1.Sortase:New Target Enzyme in Gram-positive Pathogens

Many surface proteins of Gram-positive bacteria are anchored to the cell wall by the action of sortase enzymes, a recently discovered family of cysteine transpeptidases. As the surface proteins of pathogens are frequently required for virulence, the sortase might be a suitable target for the development of anti-Gram-positive drugs. Recently, the mechanism and active sites of sortase was elucidated by the research of sortaseA(SrtA) in Staphylococcus aureus and a series of SrtA inhibitors have been identified for providing the basis for further development of potent inhibitors.
2.Establishment and Application of Hyperinsulinemic-Euglycemic Clamp Technique in Conscious Rats

Objective To establish the hyperinsulinemic-euglycemic clamp(HEC) technique in conscious rats, and to explore the effect of acute infusion of lipid on glucose infusion rate (GIR) in rats. Methods Ten SD rats were random-ly divided into two groups, 5 rats for each group. The right jugular vein and left carotid artery were catheterized and under-went a HEC with infusion of lipid (intralipid group) for 6 hours, and with continuing infusion of 5%glucose (control group). The plasma levels of free fatty acid(FFA) and GIR were measured by HEC method. Results The level of FFA concentration increased by 17.6-fold, and GIR was reduced by 27%in the intralipid group compared to those of control group (P<0.001). Conclusion The rat model of HEC has been successfully established by intravenous intralipid infusion, which can be con-firmed by HEC technique.
3.Analysis of actual mismatch risk for KIR genes and KIR ligand related genes in kidney transplant recipients

Objective To evaluate the actual mismatch risk for KIR genes,KIR ligand HLA-C gene and KIR ligand HLA-Bw4I80 related genes in kidney transplant recipients and their donors.Method KIR genes,KIR ligand HLA-C gene and KIR ligand HLA-Bw4I80 related genes were analyzed in 322 recipients and their donors who received kidney transplantation.The effect of mismatches on acute rejection after transplantation was studied as well.Result In 322 cases of recipients and their donors,average mismatch risk for KIR-L gene was 4.73%,9.10% for KIR-S gene and 1.95% for KIR ligand HLA-C gene respectively.There were 245 cases (76.09%) of KIR ligand HLA-Bw4I80 related genes mismatch and 77 cases (23.91 %) of match resepctively.There was no statistically significant difference in AR rate between KIR ligand HLA-Bw4I80 related genes mismatch group and match group (8.16% in 245 vs.7.79% in 77) (P>0.05).AR reversal rate in mismatch group and match group was 95.0% and 66.7% (P>0.05).Conclusion The mismatch risk for KIR ligand HLA-Bw4I80 related genes was even common between kidney transplant recipients and their donors,and this specific mismatch may be of benefit to patients who undergo actuate rejection.Further study is still required.
4.Clinical observation on the effect of different pharmacotherapy after lacrimal passage plastic laser operation

Objective To investigate the effect of different pharmacotherapy after lacrimal passage plastic laser operation. Methods Divided 90 patients (103 eyes) with nasolacrimal duct obstruction into group A (33 eyes), group B (36 eyes) and group C (34 eyes) randomly, each group consisted of 30 cases. Tetracycline cortisone ophthalmic ointment (TCO) was used to perfuse lacrimal duct in the end of the operation in group A, while TDO was used in group B and group C. In the first three postoperative irrigation, TDO was used only in group C, there are no no ointment during the course of irrigation in the other two groups. 5.Problem and development of the hospital medicine preparation in Beijing

Objective To realize the actuality and analyze the problem of the hospital medicine preparation in Beijing,then to discuss its developing methods.Methods By summarizing questionnaire of the hospital medicine preparation and combining with the problem showed in daily supervision,we analyzed the main existing problems and reasons for hospital medicine preparation and put forward the developing methods.Results The main existing problems of hospital preparation are the decreasing variety of medicine preparation,insufficient infrastructure and software.Conclusion Therefore we should enhance the input on the infrastructure,set up the software,improve the quality of the staff,and consummate the regulation means and system.
6.Study on Development Strategies for Promoting TCM Service Trades in Gansu Province

7.The correlation between human leukocyte antigens-A, B, DRB1 high-resolution alleles and chronic renal failure caused by immunoglobulin-a nephropathy

Objective To investigate the correlation between human leukocyte antigens-A,-B,-DRB1 (HLA-A,-B,-DRB1) high resolution alleles and chronic renal failure (CRF) caused by immunoglobulin-a nephropathy (IgAN).Method The polymerase chain reaction-sequence-based typing (PCR-SBT) method was used to investigate the genotypes of HLA-A,-B and-DRB1 high-resolution alleles in 191 cases of CRF caused by IgAN (experimental group) and 503 healthy blood donors (control group).The alleles frequencies between two groups were compared and the association between CRF caused by IgAN and the polyrnorphism of HLA was analyzed.Result (1) There were 25 alleles at A locus,48 alleles at B locus and 32 alleles at DRB1 locus in experimental group.(2) The genetic frequency of HLAA * 2901 [Pc =0.033,OR =10.738,95% CI (1.193,96.691)],HLA DRB1 * 1106 [Pc =0.0001,OR =0.969,95% CI (0.944,0.994)],HLA-DRB1 * 1202[Pc =0.002,OR =1.859,95% CI (1.259,2.745)],HLA-DRB1 * 1401 [Pc =0.021,OR =0.984,95% CI (0.967,0.998)],HLA-DRB1 * 1602[Pc=0.015,OR=1.915,95% CI (1.157,3.17)] in experimental group was higher than in control group (P<0.05).Conclusion There is susceptibility association of HLA-A * 2901,HLA-DRB1 * 1106,HLA-DRE * 1202,HLA-DRB1 * 1401,HLA-DRB1 * 1602 with CRF caused by IgAN.It is concluded that there is a close genetic and immunological correlation between HLA alleles and the pathogenesis of CRF caused by IgAN.
8.Preoperative treatment and prognosis observation in sensitized recipients of kidney transplantation

Objective To study the preoperative treatment and prognosis observation? in sensitized recipients of kidney transplantation.Method Forty-one recipients positive for preoperative PRA accepted renal allograft transplantation from January 2007 to July 2012.All recipients were given immunosuppressant or immune induction by anti-CD25rnAb in advance,and plasma exchange,immunoadsorption and intravenous high-dose immune globulin were administered.Meanwhile,donor HLA antigens had to avoid all stored HLA antibodies of the recipient,and lymphocyte cytotoxicity cross test (CDC) had to be negative.Anti-human lymphocyte globulin (ATG) was used to strengthen the immune induction,and tacrolimus + mycophenolate mofetil (MMF) + corticosteroids triple immunosuppressive regimen was adopted after transplantation.Then intravenous micafungin would be given after transplanted kidney function was normal,and ganciclovir and sulfamethoxazole were taken orally to prevent infection.Result In 41 recipients positive for preoperative PRA,13 cases were positive for only HLA class Ⅰ antibodies,15 cases for only HLA class Ⅱ antibodies,and there existed 13 cases of both HLA class Ⅰ and class Ⅱ antibodies also with PRA≥50%.Fifteen patients achieved normal serum creatinine in one week,and no hyperacute rejection and accelerated rejection occurred.Fourteen recipients experienced an episode of acute rejection (34.15%,14/41): 12 recovered by steroids bolus therapy,and the other two reversed in 3-5 days by cyclophosphamide or ATG treatment.One case died of mycotic pneumonia in 4 months later.One-year recipient/kidney survival rate was 97.6% (40/41).Conclusion The recipients positive for preoperative PRA only can accept renal allograft transplant while the donor's HLA antigens had to avoid all stored HLA antibodies of recipients themselves and CDC test was negative.After that the combination of desensitization therapy,immune induction therapy and postoperative potent immunosuppressant can prevent acute rejection effectively and increase postoperative recipient/kidney survival rate.
9.Tetrandrine treatment of severe type acute pancreatitis

Objective To study the effect and mechanism of tetrandrine (Tet) treatment of severe acute pancreatitis (SAP). Methods 45 SD rats were randomly divided into 3 groups: control group, severe acute pancreatitis group (SAP group) and tetrandrine group (Tet group). 5 rats were randomly taken from each group 3, 6, and 12 h after operation, and then underwent laparotomy for testing. Detection targets included calciumion concentration, pancreatic acinar calcium fluorescence intensity and pancreas pathological evaluation. Results In terms of the value of all detection targets, there was no statistical difference for the control group between different time points (P >0.05 )while there was statistical difference for SAP group and TET group in different time points (P<0.05). Conclusion The experiment confirms that Tet, as a calcium blocker, can inhibit calcium to penetrate into pancreatic acinar cells, thus effectively reduce the pathological damage to pancreas of the experimental rats. Calcium overload plays an important role in SAP development.
10.Effect of thymosin beta4 on matrix metalloproteinase-2,9 of hypertrophic scar

Objective To study the effect of thymosin beta4 on the expression of matrix metalloproteinase‐2(MMP‐2) and MMP‐9 of human hypertrophic scar fibroblasts and explore mechanism of thymosin beta4 on hypertrophic scar .Methods Fibro‐blasts were cultured in vitro from the orthopaedic patients(within 6 months after burns) admitted in the first affiliated hospital of Nanchang university from June to September 2013 .The Fibroblasts were divided into experiment group :0 .05 ,0 .1 ,1 .0 μg/mL and 5 .0 μg/mL ,and control group :no thymosin beta4 .The expression levels of MMP‐2 ,MMP‐9 were observed by RT‐PCR and West‐ern blot .Results The expression levels of MMP‐2 and MMP‐9 were increased after the thymosin beta4 took effect ,and it was espe‐cially significant in 1 .0 μg/mL and 5 .0 μg/mL group ,there was statistical difference between experiment group and control group (P<0 .05) .Conclusion Thymosin beta4 can dose‐dependently promote the expression of MMP‐2 and MMP‐9 and inhibit the pro‐liferation of hypertrophic scar .
