1.Observation on the effect of clostridium butyricum powder in treating infantile indigestion diarrhea

Objective To investigate the clinical effect of Clostridium butyricum powder in treating infantile indigestion diarrhea.MethodsFrom February 2013 to October 2015 in our hospital 76 cases of pediatric indigestion diarrhea as the research object, randomly divided into experimental group and control group with 38 cases in each group, two groups were given Bao,er ning granule, combined group plus live bacteria powder for the treatment of Clostridium butyricum, three days for a course of treatment, continuous treatment of two courses.ResultsBefore treatment, there was no significant difference in clinical symptom scores between the experimental group and the control group,after treatment,the scores of loss of appetite, abdominal pain, diarrhea, abdominal distension and stool of the experimental group were lower than those of the control group, the difference was statistically significant (P<0.05);Before treatment, there was no significant difference in serum motilin, gastrin and somatostatin levels between the experimental group and the control group.After treatment, the levels of serum motilin and gastrin in the experimental group were lower than those in the control group, and the level of serum somatostatin was higher than that of the control group, the difference was statistically significant (P<0.05);After treatment, the total effective rate of experimental group was 92.11% higher than that of control group 73.68%, the difference was statistically significant (P<0.05).ConclusionClostridium butyricum powder with Bao,er ning granule in the treatment of infantile dyspeptic diarrhea effect is good, worthy of clinical application.

3.Effect of TGF-? on the expression of type Ⅰ inositol 1,4,5-triphosphate receptor in WB rat liver epithelial cells

Objective:To investigate the effect of TGF-? on the expression of type Ⅰ inositol 1,4,5-triphosphate receptor in WB rat liver epithelial cells.Methods:The expression of type Ⅰ inositol 1,4,5-triphosphate receptor protein and mRNA were detected by Western blot and RT-PCR in WB rat liver epithelial cells in vitro stimulating with TGF-?.Results:The expression level of type Ⅰ inositol 1,4,5-triphosphate receptor protein and mRNA in WB rat liver epithelial cells were both increased after stimulating with TGF-? in several time points and reached the highest at 8 h stimulated, and then decreased.Conclusion:TGF-? enhance the IP_3R1 protein and mRNA expression in WB rat liver epithelial cells.

Objective To evaluate the relationship between Helicobac ter pylori (Hp) related gastritis and gastric mucosa content of interleukin - 8 (IL-8) and granuloeyte colony-stimulating factor ( G-CSF). Methods ELISA was used to detect the amount of IL-8 and G-CSF in supernate fluid of gastromucosal tissue culture from 60 patients, and to examine the difference between Hp infected and non-Hp infected cases. These 60 cases include normal gastric mucosa (n =20) , chronic gastritis mucosa (n = 14) , gastric ulcer mucosa (n = 13) , duodenal ulcer mucosa ( n = 13 ). 5.Determination of Ginsenoside Rb_1 in Compound Chanshu Granule by RP-HPLC

Objective To determine the content of Ginsenoside Rb1 by RP-HPLC. Methods HPLC was employed with a column of Diamonsil ODS-C18 and a mobile phase of Acetonitrile-H2O (29∶71), the detection wavelength at 203 nm and the column temperature at 25 ℃. Results The linear correlation is available between 0.776 and 7.760 ?g/mL of Ginsenoside Rb1, and the correlation coefficient was 0.999 99, the average recovery was 99.69% and RSD was 2.04% (n=6). Conclusion The method is simple and accurate, and applicable to control the quality of Chanshu Granule.
6.The expressional changes of mitogen-activated protein kinase signaling transduction associated genes and their significance in gastric cancer cells induced by parvovirus H-1 infection

Objective To explore the antineoplastic parvovirus H-1 induced cell death signaling pathways and their possible mechanisms in gastric carcinoma cells. Methods By using RT-PCR method, the mRNA expressional changes of mitogen-activated protein kinase (MAPK) signaling transduction pathway related genes were measured in gastric adenocarcinoma HGC27 cells infected by H-1 virus. Results The RT-PCR amplification results displayed that 48 h after HGC-27 cells were infected by H-1 virus, the expression of CREB was increased, the expression of ERK1, STAT2, p38-?, MEK2, ?-RAF and MTK1 were remarkably decreased, however, the expression of JNK2, ETS and ERK2 had no apparently change. Conclusions The cytotoxic effects of parvovirus H-1 might affect the gene expression involved in MAPK signal transduction pathway in gastric carcinoma HGC27 cells. It indicates that H-1 virus might interfere with specific cellular signal transduction pathways of gastric carcinoma cell to induce cell death. In conclusion, it was considered that modified and reconstructed parvovirus H-1 would be a very valuable tool in the anti-tumor study.
7.Investigation on the sensitivities of distinct gastric cancer cells to parvovirus H-1 induced cytotoxicity

Objective To investigate the sensitivities of distinct gastric cancer cells to parvovirus H-1 induced cytotoxicity and its possible mechanisms. Methods Six distinct differentiated gastric cancer cell lines were employed in this study, including HGC27 (undifferentiated), BGC823(undifferentiated), MKN45(poor differentiated), AGS (poor differentiated), SGC7901(moderate differentiated) and MKN28 (well differentiated). The cell cycle distributions were measured by flow cytometry. The differential sensitivities of six distinct gastric cancer cells after H-1 virus infection were detected by MTT analysis. The RT-PCR was employed to detect viral NS-1 gene expression in all six gastric cancer cell lines. Results The S phase ratios of HGC27, BGC823, MKN45, AGS, SGC7901 and MKN28 were 24.72%, 30.15%, 27.10%, 29.03%, 31.82% and 33.73% respectively. HGC27 cells were sensitive to H-1 virus induced cytotoxicity followed by SGC7901 cells. MKN45 and AGS cells were moderately sensitive. MKN28 cells were insensitive. However, BGC823 cells were resistant to H-1 virus induced cytotoxicity. The expressions of viral NS-1 were higher in HGC27, BGC823, MKN45 and SGC7901 cells, whilst NS-1 gene expressions were lower in AGS and MKN28 cells. Conclusions The sensitivities of distinct gastric cancer cells to H-1 virus induced cytotoxicity could be markedly different. In general, the poorly differentiated cells showed an enhanced sensitivity to H-1 virus attack as compared to well differentiated ones. The enhanced sensitivity of poorly-versus well-differentiated gastric cancer cells to H-1 virus is due to at least in part, to the enhanced capacity of the former cells for NS-1 protein production and accumulation. The undifferentiated BGC823 cells were resistant to H-1 virus triggered cytotoxicity. It may further verify that not all the tumor cells were sensitive to H-1 virus lytic effects.

10.Effects of straw direct vapor nitrogen method on the motility of post-thaw human sperm

ObjectiveTo study the effects of two cryopreservation methods ( direct vapor nitrogen method and program freezing method) on the motility of post-thaw human sperm.MethodsThe 25 semen samples were taken from 25 donors from Shanghai Human Sperm Bank during May to July 2011,using straw as a frozen carrier, the semen samples contained modified glycerol-yolk-citrate ( GYC ) ( 1 ∶1 ) as cryoprotectant,after the liquefaction, each semen sample was divided into 2 groups,and separately cryopreserved by direct vapor nitrogen method and program freezing method.According to the recommended standardoftheWorldHealthOrganization, semensampleswereanalyzedbeforeandafter cryopreserving.Density,total motility,PR,motility parameters ( including VAL,VSL,VCL,BCF) were determinated by the IVOS sperm analyzer.Semen samples were stained by modified papanicolaou stain and 200 sperms of each sample were counted under microscope by manual analysis to calculate the rate of normal morphology sperm.The paired t test was used to compare the above indictors before and after cryopreserving in one method and after thawing between two methods.Results Before cryopreserving,the total motility,PR,VAL,VSL,VCL,BCF were(91.36±4.96)%,(81.28 ±6.41)%,(59.29 ±9.30) μm/s,(49.80±8.40) μm/s, (83.39 ± 15.70)μm/s, (16.68 ± 3.36)Hz respectively, and after cryopreserving by direct vapor nitrogen method the results were (46.04 ± 20.14)%,(40.22 ± 18.42 )%,(48.00± 11.60)μm/s,(41.11 ±8.50) μm/s,(71.55 ±17.19) μm/s,(15.94 ± 14.30) Hz respectively,the differences of the indictors above were observed with statistic significance except BCF ( t =11.324,11.958,4.266,3.964,3.620,0.258,P of BCF >0.05,P of others <0.05); and after cryopreserving by program freezing method,the results were(48.05 ± 18.27)%,(42.47 ± 15.62)%,(45.30 ± 6.51) μm/s,(38.08 ± 4.99) μm/s,(64.23 ± 11.75) μm/s,(14.00 ± 2.99) Hz respectively,the differences of the indictors above showed statistic significance (t =10.337,10.874,7.154,5.510,6.494,3.244,P <0.05).No significant differences were observed in the revival rate,total motility,PR and normal morphology sperm rate by using different methods (t =0.425,0.364,0.622,1.106,P >0.05).The results of VAL,VSL,VCL,BCF in direct vapor nitrogen method were higher than that in program freezing method,when the differences of VSL and VCL were observed with statistic significance(t =2.489,2.822,P < 0.05 ).Conclusions Using straw as carrier is practicable.Direct vapor nitrogen method is easier to operate than program freezing method.Post-thaw sperm cryopreseved by direct vapor nitrogen method has higher motility parameters,thus making this method an ideal one for fast freezing.
