1.Protective Effect of Tongsaimai Pellets and Butylphthalide to PC12 Cell in Hypoxia/Hypoglycemia Model

Objective To discuss the protective effect of Tongsaimai pellets(TSMP) and butylphthalide(NBP) on PC12 cell in the hypoxia/hypoglycemia model.Methods Two ischemia models including hypoxia,hypoglycemia models were used to assay the anti-ischemic roles of TSMP and NBP in cultured PC12.Results TSMP and NBP possessed obvious protective effects on PC12 cells from two injured models.Both of them could increase the number of living cells and decrease LDH activity significantly,particularly in hypoglycemia injured model.Conclusion TSM and NBP have protective effect on PC12 cells from two injured models effectively in vitro.
2.Influence of material and configuration of microfiltration membranes on preparation of mono-ammonium glycyrrhizinate liposome by membrane extrusion method

AIM:To study the influence of material and configuration of microfiltration membranes on preparation of mono-ammonium glycyrrhizinate liposome(MGL) by membrane extrusion. METHODS: MGL were preparated by conventional rotary-evaporated film dispersion method and sonication,and MGL were extruding through 9 kinds of microfiltration membranes with 3 kinds of aperture specs of 0.1,0.2,0.45 ?m.The methods were evaluated by the filtration efficiency and the leakage rate of drug from liposomes. RESULTS: MGL were easily extruding through microfiltration membranes of NY6,PTFE,PVDF,PP and PC.MGL could not extruding through microfiltration membranes of PES,CN,CA and CA-CN.MGL become hardly to extrude through microfiltration membranes when increasing in the concentration of liposomes or the content of cholesterol.The leakage rate of drug from MGL was small when extruding through microfiltration membranes. CONCLUSION: The material and configuration of microfiltration membranes can remarkably influence the preparation of MGL by membrane extrusion method,and PC microfiltration membrane was selected.
3.Effect of high glucose or angiotensin Ⅱ on the expression of toll-like receptor 4 signal pathway,inflammatory and fibrotic factors in human tubular epithelial cells

Objective To investigate the effects of angiotensin Ⅱ (Ang Ⅱ) or high glucose on the toll-like receptor 4 (TLR4) expression,inflammatory cytokines and fibrotic factors in human tubular epithelial cells (HK-2),revealing the innate immune-related pathogenesis of diabetic nephropathy (DN) which may have clinical implications.Methods Three TLR4 siRNA sequences were designed and synthetized.After transfection,the most effective siRNA was selected to use for further expriments.The experiment consisted of 2 parts.Part 1:Cells were divided into three groups:normal-glucose group (NG,5.5mmol/L glucose),mannose group (M,5.5 mmol/L glucose + 19.5 mmol/L mannose),High-glucose group (HG,25 mmol/L glucose),preliminary validated the effects of high glucose and high osmotic pressure.Part 2:Cells were divided into seven groups:NG group,HG group,Ang Ⅱ group,Ang Ⅱ + negative group,HG+ negative group,Ang Ⅱ + siRNA group and HG+ siRNA group.Real time PCR was used to analyze the mRNA expression of TLR4,myeloid differentiation factor 88 (MyD88),heat shock protein 47 (HSP47).Western blotting was used to observe the protein expression of TLR4,MyD88,HSP47,NF-κB,type Ⅳ collagen (ColⅣ).ELISA was used to detect the expression of monocyte chemotactic protein-1 (MCP-1) and interleukin-6 (IL-6).Results Compared with NG group,TLR4,MyD88,HSP47 mRNA and TLR4,MyD88,NF-κB,ColⅣ,HSP47 protein were highly expressed under high glucose or Ang Ⅱconditions (P < 0.01),and the expression levels of MCP-1 and IL-6 also increased significantly (P < 0.01).Compared with HG or Ang Ⅱ group,the above indicators were obviously inhibited in the TLR4 siRNA groups (P<0.01).Comparison between blank vector transfected groups and HG group as well as Ang Ⅱ group indicated no statistic significance (P > 0.05).Conclusions Both Ang Ⅱ and high glucose stimulate TLR4 expression,which result in the up-regulation of inflammatory and fibrotic factors in HK-2.Specific target of TLR4 gene silencing can block the TLR4 pathway that is activated by high glucose and Ang Ⅱ,and thus reduce the inflammatory and fibtogenic factors' release.TLR4 signal is the common innate immune response pathway which induces the release of inflammatory and fibrotic factors in HK-2 under high glucose or high angiotension conditions.
4.24 hours in the practice of the management mode of hernia district hospi-tals

Objective To study on 24 hours hernia grassroots level in the district hospital management model practices. Methods The subjects were 283 cases of inguinal hernia patients selected from January 2008 to December 2010 in our hospital as the traditional model of the group , the treatment in the traditional management model , our hospital im-proved the management mode after January 2011 , 326 cases of inguinal hernia patients selected from January 2011 to December 2013 in our hospital as a new model group , that under the new model in 24 hours hernia treatment mode. Results Under the new model,patients waiting for surgery, hospitalization time and the days of antibiotic use were sig-nificantly shorten than the traditional model , the cost of hospitalization was significantly reduced , the difference was statistically significant (P<0.05). Under the new model, patient satisfaction was significantly higher than the traditional model, and the complication rate was significantly lower than the traditional model, the difference was statistically sig-nificant (P<0.05). Conclusion 24 h hernia is safe, effective, and economic characteristics, enabling a more rational use of health care resources and deployment , significantly reducing health care costs , can promote the application in district hospitals.
5. Assessment of comprehensive nutritional status and eating behavior among 311 middle aged and aged women with osteoporosis in Chengdu

Objective:
To investigate the comprehensive nutritional status and diet behavior of middle aged and elderly women with osteoporosis, and thereby to explore the relationship between diet behavior and comprehensive nutritional status.
Methods:
311 middle-aged and elderly women with osteoporosis in Chengdu were included in this study. Mini Nutritional Assessment (MNA) was applied to assess their comprehensive nutritional status. Information of social-demographic characteristics and diet behavior (about meals, snacks and water drinking, etc.) of the subjects was collected by questionnaire. Chi square test was used to assess the differences in nutritional status among patients who have different eating behaviors. Logistic regression analysis was performed to evaluate the relationship between diet behaviors and comprehensive nutritional status.
Results:
The mean MNA score of subjects was 25.8±2.5. 20.3% (63/311) of the subjets were at risk of potential malnutrition, but there was no malnourished subjects found. 46.9% (46/311) of the subjects were in good appetite. 95.2% (296/311) of them had a fixed food intake each meal. 65.8% (198/311) of them had snacks every day, and the most common choice was fruit (86.4% (248/287)). 54.8% (165/311) of them had initiative drinking water habits, and the most common choice was plain boiled water (79.9%, 246/308). 76.5% (238/311) of them had daily portable water less than 1 500 ml. 6.Effect of Dendrobium officinale Polysaccharides in Reversing Hepatic Fibrosis in Rats via Notch Signaling Pathway and Ultrasound Evaluation

ObjectiveTo investigate the effect of Dendrobium officinale polysaccharides (DOP) in reversing carbon tetrachloride (CCl4)-induced hepatic fibrosis (HF) in rats via the Notch signaling pathway and evaluate the therapeutic effect of DOP by ultrasound elastography. MethodFifty-six male SD rats were randomized into normal, model, colchicine (1×10-4 g·kg-1), Fuzheng Huayu powder (0.45 g·kg-1), and low-, medium-, and high-dose (0.05, 0.1, 0.2 g·kg-1) DOP groups (n=8). The rats in the model group and each treatment group were injected subcutaneously with a mixture of CCl4-olive oil (2∶3) once every 3 days for 10 weeks. After 6 weeks of modelling, the rats were administrated with corresponding drugs once a day for 4 weeks. Hematoxylin-eosin (HE) staining and Masson staining were employed to observe the pathomorphological changes of the liver tissue. An automatic biochemical analyzer was used to measure the serum levels of alanine aminotransferase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), and total bile acids (TBA). Enzyme-linked immunosorbent assay (ELISA) was employed to measure the serum levels of hyaluronic acid (HA), laminin (LN), type Ⅲ precollagen (PC-Ⅲ), and type Ⅳ collagen (Col-Ⅳ). The mRNA and protein levels of α-smooth muscle actin (α-SMA), Notch1, Jagged1, and Hes1 in the liver tissue were determined by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) and Western blot, respectively. The Young's modulus (YM) of the rat liver was measured by acoustic radiation force impulse (ARFI) elastography before and after treatment. Then, the correlations of YM with the serum levels of HA, LN, PC-Ⅲ, and Col-Ⅳ and the protein levels of α-SMA and Notch1 signaling pathway-related factors in the liver tissue were analyzed. ResultCompared with the normal group, the model group showed disordered arrangement of liver cell cords, obvious infiltration of inflammatory cells, appearance of a large number of fat vacuoles, and fibrous proliferation, elevated levels of ALT, AST, TBA, ALP, HA, LN, PC-Ⅲ, and Col-Ⅳ in the serum, and up-regulated mRNA and protein levels of α-SMA, Notch1, Jagged1, and Hes1 in the liver tissue (P<0.01). Compared with the model group, drug interventions alleviated the pseudolobule formation and the collagen deposition in confluent areas. Except that the serum level of ALT in the low-dose DOP group had no significant changes, drug interventions, especially high-dose DOP, lowered the levels of ALT, AST, TBA, ALP, HA, LN, PC-Ⅲ, and Col-Ⅳ in the serum and down-regulated the mRNA and protein levels of α-SMA, Notch1, Jagged1, and Hes1 in the liver tissue (P<0.05, P<0.01). The results of ARFI and correlation analysis showed that the YM of the liver tissue was increased in the model group (P<0.01) compared with that in the normal group, Compared with the model group, drug interventions decreased YM (P<0.01). YM was positively correlated with the expression levels of HA, LN, PC-Ⅲ, α-SMA, Notch1, Jagged1, and Hes1s (r=0.754, 0.734, 0.801, 0.885, 0.896, 0.757, and 0.800, respectively, P<0.01), and it had a moderate correlation with Col-Ⅳ (r=0.688, P<0.01). ConclusionDOP can reverse HF by down-regulating the Notch1/Jagged1/Hes1 signaling pathway. YM can be used as an indicator in the assessment of the efficacy of DOP against HF.
7.Dendrobium Officinale Polysaccharide Alleviates Hepatic Fibrosis via Epithelial-Mesenchymal Transition

ObjectiveTo investigate the effect of Dendrobium officinale polysaccharide (DOP)on CCl4-induced hepatic fibrosis(HF)and its mechanism. MethodsA total of 56 male SD rats were randomly divided into seven groups: normal group(NG),model group(MG),colchicine group(CG, 0.1 mg/kg), Fuzheng Huayu group(FG, 0.45 g/kg),low-dose DOP group(LDG, 0.05 g/kg),middle-dose DOP group(MDG, 0.1 g/kg)and high-dose DOP group(HDG,0.2 g/kg),with 8 rats in each group. HF rat model was established by subcutaneous injection with 40% CCl4 olive oil mixture, every 3-day for 10 weeks. At the end of the sixth week, the drug groups were treated with colchicine, Fuzheng Huayu and DOP solution by gavage respectively, once a day for 4 weeks. NG and MG groups were similarly handled with an equal amount of 0.9 % normal saline. Liver histopathology was detected using hematoxylin-eosin (HE), Masson and Sirius red staining; blood biochemistry was tested for liver function and four indicators of HF; RT-qPCR and Western Blot were used to measure the expression of α-SMA, Col-I, E-cadherin, and ZEB1 genes and proteins in the liver tissues of rats, respectively. ResultsHE, Masson, and Sirius red staining showed that the liver tissue of MG rats had typical pathologic features of HF, and the degree of HF was alleviated in LDG, MDG, and HDG rats, respectively. Liver function test results showed that the serum AST, TBIL, and AKP levels were significantly lower in LDG, MDG, and HDG, compared with those of the MG (P < 0.05 or < 0.01). Meanwhile, ALT levels in serum deceased remarkably except in LDG (P < 0.05 or < 0.01). The four results of HF showed that the serum HA, LN, PC-Ⅲ, and COL-Ⅳ levels in LDG, MDG, and HDG rats were significantly decreased compared with those of the MG (P < 0.05 or < 0.01). The relative expressions of α-SMA, COL-I, and ZEB1 genes and proteins were significantly decreased in the liver tissues of LDG, MDG, and HDG (P < 0.05 or < 0.01), and the relative expression of E-cadherin gene and protein increased (P < 0.05 or < 0.01). In addition, the expressions of HA, α-SMA, COL-I, ZEB1 and E-cadherin were dependent on the dose of DOP. ConclusionDOP alleviated the degree of CCl4 induced HF in rats by inhibiting the epithelial-mesenchymal transition in liver tissue.
