1.Protective effect of antihypertensive drug NO.1 on brain cortical tissue in spontaneously hypertensive rat

Objective To investigate the protective effect of antihypertensive drug NO.1 on brain cortical tissue in spontaneously hypertensive rats.Methods A total of 60 spontaneously hypertensive rats (SHR,30 males and 30 females) and 12 healthy Wistar rats (6 males and 6 females) weighed (200 20) g,were randomly divided into 6 groups.Model group (SHR,n=12) and control group (healthy Wistar rats,n=12) were given the same dose of placebo.Captopril group (SHR,n=12) were given captopril 5 mg · kg-1 · d-1.The low-,median-and high-dose groups of antihypertensive drug NO.1 were given 0.25,0.5 and 1 g · kg 1 · d-1 respectively.After 4 weeks of treatment,carotid artery blood pressure was detected.All rats were sacrificed,and brain tissue samples were taken.The expressions of Bcl-2,Bax,necrosis factor NF-κB P65,chloride channel 2 and 3 (CLC-2 and CLC-3) were determined by RT-PCR and Western blotting.Results After 4 weeks of antihypertensive NO.1 treatment,SHR carotid artery blood pressure was (182.8 ± 7.3)mmHg in low dose group,(170.3±9.4) mmHg in medium dose group,and (163.9±10.6) mmHg in high dose group; and (205.4 ± 11.3)mmHg in the model group.Antihypertensive drug NO.1 significantly reduced blood pressure in spontaneously hypertensive rats,and had a concentrationresponse relationship (P<0.05).Antihypertensive drug NO.1 decreased the expressions of Bax,CLC-2 and CLC-3,and increased the expressions of Bcl-2 and NF-κB P65.Conclusions Antihypertensive drug NO.1 plays a protective role in hypertension-induced cell injury by changing the metabolic enhancement in hypertension-induced cell volume decrease.
2.The Effects of Thalidomide on Angiogenesis, Growth and Metastasis of Human Hepatocellular Carcinoma in Nude Mice

Objective To obsrve the effects of thalidomide on angiogenesis, growth and metastasis of human hepatocellular carcinoma (HCC) in nude mice. Methods The model bearing HCC was set up in 24 nude mice, and then the rats were randomly divided into therapy group and control group, each containing 12 animal. Thalidomide (200mg/kg/d) dissolved in 0.5% CMC suspension was administered intraperitoneally in the therapy group, and equivalent 0.5% CMC was administered in the control group. Objective To study association of hepatitis B virus(HBV) precore (pre c)/basic core promoter(BCP) mutations with the genotype or the progression of liver disease. Methods The serum samples from 148 patients with HBV-relative diseases were collected, including 31 asymptomatic carriers, 32 with chronic hepatitis B (CHB), 40 with liver cirrhosis(LC) and 45 with hepatocellular carcinoma(HCC). The genes covering HBV pre c and BCP were amplified by nested polymerase chain reaction (nPCR). The PCR products were subjected to direct sequencing and the mutations in pre c 1896 and BCP 1762/1764 were determined by sequence analysis. HBV genotypes were also detected in the sera by restriction fragment length polymorphism based on S-gene PCR products. Results Of 148 serum samples of HBV, 128 were successfully genotyped and sequenced. There were 60 genotype B and 68 genotype C. 5.Treatment of humeral proximal fracture with locking proximal humeral plate

Objective To summarize the clincal effect of open reduction and internal fixation with locking-proximal-humerus-plate(LPHP)to treat proximal humeral fractures.Methods A total of 19 eases with proximal humeral fractures were treated with LPHP from August 2003 to February 2006 in this study.Their mean age was 58.6 years old.According to Neer classification,two cases were with two- part fractures,nine with three-part fractures and eight with four-part fractures including seven cases with osteoporosis.Redaction and fixation was done via deltopeetoral-groove approach.Bone graft was applied for five eases.Results The follow-up period was 3 to 32 months(mean 17 months).All cases were healed and the union period of 3-5 months.According to the Near shoulder score,the results showed that ten cases were excellent,6 cases good,with excellent rate of 84%.Conclusion The LPHP method is suitable for osteoporosis and comminuted proximal humeral fractures and enhances the treatment effect.

8.Sanqi sodium hyaluronate gel inhibits alpha-smooth muscle actin expression in the epidural scar

BACKGROUND:Epidural scar after laminectomy is one important reason for the secondary spinal stenosis, and local application ofSanqi sodium hyaluronate gel can prevent epidural scar adhesion after laminectomy. OBJECTIVE: To study the effects ofSanqisodium hyaluronate gel on α-smooth muscle actin expression in the process of rabbit's epidural scar formation. METHODS: In this study, there were ninety-six rabbits which were randomized into four groups and given 0.5 mL normal saline, 0.5 mLSanqi concentrated solution, 0.5 mL sodium hyaluronate and 0.5 mLSanqisodium hyaluronate gel around the dura. At 1, 2, 4, 8 weeks after treatment, immunohistochemistry staining was employed for analysis of α-smooth muscle actin expression. RESULTS AND CONCLUSION:At the end of weeks 1 and 2, the expression of α-smooth muscle actin antibody in the normal saline group was significantly higher than that in the other three groups (P < 0.01 orP < 0.05), but there were no significant differences among the Sanqi, sodium hyaluronate andSanqisodium hyaluronate gel groups (P> 0.05). At weeks 4 and 8, the expression of α-smooth muscle actin antibody in theSanqi sodium hyaluronate gel group was significantly lower than that in the other three groups (P < 0.01 orP < 0.05), and there was no significant difference among the latter three groups (P > 0.05). These findings suggest thatSanqi sodium hyaluronate gel can inhibit the expression of α-smooth muscle actin, and thus ease scar contracture.
9.Application of problem-based learning in clinical anesthesiology demonstration for clinical un-dergraduates

Objective To explore the effect of applying problem-based learning(PBL)in clin-ical anesthesiology demonstration for clinical undergraduates. Methods Eighty-nine 2009 clinical undergraduates were divided into two groups:experimental group and control group. Students in ex-perimental group received PBL while those in control group received traditional teaching. The effect was evaluated according to the results of case examination and investigation table. The results were analyzed by X2 test. Results Most students (42/45) answered questions correctly in experimental group while only 30 of 44 students(30/44)did in control group(P<0.05). Concerning the results of investigation,PBL had more advantages than traditional teaching method in the following 5 aspects:self-studying ability(86.7%,39/45),clinical thinking ability(73.3%,33/45),information seeking ability (75.6%,34/45),expressing ability (82.2%,37/45),communicating between students (75.6%, 34/45)(P<0.05). Conclusions PBL has evident advantages in anesthesiology clinical demonstration. It can inspire students,cultivate abilities,enhance critical thinking and finally improve teaching effect.
10.Investigation of androgen receptor gene expression in human peripheral leukocytes

Objective: To investigate androgen receptor (AR) mRNA expression in human peripheral leukocytes. Methods: Total RNA was isolated from peripheral leukocytes of healthy men and women, AR mRNA was determined by RT PCR and Northern blot. Results: An AR cDNA fragment of the expected size of 390 bp was determined by RT PCR. 9.4 kb AR mRNA was detected by Northern blot, which was consistent with AR mRNA in human prostate reported by Lubahn et al . Conclusion:AR mRNA expresses in human peripheral leukocytes, this provides basis for studying the effect of androgen on the function of the human peripheral leukocytes and the changes of AR in pathological stages.
