An internal amplification control, which could be co-amplified with the invA target gene of Salmonella in the PCR system, was constructed in order to indicate possible PCR inhibitors derived from food samples. Specificity of this PCR system was tested with 9 Salmonella strains and 15 non-Salmonella strains, and the results showed that there was a 374 bp amplicon resulted from all Salmonella strains, while only a 513 bp IAC amplicon appeared after the amplification for all non-Salmonella strains. The detection sensitivity of this PCR system was 12.8 fg/?L for purified target DNA, and the detection limit for artificially inoculated milks was 8 cfu /25g if they were enriched for 8h in buffered peptone water. Salmonella in 80 samples of seriously contaminated milks was detected by the PCR method developed in this study, and the experiments demonstrated that it could successfully eliminate false-negative results.

Objective: To study the extraction process of the total flavones of Pollen Typhae ( Typha angustifolia L.). Methods: The optimum extraction process was selected with the orthogonal design. The content of total flavones was determined by UV spectrophotometry. Results: The significant effects of alcohol concentration, extraction time and extraction times on the extraction of total flavones were discovered. Conclusion: The optimum extraction process was as follows: Pollen Typhae was extracted with 70% alcohol for 3 times, with the solvent volume 8, 6, 6 times amount and extraction time 60, 60, 30 minutes in turn.

Orthogonal design was used to study the extraction technology of Qingyan Xiehuo Granule. The solvent volumn, extracting time, extracting times and smashing degree (only for Fructus Canarii) were treated as the factors and examined at various levels. The extraction technology for Folium Polygoni tinctorii and Radix Scutellariae was determined. The methonal extract absorption (at 310nm) of the dried water extract of Folium Polygoni tinctorii and the content of Baicalin were used as the criteria.

Objective To evaluate the effectiveness of cryosurgery by using fiber optic bronchoscope for the treatment of middle or late stage central lung cancer.Methods Cryosurgery was performed on 31 patients with middle or late stage central lung cancer,who could not received open surgery,with liquid CO2 by using fiber optic bronchoscope.The effectiveness was monitored.Via the bronchoscope,a cryo-probe was inserted to the center or margin of the tumor.The cryotherapy was persisted for 30 to 120 seconds at-50 to-70 ℃.And then,the tumor was removed before the ice-ball on the point of the probe thawed.The procedure was repeated for several times till the airway was reopened.Results After 1 to 6 times therapies(2.5 times on average),the improve rates of cough,hemoptysis,dyspnea,and chest pain were 74%(23/31),87%(27/31),87%(27/31),and 58%(18/31),respectively.The rates of "markedly effective"and "effective" were 61%(19/31)and 39%(12/31)respectively.The pulmonary function of the patients was also improved:the FEV1 rose from(1.21?0.22)L to(1.72?0.35)L(t=21.843,P=0.001),and the FVC was increased from(1.86?0.31)L to(2.26?0.43)L(t=33.703,P=0.001).Conclusions Cryosurgery by using a fiber optic bronchoscope is an effective and minimally invasive method to reopen the airway,control the obstructive pneumonia,and improve dyspnea and hemoptysis.

AIM: To investigate the constituents of the effective fractions from Shenshao Xinxin Prescription(Radix et rhizoma ginseng;Radix paeoniae rubra;Radix puerariae lobatae). METHODS: The compounds were isolated by column chromatography on silica gel,polyamide,sephadex gel and their structures were determined on the basis of their spectroscopic evidence including UV,FAB-MS and NMR data. RESULTS: Eight compounds had been isolated and identified as daidzein,4′-O-methyl-daidzin,genistin,PG-3,puerarin,paeoniflorin,daidzein 8-C-Apiosyl(1→6) glucoside,4′,7-diglucoside. CONCLUSION: The compounds obtained were the alcohol-soluble compounds from Shenshao Xinxin Prescription.

Objective To investigate the value of Helicobacter pylori (HP),blood carcinoembryonic antigen (CEA),carbohydrate antigens 72-4 (CA72-4) and CA24-2 in the screening of gastric cancer.Methods The contents of HP,blood CEA,CA72-4 and CA24-2 in 50 healthy controls,45 patients with benign gastric lesions mnd 70 patients with gastric cancer were detected.According to the number of positive cases,the positive rate of single detection and the positive rate of combined detection were calculated.Sensitivity,specificity,and accuracy of serum tumor markers were calculated according to HP single and combined detection.Results The positive rate of each individual detection and 4 joint detection in gastric cancer group was significantly different from that in the benign lesion group and the normal control group (P ＜0.01).The positive rate of 4 joint detection in gastric cancer group was 89.25％,and the difference was statistically significant (P ＜0.01).There was no significant difference in the positive rate of the single test and the 4 joint detection in benign stomach group and normal control group (P ＞ 0.05).The sensitivity,specificity and accuracy of HP,blood CEA,CA72-4,CA24-2 joint detection were significantly higher than single detection (P ＜ 0.05).Conclusions Joint detection was significandy better than single detection,and can significantly improve the detection rate of gastric cancer.

Objective To investigate the clinical and pathological features of colonic polyp canceration and related risk factors of colon polyps,and explore clinicopathologic features and major risk factors of the canceration of polyps of colon polyps.Methods Totally 80 patients with colon polyps including 6 cases of cancerations and 20 patients with colon cancers were selected from Xuzhou Tumor Hospital Admissions.Those patients were divided into 74 cases of non cancer group and 26 cases of cancer group.The clinical data,the detection index and the statistical analysis were performed.Results The clinical features of patients included 51 cases of multiple polyps,30 cases of fatty liver,13 cases of fecal occult blood positive,and 30 patients with colonoscopy.There were 218 polyps,191 data intact,and 6 cases showed cancer (3.14％).Inflammatory polyps mainly occurred in rectum,cecum,and significant risk other than the colon.Tubular adenoma occurred mainly in the transverse colon.Sigmoid colon cancer colon polyps of the highest risk showed OR =2.30 (P ＜ 0.05).The cancer risk from high to low was from tubular villous adenoma to villous adenoma hair (P ＜0.01),and no difference in tubular adenoma cancer risk (P ＞0.05).There was no difference between the indexes of the canceration group and the non cancerous group (P ＞ 0.05).The single factor analysis showed pathological type,and the lesion were the main risk factors of malignant colonic polyps (P ＜ 0.05).Multi-factor regression analysis showed WBC,pathological type,and the lesion were the main risk factors of malignant colonic polyps (P ＜ 0.05).Conclusions The colonic polyps in male have higher incidence with fatty liver,hypertension and other pathological features,and are mainly tubular adenomas,inflammatory and hyperplastic polyps,with different lesions,pathological types occur and different cancer risk.

[Objective]To explore the effects of extracorporeal shock waves(ESW) therapy on tendon-bone healing after reconstruction of the anterior cruciate ligament(ACL).[Method]Animal model of reconstruction of the anterior cruciate ligament in 18 healthy F/M New Zealand white rabbits(2～3 months old) were established using digital long extensor tendon and randomly classified into 2 groups.The ESW group was given extracorporeal shock waves therapy on 24 hours after reconstruction of the anterior cruciate ligament while the control group with only reconstruction of the anterior cruciate ligament but no more therapy.All animals were sacrificed at 2、4、8 weeks after operation for histological examinations.The specimens were stained with hematoxylin-eosin and Masson trichrome to investigate pathology change of the tendon-bone healing tissue.The ink intravascular injection was used to observe the neo-vascularization of tendon-bone healing tissue.[Result]The histological examination revealed that at 4 weeks after operation,the ESW group,tendon-bone interface was filled with proliferated fibroblasts and chondroblasts,the collagen fibers were obviously increased and regularly arranged.At 8 weeks after operation,the healing tissue at the bone-tendon interface had developed into dense connective tissue,the collagen fibers were formed in abundance and the perpendicular collagen fibers were regularly and longitudinally arranged.The transition zones of collagen fibers,fibrocartilage and bone(the structure alike a direct ligamentous insertion) occurred in the local region of tendon-bone interface.The histological observations showed that the tissue healing at tendon-bone interface of the ESW group were quicker than those of the control group.The proportion of neo-vessels at the tendon-bone interface of the ESW group were significantly higher than those of the control group at 4 and 8 weeks after operation(respectively P=0.028,P=0.008).[Conclusion]Extracorporeal shock wave treatment can significantly improve the tendon-bone early healing after reconstruction of the anterior cruciate ligament in rabbits.

The study aimed at investigation the effect of the transgenic biocontrol bacteria 308R(pCPP430) on the tomato rhizosphere bacterial communities. Strain 308R (Pantoea agglomerans), an adnascent bacterium isolated from tomato, was a biocontrol bacterium. The recombinant cosmid pCPP430 contained the hrp gene cluster of Erwinia amylovora was transformed into 308R Strain. Transgenic biocontrol bacterium 308R(pCPP430) produced Harpin protein which induce hypersensitive response and disease resistance in plants and, surprisingly, increase plant growth. Two complementary methods, sole-carbon-source utilization tests (SCSU) and ERIC-PCR, were used to compare the characterization of bacterial communities from three groups of tomato roots dipped in sterilized water, 308R suspended liquid and 308R(pCPP430) suspended liquid. The cluster analysis and principal components analysis of the number of colonies from SCSU indicated consistent differences in the rhizosphere of each group. Cluster analysis of SCSU data showed that there were consistent differences in the rhizosphere of each group. The rhizosphere bacteria communities of root dipped with 308R(pCPP 430) and 308R have a relative good replication, but the differences of corresponding replicates in dipped with distilled water was increased by degrees. Cluster analysis of ERIC-PCR fingerprints revealed that most of results (eight in ten) have overlaps between roots dipped in water and those dipped in 308R liquid. The methods used in this study may prove a useful approach for the comparison of bacterial communities.

Objective To investigate the effect of cyclooxygenase-2(cox-2)on prostaglandin I2(PGI2)/thromboxane A2(TXA2)system in ischemia reperfusion(I/R)injury to small bowel in rat.Methods Forty-eight male SD rats were randomly divided into three groups:control group(n=8),I/R group(n=20)and intervention group(n=20).Small intestine I/R model was established with the animals of I/R group:after anesthesia,rats were split along the middle abdominal incision,the mesenteric artery was then bluntly separated and clipped with a blood vessel forceps at its root for 45 minutes to block the blood flow,then releasing the forceps for reperfusion.Animals of intervention group were lavaged with Nimeshulide 1h before operation,the remaining steps were the same as did in I/R group.The samples of blood and intestinal tissue were collected at 3,6,12 and 24 hours after operation.The content of plasma D-lactate was detected by ultraviolet spectrophotometric method.Radioimmunoassay was used to detect 6-keto-PGF1?(the stable metabolite of PGI2,reflected the level of PGI2)and thromboxane B2(TXB2,stable metabolite of TXA2,reflected the level of TXA2)in serum and tissues of small bowel.The expression of cox-2 mRNA was detected by RT-PCR.Results Compared with the control group,the levels of plasma D-lactate,cox-2,6-keto-PGF1? and TXB2 were significantly increased in I/R group,with more obvious increase of TXB2,and the value of 6-keto-PGF1?/TXB2 decreased significantly(P
