Objective To investigate whether HA affects the inflammatory cytokines and chemokines of synoviocytes. Methods FLS were derived from the SD mice with knee OA by papaya enzyme and stimulated by IL-1. Then, FLS were cultured with 600 ~ 800 kDa HA. Anti-CD44 blocking experiments were determined. The expressionsof TNF-γ, IL-1 , IL-6 , MMP-3 , MMP-9 , CX3CL1 , CCL11 , CCL2 , CXCL12 were detected by RT-PCR. In addition, the tunel test was used to detect the apoptosis rate. Results MMP-3, CX3CL1 and CCL2 gene expression were down regulated by HMW-HA in unstimulated FLS and TNF-α,MMP-3, CX3CL1 and CCL2 expression were decreased by HMW-HA in IL-1-stimulated FLS. CD44 blocking inhibited the down-regulatory effects of HMW-HA. Besides, HMW-HA promoted cell apoptosis under condition of inflammation. Conclusion HMW-HA has an anti-inflammatory effect by down-regulating of pro-inflammatory cytokines and chemokines and promoting apoptosis, possibly through the interaction of CD44 and HMW-HA.

Objective To observe the effect of psoralen on the proliferation of osteoblasts induced by alcohol, thus to explore the mechanism of psoralen in preventing and treating alcohol-induced osteoporosis. Methods The osteoblasts separated from neonatal rat skull were identified by alkaline phosphatase (ALP) staining method. The in-vitro confirmed osteoblasts were randomly divided into 4 groups, namely blank control group, alcohol group (model group), psoralen group, and psoralen plus alcohol group. The alcohol-induced osteoblast proliferation in various groups was determined by methyl thiazolyl tetrazolium (MTT) method. Results Compared with the blank control group, the alcohol-induced osteoblast proliferation at culturing hour 24, 48, 72, 96 in alcoholgroup was significantly decreased (P<0.05), and the proliferation in psoralen group at various time points only showed an increasing trend, the differences being statistically insignificant (P>0.05). Compared with the alcohol group, osteoblast proliferation at culturing hour 24 in psoralen plus alcohol group was significantly increased (P <0.05) , but the differences of osteoblast proliferation at culturing hour 48, 72, 96 h between the two groups were insignificant (P > 0.05). Conclusion Psoralen has certain effect on promoting alcohol-induced osteoblast proliferation in vitro.

OBJECTIVE:To provide unified standard for assessing the comprehensive ability of food and drug inspection and testing institutions in Guangxi Zhuang autonomous region. METHODS:The comprehensive ability evaluation index system for food and drug inspection and testing institutions in Guangxi Zhuang autonomous region was drafted by literature review. Indexes of the evaluation index system were determined by Delphi expert consultation, and the weights were calculated by analytic hierarchy pro-cess (AHP). RESULTS:Literature review and expert consultation had constructed the comprehensive ability evaluation index sys-tem for food and drug inspection and testing institutions in Guangxi Zhuang autonomous region, including 6 first-level indexes,16 second-level indexes and 35 third-level indexes. AHP had determined the weights of indexes at all levels, and the weights of the 6 first-level indexes as basic conditions, management system, testing ability, work performance, scientific research and satisfaction degree, were respectively 0.1898,0.1233,0.2280,0.1915,0.1551 and 0.1124,in which testing ability held the highest weight. CONCLUSIONS:A set of scientific and suitable evaluating index system for food and drug inspection and testing institu-tions in Guangxi Zhuang autonomous region has been constructed, and the weights of indexes at all levels have been determined, which can provide clear and unified standard for conducting comprehensive ability system evaluation in each inspection and testing institution in Guangxi Zhuang autonomous region.

Objective:To preliminarily observe the clinical efficacy of microwave hyperthermia combined with intensity-modulated radiotherapy (IMRT) and chemotherapy for patients with locally advanced gastric cancer. Methods:Forty patients who could not been operated or refused operation were enrolled in this clinical trial, who were confirmed as locally advanced proximal or distal gastric cancer by gastroscopy pathology and imaging. Radiotherapy was delivered by IMRT technology for 5 times per week with a total dose of 46 to 56 Gy (median dose of 50 Gy) in 25 to 28 fractions. Synchronous hyperthermia was given at 42 to 44℃ twice a week, 45 min/time. S-1 or capecitabine-based synchronous chemotherapy was performed, d1-14/3 weeks. The symptom remission rate, adverse reactions, objective remission rate (complete and partial remission) and survival were observed. Results:A total of 40 patients, aged between 56 and 83 years (median age of 71 years), were enrolled in this study. The male-to-female ratio was 7: 1. Among them, 38 cases (95%) showed symptom remission. The most common adverse reactions were grade 1-2 gastrointestinal reactions and leukopenia. The objective remission rate was 87.5%, the 2-year progression-free survival and overall survival rates were 68.6% and 70.5%, respectively. Conclusion:Preliminary findings demonstrate that microwave hyperthermia combined with chemoradiotherapy achieve satisfactory outcomes and yield tolerable toxicity in patients with locally advanced gastric cancer.

Objective:To establish the quality evaluation method of Prunellae spica dispensing granules based on three quality indexes of standard decoction. Methods:Fourteen batches of Prunellae spica were collected from different habitats. According to technical requirements, fourteen batches of Prunellae spica standard decoction and three batches of formula granules were prepared and the paste-forming rates were calculated. The fingerprints of Prunellae spica standard decoction and formula granules were established by Ultra High Performance Liquid Chromatography (UPLC). The similarity values of fingerprints between dispensing granules and standard decoction were calculated. The content and transferring rate of Rosmarinic acid were determined and calculated. Results:The average paste-forming rate of Prunellae spica was (12.59±2.32)%. The paste-forming rates of the three batches were 11.14%, 10.78% and 10.39% respectively. The average content of Rosmarinic acid in standard decoction was (18.99±9.74)mg/g. The average transferring rate was (60.58±7.87)%. The contents of three batches were 7.40 mg/g, 7.49 mg/g and 7.09 mg/g. The transferring rates were 52.06%, 50.10% and 50.40% respectively. Nine common fingerprint peaks were identified in the fingerprints of standard decoction and formula granules, two of which were identified as Rosmarinic acid and Caffeic acid by comparison of reference substance. The fingerprints similarity of Prunellae spica dispensing granules and standard decoction were 0.954, 0.973 and 0.952, respectively. Conclusions:The quality indexes of three batches of formulation granules are consistent with standard decoction. This method could provide reference for the establishment of quality standard of Prunellae spica dispensing granules.
