1.Study on the protective effect of MgSO4 on the radiation-induced neural stem cell injury

Objective To explore the neuroprotective effect of magnesium sulfate on radiation induced neural stem cell injury. Methods Brain tissue was obtained from new-born sprague-dawley rats within 24 hours, and the cerebral hemisphere was dissociated to culture the neural stem cells. After being identified by immunofluorescence method, the neural stem cells were randomly divided into 3 groups as blank control group, experimental control group and experimental group. The neural stem cells of experimental control group and experimental group were irradiated with 2 or 4 Gy of gamma rays. The proliferation and the cell cycle of neural stem cells were detected at different time-points ranging from 24 h,48 h, 72 h after irradiation with CCK-8 and FCM. Results Compared with the blank control group, the proliferation rate of experimental control group was significantly reduced ( t = 5. 33-8.44, P < 0. 05 ), and the G1 phase arrest of experimental control group was significantly enhanced ( t = 30. 60-71.22, P < 0. 05 ).Compared with the experimental control group, the proliferation of experimental group significantly increased excluding that of 24 h ( t = 2.45-4. 71, P < 0.05 ), the apoptosis rate of experimental group significantly decreasd (t = 6.73-41.12, P < 0.05 ), which was closer to the blank control group.Conclusion Magnesium sulfate can alleviate the injury of proliferation and decrease the cell apoptosis in the early stage after irradiation.
3.Effect of Resveratrol on the Expression of Survivin and p27 Protein in Human Renal Cell Carcinoma 786-0 Cells

Objective To investigate the effect of resveratrol ( Res ) on the expression of survivin, p27 protein in human renal cell carcinoma 786-0 cells. Methods The cell cycle and apoptosis of 786-0 cells were detected by flow cytometry, and the expression of survivin and p27 protein of 786-0 cells was detected by Western blotting at time point of 12, 24 and 48 h after human renal cell carcinoma 786-0 cells were cultured with Res ( 25μmol·L-1 ) and blank medium, respectively. Results The ratio in G1 phase [(64.87±3.76)%] and G2 phase [(9.91±0.82)%] in the experimental group decreased significantly (P<0.05), and the ratio of S phase [(25.05±3.12)%] increased significantly (P<0.05).The apoptosis rate of experimental group at 12, 24 and 48 h [(5.12±0.64)%, (5.71±0.49)%, (5.84±0.30)%] was significantly higher than that of the control group [ ( 0. 72 ± 0. 27 )%, ( 0. 93 ± 0. 25 )%, ( 1. 12 ± 0. 21 )%, P<0. 05 ] . The expression of survivin protein in experimental group was significantly higher than that of the control group ( P<0. 05 ) , and the expression was significantly increased with time prolonging (P<0.05).The expression of p27 protein in experimental group was significantly lower than that of the control group (P<0.05), and the expression level was significantly decreased in time-dependent manner (P<0.05).There was a negative correlation between the expression of survivin and p27 protein in the experimental group ( P<0.05) . Conclusion Res could inhibit the expression of p27 protein and up-regulate survivin expression level, and promote the apoptosis of renal cell carcinoma 786-0 cells.
4.The role of bilateral internal iliac arterial embolization in the treatment of hemorrhagic cystitis after bone marrow transplantation

Objective To evaluate the usefulness of bilateral internal iliac arterial embolization in the treatment of hemorrhagic cystitis after bone marrow transplantation. Methods Among 28 patients with hemorrhagic cystitis, 7 patients experienced bilateral internal iliac artery embolization because of serious hemorrhage that couldn′t be controlled by conservative treatments,including CML (n = 3) , AML(n = 2) , ALL (n = 1),acute myelomonocytic leukemia(M4, n = 1). Embolization of the target vessels was performed by using gelfoam particleas for bilateral internal iliac arterial visceral branches. Results Gross hematuria disappeared in 4 cases after embolization by gelfoam particles. The time of bleeding controlled continued 2-10 d(mean 6 d). No rebleeding was observed in all of the cases during the 12 - 82 days follow up.Gross hematuria markedly reduced in another 3 cases after embolization. But failure occurred in other two patients. No definite complication related to the procedure was noted. Conclusion Bilateral internal iliac arterial embolization is an effective, less invasive, and safe method for serious hemorrhagic cystitis after bone marrow transplantation. 5.Preliminary observation on the effects of implantation of BMSCs-root-CHA complex in dog

Objective:To evaluate the periodontal tissue regeneration effect of implantation of bone marrow stromal cells(BMSCs)-root-coralline hydroxyapatite(CHA) complex.Methods:BMSCs of beagle dogs were in vitro cultured on CHA scaffold and on tooth root surface respectively.Then BMSCs-root samples were inserted into BMSCs-CHA scaffolds to form BMSCs-CHA-root complexes.5 complexes were implanted on one side of mandible in each of the dogs 1 month after the 4th,5th and 6th teeth had been extracted.Root-CHA without BMSCs was implanted on another side as the control.All dogs were killed 16 weeks after implantation,and the periodontal tissue regeneration were observed histologically.Results:More new bone formation and more mature alveolar bone were observed in the test group than that in the control,and there was a clearly periodontal ligament-like tissue formation only in the test group.Conclusion:The complex of BMSC-root-CHA can improve the periodontal tissue regeneration.
6.Research status and analysis of English translation of package instruction of Chinese medicine

Through approaches of literature statistics and content analysis, the English translation studies of package instruction of Chinese medicine from Chinese literature database were summarized and analyzed, including China National Knowledge Infrastructure (CNKI), Wangfang Data and VIP Database. It was found that there existed four main problems in English translation of package instruction of Chinese medicine. First, there were relatively few studies in this field. Second, the papers were distributed unevenly in the relevant periodicals. Third, most studies were practice-based studies. Fourth, the research methods were simple. Theoretical guidance, research system and empirical research should be paid attention in English translation studies of package instruction of Chinese medicine.

Four compounds were isolated from the ethanolic extract of the bark of Alseodaphnehainanensis Merr. , The structures were identified as: a neolignan eusiderin A [(7R, 8R)-3,4,5,3′-tetram-ethox-△8’,9’-8-o-4’,7-o-5’lignan](I)two benzylisoquinoline alkaloids (6,7-dimethoxyisoquinolinyl)-(4′-methoxyphenyl) methanone(Ⅱ), and( 6, 7-methylenedioxyisoquinolinyl )-( 4′-methoxyphenyl ) methanone (Ⅲ), and 4-hydroxy-3-methoxy benzoic acid ( Ⅳ ) on the basis of HR-SIMS,1HNMR,13CNMRand 2D-NMR spectroscopic analysis. CompoundsⅠ~Ⅲ were obtained from the Alseodaphne genus forthe first time.
9.Effects of combination of dexmedetomidine and sufentanil preconditioning on ischemia-reperfusion injury in isolated rat hearts

ObjectiveTo investigate the effects of combination of dexmedetomidine and sufentanil preconditioning on ischemia-reperfusion (I/R) injury in isolated rat hearts.MethodsForty male SD rats weighing 180-220 g, aged 8-10 weeks, were anesthetized with intraperitoneal 3%pentobarbital 60 mg/kg and heparin 3000 U/kg.The hearts were excised and perfused in a Langendorff apparatus with K-H solution saturated with 95%O2-5%CO2 at 36.5-37.5 ℃.Forty isolated rat hearts were randomly divided into 5 groups ( n =8 each):control group (group C),I/R group,dexmedetomidine preconditioning group (group DP),sufentanil preconditioning group (group SP),and dexmedetomidine + sufentanil preconditioning group (group DS).I/R injury was induced by 30 min of ischemia followed by 120 min of reperfusion.Groups DP,SP and DS received 30 min of perfusion with K-H solution containing dexmedetomidine 2.3 ng/ml,sufentanil 3.77 ng/ml and dexmedetomidine 2.3ng/ml + sufentanil 3.77 ng/ml respectively at 30 min before ischemia.Coronary flow (CF),left ventricular developed pressure (LVDP),± dp/dtmax and HR were measured at 15 min of equilibration,immediately before ischemia,at 30 min of ischemia and at 120 min of reperfusion.The superoxide didmutase (SOD) and myeloperoxidase (MPO) activities and the malondialdehyde (MDA) content in myocardial tissues and infarct size were determined at 120 min of reperfusion.Results Compared with group C,CF,LVDP, ± dp/dtmax,HR and SOD activity were significantly decreased,and MPO activity,MDA concent and infarct size were significantly increased in the other 4 groups ( P < 0.05).Compared with group I/R,CF was significantly decreased in groups DP and DS,HR was significantly decreased in groups SP and DS,and LVDP, ± dp/dtmax and SOD activity were significantly increased,and MPO activity,MDA content and infarct size were significantly decreased in groups DP,SP and DS ( P <0.05).Compared with group DS,CF was significantly decreased and HR increased in group DP,and SOD activity was significantly decreased,CF,MPO activity,MDA content and infarct size were significantly increased in group SP (P < 0.05 ),and no significant change was found in SOD and MPO activities,MDA content and infarct size in group DP ( P > 0.05).Conclusion Dexmedetomidine combined with sufentanil preconditioning can reduce I/R injury in isolated rat hearts,but the myocardial protection is not further enhanced.
10.Protective effect of apigenin in acute cerebral injury of rats

Objective To study the inhibition of iron-dependent lipid peroxidation(IDLPO)by apigenin on rat cerebral homogenate in vitro and to evaluate the neuroprotective effects of apigenin on acute transient focal cerebral ischemia-reperfusion injury in rats.Methods In vitro IDLPO on rat cerebral homogenate was induced with ferrous sulfate,the incubation mixture was observed at three various concentration of apigenin and deferoxamine(DFX),and malondialdehyde(MDA)level was assayed by the 2-thiobarbituric acid(TBA)test.In vivo,the transient focal cerebral ischemia-reperfusion model in rats was established with insertion of thread embolish into middle cerebral artery.In experiment groups the neurological behavior scores,TTC stain of brain slices,and neurocyte morphology were observed.The homogenate of left hemisphere was collected for investigating the effect of apigenin on content of MDA and activity of superoxide dismutase(SOD)in 24,48,and 72 h.Results MDA was reduced in three various concentration of apigenin on rat cerebral homogenate(P0.05).In vivo,abnormal neurological behavior scores existed and typical cortical infarct lesions were found by TTC stain in both apigenin and model groups.An obvious intracellular and intercellular edema and vacuolization were found in the cerebral cortexes and hippocampuses in model group.There was karyopycnosis in glias and neurons.However lesion was alleviated in apigenin group.The MDA contents in both apigenin and model groups were increased greatly compared to ones of Sham-operated group,respectively(P
