1.Insulin-like growth factor 1 affects the apoptosis of rat condylar chondrocytes

BACKGROUND:Insulin-like growth factor 1 is the key factor during cartilage development, which is involved in the growth and reconstruction of condylar cartilage.
OBJECTIVE:To study the effect of insulin-like growth factor 1 on cel apoptosis and the apopotosis-associated factors of Bcl-2, Bax mRNA and protein expressions of rat condylar chondrocytes.
METHODS:The 1-day-old and 28-day-old rat condylar chondrocytes were cultured and identified in vitro. The condylar chondrocytes with different ages were divided into experimental group and control group. After being starved for 24 hours, chondrocytes in the experimental group were incubated with 100μg/L recombined rat insulin-like growth factor 1 for 48 hours, while the chondrocytes in the control group were incubated normal y. RESULTS AND CONCLUSION:Compared with the control group, after being incubated with recombined
insulin-like growth factor 1, the number of condylar chondrocytes was increased with high speed proliferation (P<0.05). Real-time RCR and western blot analysis revealed that the expression levels of Bcl-2 mRNA and protein were increased after added with recombined rat insulin-like growth factor 1, while the expression levels of Bax and protein were decreased (P<0.05). The results indicate that insulin-like growth factor 1 can promote the
proliferation and reduce cel apoptosis of newborn and adolescent rat condylar chondrocytes, which may be mediated by Bcl-2 and Bax.
2.CT Diagnosis of Mesenteric Panniculitis

Objective To analyze the CT manifestations of mesenteric panniculitis,so that to improve CT diagnosis of mesenteric panniculitis.Methods Seven patients with mesenteric panniculitis,which were diagnosed according to the CT criteria,were analyzed.Results The fatty masses with defined margins and around the root of the jejunum were found in all patients at CT scan,and there were no enhancement after administration of contrast medium.The masses were sharply defined with abdominal and retroperitoneal fat.There were pseudo-capsule in 7, "arcus adiposus sign" in 5. Enlargement of lymph nodes in the masses and retroperitoneum in 2 cases were seen,respectively.4 patients had abdominal operations,one patient was in combination with carcinoma of head of pancrease.Conclusion Certain features of the CT manifestations of mesenteric panniculitis can be found,CT is effective for the diagnosis of mesenteric panniculitis.
3.Three-dimensional structure and morphology of the mandible in type 1 diabetes mellitus mice

BACKGROUND: Diabetes mel itus is one of the most common systemic diseases, which often leads to the changes of the jaw and other bone structure, as wel as the abnormal changes of mineral metabolism. OBJECTIVE: To observe the three-dimensional structure and histopathological changes of the mandible in type 1 diabetes mel itus mice. METHODS: The mice were randomly divided into control group and diabetes mel itus group. The diabetes mel itus group received intraperitoneal injection of 50 mg/kg streptozotocin for 5 days to establish a type 1 diabetes mel itus model, and the control group received intraperitoneal injection of citrate buffer. RESULTS AND CONCLUSION: At 3 weeks after modeling, the micro-CT technique was used to observe the three-dimensional structure of the mandibles in the two groups. The quantitative analysis on the microstructure of cancel ous bone and cortical bone showed that the bone mineral density, bone volume fraction, trabecular number and trabecular thickness of cancel ous bone in the interest region in the mandible of type 1 diabetes mel itus mice were significantly decreased when compared with that in the control group (P < 0.01, P < 0.05), while the structure model index was increased significantly (P < 0.05); the mineral density and area of cortical bone were decreased in the diabetes mel itus group (P < 0.05). Hematoxylin-eosin staining showed that the number and volume of mandibular trabeculae of type 1 diabetes mel itus mice were decreased. The results suggest that the three-dimensional structure of the cancel ous bone and cortical bone in the streptozotocin-induced type 1 diabetes mel itus mice are changed significantly, and the microstructure change of the cancel ous bone is more obvious.
4.Development of Medical Box for Debriding and Change Dressings in Sanitary Train with Application Module Theory

Objective To establish a kind of medical box for wound debriding and change dressing in sanitary train,which could be used to satisfy the requirements of natural and manmade disasters for medical service.Methods With tri-grade modularization theory,the medical box were divided into four functional sections,including disinfection cleaning solution section,debriding and change dressings common section,contaminated wound change dressings section and infected wound change dressing section.Through the four sections,the medical box could be provided 5 wound debirding and 50 change dressings.Results The medical box can satisfy all requirements for wound treatment with useful and convenient function.Conclusions The invented medical urgent box has so many advantages of modularization,complete functions and expansionary,which could satisfy all requirements for sanitary train.
5.Effects of bisphosphonates on the expression of Bcl-2, Bax and Caspase-3 in condylar cartilage of osteoporosis rats

RESULTS AND CONCLUSION:(1)The number of apoptotic cels in rat condylar cartilage and subcondylar region: the sham operation group < the treatment group < the model group (alP< 0.05). (2)Expression of Bcl-2: The trend of the model group was lower than that in the sham operation group, although there was no statisticaly significant difference between the two groups; Bcl-2 expression in the treatment group was statisticaly higher compared to the model group (P< 0.05).(3)Expression of Bax and Caspase-3: The expression levels of Bax and Caspase-3 were higher in the model group than in the sham operation group (alP< 0.05), while Bax and Caspase-3 expression was lower in the treatment group than that in the model group (alP< 0.05). The results suggested that bisphophonates can regulate apoptosis in condylar cartilage from osteoporosis rats by changing the expression of Bcl-2, Bax and Caspase-3.
6.Effects of bisphosphonate on dental mineralization protein 1 expression in condylar cartilage of osteoporotic rats

Objective To examine the distribution and expression of dentin matrix protein1 ( DMP1 ) in the condylar cartilage and subchondral bone of osteoporosis rats. Methods Female Sprague-Dawley rats aged 6 months (n=30)wererandomlydividedinto3groups. TheShamgroupunderwentshamoperationonly(n=10),theOVX group ( n = 10 ) received a bilateral ovariectomy first and then saline solution treatment subcutaneously for 3 months. The RIS group ( n=10 ) also received a bilateral ovariectomy and then with risedronate treatment ( 2. 4μg/kg) subcutaneously for 3 months. Three months after the operation, the animals were sacrificed. Toluidine blue staining showed the structure changes of rat condylar cartilage region. The changes of osteoclasts in the bony subcondylar region were evaluated by tartrate-resistant acid phosphatase ( TRAP) staining. The expression of DMP1 was analyzed immunohistochemically and then performed by semi-quantitative imaging analyses. Results Toluidine blue staining showed a thickened hypertrophic layers of condylar cartilage in RIS group. The results of TRAP staining indicated that the number of osteoclasts was significantly greater in OVX group than RIS group (P<0. 05). Immunohistochemistry showed that DMP1 localized mainly in the chondrogenic layers and osteocytes, bony subcondylar region in three groups. The expression levels of DMP1 proteins statistically decreased in OVX group than the other two groups(both P<0. 05). Conclusion Bisphophonates may reduce the the number of osteoclasts in the condyle from osteoporosis rats, with increasing of the expression of DMP1, which may influences condylar cartilage biomineralization.
7.The feasibility study of Ion Proton semiconductor sequencing platform in the non-invasive prenatal genetic diagnosis

Objective To evaluate the feasibility of apply Ion Proton semiconductor sequencing platform in non-invasive prenatal genetic diagnosis .Methods Totally 1 000 pregnant women with a singleton pregnancy of 12-32 weeks gestation were selected from the Third affiliated Hospital of Zhengzhou University from Jan to Dec 2013.Using noninvasive prenatal genetic diagnosis based on Ion Proton semiconductor sequencing platform to study their cffDNA .In parallel, 72 pregnant women received invasive prenatal diagnosis by traditional chromosomal analysis with amniocentesis chorionic villus sampling .Results It′s shown that 18 out of 1 000 (1.8%) pregnant women underwent the noninvasive prenatal genetic testing had a high risk for aneuploid chromosomes , including 7 cases of 21-trisomy, 4 cases of 18-trisomy, 2 cases of 13-trisomy, 4 cases of sex chromosomal abnormality , and 1 case of 15-trisomy.It demonstrated that the rate and accuracy of fetal 21-trisomy, 13-trisomy and 18-trisomy by non-invasive prenatal genetic testing were both 100%without misdiagnosis , the rate of detection for sex chromosomal abnormality was 2/2 with a false positive rate of 1/3.However, the 15-trisomy predicted by the non-invasive prenatal diagnosis in a woman was finally proved to be a false positive .Based on the results by karyotyping (55/55) as well as follow-ups (493/493), the specificity of the non-invasive prenatal diagnosis for detection of 21-trisomy, 18-trisomy and 13-trisomy was 100%.One Ion PITM chip could detect 12 to 15 samples in 1.5 h and the whole process of noninvasive detection could be completed in 1 to 1.5 days.Conclusions The non-invasive prenatal diagnosis by Ion Proton semiconductor sequencing platform could provide fast and accurate detection of fetal aneuploidy .The benchtop high-throughput sequencing platform has laid the foundation for the independent application in clinical settings for fetal aneuploidy detection .
8.Feasibility analysis of independent extracorporeal cardiopulmonary resuscitation treatment for adult cardiac arrest in county-level hospitals

Objective:To summarize the experience and outcomes of independent extracorporeal membrane oxygenation (ECMO) assistance for adult cardiac arrest patients conducted by a county-level hospital.Methods:Clinical data of 23 adult cardiac arrest patients treated with extracorporeal cardiopulmonary resuscitation (ECPR) at Tiantai County People's Hospital from January 2020 to October 2023 were retrospectively reviewed. Data, including initial cardiac rhythm, ECMO initiation time, cardiopulmonary resuscitation (CA)-Pump On time, ECMO initiation-Pump On time, ECMO cannulation-Pump On time, complications, neurological function prognosis, mortality rate, and survival rate, were collected and analyzed. Collect and analyze the mortality and survival rates of 33 adult cardiac arrest (CA) patients meeting the criteria for extracorporeal cardiopulmonary resuscitation (ECPR) but receiving only conventional cardiopulmonary resuscitation (CCPR) from January 2020 to October 2023, and compare these rates with those of patients who underwent CA-ECPR.Results:Among the 23 cardiac arrest patients, 16 patients achieved spontaneous heart rhythm recovery, 15 patients experienced death, and 8 patients showed improved conditions upon discharge, with 6 patients exhibiting good neurological function prognosis. Compared to CA-CCPR, patients who received CA-ECPR showed a significant decrease in mortality rate (65.21% vs. 90.91%, P=0.017) and a significant increase in survival rate (34.78% vs. 9.09%, P=0.017). After gradual optimization of the ECPR process, the 2022-2023 group showed a significantly increased survival rate compared to the 2020-2021 group(46% vs. 20%). ECMO initiation-Pump On time [41( IQR36.5-44.5)min vs.43( IQR32.75-58.5)min, P=0.709] and ECMO cannulation-Pump On time[30( IQR24.0-37.0)min vs. 33( IQR27.25-55.00)min, P=0.575] decreased, although the differences between the two groups were not statistically significant. In the comparison between survival and death groups, the proportion of initial shockable rhythm was significantly higher in the survival group (75% vs. 20%). CA-Pump On time [61( IQR49.25-69.25)min vs.69( IQR58.0-89.0)min, P=0.287]and ECMO initiation-Pump On time[39( IQR29.25-51.75)min vs.43( IQR34.0-52.0)min, P=0.539] were lower in the survival group, but the differences were not statistically significant. Conclusions:Independent implementation of ECPR for adult cardiac arrest at the county-level primary hospital improves the success rate of resuscitation and enhances patient prognosis. The promotion of ECPR rescue technology in county hospitals is feasible and significant, benefiting a larger population of cardiac arrest patients.

A lot of evidence have proved that low level of cardiopulmonary fitness (i.e.functional capacity ) is related to high risk of cardiovascular diseases (CVD) ,high all‐cause mortality and high mortality of cancer .Cardiopulmonary fitness low level combined traditional risk factors can improve risk prediction for CVD .The present article aimed at reviewing cur‐rent knowledge about correlation among CVD ,other diseases and cardiopulmonary fitness ,improving people's consciousness for impact of cardiopulmonary fitness on risk prediction in order to optimize prevention and treatment of diseases .
10.Screening for scoliosis and associated factors among primary and middle school students in Shanghai

Objective:
To understand the prevalence and influencing factors of scoliosis among primary and middle school students in Shanghai, so as to provide reference for the prevention and control strategies of scoliosis in children and adolescents.
Methods:
From September to December 2021, a total of 4 531 students from grade four of primary school to grade three of junior high school from 16 primary and middle schools in 4 districts of Shanghai were selected by multi stage stratified cluster sampling to carry out scoliosis screening and questionnaire survey. Independent sample t-test, Chi-squared test, Fisher s exact probability method and nonparametric Mann Whitney U-test were used for statistical analysis, and multivariate Logistic regression was used to analyze the influencing factors of scoliosis.
Results:
The on the spot detection rate of scoliosis abnormality was 7.4% and the prevalence was 2.2%, mainly for thoracolumbar scoliosis (60.4%) and the scoliosis angle of 10° -<20°(88.1%). The results showed that the detection rate of scoliosis in boys was 1.6% and in girls was 3.3%, with a statistically significant difference ( χ 2=11.84, P < 0.01). There were significant differences in weight and BMI between scoliosis students and students with negative initial screening( t = -3.77, -5.30, P <0.01). And there was a statistically significant difference in the detection rate of scoliosis with different frequency of classroom seating arrangements ( Z =2.02, P <0.05). The results of multivariate Logistic regression showed that the risk of scoliosis in girls was higher than that in boys ( OR =1.87, 95% CI =1.21-2.88). BMI ( OR =0.89, 95% CI =0.83-0.95) and frequency of classroom seating arrangements ( OR =0.49, 95% CI =0.25-0.93) were correlated with lower rate of scoliosis. After stratification by sex, the frequency of classroom seating arrangements ( OR =0.26, 95% CI =0.10-0.67) in boys and the educational stage ( OR =2.35, 95% CI =1.26-4.41) in girls ( P <0.05) was correlated with higher rate of scoliosis, while BMI ( OR =0.86, 95% CI =0.79-0.94) in girls were correlated with lower rate of scoliosis.
Conclusion
Regular screening of scoliosis is an effective method for early detection of scoliosis. Girls in junior high school and low BMI should be paid close attention to. According to the occurrence and influencing factors of scoliosis, comprehensive prevention and control based on family, school, society, and other aspects can be carried out.
