Chinese Journal of Behavioral Medicine and Brain Science 2008;17(10):924-925
Objective To understand the effect of environment variables on anxiety in the soldiers at high altitude. Methods A total of 267 soldiers at high altitude were investigated with the self-rating anxiety scale, the environmental scale and Eysenck personality questionnaire. Results Compared with norm(29.78±0.46),the soldiers at high altitude had higher sore of anxiety(37.22±6.64;t=17.92,P<0.01).One-way ANOVA and independent.samples t test showed that the anxiety(38.37±5.93,36.65±6.951,36.75±6.88)of military people at high altitude had not evidently difference in different altitude(≥3587.5m,3587.5~2767.9m,≤2767.9m).Parental culture.length of military service(F=3.25,P<0.05)and neuroticism(R=0.52,P<0.01)had evidently effect on the depression of soldiers. Conclusion The occurrence of anxiety in the soldiers at high altitude has a higher level.Parental culture.1ength of military service and neuroticism had predictive value for depression.
3.Study of hand hygiene requirements in the operation skill of physician qualification examination
Chinese Journal of Medical Education Research 2014;(9):967-969
Objective To explore the difference of hand hygiene requirement between guide-line of skill practice for national examinations of doctors' qualification(clinical practitioners) and stan-dard for hand hygiene for healthcare workers in health care settings for some sterile techniques. Methods Eight sterile techniques in guideline of skill practice for national examinations of doctors' qualification(clinical practitioners) such as changing a dress and taking out stitches, aspiration, ure-thral catheterization, arteriovenous puncture, thoracentesis, abdominal puncture, lumbar puncture and bone marrow puncture were chosen and the results were compared with standard for hand hygiene for healthcare workers in health care settings. Results Three of eight sterile technics mentioned hand-washing, six of eight mentioned wearing sterile gloves, and zero of eight mentioned antiseptic han-drubbing; however, only one of six (16.67%) before wearing sterile gloves mentioned handwashing. Conclusion The requests of hand hygiene between guideline of skill practice for national examina-tions of doctors' qualification (clinical practitioners) and standard for hand hygiene for healthcare workers in healthcare settings are not concordant , which from the root leads to the inconsistence of doctors' learning at different stages of education and training . It is suggested that the examination organization departments should refer to the relevant provisions of the state in the preparation of guidelines, and regulate medical practice behavior with consistent standards.
4.Expression of TAT-Smad7-HA fusion protein and validation of its transduction activity
Journal of Third Military Medical University 2003;0(23):-
Objective To construct, express, purify, identify and label the TAT-Smad7-HA fusion protein (protein transduction domain of trans-activator,human Smad7 and hyaluronic acid) and to validate its transduction activity in the cultured human primary keloid fibroblast cells. Methods TAT-PTD,Smad7 and HA fragments were sequentially inserted into pET32a(+) to construct the pTAT-Smad7-HA prokaryotic expression vector. After the expression of target fusion protein was induced to express by IPTG,affinity purification,Western blot analysis, enterokinase cleavage, target protein capture and FITC labeling were subsequently performed by turns to obtain the FITC-TAT-Smad7-HA fusion protein and to further observe its transduction activity in the human primary KFB cells in vitro. Results The prokaryotic expression vector for the TAT-Smad7-HA fusion protein, named as pTAT-Smad7-HA was successfully constructed, and the target fusion protein was efficiently induced to express, covering more than 25% of the total bacterial proteins, successfully purified with a purity of more than 95% purity, desalted by desalting column, identified by Western blotting, thioredoxin removed and FITC labeled. Finally,a fusion protein of FITC-TAT-Smad7-HA with the approximately molecular weight of 50?103 was successfully purified and its high transduction activity in KFB cells was validated. Conclusion The highly-purified FITC-TAT-Smad7-HA fusion protein and the validation of its high transduction activity in KFB cells have provided an experimental foundation for further studies on the role the human Smad7 protein playing in the TGF-?/Smads signal transduction pathway and further elucidation of the pathogenesis of keloid formation.

Acta Pharmaceutica Sinica 2016;51(1):9-17
Parkinson's disease (PD) is a common neurodegenerative disorder associated with aging. Great progresses have been made toward understanding the pathogenesis over the past decades. It seems that both genetic factors and environmental factors contribute to PD, while the precise pathogenesis still remains unknown. Recently, increasing evidence has suggested that autophagy dysregulation is closely related to PD. Dysregulation of the autophagic pathways has been observed in the brains of PD patients or in animal models of PD, and a number of PD-associated proteins, such as a-synuclein, Parkin and PINK1, were found to involve in autophagy, suggesting a link between autophagy and pathogenesis of PD. In this review, we summarized the role of PD-associated proteins in autophagy pathways. In addition, we described the efficacy of autophagy-modulating compounds in PD models and discussed promising strategies for PD therapy.

Chinese Journal of Infectious Diseases 2012;30(7):402-406
Objective To establish a model of isoniazid induced necrosis and apoptosis in HepG2 cell and to observe the expressions of Fas/Fas ligand (FasL) in this model. Methods HepG2 cells were treated with different dosages of isoniazid (0,1,2,4,6 and 8 mg/mL) or blank control for 24 hours.Flow cytometer was used to observe the cellular morphology of the HepG2 cell.Annexin V/propidium iodide double staining and flow cytometry were employed to detect the necrosis and apoptosis of HepG2 cells. The expressions of Fas/FasL on the cells were also determined by flow cytometry. The data were analyzed by one-way ANOVA.The comparisons between the drug groups and the control group were performed by using Dunnett t test. Results The higher the dose of isoniazid (4,6,8 mg/mL) was,the more necrosis and apoptosis were observed. In the 4,6 and 8mg/mL isoniazid arms, the total mortality rates were all higher than the control group [(32.1 ±7.5)%,(34.9±8.1)%,(38.2±9.4)% vs (7.2±1.5)% respectively](t=4.62,5.14 and 5.75,respectively; all P<0.01 ).The expression levels of Fas increased along with the dose of isoniazid increasing [(8.7±2.2)%,(11.5±2.8)%,(12.3±3.0)% and (10.6±2.9)% in isoniazid 2,4,6and 8 mg/mL arms, respectively],which were all higher than that in control arm [(3.1 ±0.8) %](t=2.97,P<0.05; t=4.46,P<0.01; t=4.88,P<0.01; t=3.98,P<0.05).Furthermore,the expressions of FasL increased as well when the dose of isoniazid increased. The expression levels of FasL were (16.2±3.5)%,(21.7±4.8)% and (18.7±4.9)%,respectively in isoniazid 4,6 and 8 mg/mL arms, which were all higher than that in the control group [(7.4±1.4)%](t=3.11,P<0.01; t=5.06,P<0.01; t=3.99,P<0.05).HepG2 cell necrosis increased with isoniazid of 8 mg/mL.However,the increase of apoptosis was not observed. Conclusion Isoniazid can induce HepG2 cell necrosis and apoptosis, and the apoptosis may be related with the increased expressions of the Fas/FasL on the cells.
