1.Research and clinical application of nano-drug carriers

BACKGROUND: Compared with ordinary preparation, nano-drug carriers have the advantages of prolonged drug effects, increased therapeutic effects and small toxicity. However, reports concerning the application of nanoparticles, nano-liposomes, polymer micelles, nano-smart drug, polymer nano-body medicine, Dendrimers and other nanoscale drug carriers are few. OBJECTIVE: To summarize the characteristics, types and preparation methods of nano-drug carriers, and to elucidate its research progress. METHODS: The Medline database was searched by computer with key words of "Nano-drug carriers, nanoparticles,nano-liposomes, clinical application" to find literatures published between January 1991 to December 2009, and the language was limited to English. Simultaneously, articles published from January 1994 to December 2009 were searched in Chinese Journal Full-text database with the same key words,and thelanguage was limited to Chinese.Inclusion criteria: ①Articles are closely related to the research of nano-drug carrier. ②Related researches published on core journals in the lates years were preferential in selection.Exclusion criteria: ①Repeatability study. ②Meta analysis. RESULTS AND CONCLUSION: A total of 360 documents were seized by primary screen, finally 28 sub-standard literatures entered further analysis, including 25 reviews and 3 clinical or basic experiments. The nano-drug carriers are characterized by specificity, targeting, accurate and absorbable, which can be used in preventing and curing disease. The combination of nano carriers and specific drugs can promote the development of nano-biotechnology, and provide a reliable basis for clinical diagnosis and treatment.
2.IL-13Rα2 as a functional receptor mediates signal transduction

[ABSTRACT]IL-13isapleiotropiccytokinemainlysecretedbyactivatedTh2cells.Ithas2receptors,IL-13Rα1 and IL-13Rα2.The latter had been thought to serve exclusively as a decoy receptor for a long period of time due to its short cytoplasmic tail and lack of signal transduction structure .Since Fichtner-Feigl reported in Nature Medicine that IL-13 is in-volved in induction of TGF-βproduction and tissue fibrosis through IL-13Rα2-mediated signaling pathway , it was found that IL-13Rα2 has more sophisticated functions than just a simple decoy receptor as more and more researches have explored its signaling functions .This review combines the most advanced research results with previous investigation and discusses the gene structure, expression, production, distribution, subtype conversion and possible signal pathways mediated by this re-ceptor.More importantly, the connection with human diseases and the applications in disease diagnosis and molecule targe -ted therapy for cancer are also discussed .
3.Study on the activity of liposome doxorubicin for reversing multidurg resistance of tumor cells and its mechanism

ObjectiveTo explore the multidrug resistance(MDR) reversal activity of a novel compound liposome doxorubicin(PLD) and its mechanism.MethodsMTT assay was used to evaluate MDR reversal activity of PLD in P-gp expressing tumor cell lines,KBv200 and MCF-7/ADR.ResultsPLD had a strong reversal in vivo activity,recognized the strong reversal activity than verapamil reversal activity,in 5.0μmol/L concentration of multi-drug resistant cell KBv200 increased sensitivity to vincristine of 45 times.KBv200 PLD-dependent increased in intracellular concentration of rhodamine accumulation(0,2.5,5.0,10μmol/L).Mainly to its cardiac toxicity,bone marrow suppression and hair loss and other side effects were significantly reduced.ConclusionPLD was an efficient modulator,mainly through the continuous accumulation to tumor tissue,tumor local drug concentration,enhanced anti-tumor activity,
4.Expression level and clinical significance of LncRNA RP11-147L13.8 in peripheral blood of patients with acute cerebral infarction

Objective To investigate the expression level and clinical significance of LncRNA RP11-147L13.8 in peripheral blood of patients with acute cerebral infarction.Methods Fifty patients with acute cerebral infarction (case group) and 50 patients without cerebral infarction (control group) hospitalized in our department were randomly selected,and the expression level of LncRNA RP11-147L13.8 in peripheral blood was detected by real-time fluorescence quantitative PCR in case group and control group,and the correlation between LncRNA RP11-147L13.8 and cerebral infarction was investigated.Results 〖JP2〗The expression level of LncRNA RP11-147L13.8 in peripheral blood of patients with cerebral infarction was significantly higher than that of control group (P<0.05).The expression level of LncRNA RP11-147L13.8 in peripheral blood was positively correlated with the occurrence of acute cerebral infarction,and LncRNA RP11-147L13.8 was an independent risk factor for the occurrence of acute cerebral infarction.LncRNA RP11-147L13.8 in the diagnosis of acute cerebral infarction had an area under the curve of 0.750,a critical value of 0.40,a sensitivity of 0.66 and a specificity of 0.74.Conclusion LncRNA RP11-147L13.8 is differentially expressed in cerebral infarction,which is expected to become a potential biomarker for the diagnosis of acute cerebral infarction and provide new ideas for clinical diagnosis and treatment.
5.Nursing of patients with mandibular fractures treated with micro titanium plate rigid internal fixation

Objective To summarize the experience of nursing patients with mandibular fractures treated by micro rigid internal fixation.Method The following nursing methods including psychological interventions,pain intervention,dietary intervention, rehabilitation,intervention and prevention of complications were applied in the nursing care to 56 patients with mandibular fractures treated by micro rigid internal fixation.Results The surgical procedures of micro rigid internal fixation were successful,and all of them were cured and discharged.One case contracted gingivitis and another one oral ulcers,both cured by rinse with mouthwash. Conclusion Nursing intervention can improve the rehabilitative effect on patients with titanium mandibular fracture fixation,reduce the complications and improve clinical outcomes.
6.Preliminary study on the molecular mechanism for MPTP-induced dopaminergic neuronal degeneration in the substantia nigra of C57BL mice

Objective To study the molecular mechanism of MPTP-induced neuronal degeneration of substantia nigra in C57BL mice. Methods The C57BL mice model of Parkinson's disease were prepared by peritoneal administration of MPTP. These differentially expressed sequenced tags (EST) were obtained with mRNA differential display technique. Reverse Northern blotting was employed to eliminate the false positive ESTs. The true positive EST were cloned and sequenced. Results One of the two specific EST in degenerative substantia nigra was sequensed to be 121 bp and showed low homology with human v-abl Abelson murine leukemia viral oncogene (abl) exon 1b. Conclusions Our data suggested that MPTP may induce altered expression of promoting apoptotic genes to cause neuronal degeneration in substantia nigra.
8.The relationship among self-esteem, putting into perspective and social anxiety of college students

Objective To investigate the relationship between self-esteem and social anxiety,and the role of putting into perspective played in this relationship in university students.Methods Sample consisted university students of 376 females and 164 males,data were obtained on cognitive emotion regulation questionnaire,social anxiety subscale of the self-consciousness scale,and self-esteem scale.Results Cognitive emotion regulation strategies of putting into perspective is significantly and positively related to self-esteem(r=0.12,P< 0.01),it was also significantly and negatively related to social anxiety(r =-0.01,P < 0.05) ; self-esteem was significantly and negatively related to social anxiety(r =-0.35,P < 0.001).Cognitive emotion regulation strategies of putting into perspective played both a moderating(AR2 =0.124,P<0.05) and mediating role(F=30.717,P< 0.01) in the relationship between self-esteem and social anxiety.Conclusion Self-esteem is not only directly but also indirectly associated with social anxiety because of the moderation and mediation model of putting into perspective.
9.Effects of valproic acid on rats with experimental autoimmune encephalomyelitis and its immuno-modulatory mechanism

Objective To investigate the effects of valproic acid ( VPA ) on SD rats with experimental autoimmune encephalomyelitis ( EAE) and its possible immunomodulatory mechanism .Meth-ods Fifty female Sprague-Dawley ( SD) rats were randomly divided into five groups by random digit table , including control group (n=10), EAE group(n=10), low dose VPA treated group (100 mg/kg, n=10), median dose VPA treated group (300 mg/kg, n=10) and high dose VPA treated group (600 mg/kg, n=10).The SD rat model of EAE was induced by immunizing with a guinea pigs′spinal cord homogenate (GPSCH).Normal saline and various doses of VPA were given to rats in according groups twice a day from day 0 to day 19 ( close to the peak stage of EAE ) .The severity of EAE was scored according to the signs and symptoms.Pathological changes were observed through Hematoxylin-Eosin staining, and then the degrees of inflammatory infiltration were evaluated .The numbers of activated neuroglia that expressed Iba-1 in cerebral and lumber cords were counted by immunohistochemistry .The expression of IFN-γ, IL-17 and IL-10 in cer-ebral and lumber cords were measured by ELISA .Results Compared with EAE group , rats in the low, me-dian and high dose VPA treated groups had lower incidence of EAE and prolonged latency , but only the me-dian dose treated group showed significant alleviation in clinical symptoms (P<0.05).Both the median and the high dose treated group showed decreased inflammatory cell infiltration in CNS (P<0.05).Immunohisto-chemistry results showed that the numbers of activated microglia were significantly inhibited in rats treated with median and high dose of VPA in comparison with those in EAE group (P<0.05).Results of ELISA demonstrated that the expression of IFN-γand IL-17 in both median and high dose VPA treated groups were significantly decreased compared with those in EAE group (P<0.05), but only the median dose treated group showed a remarkably increased expression of IL-10 (P<0.05).Conclusion VPA, especially medi-um dose of VPA ( 300 mg/kg ) , had neuroprotective effects on rats with EAE .The possible mechanism might be associated with the inhibited activation of microglia and the increased percentage of anti -inflammato-ry cytokines .
10.Anterior Cruciate Ligament: Evaluation with Oblique Coronal MR Images

Objective To investigate the value of obli que coronal MR images in visualization of the anterior cruciafe ligment(ACL).Methods Conventional axial,oblique sagittal and coronal MR images were performed in 30 normal knees.Then oblique coronal T 2-weighted i mages,which was parallel to ACL,based on oblique sagittal T 2 -weighted images ,were acquired.The visualization of ACL on oblique coronal,oblique sagital and coronal T 2-weighted images was analysed.Results The visualization of the whole ACL was 100%(30/30 ) on one image of the oblique coronal T 2-weighted images, 83.3%(25/30) on on e image of the oblique sagaital T 2-weighted image. On the coronal T 2-weighte d images,the ACL appeared as segmental bundle.The whole ACL could not be visua lized on one image of coronal T 2-weighted images.Conclusion The whole ACL can be showed on one image of ob lique coronal T 2-weighted image,which is helpful in evaluation of the injure o f ACL.
