1.Application of DNA ploidy analysis in diagnosis of serous cavity effusion

Objective To explore the application value of automated cell DNA ploidy analysis system in the diagnosis of benign and malignant serous cavity effusion.Methods 262 cases of serous cavity effusion(169 cases of pleural effusion,78 cases of ascites,15 cases of pericardial effusion)were treated by centrifugation,2 slices of each sample were made.One of them used for dyeing Feulgen,which given automatic cell DNA ploidy analysis,another one for Papanicolaou staining,with a conventional cytology.The positive detection rate of these 2 kinds of different detection methods for malignant serous cavity effusion were compared.Results 119 cases(45.4%)of 262 cases abnormal were detected by conventional cytology of serous cavity effusion.Meanwhile,113 cases (43.1 %)were detected abnormal by DNA ploidy analysis in the same samples.73 cases of tumor cells and suspicious tumor cells were found by conventional cytology,and different ploidy cells were found in all of these samples In conventional cells,46 cases of nuclear heterogeneous cells were found,while only 34 cases exist different ploidy cells.Conclusion Automated cell DNA ploidy analysis system is helpful to improve the positive diagnosis rate of serous cavity effusion,which can be used as an important auxiliary means of cytology.
2.Study on Mechanism of Sophora flavescens against hepatitis B virus

Objective To study on Sophora flavescens against hepatitis B virus (HBV) and its mechanism.Methods HepG2.2.15 cells were cultured in vitro, respectively with the negative control group without drug intervention, positive control group (5-Fu group) which was given 0.77 mmol/L 5-Fu, Matrine group treated with 0.77 mmol/L Matrine intervention.HBSAg, HBeAg inhibition rate and cell survival rate, apoptosis rate, cell cycle as observation indexes, comprehensive evaluation of matrine effect on anti 2.2.15 cell HBV in vitro.Results The positive control group, matrine group compared with the negative control group, the inhibition rate of HBsAg and HBeAg were statistically difference ( all P<0.05 ) , when positive control group and Oxymatrine group compared, the inhibition of HBsAg and HBeAg, the difference were statistically (all P<0.05).Cell survival rate, when the positive control group and the negative control group compared, had no significant difference (P<0.05), when matrine group compared with the negative control group, there were significant differences in the survival rate of the cells (t=4.25, P<0.05).Rate of apoptosis cells in negative control group and positive group had not statistically significant, matrine group apoptosis rate was higher than that of the negative control group and the control group (P<0.05).The cells in G0/G1 phase, had no significant differences of distribution in each group, the cells in G2/M phase, the positive control group and matrine group were lower than those in negative control group (P<0.05), matrine group was lower than that of the positive control group (P<0.05).The S phase cells of positive control group and matrine group were higher than those in negative control group (P<0.05), matrine group was higer than that of the positive control group (P<0.05).Conclusion Matrine has obvious effect of anti HBV, inducing cell apoptosis, cell proliferation arrest in S phase.

4.Prevalance of HIV Infection in Hospitalized Patients and Prevention Measures

OBJECTIVE To understand the prevalence of HIV infection in hospitalized patients and the prevention measures.METHODS The prevalence of HIV infection of the hospitalized patients in our General Hospital from Jan 2005 to Jun 2009 was investigated retrospectively.The time,population and endemic distribution of the HIV infected patients were analyzed.RESULTS Forty-eight HIV infected patients(19 HIV patients),were screened out from 271 955 hospitalized patients from Jan 2005 to Jun 2009 with year by year increasing.Most of HIV infected patients(62.5%) came from Beijing,Henan and Hebei provinces.Some sporadic patients came from other areas.CONCLUSIONS The prevalence of HIV infected patients among hospitalized patients in our hospital is increasing in recent years.The prevention measures in diagnosis and therapy process for these patients should be strengthened to reduce hospital infection.
5.The design of high-efficiency exhalation disinfection device for the patient infected with SARS

High-efficiency exhalation disinfection device for the patient infected with SARS can also meet the disinfection requirements of the patients with severe respiratory communicable diseases. Connected with the exhaust vent of the breathing mask or the respirator by corrugated tubes, this device gathers exhalation containing a large amount of causative agents through the completely leakproof ductwork and then expels purified air after multiplicate disinfection by high temperature, high pressure and chemical disinfectant. The device can prevent the patient with severe respiratory communicable disease from polluting their surrounding air and thus cut off the route of transmission, reduce the infection rate and the death rate of the person and medical personnel who has made intimmate contact with the patient.

8.Expression of hMLH_1 gene in gastric cancer and its significance

Objective To investigate the expression of hMLH_1 in gastric cancer and its significance,Methods The expressions of hMLH_1 protein and mRNA were detected by immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR) in 68 cases of gastric cancer tissues,33 cases of para-cancer tissues and(35 cases) of normal gastric tissues.Apoptosis was assayed by terminal deoxytransferase mediated dUTP nicked labeling.Results The positive expression rates of hMLH_1 mRNA were 63.3%(43/68),100%(33/33) and 100%(35/35),and those of protein were 61.8%(42/68),93.9%(31/33),100%(35/35),respectively,in gastric cancer tissues,para-cancer tissues and normal gastric tissues.There was a correlation between the expression of hMLH_1 and clinical grade of gastric cancer(r=0.531,P
10.The changes of main upper gastrointestinal diseases and Helicobacter pylori infection status in the past thirty five years

Objective To study the spectrum of upper gastrointestinal diseases and infection rate of Helicobacter pylori(Hp) in our hospital during the past 35 years.Methods Patients who were 16 or older with duodenal ulcer,gastric ulcer,reflux esophagitis,gastric cancer and esophageal cancer diagnosed by gastroscopy and pathology were retrospectively enrolled in our study from January 1980 to December 2014.Patients with chronic superficial gastritis,chronic atrophic gastritis or Hp infection from January 1989to December 2014 were also included in our study according to the same diagnostic criteria.The incidences of diseases and the infection rates of Hp were studied.Results A total of 213 495 patients underwent gastroscopy in our department during the past 35 years.The overall diagnostic rates of duodenal ulcer,gastric ulcer,reflux esophagitis,gastric cancer and esophageal cancer were 9.87%,3.79%,6.66%,1.59% and 0.66% respectively.There were 183 426 patients receiving gastroscopy in our department from January 1989to December 2014.The overall endoscopic diagnosis rates of chronic superficial gastritis and chronic atrophic gastritis were 49.83% and 22.43% respectively.The overall infection rate of Hp was 36.18%,which had a declining trend consistent with peptic ulcer (all P =0.000).Yet,the prevalence of reflux esophagitis,chronic superficial gastritis and chronic atrophic gastritis were increasing (all P =0.000).The diagnostic rates of gastric cancer and esophageal cancer were persistent (P =0.266,P =0.156).Conclusions The Hp infection during years has been decreasing,consistent with the declining tendency of peptic ulcer.On the other hand,reflux esophagitis,chronic superficial gastritis and chronic atrophic gastritis show an ascendant trend.The proportion of patients with gastric cancer and esophageal cancer is relatively stable.
