1.Effects of rosiglitazone on pigment epithelium-derived factor, matrix metalloproteinase-2, and transforming growth factor-β1 expressions in diabetic rats kidney

Forty-five male SD rats were divided into normal group, diabetic control group, and rosiglitazone treatment diabetic group.By the end of 12 weeks, the expressions of pigment epithelium-derived factor(PEDF), matrix metalloproteinase-2 (MMP-2), and transforming growth factor-β1 (TGF-β1) in the kidney were determined by immunohistochemistry and RT-PCR.The results showed that rosiglitazone decreased the increased kidney weight/body weight ratio, serum creatinine, blood ureanitrogen, urinary albumin excretion, triglyceride levels in diabetic rats (all P<0.01).Rosiglitazone prevented the decreasing of protein expressions of PEDF and MMP-2 and the increasing of protein expression of TGF-β1 (all P<0.01).PEDF mRNA showed a similar change,suggesting that renoprotection of rosiglitazone on diabetic rats may be mediated through regulating the expressions of PEDF, MMP-2, and TGF-β1.
2.Effect of periodontal mechanical therapy on periodontal health and interleukin-6 levels in gingival crevicular fluid in type 2 diabetic patients with chronic periodontitis

Objective To evaluate the effectiveness of periodontal mechanical therapy on periodontal health and interleukin-6 (IL-6) levels in gingival crevicular fluid (GCF) in type 2 diabetic patients with chronic periodontitis. Methods GCF samples were obtained with filter papers in first molar sites from 37 type 2 diabetic patients with chronic periodontitis. The randomized controlled and blinded clinical trial with a duration of six months was designed to compare effects between three groups of professional mechanical tooth cleaning (PMTC), i.e. coronal scaling (group Ⅱ) with periodontal initial therapy (group Ⅰ) and without clinical therapy (control group). The clinical periodontal index such as probing depth (PD), attachment loss (AL), modified bleeding index (mBI) and bacteria plaque index (PLI) were obtained with a Williams type periodontal probe. Laboratory examination including GCF volume, concentrations and total amounts of IL-6 in GCF were detected with ELISA method. Results Significantly decreasing trends of PD, AL, mBI, GCF volume and total amounts of IL-6 were observed in group Ⅰin all test period. The group Ⅱ had a significant reduction of PD at the third month as compared with baseline (⊿=0.36 mm, P<0.05), and the other figures showed descending trends but didn′t present statistical significances. At the end of study, the group Ⅰand Ⅱ had significant reductions of GCF volume compared with increases of control group (0.96 μ1>0.03 μ1mm>-0.20 μ1, P<0.05). Conclusions The sequential periodontal supra- and sub- gingival scaling has definitive effects on periodontal health improvement and on reducing the IL-6 level in GCF in type 2 diabetic patients.
3.Obesity and diabetic nephropathy in type 2 diabetes

Obesity is not only the high risk factor of diabetes mellitus,coronary heart disease and hypertension,but also a cause of renal damage.Diabetic nephropathy is one of the most frequent chronic microvascular complications of diabetes mellitus,and one of the leading causes of end-stage kidney failure.It is documented that the obese patients with type 2 diabetes may have obviously multiple metabolic abnormalities and are more prone to develop diabetic nephropathy.This article summarized the pathogenetic mechanisms of kidney injury in diabetic obese patients and provided some ideas in the medical therapy of diabetic nephropathy.
4.Comparison of two different workflows of crossmaching by full-automatic blood grouping instrument

Objective To provide an optimal working process for crossmatching by full-automatic blood typing instrument.Methods Two workflows were applied to crossmatch test by full-automatic blood typing instrument.One was diluting red blood cells of donor samples to concentrate of 1%,the other was detecting directly the donor′s packed red blood cells.Compared consistency and processing time of the two different workflows.Results Cross match results of two workflows were consistent well(U=0,P>0.05).The average processing times before testing and undergoing testing were not significantly different(t=0.692,t=0.562,P>0.05),whereas the average processing times after testing and throughout testing were significantly different(t=146.485,t=67.053,P<0.05).Conclusion The workflow of diluting donor′s sample before testing saved processing time and better suits hospital having a large quantity of specimens.
5.Relationship between serum matrix metalloproteinase-9 level and coronary heart disease in type 2 diabetic patients

Twenty-seven cases of simple type 2 diabetes mellitus,30 cases of coronary heart disease,and 32 cases of type 2 diabetes with coronary heart disease were enrolled in this study according to the results of coronary angiography.Meanwhile,32 healthy subjects were taken as a control group.The serum matrix metalloproteinase-9 (MMP-9) and other clinical and laboratory parameters were determined.The results showed that serum MMP-9 may play a minor role in the progression of coronary heart disease in type 2 diabetic patients.
6.Clinical efficacy of circular wound edge protector in preventing surgical site infection following open abdominal surgery

Objective To explore the efficacy of circular wound edge protector(CWEP)in preventing surgical site infection (SSI)following open abdominal surgery,analyze the related risk factors for postoperative SSI,and provide scientific basis for preventing SSI following open abdominal surgery.Methods 162 patients who underwent open abdominal surgery in a hospital from March 2014 to December 2015 were randomly divided into intervention group (used CWEP during surgery)and control group (used sterile gauze pad to protect incision).Length of hospital stay,cost of dressing change,and SSI between two groups were compared,risk factors for postoperative SSI were analyzed.Results Incidence of SSI in intervention group was lower than control group (14.81%[12/81]vs 34.57%[28/81],P <0.05);grade-A incision healing rate of intervention group was higher than control group(86.42% vs 66.67%,P <0.05).The cost of dressing change in intervention group was lower than control group([305.5 ± 176.7]yuan vs [431.6 ± 381.4 ]yuan,P = 0.008 ). Multivariate logistic regression analysis indicated that hypoproteinemia(OR,2.88[95%CI ,1.21-6.87]),body mass index ≥24(OR,3.11[95%CI ,1.12-8.66]),and blood loss≥400 mL(OR,3.98[95%CI ,1.36- 11.64])were independent risk factors for postoperative SSI,while CWEP use was the protective factor(OR,0.24[95%CI ,0.09-0.59]).Conclusion CWEP can effectively reduce the incidence of SSI following open abdominal surgery.
7.Analysis of Prognosis Influencing Factor of Children with Tourette Syndrome

Objective To explore the related factors influencing the prognosis of children with Tourette syndrome ( TS) . Methods We collected 420 outpatient cases of TS children in the department of pediatrics of Guangdong Provincial Hospital of Traditional Chinese Medicine from January of 2007 to October of 2010 as the research objects. Using the unified survey questionnaire, we observed the influencing factors of TS prognosis such as sex, onset age, mother’s pregnancy situation, the perinatal period, partiality for food intake, family relationship, the first symptom, the severity of disease, underlying diseases, comorbidities ( such as attention deficit hyperactivity disorder, anxiety disorder, and obsessive-compulsive disorder) , family history of psychiatric or neurological diseases, trace elements levels, electroencephalogram (EEG), antistreptolycin O (ASO), course of disease, and maintenance treatment. The related factors of prognosis was analyzed with single factor and multiple factors Logistic regression analysis. Results Among the 420 cases of TS children, 396 cases were included into the final analysis, 24 were lost and the follow-up lost rate was 5.7%. The remission rate of TS was 78.3%, and the uncured rate was 21.7%. The results of preliminary screening of the influencing factors by single factor Logistic analysis showed that the related influencing factors for TS prognosis were 12, and they were course of diseases, abnormal birth history, father’s education level, mother’s education level, upbringing methods, family history of psychiatric or neurological diseases, underlying disease history, comorbidities history, abnormal ASO, the severity of disease, the frequency of disease relapse, and the medication history of western medicine (P<0.05) . And then the obtained 12 factors were analyzed by the multiple fac tors Lo gistic regression analysis, the results showed that upbringing methods, comorbidities history, the severity of disease, and the frequency of disease relapse were correlated with TS prognosis ( P < 0.05) . Conclusion TS children will have poor prognosis when their parents spoil, indiscipline, beat and scold, and dictate them, or when the children have severe illness state, frequent recurrence of the disease, and the history of comorbidities ( atten tion deficit hyperactivity disorder, anxiety disorder, obsessive-compulsive disorder, etc.) .

The vehicle-mounted wireless tele-consultation system involves computer network technology, satellite communication technology and multimedia video technique in, which is composed of GPS, vehicle -mounted satellite communication sub-system, automatic satellite-tracking sub-system and computer control system. The system can be applied to medical command, operation tele-instruction, tele-education, tele -consultation, uploading medical service data and searching electronic medical information,and thus the field medical treatment and medical service are both improved. This paper introduces such aspects of the system as its principle, components, functions and application to medical service support.

10.Ovine Follistatin gene expression and functional analysis of follistatin domains.

In order to study ovine follistatin function, we amplified the total of 1038 base pair of ovine complete follistatin cDNA and cloned into pGEM-T vector by RT-PCR from ovine ovary RNA. After removal of the signal peptide it was subcloned into the pET41a to construct the prokaryotic expression vector, named pFSsig-. SDS-PAGE and Western blotting identified the 66 kDa product of the expression of follistatin cDNA. Based on the complete CDS sequence, we cloned follistatin N-terminal domain and domain 1 with PCR and inserted into pLEX-MCS lentiviral vector, named pFS-N+D1. After package and passage of lentivirus in 293T cells, and then infected sheep primary muscle cells (SPMC). The expression of FS N+D1 in SPMC was assayed by Western blotting. The cell growth curve of the infected SPMC and noninfected control cells displayed that FS N+D1 stablly transfected SPMC proliferated significantly faster than the control cells (P < 0.01). Our data inferred that ovine FS N+D1 domain had the function to stimulate sheep muscle cell growth.
