OBJECTIVE: To establish a RP-HPLC method for the determination of pamidronate disodium. METHODS: The determination was performed on C18 column with acetonitrile-0.4% sodium hydroxide solution of EDTA-Na2 (14∶86) as mobile phase at a flow rate of 0.6mL?min-1,the sample size was 10?L and the detection was performed by fluorescence detector with excitation wavelength at 395nm and emission wavelength at 480nm. RESULTS: The linear range of pamidronate disodium was 7.2~16.8?g?mL-1(r=0.999 8,n=9),the recovery rate was 100.11%(RSD=0.7%, n=9). CONCLUSIONS: The established method is simple and accurate.
3.Establishment of segmentational data set of larynx based on Chinese visible human

Objective To establish a digital segmentational data set of larynx based on Chinese visible human (CVH). Methods Magnetic lasso and polygon tools of Photoshop were used to segment the small organs and structures of larynx of CVH to establish the segmentational data set of larynx. After conversion of the image format, the segmentational structures were extracted automatically with Thresholding Method and presented 3-D visualized, and then were checked up by its result of 3D reconstruction with Amira 4.1 software. Results Many small structures of larynx were segmentated, such as laryngeal cartilage, laryngeal muscles, vocal cords and so on. Then the segmentational data set of larynx based on CVH was established, which can be used to 3-D reconstruction accurately. Conclusion The segmentational data set of larynx is accurate and integrated, which is helpful to establish the elaborate model of larynx and can provide the method of color image segmentation.
4.Cross-section study of early renal damage in patients with chronic hepatitis B

Objective To investigate early renal damage of chronic hepatitis B (CHB) patients and the risk factors related to their renal function. Methods CHB patients who visited the second people’s hospital but did not receive systemic treatment were enrolled in our study. Those who visited for general check-up with no hepatic findings during the same period were selected as control group. Glomerular filtration rate (GFR) of all the participants were estimated by simplified MDRD equation and CKD-EPI equation (designated as M-eGFR and C-eGFR respectively). Influence factors of eGFR were statistically analyzed. Results In the total 528 cases in CHB group, 88 (16.67%) and 62 (11.74%) suffered declined M-eGFR and C-eGFR respectively. By contrast, 10 (8.77%) and 6 (5.26%) cases in the total 114 cases in control group present declined M-eGFR and C-eGFR ac?cordingly. Percentages of renal function impairment, estimated by both M-eGFR and C-eGFR, were higher in the CHB group than those in control group. The difference was statistically significant (χ2=4.518, P<0.05;χ2=4.156, P<0.05). Multiple linear regression analysis indicated that age, HBsAg and body mass index (BMI) were risk factors of M-eGFR while age, HBsAg, gender and serum albumin were risk factors of C-eGFR. On the other hand, HBV-DNA and HBeAg were not risk factors for M-eGFR or C-eGFR. Conclusion HBV infection can lead to early renal damage. Age and HBsAg are main risk factors of renal function impairment. Therefore, renal function should be scrutinized in CHB patients.
5.Practice and experience of training model based on automated breast ultrasound system

The new ultrasound device, automated breast ultrasound (ABUS), has the advantages of repeatable reading of standardized whole breast images, and can separate ultrasound scanning and diagnosis. ABUS is more suitable for breast ultrasound training. This study compared the breast ultrasound imaging training model with ABUS diagnosis practice as the core and the traditional breast ultrasound imaging training model with handhold ultrasound operation and diagnosis as the core, and found that the new training model with ABUS diagnosis practice as the core had a short training period. The diagnostic level of doctors have improved rapidly, and the new training model could efficiently train more technically qualified ultrasound doctors for breast disease.
6.Expression and location of YB-1 protein in hepatocellular carcinoma and their clinical significance

ObjectiveTo study the expression levels of YB-1 protein in hepatocellular carcinoma (HCC) tissues,tissues adjacent to tumour and normal liver tissues,and to explore the clinical significance.MethodsTo study the expression levels of YB-1 protein,immunochemistry was carried out on 58 HCC tissues and their corresponding adjacent tissues to tumour and on 20 normal liver tissues.The YB-1 protein with nuclear translocation was detected by Western blot in HCC tissues,tissues adjacent to tumour,normal liver tissues,HCC cell lines including QGY-7701 and SMMC-7721,and a normal hepatic cell line LO2.ResultsPositive signals of YB-1 protein were detected at a high level in HCC tissues (72.4%,42/58) when compared to tissues adjacent to tumour (41.4%,24/58) and to normal liver tissues (35 %,7/20) (P>0.05).The nuclear expression of YB-1 (31%,18/58) was significantly correlated with the pathologicalgrade,tumour size and portal venous invasion (P<0.05).Using Western blot,the YB-1 protein with nuclear translocation was expressed at a higher level in HCC tissues (0.474±0.107) than in tissues adjacent to tumour and in normal liver tissues (P<0.05).Similar results were obtained in HCC cell lines and the normal hepatic cell line (P<0.05).ConclusionThe high expression of YB-1 in HCC and its nuclear translocation may be involved in human HCC progression.YB-1 may be a potential target for HCC treatment in the future.
7.Differential protein expression after acute spinal cord injury

BACKGROUND:In mass spectrometry analysis, the same protein in different samples labeled with isobaric tags for relative and absolute quantitation presents the same mass-to-charge ratio, while in the tandem mass spectrometry analysis, the ion signal shows different mass-to-charge ratio (114-121). Thus the quantitative information of the related proteins can be obtained. OBJECTIVE:To establish the protein spectrum of spinal cord tissue differences proteins after acute spinal cord injury, and to explore the spinal differential protein expression on the molecular level using isobaric tags for relative and absolute quantitation combined with LC-MS/MS mass spectrometry technique. METHODS:Eight Sprague Dawley rats were selected to establish the acute spinal cord injury models using Al en’s method. The rats were randomly divided into 0 hour spinal cord injury group and 8 hours spinal cord injury group, four rats in each group. The spinal cord tissues were col ected after injury, and the spinal cord tissue differences proteins were analyzed with isobaric tags for relative and absolute quantitation technique after acute spinal cord injury. RESULTS AND CONCLUSION:Total y 220 differential y expressed proteins were identified in this research, the number of up-regulation proteins was 116 and the number of down-regulation proteins was 104. There were 12 differential proteins related to neural regeneration, and among the 12 proteins, there were seven up-regulation proteins and five down-regulation proteins. The various identified differential proteins and significantly expressed nerve growth factors in this experiment can be used as the biomarkers of acute spinal cord injury or used as the strong evidence for the clinical management and monitoring of the injury process and target therapy of acute spinal cord injury, as wel as the effect evaluation.
8.Comparison of ultrasonography and X-ray in the assessment of peripheral joints invovement in psoriatic arthritis

Objective To compare and investigate the role of ultrasonography and X-ray examination in the assesment of peripheral joints in psoriatic arthritis (PsA).Methods One hundred and twenty-five groups joints of twenty-five patients with PsA underwent ultrasonography and X-ray examination,including knee joints,wrist joints,ankle joints,metacarpophalangeal joints and interphalangeal finger joints,metatarsophalangeal joints and interphalangeal toe joints.The results of the two examinations were analysed and compared.Results The numbers of patients with abnormal joints in knee joints,wrist joints,ankle joints,metacarpophalangeal/interphalangeal finger joints,and metatarsophalangeal/interphalangeal toe joints were 9,9,9,10,and 11,respectively.Ultrasonography found more abnormal joints than X-ray ( P <0.01 ).The cardinal ultrasonographic manifestations of PsA were synovitis (29),tendonitiS (16),and bone erosion (7).The cardinal X-ray manifestations of PsA were bone destruction (11 ),periarticular osteoporosis (5),periosteal reaction (3) and periarticular soft tissue swelling (4).Ultrasonography found more synovium and tendon abnormalities than X-ray (P < 0.01 ),and X-ray found more bone abnormalities than ultrasonography ( P <0.01).Conclusions Ultrasonography has the ability of demonstrating early changes of peripheral joints in PsA.The combination of ultrasonography and X-ray can make the diagnosis of PsA more correct and comprehensive.
9.The study of proteomics in acute spinal cord injury rats by iTRAQ technology

BACKGROUND:Isobaric tags for relative and absolute quantitation (iTRAQ) mass spectrometry technology studys the information of relevant protein according to the ion signal shows different mass-to-charge ratio in the tandem mass spectrometry analysis. OBJECTIVE:To establish the protein spectrum of differential proteins in cerebrospinal fluid of acute spinal cord injury rat model, study the secondary injury mechanism and find an effective method of treating acute spinal cord injury from molecular level. METHODS:Acute spinal cord injury was produced in Sprague-Dawley rats and iTRAQ technology was applied to analyze the differential proteins in cerebrospinal fluid of acute spinal cord injury rat model. RESULTS AND CONCLUSION:Total 722 proteins have been identified in this study, including 107 differentialy expressed proteins, 63 downregulated proteins and 44 upregulated proteins. There were 19 proteins related to neurogenesis, including 14 up-regulation proteins and 5 down-regulation proteins. Seven proteins contributed to the regulation of neurogenesis. The differential proteins and growth factor identified in this study can be taken as the biomarkers of acute spinal cord injury or indicators of clinical monitoring of the progression, target treatment and efficacy assessment after acute spinal cord injury.
10.Feasibility of anticoagulation therapy after mechanical valve replacement in grass-root health institutions

Objective To assess the feasibility of anticoagulation therapy after mechanical valve replacement in grass-root health institutions.Methods One hundred and sixty one patients with mechanical valve replacement received anticoagulation therapy with warfarin,including 79 cases receiving the therapy in grass-root health institutions (test group) and 82 cases in the tertiary hospitals (control group).The patients were followed up for 12 months after operation;the rate of anticoagulation efficacy,the anticoagulationrelated complications,and the anticoagulation-related cost were documented and compared between two groups.Results The international normalized ratio (INR) tests were performed for 1 021 times in test group and 717 times were up to anticoagulation standard (70.2 %,717/1 021),while INR tests in control group were performed for 965 times and 688 times were up to standard (71.3%,688/965);there were no significantly differences in efficacy rate between two groups (P > 0.05).There were no significant differences in rate of bleeding events and thrombosis between two groups [16.5% (13/79) vs.12.2% (10/82),6.3%(5/79) vs.4.9%(4/82),respectively,x2 =0.596,P=0.44,x2 =0.161,P=0.69].The anticoagulation-related cost per month and per patient in test group was significantly lower than those in control group [(63.1 ±.12.8) vs.(176.6 ± 16.4) yuan,t =48.716,P <0.05].Conclusion Compared with the tertiary hospital,the anticoagulation therapy in grass-root institutions can accomplish the similar clinical outcomes and significantly reduce the medical cost in patients with mechanical valve replacement.
