Objective To explore the effect on the differentiatiation of bone marrow mesenchymal stem cells(BMSCs) induced by Ganglioside(GM1) into neural cells. Methods Rat BMSCs were isolated on the basis of its ability to adhere to the culture plate, passaged three times, and finally added GM1 to induce its differentiation. The morphologic changes of BMSCs were observed under phase-contrast microscopy, then neuron specific enolase(NSE) and glial fibrillary acidic protein(GFAP) expression were determined by immunocytochemistry way.Never growth factor(NGF) mRNA and brain-derived neurotrophic factor(BDNF) mRNA expression were detected with RT-PCR and then compared with those of FBS control group and blank control group. Results In GM1 induction group,BMSCs appeared round-shaped,with some dendrite and axon interlaced.(29.47%?3.26)% BMSCs showed positive staining to NSE,while(2.32?0.18)% were positive to GFAP.(6.97?0.56)% and(10.6?0.75)% BMSCs were positive to NSE in FBS control group and blank control group respectively,while(1.41?0.35)% and(1.21?0.35)% BMSCs were positive to GFAP in FBS group and blank control group respectively.NGF mRNA and BDNF mRNA levels in GM1 induction group were significant higher than those in the FBS group(all P
6.Progress in the mechanism of phospholipase CE1 in children with primary nephrotic syndrome
Chinese Journal of Applied Clinical Pediatrics 2021;36(5):387-390
Phospholipase CE1 (PLCE1) is a special phospholipase C isoenzyme in the phospholipase family. It is expressed in podocytes of mature glomeruli and participates in cell signal transduction through a variety of pathways, thereby promoting its growth and expression. It has been reported that the single gene mutation of PLCE1, the heterozygosis mutation with other pathogenic genes and the interaction between PLCE1 and other factors are likely to cause primary nephrotic syndrome (PNS) in children. There have been children with PLCE1 mutation-induced steroid-resistant nephrotic syndrome cured after taking calmodulin inhibitors. They bring hope to treat PNS.In this article, the expression of PLCE1, its biological function, the mechanism of PLCE1 leading to PNS in children and its treatment were reviewed.

Objective To comparatively analyze the cognition and feelings of professional self-con-cept of nurse (PSCN) between nursing students and nursing staff and and to propose recommendations for the education of nursing students. Methods Professional self-concept of nurses instrument (PSCNI) scale was used to investigate the self evaluation of nursing students and nursing staff in the aspects of nursing skills, leadership, flexibility, satisfaction degree and communication skills. SPSS 19.0 was used to analyze the total scores and scores of each aspects; F test and t test were used to analyze the difference between two groups. Results Total score for nursing students and nursing staff were (81.27±7.40) points (67.7%) and (85.00±7.96) points (70.0%), with significant differences between the two groups (P=0.003). Scores in as-pects of nurturing skills, communication, flexibility were higher in nursing staff than in nursing students, with significant differences between the two groups (P<0.05). Conclusion The cognition and feeling of nursing staff is better than nursing students in aspects of nurturing skills, communication, flexibility. Nursing teaching can correspondingly improved based on the differences between the two groups.
8.Neutrophil extracellular traps-induced endothelial cell damage in the pathogenesis of dermatomyositis-associated interstitial lung disease
Chinese Journal of Internal Medicine 2017;56(9):650-654
Objective To explore the role of neutrophil extracellular traps (NETs)-induced endothelial cell damage in the pathogenesis of dermatomyositis (DM)-associated interstitial lung disease (ILD).Methods Serum free DNA and krebs von den lungen-6 (KL-6) level were tested in healthy controls, dermatomyositis patients with or without interstitial lung disease (DM-ILD and DM-NILD).Subjects' peripheral blood neutrophils were stimulated with phorbol 12-myristate 13-acetate (PMA),then human umbilical vein endothelial cells (HUVECs) were co-cultured with NETs.The cell morphology was observed by the inverted phase contrast microscope.Cell viability was detected by cell counting kit-8 (CCK8).Results The concentration of serum free DNA in DM patients [(271.27 ± 76.53) μg/L] was significantly higher than that in health control (HC) [(152.89 ± 37.34) μg/L,P < 0.001].Moreover,free DNA level in DM-ILD patients [(302.67 ± 74.15) μg/L] was higher than that in DM-NILD patients [(235.59 ±63.55) μg/L,P <0.005].The concentration of KL-6in DM patients [(3.08 ±2.07) μg/L] was higher than that in HC [(0.87 ± 0.51) μg/L,P < 0.001].Similarly,KL-6 in DM-ILD patients [(4.00 ± 2.44) μg/L] was higher than that of DM-NILD patients [(2.03 ± 0.61) μg/L,P < 0.005].Free DNA and KL-6 were positively correlated (r =0.251,P < 0.05).The survival of endothelial cells in DM group (53 ± 11) % was lower than that of HC group [(70 ± 5) %,P < 0.001].Not surprisingly, the survival of endothelial cells in DM-ILD group (44 ±4) % was lower than that in DM-NILD group [(61 ± 8) %,P < 0.01].Conclusion NETs could play an important role in the pathogenesis of dermatomyositis associated interstitial lung disease, suggesting that NETs may be the potential therapeutic target.

China Journal of Endoscopy 2016;22(9):47-51
Objective To investigate the clinical manifestations, endoscopic manifestations, histological characteristics, diagnosis and treatment of the primary gastrointestinal melanoma in China. Methods The gender, age, position, endoscopic manifestations, histological characteristics, treatment, prognosis of the primary gastrointestinal melanoma were gathered and analyzed by reviewing retrospectively the data of 2 cases seen at our hospital and 449 cases searched in CNKI from 1983 to 2015. Results The primary gastrointestinal melanoma was mostly seen in the rectum and anus, and secondly in the esophagus. A typical endoscopic finding was a protuberant or nodular tumor, with erosion or dirty surface on it. Most lesions were friable and easily hemorrhagic. The maximum diameters of most lesions, accounted for 83.3 %, were larger than 2.0 cm. The average value of the maximum diameters was (4.79 ± 2.67) cm. Most cases presented brown or black, followed by hoar. However, some lesions presented various colors, such as blue, red, even co-exited color or achromatic color. Preoperative misdiagnosed rate was 73.64 % and 1-year mortality was 54.19 %. Negative correlation was observed between survival time and lesion size, infiltration depth. Positive correlation was observed between lesion size and metastasis infiltration depth. Survival time had nothing to do with metastasis and age. Conclusion The primary gastrointestinal melanoma was highly aggressive, and usually found in the terminal age, usually together with extensive metastasis, misdiagnosed and poor prognosis. Knowing the clinical features, endoscopic manifestations, histological and immunohistochemical characteristics is the key to early diagnosis.
