1.Analysis of HIV and HCV infection of heroin dependent patients

Objective To understand the situation of HIV and HCV infection of heroin dependent patients .Methods HIV and HCV serological detection were proceed among 565 heroin dependent patients ,and the results were compared according to genders and nationalities .Results Among 565 heroin dependent patients ,the infection rates of HIV ,HCV ,and HIV/HCV for males were 14 .57% (72/494) ,59 .72% (295/494) ,and 13 .36% (66/494) respectively ,and those for females were 19 .72% (14/71) ,77 .46%(55/71) and 16 .90% (12/71) respectively .The infection rates for males were significantly lower than females (P<0 .05) .There were significant difference of the infection rates of HIV ,HCV and HIV/HCV between Han patients and Uyghur patients(P<0 .05) .Conclusion The heroin dependent patients are high risk population for HIV and HCV infection .Education and publication of drugs and infectious should be enhanced in order to control the spread of disease and reduce disease incidence .
2.The changes of mitochondrial membrane potential detected by confocal microscopy in spermatocyte induced by bisphenol A

Objective:To detect changes in mitochondrial membrane potential after sperm mother cell damage induced by bisphenol A by laser scanning confocal microscopy(LSCM) and underline its potential action mechanism.Methods: The cultured spermatogenic cells were divided into 3 groups, respectively. Then 0, 10μmol/L, 100μmol/L of Bisphenol A(BPA) were added into the culture, after 3 hours culture, fluorescence probe JC-1 was used to lable the three groups. The fluorescence intensity of JC-1 in mitochondrial was then detected by LSCM. LSCM software was used to analyze the fluorescence intensity. Change of the mitochondrial membrane potential was represented by the change of fluorescence colors (relative proportion of red and green fluorescence is commonly used to measure mitochondrial depolarization ratio). Results:The ratio between the red and green fluorescence in the control group, low dose group and high dose group had significant difference. Intracellular mitochondrial membrane potential of Bisphenol A treatment group was lower than that of the control group, while mitochondrial membrane potential of high dose group was lower than that of the low dose group. Conclusion: Bisphenol A could damage spermatocytes, and as the dose increased, the more serious the injury. The method could real-time monitor with high sensitivity the change of intracellular mitochondrial membrane potential.
3.Effect of Pioglitazone on Interleukin-18 and Toll-Like Receptor 4 Expression in OLETF Rats with Insulin Resistance

Objective To investigate change of TLR4 in OLETF (Otsuka Long-Evans Tokushima fatty) rats with in-sulin resistance (IR), and to study the effect of pioglitazone (PIO) on the expression of TLR4, and to explore the possible mechanisms of the PIO reducing the risk of cardiovascular diseases. Methods Twenty four OLETF rats were fed with high-fat diet for 20 weeks to establish the IR model then they were randomly assigned into two groups:the model group (group M), in which the rats were fed with high-fat diet;the PIO group (group P), in which the rats were fed with PIO in addition to high-fat diet . Control group include 12 OLETF rats fed with normal diet (group NC). After 20 weeks of drug intervention, plasma levels of FINS (Fasting INSulin), FBG (Fasting Blood Glucose), blood lipid, IL-18 and TLR4 were assessed in every group. Results Comparing with group NC, FBG, Blood lipid, IL-18 and TLR4 were significantly increased in group M(P<0.05 or P<0.01), comparing with group M, FBG, Blood lipid were improved in group P, and serum IL-18, TLR4 were signifi-cantly lower in the group P than that in group M(P<0.05 or P<0.01). Conclusion TLR4 may be involved in IR by pro-moting inflammatory response, and PIO can significantly improve IR and inflammatory, and reduce the risk of cardiovascular diseases by inhibiting the expression of TLR4.

Myelodysplastic syndromes (MDS) are a heterogenous group of hematologic malignancies characterized by clonal expansion of BM myeloid cells with impaired differentiation. Of particular interest mutations is the recent recognition that genes involved in the regulation of histone function (EZH2, ASXL1,and UTX) and DNA methylation (DNMT3A,IDH 1/IDH2,TET2) are recurrently mutated in MDS,providing an important link between genetic and epigenetic alterations in this disease. Ongoing analysis of the seminal AZA-001study has taught many important lessons in the use of DNA methyltransferase (DNMT) inhibitors.Improved survival in patients with high-risk MDS treated with azacitidine extends to patients with any International Working Group-defined hematologic response.New information on the impact of DNMT inhibitors on the immune system and on stem cells will likely lead to novel uses of these drugs in MDS and other hematologic and nonhematologic malignancies. The immunomodulating drug thalidomide and its derivative lenalidomide have been used in the treatment of MDS,principally in lower-risk MDS.
5.Comparative Study on the Preventive Use of 3 Kinds of Cephalosporins for 1 066 Patients with Breast Lesion Resection in Shaanxi Province

OBJECTIVE:To compare the effects of prophylactic application of 3 different antibiotics on prognosis in patients underwent breast lesion resection. METHODS:1 066 patients with breast lasion resection from 12 hospitals of Shaanxi province were divided into trial group(360 cases),control group A(352 cases)and control group B(354 cases)according to random num-ber table. Trial group was given first generation cephalosporin cefazolin;control group A was given second generation cephalospo-rin cefuroxime;control group B was given third generation cephalosporin cefoperazone sodium and tazobactam sodium. The dosage regimens of 3 groups were as follows:relevant drug 2 g added into 0.9%Sodium chloride injection 100 ml,ivgtt,0.5 h before sur-gery,medication course≤24 h after surgery in trial group. Those indexes of 3 groups were observed,such as post-operative ADR, incision healing,infection,hospitalization duration,phamaceutical costs per capita. RESULTS:There was no statistical signifi-cance in the rate of incision healing and the rate of post-operative infection among 3 groups(P>0.05). The incidence of post-opera-tive ADR,hospitalization duration and phamaceutical costs per capita in observation group were significantly lower or shorter than in control group A and B,with statistical significance(P<0.05). CONCLUSIONS:Cefazolin is better than cefuroxime and cefo-perazone sodium and tazobactam sodium to reduce the postoperative adverse reaction,antibiotics cost per capita and hospital drug cost per capita,shorten the hospitalization duration.
6.Correlation Study Between Angiotensin II Type1 Receptor Autoantibodies and the Onset Risk in Patients With Essential Hypertension Combining Acute Coronary Syndrome

Objective: To study the correlation between angiotensin II type1 receptor autoantibodies (AT1-AAs) and the onset risk in patients with essential hypertension (EH) combiningacute coronary syndrome (ACS). Methods: Our research included in 4 groups: EH+ACS group,n=28, ACS group, n=29, EH group,n=26 and Control group,n=23 normal subjects. Serum levels of AT1-AAs were examined by ELISA; high sensitivity C-reactive protein (hs-CRP), blood levels of lipids and glucose were also measured and compared among different groups. The correlations between AT1-AAs and blood lipids, glucose, hs-CRP were studied by multiple linear regression analysis. Results: Compared with Control group, ACS group had similar level of AT1-AAs (0.26±0.09) vs (0.21±0.06),P=0.105; while EH+ACS group and HE group had increased AT1-AAs level as (0.40±0.005) and (0.33±0.10),P=0.001 andP=0.02 respectively; AT1-AAs level was higher in EH+ACS group than HE group,P=0.044. In addition, serum AT1-AAs level was positively related to hs-CRP in EH+ACS group, ACS group and EH group (r=0.589,r=0.503 andr=0.273, allP<0.01). Conclusion: Serum AT1-AAs level was positively related to the onset risk in patients with EH combining ACS; AT1-AAs was also related to hs-CRP at certain degree.
7.Internal component analysis on the event-based prospective memory of schizophrenia patients

Objective To compare the difference of prospective memory(PM) between schizophrenia patients and normal people,and to analyze the internal component of prospective memory using multinomial processing tree(MPT) model.Methods 17 schizophrenia patients and 17 age-and education-matched control participants completed an event-based PM task which was embedded within an ongoing computer-based color-matching task.Internal component of prospective memory was analyzed using multinomial processing tree(MPT) model.Results The scores of prospective memory performance in schizophrenia patients and normal controls were (21.83± 2.46) % and (38.81±2.26) %, and the difference was statistically significant (t=2.11, P<0.05).The difference of ongoing task between schizophrenia patients ((75.88±0.43) %) and controls ((71.44±0.45) %) was statistically significant (t=2.79, P<0.05).The reaction time of schizophrenia patients was significantly longer than that of normal controls((2.34±1.41) s vs (1.81± 1.19) s, t=11.24, P< 0.05).The MPT model revealed a significant group difference in the prospective component (0.82 vs 0.97, G2(1)=14.17, P< 0.01) and retrospective component (0.54 vs 0.41 ,G2(1) 14.17, P<0.01).Conclusion The prospective component of event-based prospective memory of schizophrenia patients was lower than normal controls,which may result in the impairment of PM performance.
8.Change in hippocampal voltage-gated Ⅰ sodium channel of Lithium chloride-Pilocarpine epileptic rat model

Objective To research the changes in hippocampal voltage-gated sodium channel of Lithium chloride-Pilocarpine epileptic rat models,including Ⅰ sodium channel α subunit protein (Nav1.1),mRNA of Ⅰ sodium channel alpha subunit protein gene and function of sodium channel.Methods Epileptic rat models of Lithium chloride-Pilocarpine were established.Nav1.1 expression in the hippocampus of experimental rats was detected by immunohistochemical staining method,and the changes in voltage-gated sodium channel function (the current-voltage curves,activation and inactivation curves and the recovery curve) of hippocampus nerve cells were detected by whole cell patch-clamp technique.Results (1) The Lithium chloride-Pilocarpine rat models were successfully reproduced.Three stages of behavior (acute,latent and chronic) of rat models were observed.The blank control group was free of seizure.(2) Immunohistochemistry results:neurons in CA1 and DG regions of hippocampal of epileptic rats were normal,and there was no obvious change in the expression of Nav1.1.In CA3 area,the degeneration and necrosis of neurons were obvious.Staining of Nav1.1 became superficial and even disappeared in these areas,but the normal tissues were enhanced around degenerative and necrotic neurons.Compared with the blank control group,the expression of Nav1.1 in the model group was higher(0.235 ±0.008 vs.O.210 ±0.002),and there was statistically significant difference (t'=-7.426,P < 0.05).(3) The whole-cell patch-clamp technique showed that the sodium current density of the model group increased significantly compared with that of the blank group [(-319.70 ± 28.24) pA/pF vs.(-229.06 ± 26.01) pA/pF,t =8.178,P < 0.05],the threshold value of activation curve decreased (4.15 ± 0.80 vs.4.50 ±0.85,t =11.020,P < 0.05),the threshold value of inactivation curve increased (7.47 ± 0.53 vs.6.24 ±0.31,t =6.940,P < 0.05),and the recovery time after inactivation shortened [(1.36 ± 0.15) ms vs.(1.86 ± 0.21)ms,t =6.712,P < 0.05],and there were all statistically significant differences.Conclusion Repeated seizures can lead to increase Nav1.1 compensatory expression of,and significantly increase sodium channel current density,while the threshold value of activation curve decreases,the threshold value of inactivation curve rises,and the recovery time after inactivation is shortened,which eventually leads to increased neuron excitability and is more likely to cause seizures.

Abstract: S-palmitoylation, a reversible and dynamic post-translational modification in cells, is involved in regulating the transcription and expression of downstream target genes as well as signal transduction, thereby affecting cell life activities. Studies have shown that thousands of human proteins undergo S-palmitoylation modification, suggesting that S-palmitoylation is closely related to the progression and treatment of diseases. T cells play central roles in anti-tumor immune responses. A variety of T cell immune-related proteins are regulated by S-palmitoylation. In the present study, we focus on the impact of S-palmitoylation on T cell signal transduction and its application in T cell immunotherapy, aiming to provide new ideas for the development of new targets and peptide inhibitors for T cell immunotherapy.
