2.The drug resistance mutation detection and relevant factors analysis of HBV P region in chronic hepatitis B patients in Weifang City, Shandong Province.

In order to investigate the mutation of HBV polymerase gene reverse transcription conserved region (P region) in chronic hepatitis B (CHB) patients, 212 CHB patients who took antiretroviral treatment with nucleotide analogues were chosen. The drug resistance mutations of HBV P region and HBV genotype were detected by Pyrosequencing. Sequence analysis showed that the drug resistance sites of HBV P region located at sites 173; 180; 181; 184; 204; 236 and 250. The main site of HBV P region drug resistance was 204 and 180, accounting for 35.8% and 23.5%, respectively. There were significant differences in the mutation rate of site 180 among different age groups. There were also significant differences in the mutation rate of site 204 among younger than 30 age group, 41 to 50 age group and 51 to 60 age group. (P < 0.05, P < 0.01). The mutation rate of site 180 combined with site 204 was 66.6%. The mutation rate of site 181 combined with site 236 was 23.3%. The age of C genotype infected patients was significantly older than B genotype infected patients (P < 0.01). M204V/I mutation mostly existed in the form of joint L180M mutation, the mutation rate was age-related. The detection of HBV genotypes and drug resistance sites of HBV P region have important clinical implications for the treatment and prognosis of patients with CHB.

4.Effect of tetrandrine on murine hemangioendothelioma cell and its molecular mechanisms

Objective To explore tetrandrine on murine hemangioendothelioma cell and its mechanisms.Methods Explore the effect of tetrandrine on EOMA cells in time and concentration MTT experiment, the content of tetrandrine treated EOMA cells in different cell cycle detected by flow cytometry, speculated effect of tetrandrine on G1/S phase checkpoint for EOMA cells blocking.Western blot analyzed the molecular mechanism of tetrandrine induced EOMA cell block, and intracellular reactive oxygen species level were detected byflow cytometry.Then used the active oxygen scavenger NAC pretreated cells after drug incubation.Results Tetrandrine could inhibit the cell reproduction, and showed a concentration and time dependence, role in tetrandrine 20μM was significantly inhibited after 48 h.Tetrandrine caused mainly by EOMA cells during the cell cycle of G1/S phase cell block, so as to achieve the effect of inhibiting cell proliferation.Tetrandrine increased intracellular reactive oxygen species ( ROS) blockade effect of elevated levels of cell cycle to achieve, and the regulation of AKT, GSK-3 βand p53 cell cycle upstream protein level, finally maked the expression changes.Further research proved that, tetrandrine induced up-regulation of ROS level in cells to induce EOMA cell cycle arrest.Moreover, regulated by the ROS inhibitor NAC could significantly inhibit EOMA cell cycle factor.Conclusion Tetrandrine can inhibit tumor angiogenesis, which provides theory basis for clinical anti-cancer drugs in inhibiting tumor growth.

6.Diagnosis and treatment of primary malignant melanoma of the foreskin

Objective To discuss the biological and pathological features, diagnosis, treatment and prognosis of primary malignant mel-anoma of the foreskin. Methods The diagnosis, treatment, and follow-up of one case of primary malignant melanoma of foreskin treated in our hospital was reported, and the related literatures were reviewed. Results The tumour stage of the patient was stage I, and the patient was given partial amputation of the penis and bilateral inguinal lymph node dissection. Chemotherapy and immunotherapy were conducted at the same time. And there was no recurrence metastasis of during the postoperative follow-up of 54 months. Conclusion Primary malignant melanoma of the foreskin is rare in clinical and it is of high malignant degree. Its diagnosis depends on pathology and surgery is the main treatment. Postoperative chemotherapy and immunotherapy can prolong the survival time of patients, but the overall prognosis is poor.
7.Image reconstruction of positron emission tomography

8.Research progress in diagnosis of intensive care unit patients with invasive pulmonary aspergillosis

Invasive pulmonary aspergillosis is valued in intensive care unit (ICU) patients, but in ICU patients, the diagnosis standard of the European Organization for Research on the Treatment of Cancer/Mycosis Study Group (EORTC/MSG) could easily lead to misdiagnosis and missed diagnosis. It’s imaging manifestations are diverse: halo sign, nodules or micro nodules, nodular shadow, pulmonary consolidation, wedge-shaped consolidation, voids, air crescent sign, ground glass shadow, and so on, and with the relative characteristic features. The sensitivity and specificity of galactomannan detection in bronchoalveolar lavage fluid (BALF) are high for diagnosis of invasive pulmonary aspergillosis in ICU patients, but the significance of the cultivation of lower respiratory tract specimens is limited.
9.Research progress of functional magnetic resonance imaging in self-referential processing of depression

Objective To review the research progress of functional magnetic resonance imaging in Self -referential Processing (SRP) of patients with Depression based on the papers published from 2004 to 2014 at abroad and domestic,with an attempted to explore the neuromechanism of depression and to provide reference for treatment of depression.Methods Major online database including PubMed,EBSCO,Ovid,Medline,Clinicalkey,Wan Fang,CNKI,Cqvip,and CBM date bases were searched in Octobor 2014.The key words we used are depressionor depressive disorder,Self-referential Processing or rumination orautobiographical memory or Self-face recognition, Functional Magnetic Resonance Inaging(tMRI)and so on.Results 28 studies were adopted in this study,findings from the study showed:①rumination,autobiographical memory (AM) retrieval,and self-face recognition(SFR) are SRP paradigms,the cortical midline structures (CMS)and default mode network (DMN) were involved in the SRP of depression.②Both medication and cognitive behavioral therapy could reduce the abnormal activation of CMS and DMN in patients with depression.Conclusioins ①Both CMS and DMN may be the neuromechanism of SRP of depression.②Both CMS and DMN may be used as objective indicators to evaluate the treatment response in patients with depression.
10.Bone cancer pain induce anxiety-like behavior and high expression of NR2B subunit in anterior cingulate cortex of rats

Objective To investigate the effect of bone cancer pain on emotion and NMDA re-ceptor NR2B subunit expression level in anterior cingulate cortex (ACC)in rats.Methods One hun-dred and fifty healthy male Wistar rats weighing 200-250 g aged 3 months old were randomly divided into 3 groups (n = 50 in each group):sham operation group (group S),bone cancer pain group (group BCP),RO25-6981 group (group RO).The BCP model was established by inoculating Walker 256 breast cancer cells into right intra-tibial.Rats in group S were given the same dose of d-hanks. Group RO was injected intraperitoneally with RO25-6981 (5 mg/kg/d)on the day of inoculation, while rats in the group S and group BCP were given the same dose of normal saline.Mechanical with-drawal threshold (MWT)and thermal withdrawal latency (TWL)of right hind legs were measured on day 7,10,14 after inoculation respectively.Elevated plus-maze test was carried out to investigate the effect of bone cancer pain on emotion in rats after pain threshold detection,then the percentage of the times entering the open arms (OE)and the percentage of the time staying in the open arms (OT) duration the total period were evaluated.Then the anterior cingulate cortex tissue was removed to e-valuate the NR2B protein and mRNA expression levels by RT-PCR,Western blot and immunofluo-rescence methods on day 14 after elevated plus-maze test.Results All the parameters did not differ with significant difference between group S and group RO.MWT decreased and TWL shortened on day 7,10,14 after inoculation in group BCP compared with those before inoculation and those of group S and group RO.OE and OT in group BCP reduced remarkably than those before inoculation and those of group S and group RO.Relative absorbance of NR2B mRNA,the expression of NR2B pro-tein,average NR2B relative fluorescence intensity value is obviously higher than that of group S and group RO (P <0.05).Conclusion Bone cancer pain can induce pain-related aversion and anxiety-like behavior of rats,and the mechanism may be related to the high expression of NR2B in anterior cingu-late cortex.
