1.The association of ND-1 gene point mutations of mit ochondrial DNA with type 2 diabetes mellitus

In type 2 diabetic patients the f re quencies of point mutations 3316G→A and 3394T→C in mitochondrial DNA (mt DNA) were 3.9% (6/152) and 5.3% (8/152) respectively, which were significantly higher than those in normal controls and patients with coronary heart disease. This fi nding suggests that mtDNA 3316G→A and 3394T→G mutations are related to type 2 diabetes.

3.Predictive effects of learning burnout on positive and negative mental health in college students

Objective:To explore the predictive role of learning burnout on positive and negative mental in college students.Methods:By convenience sampling method,786 college students were selected.They were as-sessed with the Learning Burnout Scale of Undergraduates (LBUS;including three factors:dejection,improper be-havior and reduced personal accomplishment)to investigate the learning burnout,the Mental Health Continuum Short Form (adult version)(MHC-SF;reflecting positive mental health states:languishing,moderately mentally healthy and flourishing)to investigate positive mental health,and the General Health Questionnaire-28 (GHQ-28;dividing negative mental health levels:pathological one or not)to investigate negative mental health.Results:The total scores and three factors scores of LBUS were negatively correlated with MHC-SF scores (r = -0.48 --0.14,Ps <0.01).The total scores and sub-scores of dejection and improper behavior were positively correlated with GHQ-28 scores (r =0.14 -0.41,Ps <0.01 ).The results of discriminate analysis showed that the learning burnout could predict the positive mental health,the correct prediction rates of three mental health symptoms,lan- guishing,moderately mentally healthy and flourishing were 63.2%,51.2%,and 64.1%,respectively.The total forecast was at a rate of 56.4%,and three factors of learning burnout had the contribution to predict positive psy-chological health.Logistic regression analysis showed that learning burnout of undergraduates could positively pre-dict negative psychological health,the pathological and non-pathological rate of correct prediction were 67.5% and 55.9%,and the total correct prediction rate was 62.0%.Depression could positively predict negative mental health (OR =1.11).Conclusion:Learning burnout could effectively predict both positive and negative mental health in college students.

5.The prognosis effect of different Helicobacter pylori eradication methods in patients with chronic gastritis

Objective To compare the prognosis effect of different Helicobacter pylori (Hp)eradication methods in patients with chronic gastritis.Methods One hundred and twenty patients with chronic gastritis diagnosed by gastroscopy and pathology examination were divided into sequential therapy group (36 cases),triple-combined therapy group (34 cases),Hp positive without eradication group (30cases) and Hp negative group (20 cases) according to the Hp infection status.All patients were followed up by gastroscopy,biopsy and rapid urease test before and after therapy.Results The Hp eradication rate in sequential therapy group was 94.44% (34/36),in triple-combined therapy group was 73.53% (25/34),and there was statistical difference (x2 =5.775,P =0.016).The symptoms scores and gastroscopy scores after therapy in the four groups were significantly lower than those before therapy,and there were statistical differences (P < 0.05).The symptoms scores and gastroscopy scores after therapy in sequential therapy group were significantly lower than those in triple-combined therapy group,and there were statistical differences (P <0.05).The symptoms scores and gastroscopy scores after therapy in sequential therapy group,triple-combined therapy group and Hp negative group were significantly lower than those in Hp positive without eradication group [(0.84 ± 0.60),(1.34 ± 0.59),(1.49 ± 0.62) scores vs.(2.98 ± 0.54) scores,(0.47 ± 0.37),(0.83 ± 0.35),(0.96 ± 0.75) scores vs.(1.22 ± 0.40) scores],and there were statistical differences (P < 0.05).Conclusions The Hp eradication rate of sequential therapy is higher than that of triple-combined therapy.The two therapy methods can both improve the symptoms score and gastroscopy score,but the symptoms after sequential therapy are relieved faster compared with triple-combined therapy.

7.Human mesenchymal stem cells differentiate into neuron-like cells with bFGF

Objective To investigate the differentiation from human mesenchymal stem cells (hMSC) into neuron-like cells. Methods hMSC were separated from rib marrow with Ficoll-Paque reagent and expanded in culture medium. hMSC were induced to differentiate into neurons with DMEM/BHA/DMSO or DMEM/monothioglycerol, respectively. Neuron-specific enolase (NSE), neurofilament (NF), nestin, glial fibrillary acidic protein (GFAP) were detected by immunohistochemistry. Results hMSC were expanded to be undifferentiated cells in culture for more than 10 passages. The isolated and cultured MSC comprised a single phenotypic population and displayed a fibroblast-like morphology. Simple method induced hMSC exhibiting a neuronal phenotype, with a positive expression of NSE, NF-M and nestin at 5 hours. But the neuron-like cells did not express the glial astrocyte marker GFAP. Conclusion It suggests that hMSC can be differentiated into neurons in vitro .

Objective To study the effects of puerarin on advanced glycation end products (AGEs) and expression of monocyte chemoattractant protein-1 (MCP-1) in diabetic rats. Methods The rats were randomly divided into normal control rats (CON rats), diabetic rats (DM rats), diabetic rats treated with aminoguanidine (AG rats), and diabetic rats treated with puerarin (PU rats). The diabetic rat model was induced by streptozotocin. The serum levels of AGEs and MCP-1 were quantified by fluorescence spectroscopy and ELISA respectively. Tissue sections with PAS staining and electronic microscopy were used for observation of the pathologic changes in renal tissues. Objective To study the effects of static magnetic field (SMF) on development of rat embryonic spinal cord neurons. Methods Primary cultured embryonic spinal cord neurons of Wistar rat were exposed to 1.0, 10.0, 50.0, 100.0 and 200.0 mT static magnetic field. The morphological structure, cell's differentiation and proliferation of the embryonic spinal cord neurons were observed and the contents of MDA, superoxide dismutase (SOD) and protein contents in the neurons were determined. Results Static magnetic field at density of 50-200.0 mT could inhibit the differentiation and proliferation of the cells and the phenomena such as cell aggregation, detouchment of some cells, decrease of clone-formation rate and the size of the cells were observed. The contents of MDA in the cells were increased, while the activities of SOD and the level of protein were decreased. Conclusion Static magnetic field might damage the development of embryonic spinal cord neurons by enhancing the lipid peroxidation.
10.Relationship between polymorphism of N-acetyltransferase 2 gene and susceptibility to bladder cancer

Objective To investigate the relationship between genetic polymorphism of N-acetyltransferase 2(NAT2) and susceptibility to bladder cancer.Methods Based on case-control study,NAT2 mutation alleles(NAT2*5,*6 and*7) were determined by ASPCR and PCR-RFLP in 78 patients with bladder cancer and 80 nontumorous patients.In addtion,the relationships between the genotypes and tobacco smoking,occupational exposure,high dose intake of meat or pathological characteristic of bladder cancer patients were analyzed.Results In the blood samples from 158 cases,the 4 alleles NAT2*4,NAT2*5,NAT2*6 and NAT2*7 were detected.The frequency of NAT2 slow genotypes was 29.5%(23/78) in patients with bladder cancer,which was significantly higher compared with 16.3%(13/80) in control patients(P
