1.PATERNITY TEST STUDY BY DNA FINGERPRINTS AND SEROLOGICAL TESTING

Comparison between the DNA fingerprinting and the serological method used for paternity test is presented. 32 cases of disputed paternity were tested using DNA fingererints with MYO minisatellite DNA probe and blood typing. The results of DNA fingerprinting test correlated with those of blood typing. 8 alleged fathers were excluded while 24 were confirmed. The relationship between the results from two methods was discussed.
2.Anti-hepatoma activity of nitidine chloride and its effect on topoisomerase

Aim To investigate the anti-tumor effect of nitidine chloride(NC)on human HepG2 hepatocellular transplanted tumor in nude mice and its effect on topoisomerase.Methods The subcutaneous transplantable tumor model of human liver cancer in nude mice was established and the anti-tumor effect of NC was calculated.The effects of NC on TopoⅠ/Ⅱ mediated-pBR322 DNA relaxation were measured by using agarose gel electrophoresis.Results NC inhibited significantly the growth of hepatoma,The inhibitory rate at the dose of 2.5,5,10 mg·kg~(-1) was 12.06%,35.63% and 60.91% respectively.At the concentration of 6.25 μmol·L~(-1),NC completely inhibited the pBR322 DNA cleavage mediated by TopoⅠ;at the concentration of 25 μmol·L~(-1),NC completely inhibited the pBR322 DNA cleavage mediated by Topo Ⅱ.Conclusion Nitidine Chloride can inhibit hepatic carcinoma growth in nude mice,The anti-tumor mechanism is probably related to the inhibitory effect on Topo.
3.Epithelial-mesenchymal transition and gastric cancer

Epithelial-mesenchymal transition (EMT) is that cells undergo a switch from epithelial phenotype to mesenchymal phenotype.Recent researches show that EMT can affect the occurrence and development of gastric cancer through a variety of mechanisms and promote the migration and invasion of tumor cells.The process of EMT involves a number of signal transduction pathways,and is related to microRNAs,Helicobacter pylori,transcription factors and so on.EMT is expected to be a novel therapeutic target of gastric cancer.

5.Clinical Observation on Therapeutic Effect of Breviscapine Injection on Migraine

OBJECTIVE:To observe the therapeutic effect of Breviscapine injection in treating migraine. METHODS: 81 patients with migraine were randomly divided into two groups: 42 in treatment group were treated with Breviscapine injection 20 mL added to 5% Glucose injection 300 mL iv gtt q.d, and 39 in control group with Flunarizine 10 mg po quaque nocte. The treatment lasted for 2 courses (1 course of treatment lasted 14 days) with an interval of 15 days between the two courses. The patients were followed for more than 6 months. The curative effects between the 2 groups were compared, and the variations in hemorheological indexes of the 2 groups before and after treatment were observed. [Objective]To evaluate the safety and efficacy of the surgery,which was done by a video-assisted thoracoscope to release the anteriorroute part of spine and done by posterior instrumented fusion with severe Adolescent Idiopathic Scoliosis(AIS).Clinic date:There were 10 cases of AIS(King Ⅱ 6,King Ⅲ 3,King Ⅳ 1),in which there were 6 males and 4 females.The average age was 17.2 years(range 14~21).The average preoperative primary curve in the coronal plane was 91.3?(ranged 67?~125?)and the average pliability of thoracic spine was 28.3%(ranged 19.3%~36.5%).[Method]With general anesthesia(onelung ventilation or two-lung ventilation),the disc complexes and part of the anterior longitudinal ligament of 5~6 disc near apex vertebra were excised under endoscopic visualization.In the same operation,the posterior instrumented fusion was done.[Result]The anterior releases under endoscopic visualization needed 120~150 min with average bleeding 157ml(ranged 100~200 ml).The thoracic draining was 100~150 ml After the posterior route surgery,the average coronal Cobbs angle was 47?(ranged 26?~75?)and the rate of rectification was 51.5%.The patients had good fusion and outlook without other complications.[Conclusion]This way,which is done by a videoassisted thoracoscope to release the anterior part of spine and done by posterior instrumented fusion has a good result about safety and efficacy.
7.The model of experimental autoimmune uveoretinitis in rats

Objective:To induce experimental autoimmune uveoretinitis in Wistar for investigating the pathogenesis mechanism of uveitis.Methods:18 Wistar rats were immunized by different dosage of bovine soluble antigen(S Ag),and were detected the clinic changes dayly.The animals were anesthetized and killed when they had significant clinic EAU appearance or 3~4 weeks after immunized,the eyes were removed and followed by pathologic studies.Results:The incidence of EAU in the 3 different dosage S Ag immunized rats were 2/12 in 100 ?g S Ag group(6 rats,12 eyes),6/12 in 200 ?g S Ag group(6 rats,12 eyes) and 8/12 in 300 ?g S Ag group(6 rats,12 eyes) separately and the pathology scores were 0?16 76?11.02?17 56?9 96 separately in 3 group of rats.Conclusion:The middling sensitive Wistar rats can be used to induce EAU successly with middling purified S Ag.
8.Sox2 induce mouse embryonic fibroblasts direct reprogramming into neu-ral stem cells

AIM:To study the direct reprogramming method of mouse embryonic fibroblasts (MEFs) conver-ted into induced neural stem cells (iNSCs).METHODS:Sox2-infected MEFs were cultured in NSCs culture medium for 10 d.Subsequently , repeated suspension and adherent culture were performed for 3 times for the purification of iNSCs .The iNSCs were cultured in suspension medium .Real-time PCR was used to detect the expression of neural stem cell marker genes and pluripotent marker gene .In vivo, iNSCs were microinjected into the mouse cerebral cortex .Immunofluorescence was performed to detect the expression of neural stem cell , neuron, oligodendrocyte and astrocyte markers in vitro and vivo. RESULTS:A variety of neural stem cell marker gene expression was significantly increased in iNSCs detected by real -time PCR.Immunofluorescence confirmed that iNSCs expressed nestin and differentiated into neurons , oligodendrocytes and as-trocytes in vitro and vivo.CONCLUSION:Sox2 is sufficient to trigger the direct reprogramming from MEFs to iNSCs .iN-SCs have the ability of self-renew and 3 differentiation potentials in vivo and vitro.iNSCs are the suitable seed cells of SCI .
9.Therapeutic effect of ulnar nerve with or without transposition during open reduction and internal fixation of distal humerus fractures

Objective To investigate the effect of ulnar nerve with or without transposition during open reduction and internal fixation of distal humerus fractures.Methods A retrospective case control study was made on 74 cases receiving open reduction and internal fixation of distal humerus fractures between January 2008 and December 2015.There were 28 males and 46 females,with a mean age of 63.2 years (range,32-83 years).AO fracture classification was A2 type in seven cases,A3 type in four,C1 type in 14,C2 type in 28 and C3 type in 21.According to the intraoperative management of the ulnar nerve,the patients were divided into transposition group (n =43) and non-transposition group (n =31).Two groups were compared with respect to operation time,Mayo elbow performance score (MEPS),incidence of ulnar neuritis and postoperative complications.Results Follow-up lasted for 6-24 months (mean,11.7 months).Operation time differed significantly between transposition group [(110.9 ± 20.8) minutes] and non-transposition group [(95.6 ± 18.9) minutes] (P < 0.05).MEPS in transposition group was excellent in 15 cases,good in 20,fair in six and poor in two.MEPS in non-transposition group was excellent in 10 cases,good in 15,fair in five and poor in one,with the same excellence rate of 81% in comparison with transposition group (P > 0.05).Incidence of ulnar neuritis showed no significant difference between transposition group (9%,4/43) and non-transposition group (7%,2/31) (P >0.05).Postoperatively,one case of superficial wound infection and two delayed healing were noted in transposition group;one case of superficial wound infection and one delayed healing were noted in non-transposition group.There was no significant difference in incidence of postoperative complications between groups (P >0.05).Conclusions Ulnar nerve with or without transposition can both achieve satisfactory clinical effect during open reduction and internal fixation of distal humerus fractures.But non-transposition group is associated with shorter operation time without increasing the incidence of ulnar neuritis.
10.Improvement of spinal function in patients with severe kyphosis due to upper thoracic tuberculosis by anterior decompression and fresh-frozen allograft fibular fusion

60?) in the Huaxi Hospital of Sichuan University. Three cases aged less than 15 years, but the patients or their relatives were informed of and consented the operation scheme. Without rectification, fusion segments were propped open about 5 mm via the upper and lower fixed vertebra, and anterior one was transplanted with fresh-frozen fibular allograft (offered by the donor corpse and excised extremities due to surgical trauma from the Department of Orthopaedics in Huaxi Hospital. The immunogenicity was greatly reduced by routine defat, acellular procedure, soaking sterilization and deep freezing), while rear one was transplanted with costal bone for supporting graft and filling the graft bone intervals with autogenous bone strips. Gypsum (brace) was used to assist external fixation for 8-10 month postoperation. The complications, graft incorporation, neurological function (Frankel grading scale A-E: complete paralysis-normal or no paralysis), back pain (0-3: none-severe), and mobility score (1-4: lying in bed-needing no assistance) before operation and at final follow-up of each patient were collected for analysis. RESULTS: All the patients were available for follow-ups and the mean period was 3.4 years (1.5-5 years).①Apparent fusion was observed in all cases with no graft fracture, only one patient reported mild graft shift and the kyphosis was unchanged.②There was no worsening of the neurological status in any patient. Frankel grades of 3 adult patients were unchanged after operation. All adolescent patients (n =3) had definite neurological improvement and could walk without assistance, while another was adult patient. Complete relief of back pain was observed in all patients. CONCLUSION: Fresh-frozen allograft fibular is a liable graft material for the treatment of severe kyphosis induced by inactive upper thoracic tuberculosis. Anterior decompression and fusion can promote the recovery of motive function and the alleviation of pain, especially is effective for neurological recovery of adolescent.
