2.Effects of small interfering RNA specific to stably transfected HMGA1 gene on biological characters of ovarian carcinoma cells
Chinese Journal of Obstetrics and Gynecology 2010;45(1):51-55
Objective To investigate the effects of biological characters of the human ovarian cell line (OVCAR) by stable transfection short hairpin RNA into the target HMGA1 gene. Methods Experiments were divided into two groups: transfected the OVCAR cells with pSilence4. 1-CMV-Hs plasmid as group A, while transfected OVCAR cells with pSilence4. 1-CMV-Hn plasmid as group B, in which stably transfected cells were gained by antibiotic screening. The comparative expressions of HMGA1 were detected by RT-PCR and western blot. Methyl thiazolyl tetrazolium (MTT) method was applied to measure cell proliferation and at the same time, the cell growth curve was also be mapped. Vitro invasion assay was used to observe the invasion ability of the cancer cells, and the tumor growth of the nude mice inoculated of tumor cells were compared with before and after transfection. Results In group A, the expression level of mRNA and protein HMGA1 gene in OVCAR cells were remarkably reduced before and after the stable transfected with HMGA1 siRNA, in which the percents of mRNA expression were [(86.3±2.7) % vs. (35.8± 3.1) %, P<0.05], the expression of protein were [(68.6±2.8) % vs. (22.3±4.2) %, P<0.05)]. The OVCAR cell growth in stable transfection status was more significantly decreased than that in non-transfection status (P<0.05). In group B, there were no statistical difference in the expression of HMGA1 siRNA, protein and the cell growth between before and after transfection states (P>0.05). The invasion cell numbers were reduced from before to after transfection state in group A [(53±6)vs. (21±6), P< 0.05] ,while there was no significant difference in group B [(51±6)vs. (47±8), P>0.05]. After inoculated transfected cells into nude mice, it took (6.0±0.9) days to grow the planed tumors in group A, which was much shorter than that (12.3±3.9) days in group B (P<0.05 ). After 5 weeks, the tumor weight and volume in group A was were significantly lower than those in group B [(0.8±0.3)g vs. (2.1± 0.4)g, (205±34)mm~3 vs. (987±82) mm~3, all P <0.05]. Conclusion HMGA1 siRNA could remarkably reduce the expression of HMGA1 gene in ovarian cell and also inhabit the ovarian cell growth.
3.Expression and clinical significance of matrix metalloproteinase-3 and osteopontin in cervical carcinoma
Cancer Research and Clinic 2010;22(z1):29-31
Objective To study the expressions of matrix metalloproteinase-3 (MMP-3) and osteopontin (OPN) in cervical carcinoma and their relationship with invasion and metastasis. Methods The expression of MMP-3 and OPN were detected in 54 cases of cervical cancer tissues by immunohistochemical SP method. Compared with 15 cases of normal cervical tissues, the relationship was analyzed between both protein expressions and clinical pathology. Results The expression positive rates of MMP-3 and OPN in cervical cancer tissues (70.37 % and 66.7 %, respectively) were significantly higher than those of normal cervical tissues (20 % and 0, respectively), were significantly higher with infiltration depth >1/2 and lymph node metastasis than those with depth≤1/2 and without metastasis , respectively(P <0.05). The expressions of MMP-3 and OPN were related with depth of cervical infiltration and lymph node metastasis of cervical cancer.Expressions between MMP-3 and OPN were significantly positively correlation(r =0.401, P<0.05). Conclusion The expressions of MMP-3 and OPN are correlated with invasion and metastasis of cervical carcinoma.

7.SERUM-URIN ?_2 MICROGLOBLIN CHANGE AND DYNAMIC OBSERVATION IN CHILDREN'S GLOMERULAR DISEASES
Journal of Xi'an Jiaotong University(Medical Sciences) 1982;0(01):-
This paper reports serum ?_2 microglobiln (?_2M in abbreviation) in 216 normal children of different ages. Urin ?_2M in 60 normal adults; and both in 50 cases of common glomerular disease in children were detected by RIA. The results showed that serum ?_2M is more accurate than BUN in judging the renal function. The elecating serum ?_2M is coincidence with the elevating of diastolic pressure of the sick children. The dynamic observation on delayed cases examination of serum ?_2M and urin ?_2M may be of help in monitoring the pathological changes in glomerular and tubule.

Acta Anatomica Sinica 1954;0(02):-
Rapid Golgi preparations of golden hamster dorsal lateral geniculate nucleus (LGd)show three classes of neurons with different morphological characteristics. Class Ⅰ and class Ⅲ cells are distributed throughout the LGd.Class Ⅱ cells are found more common medially at mid-genieulate levels.Class Ⅰ cells are multipolar neurons with large cell body. The smooth primary dendrites branch extensively. Spine-like and small excrescences are found along distal dendrites.Class Ⅱ cells have medium size pear-like cell body. The distal dendrites are usually derived from one primary dentrite.Dense cluster of dentritic protrusions is usually found at branch points of the dendrites.Class Ⅲ cells have the smallest cell body and are paucidendritie but the dendrites have complex protrusions including pearl-like dendritic appendages.Class Ⅰ and class Ⅱ cells possess an axon but most of class Ⅲ cells are not found to have an axon. The connections between the LGd and visual cortex was investigated using the retrograde horseradish peroxidase(HRP)histochemical method. After injection of HRP into the visual cortex,Class Ⅰ and class Ⅱ neurons were labelled but class Ⅲ celts were not suggesting that class Ⅰ and class Ⅱ cells are projecting neurons.Class Ⅲ cells do not seem to project to the visual cortex and are probably local circuit neurons.
10.Expression and Significance of Ets- 1 in Chondrosarcoma and Osteochondroma of the Jaws
Journal of Shanghai Jiaotong University(Medical Science) 2000;20(6):550-552
ObjectiveTo analyze the expression and significance of Ets- 1 (E26 transformation-specific) in chondrosareoma and osteochondroma of the jaws.MethodsEts- 1 expression was de-tected by immunohistochemistry in 20 cases of chondrosarcoma and 8 cases of osteochondroma of thejaws.Results12 of the 20 cases of chondrosarcoma (t50%) showed an Ets- 1 positive reastion. Thepositive rates of grade Ⅱ and grade Ⅲ were significantly higher than that of grade Ⅰ ( P < 0.05 ). Re-current cases showed a higher Ets - 1 positive rate than primary ones ( P < 0.05 ). Ets - 1 was presentin 12.5 % (1/8) osteochondroma. The positive rate was significantly different between chondrosarcomaand osteochondrorna ( P < 0.05 ).ConclusionThe results indicate that Ets- 1 is overexpressed inchondrosarcoma of the jaws and correlated with its pathological grade and recurrence.
