3.The relationship between the serum ghrelin level and β-cell function after treatment of patients with newly diagnosed type 2 diabetes mellitus

Objective To investigate the relationship between the serum ghrelin level and β-cell function after treatment of patients with newly diagnosed type 2 diabetes mellitus.Methods 34 patients with newly diagnosed type 2 diabetes mellitus were divided into three groups of pre-treatment(DM0),two weeks after treatment(DM1)and six months after treatment(DM2).Intravenous glucose tolerance tests(IVGTT)were performed and blood glucose,HbA1C,ghrelin,insulin were measured during the study.Insulin sensitivity was assessed by homeostasis model assessment(HOMA).Results(1)Compared with DM0 group,beta-cell function and the mean AUC of insulin were dramatically improved in groups of DM1 and DM2(all P<0.05).(2)Compared with DM1 and DM2,ghrelin levels and the mean AUC of ghrelin in DM0 groups were lower(all P<0.05).(3)Only positive correlation between ghrelin and HOMA-β(r=0.446,P<0.05),and negative correlations of ghrelin with glucose(r=-0.274,P<0.05),insulin(r=-0.35,P<0.05)and BMI(r=-0.276,P<0.05)were found after six months of treatment.Conclusions Ghrelin might play an important role in the pathogenesis and progress of type 2 diabetes mellitus.
6.Mediating effect of self-nursing ability between living condition and anxiety in patients with peptic ulcer caused by helicobacter pylori

Objective:To explore the mediating effect of self-nursing ability between living condition and anxiety in the patients with peptic ulcer caused by helicobacter pylori.Methods:By using questionnaire survey method,300 patients with a peptic ulcer caused by helicobacter pylori were assessed with the exercise of self-care agency (ESCA),Self-rated Health Measurement Scale (SRHMS) and Self-Rating Anxiety Scale (SAS).The result was compared with that of 300 healthy crowd whose general condition was well-matched.The data were processed and analyzed by using SPSS13.0 and AMOS7.0.Results:The total scores of the ESCA,SRHMS and SAS were (101 ± 13),(299 ±35),(56 ±8) respectively.The result of the correlation analysis showed there were varying degree correlation among the disease self-nursing ability,self-rated Health,and anxiety (Ps < 0.01).Bootstrap test showed the size of the mediating effect of self-test health to anxiety was 0.17,the 95% confidence intervals was 0.11-0.26,and the ratio of the mediating effect to the total effect was 0.44.Contusion:It suggests that self-nursing ability may have much strong mediating role between health condition and anxiety.

8.Pharmacokinetics of oral FK506 in patients after renal transplantation

Objective To carry out a pharmacokinetic evaluation of oral FK506 in 13 patients after renal transplant. Methods 13 patients after renal transplantation were given prograf based immunosupressive regimen 24 hours after surgery.Blood samples to determine FK506 levels were drawn in heparinized tube at 0、20、40、60、90 min and 2、3、6、8、10 hours after the first oral dosing.The whole blood concentrations were measured by MEIA and the pharmacokinetic parameters were calculated by 3P87 program.The FK506 doses were recorded in detail for the first month. Results Cmax was (13.6259?4.1117)ng/ml;T(peak) was (1.4866?1.0725)h;t1/2 ? was (0.7749?0.7791)h,t1/2 ? was (10.7267?10.4926)h;AUC was (91.0415?40.7694)ng?ml -1 ?h -1 ,CL was (0.046?0.0036)ng?ml -1 ?h -1 and MRT was (8.1540?4.2937)h.AUC was negative correlateld with prograf dose in the first month posttransplant (r=-0.53, P =0.038). Conclusions The absorption of oral administration of FK506 was rapid in patients after renal transplantation,and can achieve Cmax in (1.5?1.1)h,the mean half life time being 10.7 h.The pharmacokinetic parameters can be the guideline for FK506 application.
9.THE DISTRIBUTION OF PRIMARY AFFERENT FIBERS FROM THE GREATER SPLANCHNIC NERVE TO THE SPINAL GRAY MATTER AND NUCLEUS GRACILIS IN THE CAT

The central distribution of visceral primary afferent fibers from greater splanchnic nerve of the cat have been studied. Horseradish peroxidase (HRP) dissolved in distilled water was injected into the coeliac ganglion or greater splanchnic nerve unilaterally. The HRP was taken up by afferent fibers and transported across the spinal ganglia and then anterogradely as far as central terminals in the spinal cord and medulla. The HRP reaction-product within the central processes of the primary sensory neurons was detected ipsilaterally in the Lissauer's tract, dorsal funiculus and spinal gray matter. After entering the spinal cord via the spinal dorsal roots, the great majority of labeled fibers (viz. the central processes of primary sensory neurons) were found to run longitudinally within the zone of Lissauer, and only sparse fibers to ascend in the dorsal funiculus. The medial projection fibers (MPF) and lateral projection fibers (LPF) arose periodically from the labeled fiber bundle in the Lissauer's tract to form a thin shell surrounding both the medial and lateral sides of the dorsal horn. The terminal areas of the more prominent LPF were found at laminae I, V, VII and the region surrounding the central canal, whereas the MPF ended at the medial part of lamina V and lamina X. Some smaller fiber bundles from LPF were seen to enter the cell clusters of the intermediolateral nucleus, where these fibers divided rostrocaudally and ran parallel with the longitudinal dendritic branches of the intermediolateral cells. The few labeled fibers located at the dorsal funiculus were traced to the lower medulla and the distinct terminal area of them was situated in the ventrolateral part of the nucleus gracilis at the level below the obex.
