1.Activity of plasma lecithin cholesterol acyltransferase and its lipid metabolism in patients with ischemic cerebrovascular disease

BACKGROUND: Abnormal lipid metabolism is one of the risk factors in patients with ischemic cerebral disorders, and is correlated with the changes of lecithin cholesterol acyltransferase activity.OBJECTIVE: To observe the relationship between the changes of lecithin cholesterol acyltransferase activity and lipid content in red blood cell membrane.DESIGN: A case-control study(experimental group with control as standard level).SETTING: Department of clinical laboratory, emergency room and department of neurology of a hospital affiliated to a medical college of a university.PARTICIPANTS: Totally 105 inpatients and outpatients with cerebrovascular diseases were selected from the Department of Neurology, Affiliated Hospital of Medical College of Qingdao University, from March 2002 to December 2003. They accorded with the Diagnostic Criteria set at the Second National Conference on Cerebrovascular Diseases. A total of 42 patients with cerebral arteriosclerosis and 63 patients with cerebral infarction were selected as patients group consisting of 67 males and 38 females. Another 65 healthy people receiving physical examination in the hospital, 36 males and 29 females, were selected as control group.METHODS: Venous blood of 8 mL was drawn from the participants on an empty stomach. We assayed the activity of lecithin cholesterol acyltransferase,high density lipoprotein cholesterol, low density lipoprotein cholesterol,apolipoprotein A1 and apolipoprotein B. Red blood cell membrane cholesterol was determined by phthalyl aldehyde-acetometry and red blood cell membrane phospholipid was determined by chemical quantitative analysis.MAIN OUTCOME MEASURES: Changes of lecithin cholesterol acyltransferase activity and lipid content in red blood cell membrane in patients group and control group.RESULTS: According to intention analysis, all the 105 patients in patients group and 65 patients in control group entered the results analysis. Activity of lecithin cholesterol acyltransferase: Activity changes in cerebral arteriosclerosis group and cerebral infarction group were obvious lower than those in control group[(2.14±0.72) kat/L, (2.06±0.80) kat/L, and(2.61± 0. 74) kat/L, P < 0.01 ] . Level of high density lipoprotein cholesterol and apolipoprotein A1: The level in cerebral arteriosclerosis group and cerebral infarction group was obvious lower than that in control group[ (1.32±0.33) mmol/L, (1.37±0.33) g/L, (1.28±0.33) mmol/L; (1.27±0.31) g/L, (1.60±0.43) mmol/L, (1.60±0.43) g/L, t=2.72 to 5.01, P < 0.01 ]. Content of low density lipoprotein cholesterol and red blood cell membrane-cholesterol: The content in cerebral arteriosclerosis group and cerebral infarction group was obvious higher than that in control group [ (2.94 ± 0. 82) mmol/L, (0.63 ±0.05) mmol/g, (3.02 ±0.79) mmol/L;(0.60 ±0.07) mmol/g, (2.56 ±0. 58) mmol/L, (0.57 ±0.05) mmol/g, P < 0. 01 ] . Moreover, the activity of lecithin cholesterol acyltransferase was positively correlated with high density lipoprotein cholesterol and apolipoprotein A1(r=0.247, P <0.05; r=0.303, P <0.01), but was negatively correlated with low density lipoprotein cholesterol and red blood cell membrane cholesterol(r= -0.212, P <0.05;r= -0.346, P <0.01).CONCLUSION: In patients with ischemic cerebral disorders, the major change of plasma lipid is the decrease of lecithin cholesterol acyltransferase,but it is not secondary to cerebral infarction. The activity of lecithin cholesterol acyltransferase is positively correlated with high density lipoprotein cholesterol and apolipoprotein A1, but is negatively correlated with low density lipoprotein cholesterol and red blood cell membrane cholesterol.
2.Development of Rapidly-fixed Evacuation Stretcher

Objective To develop a suit of rapidly-fixed evacuation stretcher for safe and convenient transportation of the wounded. Methods By making use of the expansion-shaping property of a bubble-shaped material produced by the chemical reaction of two kinds of Macromolecule materials, and the restrictive effect of body shape matching cotton fabric fixation-bag, a new type of stretcher was developed. Results It was proved by the maneuver and all kinds of traffic accidents that the field stretcher could provide dependable safeguard for the wounded, and the effect was very well. It was suitable to carry out the second or the third treatment and other transportation schemes. Conclusion This new type of field stretcher is light, portable and easily-operated, and besides, it can be used repeatedly.
3.Research advances on pathogenic mechanisms of HBV-related intrahepatic cholangiocarcinoma

Intrahepatic cholangiocarcinoma is the second most common intrahepatic primary liver tumor after hepatocellular carcinoma (HCC).Epidemiological study suggests a strong correlation between HBV infection and ICC development.This review focused on the potential mechanisms of HBV-induced ICC and gives a primary summary of suggested hypothesis,which included:(1) HBV infection of liver stem/progenitor cells will indirectly lead to HBV infection of intrahepatic biliary epithelial cells and lead to the development of ICC; (2) the changed microenvironment of intrahepatic biliary epithelial cells by HBV infection eventually results in carcinogenesis ; (3) the HBV infection of hepatic sten/progenitor cell can transform into tumor-like stem cells and ultimately differentiate into ICC-like tumor cells.

AIM To investigate the effects of Tea Po lyp henols on induction of apoptosis in human liver cancer cell etc. Telomerase acti vity of BEL-7404 cells was dramatically declined during the cell incuba tion with Tea Polyphenols. METHODS MTT colorimetric assay and trypan b lue exclusion method were used to examine the growth inhibition ofTea Polyphenols on BEL-7404 cells. Telomerase activity in cell was determined by PCR-ELISA method. Cell morphology and DNA gel electrophoresis were used to observe the apoptosis. RESULTS The solution of Tea Polyphenols showed dose-dependent and time-dependent inh ibitory effects on the proliferation on BEL-7404 cell line. After treatment wit h 0 2 and 0 1 g?L -1 tea polyphenols for 48 hours, cells displayed DNA ladder bands and typical morphological change of apoptosis included: ce ll shrinkage, membrane blebbing, chromatin condensation, nuclear fragmentation etc. Telomerase activit y of BEL-7404 cells was dramatically declined during the cell incubation with Tea Polyphenols. CONCLUSION Tea Polyphenols has growth inhibiti ng effect and may induce apoptosis in BEL-7404 in a certain dose range. The ant icancer mechanism of Tea Polyphenols might be related to the inhibition of telom erase activity.
5.Development and application of a new orthopedic fixing material

The development of orthopedic fixing materials is a most basic and active aspect in orthopedic injury treatment.Their invention and application of early manual splints,gypsum bangs,hot-shaped splints,polymeric bangs and so on,greatly promoted orthopedic fixing performance and varied the strategies of orthopedic injury treatment.By repeated experiments and clinical trials,we developed a new orthopedic fixing material using polymeric materials.Convenient to operate,easy to remove,auto-shaped,X ray transmittable,water proof and durable,it not only satisfies the needs of fixing different parts of the body,but also remedies the defects of earlier fixing materials,and therefore has gained an extensive application as the best fixing material for the treatment of clinical and battle-field bone fractures.Further studies on it will surely better its properties,reduce its cost and lead it to a wider application.
6.Inhibitive effect of matrine modification X on the growth of human nasopharyngeal carcinoma CNE2 cell xenografts in nude mice.

OBJECTIVE:
To evaluate the inhibitive effect of matrine modification X on the growth of human nasopharyngeal carcinoma CNE2 cell xenografts in nude mice.
METHOD:
Tumor model was established by subcutaneous inoculation of nasopharyngeal carcinoma cell CNE2 into nude mice, which was used to evaluate the antitumor effect of matrine modification X in vivo. The expression levels of Bax, Bcl-2, Caspase3 were detected by real-time PCR and western blot.
RESULT:
The growth of xenografts in nude mice was significantly suppressed after application of matrine modification X in a dose-dependent manner. The inhibition rates were 32.55% and 44.89% when treated at medium and high dose respectively. Real-time fluorescence quantitative-PCR and Western Blot results showed that the expression of Bax and Caspase3 increased, while the expression of Bcl-2 decreased in a dose-dependent manner. The change of high dose group was obvious, and the difference was statistically significant (P < 0.05).
CONCLUSION
Matrine modification X could significantly inhibit the growth of human nasopharyngeal carcinoma CNE2 cell xenografts in nude mice, probably by inducing the apoptosis of nasopharyngeal carcinoma cells, and the possible mechanism is related to regulating the expression level of Bax/Bcl-2 and Casepase3.

7.Diagnosis and treatment of insertion obstacle(report of 32 cases).

OBJECTIVETo point out and define "insertion obstacle" (IO) as another sort of sexual dysfunction.
METHODSA retrospective review was done on 32 patients with IO. They were diagnosed by self-rating insertion scale(SIS) designed by reference to the first five questions in OLeary's "Brief Male Sexual Function Inventory for Urology" (1995), involving sexual desire and erection and insertion, and treated mainly by psychological behavior therapy.
RESULTSOf the 32 cases, 30 patients were diagnosed as IO, 29 (90.6%) were simple psychological IO, and successfully cured by psychological behavior therapy.
CONCLUSIONSIO, as a sort of sexual dysfunction, could be diagnosed by SIS, and cured by psychological behavior therapy.

8.The SLC22A5 genetic analysis in Chinese patients with systemic primary carnitine deficiency

Objective To investigate the clinical and biochemical metabolic features of 12 patients with systemic primary carnitine deficiency(CDSP) and to identify the SLC22A5 gene mutation types of the disease. Method The clinical and biochemical data were collected by retrospective analysis. DNA direct sequencing and multiplex ligation dependent probe amplification(MLPA)were applied for SLC22A5 gene analysis. Result Among 12 patients with CDSP, 3 cases had evident infection factors, 6 cases with convulsions, 5 cases manifested liver hypertrophy, 8 cases with hyperammonemia, and 9 cases showed myocardial damage. All CDSP patients were detected biallelic pathogenic mutation in SLC22A5 gene by direct sequencing. The gene types include IVS2+1G>T, c.3G>T(p.Met1Ile), c.760C>T(p.Arg254X), c.1400C>G(p.Ser467Cys), c.844dupc(p.Arg282fs), c.338G>A(p.Cys113Tyr), c.51C>G(p.Phe17Leu), c.659A>T(p.Glu220Val), and c.1365dupC(p.Thr456fs). c.659A>T(p.Glu220Val) and c.1365dupC(p.Thr456fs)are novel mutations. One female patient was maternal CDSP, her child had abnormal newborn screening. The allele frequency of c.760C>T(p.Arg254X) and c.1400C>G(p.Ser467Cys) were 37.5%(9/24)and 29.2%(7/24)respectively. The MLPA test results of all patients were negative. Conclusion The clinical manifestations are complex and various in patients with CDSP. Point and small InDel(insertions/deletions)mutation constitute the major alteration in SLC22A5 gene. c.1400C>G(p.Ser467Cys) might be another prevalence mutation type in Chinese CDSP patient.
9.Influence of nursing intervention based on "Timing It Right" on heart function and emotion in elder patients with chronic heart failure

Objective To observe the influence of nursing intervention on heart function and emotion in elder patients with chronic heart failure(CHF)based on "Timing It Right".Methods A total of 100 elder patient with CHF in Nanyang City Center Hospital from January 2016 to January 2017 were recruited,and divided into control group and observation group by random number table method,with 50 cases in each group.The control group received routine care and follow-up,while the observation group received routine care and nursing intervention base on "Timing It Right" for 12 weeks.The heart function and emotion were compared between those two group at before and after nursing intervention by LVEF%,NT-proBNP,heart output and Chinese version of Brief Profile of Mood States(BPOMS).Results The patients' heart function and emotion of both groups improved after intervention.After intervention,the LVEF%,NT-proBNP and heart output of the observation group were(59.65±2.29)%,(249.21±58.79)ng/L and(5.72±1.10)L/min,the differences compared with the control group were statistically significant(t=-28.473,8.473,-5.505;P < 0.05).The total score of BPOMS of the control group and the observation group was(25.55±4.41)and(22.62±5.40)respectively(t=2.878,P<0.05).Conclusions Nursing intervention based on "Timing It Right" can improve patient's heart function and emotion in elderly patients with CHF,which has better effect then routine nursing intervention.
10.Research and investigation in germplasm resource of Dendrobium chrysotoxum.

Collected and preserved germplasm resource of Dendrobium chrysotoxum to lay the foundation for screening fine germplasm. Through refering literatures, visiting and field survey to investigate the distribution, botanic characters and apply status of D. chrysotoxum, furthermore to collect the germplasm resource. The result show that wild germplasm resource of D. chrysotoxum has obvious differences in stem characters, leaf shape as well as flower color aspects. In addition, in recent ten years, the reserves of D. chrysotoxum germplasm resource seriously descended. Through this study, we can draw a conclusion that D. chrysotoxum germplasm resource exist diversity in biology. In these germplasm resource, there are high yield and good quality variety.
