1.Effect of maternal hypertensive disorders complicating pregnancy on brain development of the neonates

Objective To study the effect of maternal hypertensive disorders complicating pregnancy on brain development of the neonates. Methods Insular area,circle and cerebral gyri width of lobus frontals were measured in 56 neonates of the mothers with hypertensive disorder complicating pregnancy(HDCP)by ultrasound and were compared with those of 53 neonates of the mothers without HDCP(control group). Results In the control group,the cerebral gyri width of term neonates was narrower than that of preterm neonates,while the insular circle was longer and the insular area was larger in the term neonates(P<0.01).Insular circle was significantly shorter and the insular area was significantly smaller in neonates of HDCP mothers [term:(87.43±10.21)mm,(413.60±90.05)mm2;preterm:(72.59±7.50)mm,(285.71±48.62)mm2]than those of the corresponding neonates in the control group[term:(102.14±9.97)mm,(557.08±98.89)mm2:preterm:(78.42±10.14)mm,(356.01±93.07)mm2].Cerebral gyri width were significantly wider only in the full term neonates.The incidence of small gestational age neonates was 30.36%(17/56)in hypertensive disorder complicating pregnancy group.The neonatal behavioral neurological score of hypertensive disorder complicating pregnancy group was significantly lower than that of control group (P<0.01). ConclusionsMaternal hypertensive disorder complicating pregnancy may result in poor development of brain of their neonates,which could be evaluated by cerebral ultrasound.
2.Analysis on the implementation effect of nursing management intervention quality of care in critically ill patients with mechanical ventilation

Objective:To investigate the clinical application of and critically ill patients received comprehensive nursing interventions during mechanical ventilation effect value. Methods:Ninety eight patients were selected in 2012 July~2013 year in July in our hospital, were treated with mechanical ventilation, were randomly divided into observation group and control group, 49 cases in each group, the control group using ICU routine care, the observation group in the control group on the basis of comprehensive nursing intervention, observation and nursing effect of the two groups. Results:The group of patients with mechanical ventilation time, hospitalization time was shorter than the control group, comparing the data between the groups, the difference was statistically significant(t=8.3678, t=5.9103; P<0.05). The observation group patients with unplanned estuation, ventilator-associated pneumonia was lower than the control group, the mortality rate was lower than the control group, comparing the data between the groups, the difference was statistically significant (x2=5.0178, x2=10.2424, x2=5.9905;P<0.05). Conclusion:in critically ill patients were given comprehensive nursing intervention can shorten the duration of mechanical ventilation and hospitalization in patients with mechanical ventilation, to reduce the unplanned extubation and ventilator-associated pneumonia, reduce mortality, it is worth promoting in clinical use.
3.Determination of content of astragaloside A in Astragalus membranaceus powder for

To develop a method for the quality control of Astrgalus membranaceus (Fisch.) Bge. powder for injection (AMPI). Methods TLC-spectrophotometry was used to determine the content of astragaloside A in AMPI. The mobile phase was chloroform-methanol-water (65∶30∶10) and detected at the wave length of 418 nm. Results The regression equation within the spotted range of 80~240 μg was A=1.538 C+ 0.189, with good linearity, r=0.999 8. The average recovery rate was 98.68% and RSD=2.52%(n=5). Conclusion The method used simple and easily available instruments and the rusults obtained were reliable and specific, suitable for the quality control of AMPI.

5.Expression and activity of vMIP-Ⅱ in single strand of blocking to chemokine receptors

Objective:To establish an efficient method for expression and purification of vMIP-Ⅱ which encoded by a viral gene and ho-mogued human chemokine in active single strand in E.coli cells. Methods: Cloning with bi-enzyme restriction and lysising bacteria with cold osmoic shock were used for expression,MBP affinity chromatography of fusion protein and self-selective of recombinative peptide for final purification. The expressing and purifying products were detected with SDS-PAGE and Western blotting and identified with inhibitory adhesion experiment for its activity.Results: Fusion protein MBP-vMIP was effeciendy expressed in E. coli at secretive type, and self-cleaved to sparate the vMIP-Ⅱ. The final product single strand vMIP-Ⅱ was active for blocking chemokine receptor CCR5.Conclusion:This is an effective method for obtaining viral chemokine vMIP-Ⅱ of recombinant single strand.The recombinant vMIP-Ⅱ may be useful for the study of diseases involving in chemokiine receptors such as HIV infection,rejection of transplantation and chronic inflammation,etc.
6.Reasons for the decline of autopsy rate after accidental death

To investigate and analyze the factors which influence the public attitudes towards autopsy after accidental deaths.The study has carried on an investigation among 386 individuals in random,with a questionnaire named "The cognition of the public whether to carry on the autopsy after accidental deaths".Using the statistical methods of the Logistic Regression analysis and optimum regression analysis to analyze the recycling questionnaires.Through the diagnosis,we found there are four main factors which influence public opinion upon the autopsy after accidental deaths,the knowledge of autopsy,the believing in autopsy,years of schooling and family financial circumstances.
7.Influence of parathyroid hormone and estrogen on alveolar bone metabolism of castrated female rats.

OBJECTIVETo investigate the influence of parathyroid hormone and estrogen on alveolar bone metabolism of castrated female rats.
METHODSSixty-six female Wistar rats which were healthy and 4 months old were divided into two groups, with group SHAM (n = 18) and group ovariectomy (OVX) (n = 48). After 8 weeks of ovariectomy, the osteoporosis model was confirmed by examing 8 ovariectomized and sham-operated rats. The rest 10 rats in group SHAM were the control group (group A). The rest 40 rats in group OVX were divided into ovariectomized group (group B), ovariectomized and treated with estrogen (group C), ovariectomized and treated with parathyroid hormone (group D), ovariectomized and treated with estrogen and parathyroid hormone (group E) at random with 10 in each group. Group A and B injected physiological saline (1 mL x kg(-1)), group C injected estradiol benzoate (10 microg x kg(-1)), group D injected parathyroid hormone (20 microg x kg(-1)), group E injected parathyroid hormone (20 microg x kg(-1)) and estradiol benzoate (10 microg x kg(-1)). The intraperitoneal injection were maken every other day to rats in each group, which continued for 8 weeks. The bone mineral density (BMD), bone histomorphology and serum Ca, P, alkaline phosphatase (ALP) were measured after therapy.
RESULTSAfter 8 weeks of ovariectomy, the lumbar BMD of ovariectomized rats were significantly declined compared with those of the sham-operated rats (P < 0.05). Eight weeks later after the drug use, the BMD, %Tb.Ar, Tb.Th, Tb.N in group C, D, E were slightly elevated compared to group B, especially the group E (P < 0.05). Serum calcium and phosphorus values did not change significantly (P > 0.05). ALP values in group B was significantly higher than that in group A (P < 0.05).
CONCLUSIONIntermittent application of parathyroid hormone in small doses can increase alveolar BMD of castration rats and improve their bone structure. And it can have synergy effects on the treatment of osteoporosis if it is used combining with estrogen.

8.Effect of MG132 on the expression of ERK1/2 and connective tissue growth factor in rat peritoneal mesothelial cells induced by high glucose

Objective To observe the effect of MG132 on the expression of extracellular regulated kinase 1/2 (ERK1/2) and connective tissue growth factor (CTGF) in rat peritoneal mesothelial cells (RPMCs) induced by high glucose.Methods RPMCs were isolated,cultured and passaged by trypsin,then identified.The second generation of cultured RPMCs were used in the experiment.RPMCs were divided into normal control group,high glucose (1.5%,2.5%,4.25%) for 24 hours,high glucose (2.5%) for 0,12,24,48 hours,incubated with MG132 (0.5,1,2 μmol/L) for half an hour and then with high glucose (2.5%) for 24 hours.ERK1/2 protein was detected by Western blotting,and CTGF protein in supernatant was detected by ELISA.Results Compared with the control group,the expression of p-ERK1/2 was significantly increased in the groups stimulated by high glucose (P <0.01),reached the peak at 24th hour (P < 0.01),and then the expression decreased at 48th hour,but still was higher than that in the normal control group (P < 0.01).CTGF protein expression of RPMCs induced by high glucose increased,in time-and dose-dependent manner (P < 0.05).MG132 could significantly decrease the expression of ERK1/2 and CTGF induced by high glucose (P<0.05).Conclusions MG132 can decrease the expression of p-ERK1/2 and CTGF in RPMCs induced by high glucose.The ubiquitin proteasome pathway participates in the development of peritoneal fibrosis,and blocking the way may contribute to the prevention of peritoneal fibrosis.
9.Identification of rodent residues by DNA barcoding technology in plague areas

Objective To identify rodent residues using DNA barcoding technology in plague areas of Shaanxi and to analyze the feasibility of DNA barcoding method.Methods DNA extraction,PCR,electrophoresis and sequence determination and alignment were used to determine the cytochrome C oxidase Ⅰ (COI) gene sequence from 62 residues of 7 species identified by morphology in 8 different parts.COI gene sequence was analyzed using BLAST software of American National Center for Biotechnology Information (NCBI) for sequence homology comparison and the phylogenetic tree was constructed by using the neighbor joining(NJ)method of COI gene sequence.Results In addition to the hair,CO I genes of the feet,the tail,the fur,the muscle,the ribs,the ear,and the eye were amplified,respectively,and the size of amplified fragment was similar to the size of the product with an expected fragment(700 bp),and the fragment was a single band.COI genes of 59 residue specimen were obtained by DNA sequencing and there were high degree of homologies between CO I gene sequences of Meriones unguiculatus,Cricetulus barabensis,Meriones meridianus,Dipus sagitta,Phodopus roborovskii,Rattus norvegicus and Allactaga sibirica and their corresponding host genes in NCBI(99.0%,98.1%,99.8%,98.9%,99.5%,99.1%,98.3%).NJ method showed that 7 NJ phylogenetic trees were constructed with COI sequence of 59 species residues.The CO I sequences of same rodent with different residues were clustered into one group.Condusion DNA barcoding technology can identify host animal residues in plague areas,and the identification results are reliable.
10.Correlation between the uncertainty in illnessand social support in elderly patients with prediabetes

Objective To investigate the level of uncertainty of illness and social support state in patients with prediabetes,and to discuss the correlation between the two.Methods 243 cases with prediabetes were analyzed by Uncertainty in Illness Scale and Social Support Rating Scale.Results There was a middle uncertainty in the elderly patients with prediabetes.The objective support,sabjective support,social support utilization degree and social support score were negatively related to the complexity of uncertainty(r--0.419,-0.433,-0.390 and-0.421,respectively,all P< 0.05).Conclusions Medical staff should evaluate the uncertainty in illness in elderly patients with prediabetes and conduct nursing intervention accordingly,in order to reduce the uncertainty,increase social support,and ultimately improve the quality of life.
