BACKGROUND:Numerical stimulation technology is a good method to analyze blood flow changes after intracranial aneurysm stenting that oftenresult in restenosis.
OBJECTIVE:To explore the hemodynamic effect of stent implantation on patient-specific intracranial aneurysm and the distribution of the wal shear stress on the stent surface.
METHODS:Brain CT data from a patient with intracranial aneurysm were extracted and optimized to establish a patient-specific intracranial aneurysm materialization model. Meanwhile, a three-dimensional model of rectangular section spiral stent was designed to develop an aneurysm model with the stent by the method of boolean operation. Then, the possibility of restenosis was analyzed based on the distribution of wal shear stress on the surface of stent model.
RESULTS AND CONCLUSION:The wal shear stress on the stent surface which was more than 40 Pa increasedalong with the increase of blood velocity and blood viscosity at the same moment. However, the wal shear stress on the stent surface which was less than 0.5 Pa decreased along with the increase of blood viscosity, and its distribution was the largest when the blood velocity was 0. This stent provides a new insight into the controling of the aneurysm growth and rupture, but the restenosis area is too large. In order to prevent restenosis, the stent need to be optimized or reselected to keep the desired shear stress values of 0.5 to 40 Pa, and meanwhile to minimize the pulsating change of wal shear stress during a cardiac cycle.

Objective To explore transinfection of rabbit chondrocytes via chitosan microsphere with human IL-1Ra and TGF-β1 gene. Methods Chitosan-DNA microspheres carrying plasmids with IL-1 Ra and TGF-β1 genes were prepared. The encapsulation efficiency,DNA-released kinetics and lysozyme degradation in vitro were performed.Articular rabbit chondrocytes were co-transinfected with the plasmids with IL-1Ra and TGF-β1 genes via chitosan-DNA mierosphere, evaluated by fluorescence microscope,TaqMan real-time PCR assay and MTF test. Results The chitosan microspheres with IL-1Ra and TGF-β1 genes were(2.8±0.2)μm and(2.6±0.1)μm in diameters respectively. The encapsulation efficiency were(88.3±4.1)%and(87.2±2.6)%.During the degradation, significant morphological changes were noticed. The plasmids could be released in a multiphasic fashion.Enhanced green fluorescent protein and Real-Time PCR analysis showed that genes were expressed in chondrocytes.lasting near 30 days.MTT indicated that the cotransinfection promoted the chondrocytes'proliferation. Conclusion IL-1Ra and TGF-β1 genes cotransfected into chondrocytes via chitosan-DNA microsphere could be expressed in a long term and cotransinfection promoted the chondroeytes'proliferation, which is the base of inhibiting the degeneration of cartilage and promote cartilage repair.

AIM: To evaluate the specific cellular immune response in mice inoculated with the recombinant hepatitis B virus (HBV) surface vaccine (rHBs). METHODS: Spleen T lymphocyte reactivity to rHBs was assessed by a proliferation assay and cytokine secretion. BALB/c mouse were intraperitoneally inoculated with rHBs at doses of 0.65, 1.25, 2.5 or 5 ?g for once or twice. 4 weeks later, spleen lymphocytes were harvested and restimulated with rHBs in experimental group or with PBS as control. The spleen lymphocytes were labeled with [~3H]-thymidine for 3 days. The [~3H]-thymidine incorporation was measured, which expressed as the incorporation of [~3H] (counts?min~ -1 ) and stimulation index (SI) was calculated by the method of dividing the cpm obtained in the experimental group by that in control group. The content of IL-2 and IFN-? secreted from the spleen lymphocyte were measured by the method of ELISA. RESULTS: 2 weeks after primary vaccination, the SI in 0.65, 1.25, 2.5 and 5 groups was 1.55, 1.93, 2.41, 2.81 ng/L, respectively. IL-2 was 5.48?8.88, 9.28?6.98, 28.53?14.32, 64.69?20.88 ng/L, respectively. IFN-? was 8.22?8.61, 9.89?9.34, 20.27?15.50, 30.77?22.12 ng/L, respectively. 2 weeks after boost vaccination, the SI in 0.65, 1.25, 2.5 and 5 groups was 1.61, 2.05, 3.74, 3.62 ng/L, respectively. The IL-2 was 5.75?5.04, 102.53?67.52, 177.13?91.12, 332.10?124.31 ng/L, respectively. IFN-? was 3.63?4.42, 28.33?13.04, 59.66?25.75, 80.73?19.30 ng/L, respectively. CONCLUSION: Specific proliferation activity and IL-2, IFN-? secretion from the spleen lymphocytes of the mouse inoculated with rHBs are produced, that the strength is dependent on the dose of vaccination.
