1.Dynamic changes of INF-γ and IL-4 in the peripheral blood of mouse infected with Mycoplasma suis

The changes in the levels of IFN-γ and IL-4 in peripheral blood of mice infected with Mycoplasma suis were investigated in the present study, in which BALB/c mice were infected with Mycoplasma suis and the levels of IFN-γ and IL-4 in peripheral blood of mice were determined by means of ELISA assay 3, 5 and 7 days after infection. The results showed that the lower level of IFN-γ and higher level of IL-4 could be demonstrated 3 and 5 days after infection in comparison with those of the control groups respectively(P<0.05, P<0.01), but there was no significant difference in the levels of IFN-γ and IL-4 after 7 days of infection (P>0.05) . It is evident from this result that determination of the levels of IFN-γ and IL-4 in peripheral blood of mice may constitute an additional sero-diagnostic index to infection with M.suis and provide the experimental data for the studies in the serology and immunology of this infection.

AIM: To preliminarily study the molecular mechanism for the cardiac injury in rat by adriamycin and the mechanism for the acute repair in the body. METHODS: The male Sprague Dawley rats were randomly divided into four groups (n=10 in each): The first group was kept without treament and served as the control ;the second, the third and the fourth received ADR in different doses (10, 20, 40 mg?kg -1 , respectively) by injection of adriamycin. The content of malondialdehyde (MDA)in the serum was estimated with thiobarbituric acid. Cu Zn SOD was measured by its reaction with xanthine oxidase. Objectives To investigate the prevalence of heterogeneous vancomycin intermediate Staphylococcus aureus(hVISA) and the sensitivity of hVISA to novel antibiotics,and to explore the risk factors and infection attributable mortality associated with hVISA infection.Methods A total of 456 methicillin resistant Staphylococcus aureus (MRSA) isolates were isolated in Zhongshan Hospital from January,2008 to November,2010.All MRSA isolates were investigated for hVISA by two agar screening methods BHIA5T (brain-heart infusion containing teicoplanin 5 mg/L)or BHIA6V (brain-heart infusion containing vancomycin 6 mg/L),as well as macroEtest method(MET).Possible hVISA isolates were tested by modified population analysis profile-area under the curve (PAP-AUC).The minimal inhibitory concentrations(MICs) of vancomycin,teicoplanin and linezolid were determined by microbroth dilution as recommended by Clinical Laboratory Standards Institute(CLSI).The contribution difference between hVISA and vancomycin susceptible Staphylococcus aureus (VSSA) in different MIC range was compared.A retrospective case-control study of the patients with hVISA infection or VSSA infection was carried out and statistical analysis was performed using t test,Mann-Whitney test,x2 test and Fisher exact test.Results A total of 105 isolates of hVISA were screened by BHIA5T and BHIA6V (23.0%) with other 23 isolates by MET(5.0%) and 21 by PAP-AUC(4.6%).All isolates were 100% sensitive to vancomycin,teicoplanin and linezolid.The vancomycin MIC [(1.76 ±-0.16) mg/L] in hVISA group was significantly higher than that in VSSA group[(1.09 ± 0.07)mg/L,P < 0.01],which was a potential risk factor for hVISA infection.The retrospective study showed chronic obstructive pulmonary disease (COPD) was also a risk factor for hVISA infection of the lower respiratory tract.No significant difference in infection attributable mortality was showed between the hVISA group and the VSSA group.Conclusions The overall prevalence of hVISA in Zhongshan Hospital is estimated as 4.6%,while the prevalence of hVISA isolated from blood is as high as 12.5%.All isolates are 100% sensitive to vancomycin and linezolid.COPD is a risk factor for hVISA infection of the lower respiratory tract.
5.Behavior, inflammatory response and cell autophagy of amyloid precursor protein / tau protein /presenilin-1 transgenic AD mice

Objective To explore the behavior characteristics ofamyloid precursor protein / tau protein / presenilin-1 (APP/Tau/PS1) transgenic mice in different months of age and study the progress of inflammation and the changes of autophagy level in brain of these APP/Tau/PS1 Alzheimer's disease(AD) mice.Methods Four-month-old wild-type (WT) mice (B6:129SF2/J) and Tau/APP/PS1 transgenic mice at 2,4,6,8 and 12 months old (n=8) were chosen in our study.Morris water maze was used to detect the spatial learning and memory abilities ofAPP/Tau/PS1 AD mice.After the paraffin section,immunohistochemical fluorescence technology was used to detect the expression and distribution of glial fibrillary acidic protein (GFAP) in the brain of mice in the above 6 groups.The expression changes of GFAP,CD45 (inflammation marker) and LC3 (autophagy marker) in the hippocampal tissues of 6 groups were detected by Western blotting.Results In the morris water maze test,the escape latency increased obviously following the increasing of months:the latency of 6 months old mice was significantly prolonged as compared with that of 2 months old mice (P<0.05),which was further significantly increased in 12-month-old mice (P<0.05).In the exploring experiment,with the increasing of months,the times of crossing the original table in mice showed a trend of decline,time ofresidencing in the original platform quadrant was significantly reduced.As compared with that in the WT mice,the GFAP expression in the hippocampus of 8 and 12 months old AD mice was significantly increased (P<0.05); meanwhile,the GFAP expression in 8-month-old mice was also significantly higher than that in 2 months old AD mice (P<0.05).The CD45 expression in 6 months old AD mice was obviously increased as compared with that of WT mice (P<0.05).As to the level of LC3-Ⅱ/Lc-Ⅰ,there was a significant increase in 2 months old AD mice as compared with WT mice (P<0.05),which was further increased in 4 months old AD mice (P<0.05),but decreased in 6 and 8 months old AD mice.Conclusion With the gradually loss of the leaming and memory abilities,inflammatory reaction is significantly increased,and the autophagy level increases at first,and then,decreases in the brain ofAPP/Tau/PS1 AD mice.
