2.The genes related with the osmotic expansion in sugar cataract
Chinese Pharmacological Bulletin 1986;0(04):-
Lens is a perfect optical organ, which roots in the surface ectoblast and has good quality of light penetration and refraction. It consists of lens bursa, epithelia, cortex and nucleus from exterior to interior in turn. The life center of lens is the epithelia which not only excretes collagen Ⅳ to form lens bursa outside, and gradually develops to fiber cell to form cortex and nucleus of lens, but also supports all the energy of lens metabolism.Aldose reductase can be activated by high glucose in a long time. Then, the sorbitol accumulation of lens epithelial cells and fibre cells causes cortex to absorb water and expand.Besides, some research results showed that p-glyprotein,clcn3(Cl-channel 3) and aquaporin were also involved in the high osmotic expansion of the lens.

Journal of Clinical Otorhinolaryngology Head and Neck Surgery 2012;26(9):418-421
OBJECTIVE:
To explore the effects of 5-Aza-2'-deoxycitydine(5-Aza-dC) and trichostatin A (TSA) on the expression and methylation of CHFR in human laryngeal carcinoma cell line.
METHOD:
The mRNA expression and promoter hypermethylation and were detected by Realtime fluro-genetic quantitative PCR and methylation specific PCR in Hep-2 cell line, which were cultured in vitro and then treated with different concentrations of 5-Aza-dC and TSA.
RESULT:
Compared with the control team, 5-Aza-dC alone reactivated expression of the CHFR in Hep-2 cell line (1.75 +/- 0.21). TSA had no effect on gene expression (1.05 +/- 0.13). The combined treatment with 5-Aza-dC and TSA increased gene expression (2.15 +/- 0.18). The cell lines showed a characteristic DNA methylation status. 5-Aza-dC and combined 5-Aza-dC and TSA resulted in demethylation of CHFR. In contrast, TSA alone did not affect the DNA methylation status of CHFR.
CONCLUSION
Hypermethylation of CHFR gene promoter is a common event in the occurrence and development of laryngeal carcinoma. The promoter aberrant methylation of CHFR is a main cause for down-expression of CHFR. After either treatment with 5-Aza-dC alone or in combination with TSA, the expression of CHFR is up-regulated duo to the reversal methylation. It can be a new idea to the therapy of laryngeal carcinoma.

Objective To study the changes of plasma cystatin C level (PcyC),and evaluate the effects of the joint use of atorvastatin and probucol on PcyC and severity of coronary lesion in patients with borderline lesion of coronary artery. Methods One hundred and thirty consecutive patients with borderline coronary lesion assessed by quantitative coronary angiography were enrolled into borderline coronary lesion group (BCL),and another 136 subjects without coronary lesion were enrolled as controls (CTR).And in the meantime, the subjects in BCL group were randomized (closed envelope method) into routine treatment subgroup ( RTT,n =60),and combined treatment subgroup in which patients were treated with atorvastatin 20 mg plus probucol 1.0 g daily in addition to routine medication ( CBT,n =70) for 6 months. There were no statistical differences in basic clinical features between two subgroups.PcyC,high-sensitive C-reactive protein (hs-CRP),total cholesterol (TC),low-density lipoprotein cholesterol (LDL-C),high-density lipoprotein cholesterol ( HDL-C ) and triglycerides (TG) were determined.Of them,104 patients in BCL group rechecked by coronary angiography.Comparison of biomarkers carried out between two groups by using a number of independent-sample t-test and analysis of variance. For enumeration data,chi-square test was used to compare mean values of biomarkers between groups. P < 0.05 was considered statistically significant. Results PcyC levels were significantly higher in BCL group than those in CTR group ( P < 0.05 ).Compared with RTT subgroup, levels of PcyC,TC,LDL-C,TG and hs-CRP were more significantly decreased in CBT subgroup (P < 0.05,P < 0.01 ).Moreover, there was a trend of slight decrease in the mean percent of stenosis (MPS) of coronary artery with borderline lesion in RTT subgroup treated for 6 months, whereas more marked decrease in the MPS of coronary artery with borderline coronary lesion in CBT subgroup treated for 6 months ( P > 0.05 ; P < 0.05 ).Conclusions Cystatin C plays an important role in the pathogenesis of coronary artery, and PcyC is associated with severity of coronary lesion, the combination of atorvastatin and probucol decreases the PcyC level, and it may be the treatment of choice for borderline lesion of coronary artery.

Chinese Journal of Practical Nursing 2018;34(28):2182-2186
Objective To test the reliability and validity of the Chinese version of Insomnia Severity Index(ISI-C) in patients with insomnia. Methods One hundred and twenty patients with insomnia were selected from Zhongshan Hospital Affiliated to Dalian University. The subjects had completed the ISI-C scale and Pittsburgh Sleep quality Index(PSQI) scale, Epworth Sleepiness Scale(ESS) at the same time. The reliability and validity of the scale were tested by Cronbach α coefficient, item correlation analysis, exploratory factor analysis and ROC curve test. Results The Cronbach α coefficient of the ISI-C was 0.804,the test-retest reliability was 0.887 (P<0.01). The items of ISI-C scale were correlated with the corresponding items of PSQI (r=0.831, P<0.01), ESS(r =0.218, P<0.05). Two common factors were extracted by factor analysis including the severity of insomnia and the influence of insomnia, and the cumulative contribution rate was 63.117% (P<0.01).When the cutting point was 10.5 points, the sensitivity and specificity were 84.0% and 92.3%, respectively (P<0.01). Conclusions ISI-C scale is a reliable and valid instrument. It can be used to measure the insomnia of patients in hospitals.

Journal of Clinical Otorhinolaryngology Head and Neck Surgery 2010;24(15):673-677
OBJECTIVE:
To explore the relationship between the level of expression and hypermethylation of the CHFR gene and the occurrence and development of laryngeal squamous cell carcinoma (LSCC).
METHOD:
The mRNA expression and promoter hypermethylation were detected by Realtime fluro-genetic quantitative PCR and methylation specific PCR in 50 LSCCs (LSCC group) and 15 normal laryngeal tissue (control group).
RESULT:
1) CHFR mRNA was shown in the control group, while the mRNA was loss expression in the 2 LSCC (4%), and the level of mRNA expression was significantly lower in the LSCC group. The relative ratio was 0.50 +/- 0.12, which is 0.30 +/- 0.04 at the early stage of the LSCC and 0.70 +/- 0.21 at the advanced stage, respectively. The discrepancy had statistical significance (P<0.01). 2) The methylation rate of CHFR was 22% (11/50) in the LSCC tissues, which was not found in the normal tissues. The aberrant methylation of CHFR was observed in 10 of the patients at the stage I and stage II of LSCC , in 1 of the patients at the stage III, and was absent at the stage IV. There was significant difference between the aberrant methylation of CHFR and the stage of carcinoma (P<0.01). 3) The mRNA expression level of the aberrant methylation patients was 0.11 +/- 0.05, which was significantly lower than that of the unmethylation patients 0.75 +/- 0.13. Gene inactivation was observed in 2 of the 11 patients with the aberrant promoter methylation. The methylation was associated with the expression of mRNA, with the correlation coefficient 0.387 (P<0.05).
CONCLUSION
Hypermethylation of CHFR gene promoter is associated with loss or lower expression of CHFR mRNA in the LSCCs, and it may contribute to the occurrence and development of LSCC. The promoter aberrant methylation of CHFR may be one of the early diagnostic and therapeutic marker genes.

Journal of Preventive Medicine 2019;31(7):678-682
Objective:
To explore the relationship between different temperature indicators and hospital admission for childhood pneumonia.
Methods:
The hospital admissions for pneumonia in children aged 0-14 years and meteorological data in Ningbo from 2015 to 2017 were collected. A distributed lag non-linear model combined with a generalized linear model was employed to analyze the exposure-response relationships between different temperature indicators(daily average, minimum and maximum temperature;the first percentile as low temperature and the 99th percentile as high temperature)and hospital admission for childhood pneumonia.
Results:
A total of 4 542 cases of childhood pneumonia were recruited. There were obvious seasonal fluctuations found in the inpatient volume of childhood pneumonia, which peaked in winter and bottomed in summer. After adjusting for potential confounding variables such as relative humidity,PM2.5,long term trend and seasonal trend, the results suggested that after exposed to whether low or high temperature, the inpatient volume of childhood pneumonia would increase. When the daily average temperature and daily minimum temperature were employed, the effect of high temperature on the increase of inpatient volume for childhood pneumonia was statistically significant and the cumulative relative risk for a lag of 0-7 days were 1.52(95%CI:1.04-2.23)and 1.59(95%CI:1.08-2.34),respectively. When the daily maximum temperature was employed, the effect of low temperature on the increase of inpatient volume for childhood pneumonia was statistically significant and the cumulative relative risk for a lag of 0-7 days were 1.30(95%CI:1.02-1.66).
Conclusion
Our findings suggested that an increased risk of hospital admission for childhood pneumonia was associated with both low and high temperature.
