the "National Universities Micro-teaching Competition of Life Science", this paper explores the application of micro-teaching in life sciences teaching from the perspective of curriculum introduction, mode of presentation, teaching design, and other aspects of teaching. This information could serve as a guide to frontline college teachers to help them understand and master the skills of designing micro-teaching, so as to generate interest and improve learning efficiency among college students.
