2.The drug resistance mutation detection and relevant factors analysis of HBV P region in chronic hepatitis B patients in Weifang City, Shandong Province.

In order to investigate the mutation of HBV polymerase gene reverse transcription conserved region (P region) in chronic hepatitis B (CHB) patients, 212 CHB patients who took antiretroviral treatment with nucleotide analogues were chosen. The drug resistance mutations of HBV P region and HBV genotype were detected by Pyrosequencing. Sequence analysis showed that the drug resistance sites of HBV P region located at sites 173; 180; 181; 184; 204; 236 and 250. The main site of HBV P region drug resistance was 204 and 180, accounting for 35.8% and 23.5%, respectively. There were significant differences in the mutation rate of site 180 among different age groups. There were also significant differences in the mutation rate of site 204 among younger than 30 age group, 41 to 50 age group and 51 to 60 age group. (P < 0.05, P < 0.01). The mutation rate of site 180 combined with site 204 was 66.6%. The mutation rate of site 181 combined with site 236 was 23.3%. The age of C genotype infected patients was significantly older than B genotype infected patients (P < 0.01). M204V/I mutation mostly existed in the form of joint L180M mutation, the mutation rate was age-related. The detection of HBV genotypes and drug resistance sites of HBV P region have important clinical implications for the treatment and prognosis of patients with CHB.

4.Bone cancer pain induce anxiety-like behavior and high expression of NR2B subunit in anterior cingulate cortex of rats

Objective To investigate the effect of bone cancer pain on emotion and NMDA re-ceptor NR2B subunit expression level in anterior cingulate cortex (ACC)in rats.Methods One hun-dred and fifty healthy male Wistar rats weighing 200-250 g aged 3 months old were randomly divided into 3 groups (n = 50 in each group):sham operation group (group S),bone cancer pain group (group BCP),RO25-6981 group (group RO).The BCP model was established by inoculating Walker 256 breast cancer cells into right intra-tibial.Rats in group S were given the same dose of d-hanks. Group RO was injected intraperitoneally with RO25-6981 (5 mg/kg/d)on the day of inoculation, while rats in the group S and group BCP were given the same dose of normal saline.Mechanical with-drawal threshold (MWT)and thermal withdrawal latency (TWL)of right hind legs were measured on day 7,10,14 after inoculation respectively.Elevated plus-maze test was carried out to investigate the effect of bone cancer pain on emotion in rats after pain threshold detection,then the percentage of the times entering the open arms (OE)and the percentage of the time staying in the open arms (OT) duration the total period were evaluated.Then the anterior cingulate cortex tissue was removed to e-valuate the NR2B protein and mRNA expression levels by RT-PCR,Western blot and immunofluo-rescence methods on day 14 after elevated plus-maze test.Results All the parameters did not differ with significant difference between group S and group RO.MWT decreased and TWL shortened on day 7,10,14 after inoculation in group BCP compared with those before inoculation and those of group S and group RO.OE and OT in group BCP reduced remarkably than those before inoculation and those of group S and group RO.Relative absorbance of NR2B mRNA,the expression of NR2B pro-tein,average NR2B relative fluorescence intensity value is obviously higher than that of group S and group RO (P <0.05).Conclusion Bone cancer pain can induce pain-related aversion and anxiety-like behavior of rats,and the mechanism may be related to the high expression of NR2B in anterior cingu-late cortex.
5.Influence of Metabolic Syndrome on the Left Ventricular Hypertrophy in Hypertensive Patients

6.Research of Access Control Mechanism in Hospital Information System

Objective To present a solution of the hospital information management access control for more secure information sharing. Methods Several commonly used access control models were compared. Some problems of the RBAC model were analyzed in the hospital information circumstances. Results A solution of space-based data access control was presented on the base of the role-based access control (RBAC) module. Conclusion The space-based data access control method can reduce space consumption and maintain data consistency, which is more suitable for the use of the hospital management information system.
7.miR-149 inhibition of cell growth and invasion by targeting FOXM1 in human prostate carcinoma

Objective:To investigate the effects of the miR-149 on the growth and invasion of prostate carcinoma cells. Meth-ods:Real-time fluorescence quantitative polymerase chain reaction was performed to detect the expression of miR-149 on prostate car-cinoma tissues and paraneoplastic tissues. The PC3 and DU145 cells were transfected with miR-149 mimics and negative controls. The cell growth and invasion abilities were tested in terms of colony formation and via Transwell invasion assay. The cells were transfected with the siRNA of the target gene FOXM1 and siRNA control. Western blot was used to detect the expression of FOXM1. The cell colo-ny formation and invasion ability were also detected. Results:Compared with the paraneoplastic tissues, miR-149 was down-regulated in the prostate carcinoma tissues (P<0.01), and the FOXM1 mRNA was highly expressed (P<0.01). PC3 and DU145 cells with miR-149 mimics had only a few colonies and invading cells (P<0.01). Moreover, PC3 (P<0.01) and DU145 (P<0.05) with miR-149 mimics had a low protein level of FOXM1. The FOXM1 expression was knocked down by the siRNA of FOXM1 in the PC3 and the DU145 cells (P<0.01). The knocking down of FOXM1 resulted in an inhibition of the cell colony formation and invasion abilities (P<0.01). Conclusion:The miR-149 inhibits prostate carcinoma cell growth and invasion by suppressing the FOXM1. Our data suggest that miR-149 may function as an effective tool for the molecular treatment of prostate cancer.
8.Clinical evaluation of coronary angiography by prospective ECG gated sequence scanning using second generation dual source CT:no heart rate control

Objective To analyze the effects of image quality and radiation dose of the patients with no heart rate control on different acquisition window settings.Methods A total of 75 patients underwent CTCA scan,with a median age of 55 years.All patients had no heart rate control.HR≤65 beats per minute acquisition phase setting was from 65%to 75%,65 beats per minute
80 beats per minute acquisition phase setting was from 35%to 45%.Evaluated patients radiation dose and subjective image quality of coronary artery.Results 75 patients with ED was (2.44±1.21)mSv,group A was (1.54±0.41)mSv, group B was (3.57±0.53)mSv,group C was (1.64±0.27)mSv,group A vs group B(P=0.000),group A vs group C(P=0.854),group B vs group C(P=0.000).A total of 1 043 coronary artery segments of 75 patients were evaluated,with an average of 13.9 segments showed by every case,(3.66±0.51)scale.Group A was (3.75±0.44)scale,group B was (3.60±0.54)scale,group C was (3.38±0.67) scale,group A vs group B(P=0.000),group A vs group C(P=0.004),group B vs group C(P=0.032).Conclusion The prospective ECG gated sequence scan could obtain the image quality and avoid unnecessary radiation dose by adapting to the different heart rate.
9.PHOSPHORUS INTERFERENCE AND EFFECT OF DIFFERENT INTERFERENCE SUPPRESSING AGENTS ON THE DETERMINATION OF CALCIUM IN FOOD BY ATOMIC ABSORPTION SPECTROMETRY

Interference of P with determination of Ca in food and the effect of interference suppressing agents of La, Sr and 8-hydroxyquinoline were studied. The results showed that 1) interference of P with Ca related to P/Ca ratio, the relative absorbance of Ca were getting increase along with, the decrease of P/Ca ratio if P/Ca ratio about 0.6; 2) La was more effective than the other interference suppressing agents. The relative absorbance of calcium reached to about 90% when 0.5% or 1% La was used in the P/Ca ratio of 30; 3) Sr had two actions. On one hand Sr had some suppressing action on P interference, and on the other hand Sr depressed the Ca absorption signal obviously; and 4) 8-hydroxyqui-noline had some but not very effective interference suppressing action. The content of Ca determined in food SRM were lower than certified value using 0.1% 8-hydroxyquinoline as interference suppressing agent.
10.Quality of life and its relative factors in patients with chronic obstructive pulmonary disease

Objective To explore the factors associated with quality of life(QOL)in patients with chronic obstructive pulmonary diseases(COPD).Methods Questionnaire was used to investigate 206 COPD patients,including demographic information,disease-related knowledge-attitude-practice and QOL scale.Results The total score of QOL in these patients was(64.70?14.78).There was statistically significant correlation between the quality of life and frequency of hospitalization in the past 2 years,marriage,education,course of disease by stepwise multiple regression analysis.Conclusion The quality of life in COPD patients is poor,especially in those who are spouseless,of longer disease course,of lower education degree,frequently hospitalized and insufficient for healthy practice.
