1.Effect of Static Magnetic Fields on Polychromatic Erythrocyte Micronucleus, Blood Cells and Brain Tissue Lipid Peroxidation of Mouse

Objective To study the effect of static magnetic fields on the micronucleus in polychromatic erythrocytes(PCE) in the bone marrow and and the lipid peroxidation in the brain tissue in mice.Methods Fourty Kunming mice were randomly divided in to the negative control group,positive control group(cyclophosphamide:145 mg/kg) and the static magnetic fields(20,40,50,60,80 and 100 mT) exposure group,exposed twice a day,for two hours each time.After 15 days,the micronuclei rate of the bone marrow,blood cells and the activity of superoxide dismutase(SOD),the protein malonaldehyde(MDA) were determined.Results Compared with the control,the micronuclei rate in 50,60,80 and 100 mT exposure group increased significantly,the amount of leukocyte significantly decreased,the activity of SOD in the brain tissue in 60,80 and 100 mT exposure group decreased and the content of MDA increased significantly.Conclusion The present experiment demonstrats that static magnetic fields exposure at doses of 50,60,80 and 100 mT may induce the micronuclei rate increase,the amount of leukocyte decrease,the activity of SOD in the brain tissue decrease and the content of MDA increase.
2.hMLH1 promoter hypermethylation correlates with platinum-resistance in non-small cell lung cancer cells

Background and purpose:hMLH1 promoter methylation is one of the factors in the development of lung cancer.However,little is known about hMLH1 promoter methylation in association with chemotherapy resistance in lung cancer cells.This study aimed to investigate whether resistance to DDP can be overcame by restoring the expression of hMLH1 in A549/DDP cells after treatment with a demethylating agent,5-Aza-2'-deoxycytidine (5-Aza-CdR).Methods:hMLH1 mRNA was determined by RT-PCR in A549,A549/DDP and A549/DDP treated with 5-Aza-CdR;MSP was carried out to determine the methylation status of hMLH1 in A549 and A549/DDP,A549/DDP treated with 5-Aza-CdR.MTT test,Hoechst 33258 staining and flow cytometry were used to determine the change of cell proliferation,apoptotic morphology and the cell apoptosis index,respectively.Results:hMLH1 mRNA was expressed higher in A549 than in A549/DDP.The unmethylation status of hMLH1 was observed in A549 cells whereas partial methylation status of hMLH1 in A549/DDP cells.After treatment with 5-Aza-CdR,the A549/DDP cells displayed an unmethylation of the hMLH1 gene.After the treatment with cisplatin,the IC50 of A549,A549/DDP and A549/DDP treated by 5-Aza-CdR were (4.7?0.7),(30.1?1.8) and (6.9?0.6)?mol/L,respectively.Apoptotic bodies were less in A549/DDP cells than in A549/DDP cells treated with the 5-Aza-CdR.The rate of apoptosis of A549/DDP treated by 5-Aza-CdR was higher than that of A549/DDP.Conclusion:The methylation of hMLH1 may be relevant to the resistance to platinum-based chemotherapy in the A549/DDP cell line.5-Aza-CdR can inhibit hMLH1 methylation,restore the expression of hMLH1,and enhance cell apoptosis as well as the inhibition of cell proliferation of A549/DDP cells after the treatment of cisplatin.
3.Establishment of the assessment system for blood transfusion and discussions on the method of clinical transfusion monitor

Objective Discuss the methodology and significance of clinical transfusion assessment,establish an effective evaluation system for blood transfusion,identify rational indexes for the evaluation,and promote quality of clinical transfusion.Methods Development of the clinical blood transfusion assessment regulations,tightened blood transfusion approval system,enhanced medical record check for blood transfusion,better statistics for the data collected,and analysis of data in 2012 to compare changes in blood volume before and after tighter management in place.These efforts aim at improving the assessment system for optimal clinical blood transfusion.Results Compared to Jan.-June in 2012,patients discharged and surgical cases in Dec.of the same year dropped 2.15% and 0.73% respectively.However,the volume of blood transfusion decreased 22.7%,the percentage of blood transfusion for inpatients decreased from 8.78% to 7.17%,and the average use of blood for inpatients decreased from 0.73U to 0.57U.Conclusion Reasonable and scientific assessment for blood transfusion and better clinical blood use management can improve blood transfusion therapeutic efficacy and save blood resources.
4.Analysis on the activity of the dendritic cells stimulated with Coxiella burnetii antigens

Objective To investigate the change of surface molecules and cytokine expressions of dendritic cells (DC) stimulated with recombinant proteins Com1 and heat shock protein B (HspB) of Coxiella burnetii (Cb).MethodsThe DC cultured for 5 d were stimulated with 20 μg/mL recombinant protein Com1,20μg/mL HspB or 6 μg/mL E.coli lipopolysaccharide (LPS).Twentyfour hours later,the surface mature marker,CD83,and activation-associated markers of T lymphocytes,CD58,CD54,CD40,CD80 and CD86,and expression levels of interleukin (IL)-10 and IL-12 of DC were detected by fluorescence activated cell sorter (FACS).Multiple comparisons were performed by using Student-Newman-Keuls test.ResultsCom1 could induce DC maturation efficiently in vitro.Human monocyte-derived DC exhibited significantly higher expression levels of surface molecules including CD83,CD54,CD58,CD80,CD86,and CD40,which were all >80%.CD83 expression induced by Com1 was similar with that induced by 1E.coli LPS,while CD54 and CD86 expressions were slightly higher than those induced by E.coli LPS,and expressions of other molecules were significantly higher than those induced by E.coli LPS (all P<0.05).After Com1 stimulation,intracellular IL-12 level increased to 9 % from zero.HspB could not induce DC maturation in vitro.The intracellular IL-10 level was 6% after HspB stimulation.DC pulsed with Com1 and HspB exhibited intracellular IL-12 level of zero and IL-10 level of <2%.Conclusion Com1 but not HspB can efficiently activate DC and Com1-activated DC can drive T cells toward Th1 cell development due to a high level of IL-12 production.
5.Value of heart-type fatty acid-binding protein in early diagnosis of acute myocardial infarction

Objective To explore the value of heart-type fatty acid-binding protein (H-FABP) in the early diagnosis of acute myocardial infarction (AMI). Methods Sixty confirmed AMI patients were observed, the data of H-FABP, cardiac troponin T (cTnT), creatine kinase MB (CK-MB) were detected in < 6 h and 6-12 h after the symptoms appeared, and the sensitivity of the three markers was calculated. The specificity was compared with 15 uncertain AMI patients and 45 healthy subjects. Results The early diagnosis sensitivity of H-FABP in < 6 h was 94% in AMI, which was higher than that of cTnT (50%) and CK-MB (56%) (P < 0.05 ). The diagnosis sensitivity of H-FABP, CTnT, CK-MB in 6-12 h was 100%, 92%, 92% respectively (P> 0.05 ). There was no significant difference in the specificity among the three markers (P > 0.05). Conclusions H-FABP has more sensitivity and specificity in the early diagnosis of AMI. It is applicable in the screening of patients who suffered chest pain and the diagnosis of early AMI.
6.Two cases of synovial sarcoma in the head and neck.

Synovial sarcoma is a malignant soft tissue with unknown origin. Although head and neck region is the second common site of involvement, rare cases have been reported in Para nasal sinus and larynx. We presented two cases of synovial sarcoma, one of which arised from maxillary sinus and the other from laryx, and re- view the literature to sum up the diagnosis and treatment strategies. The conclusion is that synovial sarcoma in the head and neck still raises diagnostic and therapeutic issues. Surgical excision with wide margins is essential and necessary, usually associated radiotherapy. The effect of chemotherapy remains to explored.

7.Effect of Acrylonitrile on Proliferation and Differentiation of Mouse Lung Fibroblasts in Vitro

Objective To approach the effect of acrylonitrile (ACN) on the proliferation and differentiation of the lung fibroblasts of mice. Methods The purification and primary culture of the lung fibroblasts(LFb)of new born mice were conducted. The cells were treated with ACN added in the medium at the varying doses of 0.01, 0.5, 1.0, 10.0, 50.0, 100.0 and 200.0 ?g/ml. The cytomorphological methods were used, the protein, malonaldehyde (MDA) and the activity of superoxide dismutase (SOD) were determined. Results No inhibitted effect on the lung fibroblasts was observed at the doses of 0.01-10.0 ?g/ml. At the doses of 50.0-200.0 ?g/ml, acrylonitrile could inhibit the differentiation and proliferation of the cells, the volume of cells became small,the rate of cell clusters decreased, at the doses of 10.0-200.0 ?g/ml, the content of protein and the activity of SOD decreased, MDA content increased. Conclusion Acrylonitrile can inhibit the proliferation and differentiation of the lung fibroblasts in mice, protein synthesis inhibition and lipid peroxidation are considered as the related factors.
8.Effects of epidural ropivacaine labor analgesia on duration of labor and mode of delivery

Objective To study the effects of ropivacaine on the duration of labor and mode of delivery in the primigravidas using patient-controlled epidural analgesia (PCEA). Methods Retrospective analysis was performed. The 190 healthy, full-term, and single-fetus parturient primigravidas who received PCEA with 0.1% ropivacaine+fentanyl (1 ?g/ml ) were in the epidural analgesia group. Another 222 primigravidas who didnot receive PCEA were in the control group. The duration of labor and modes of delivery, and the neonatal Apgar scores in both two groups were recorded and evaluated. Results Those in the epidural analgesia group experienced a significantly longer first stage [(426?161) minutes], longer second stage [(54?27) minutes] and longer full duration of delivery [(489?166) minutes] than those in the control one [(409?170) minutes, (364?167) minutes and (37?22) minutes]. The rate of using pitocin in the epidural analgesia group (30.2 %) was significantly higher than that in the control group (4.1%). The cesarean section rate in epidural analgesia group (20.0 %) was lower than that in the control one (28.4%); while the rate of instrumental delivery in the epidural analgesia group (20.0%) was significantly higher than that in the control one (6.3%). In summary, there were significant differences between two groups in the duration of labor, the rate of using pitocin, the rate of instrumental delivery and the rate of cesarean section. But there were no differences found for those newborn who had Apgar scores less than 7 at the point of both one and five minutes (7.9% and 4.5%, 2.6% and 0.5% respectively). Conclusion Epidural ropivacaine labor analgesia lengthens the duration of labor and increases the rate of instrumental delivery, but it has no significant negative effects on the neonates.
10.Influence of heme oxygenase-1/endogenous carbon monoxide system on atherosclerotic process in rabbit

Objective To study the changes in heme oxygenase-1/endogenous carbon monoxide system in rabbit aorta with atherosclerosis induced by high cholesterol diet, and the influence of the system on the atherosclerotic process. Methods Rabbits were given 1.5% cholesterol diet (Ch group, n=8) or 1.5% cholesterol diet plus hemin(15mg?kg~ -1 ?d~ -1 , ip., Hm group, n=8) or zincprotoporphyrin IX (Znpp-IX,45?mol?kg~ -1 ?d~ -1 , ip., Zn group, n=8).After 12 weeks,serum lipids and total bilirubin, aortic CO protduction and HO activity were assessed, the area of atherosclerotic plaques in aorta were measured by oil-red-O staining and image analysis software. Results Compared with control group (C group, n=8), aortic HO activity and CO production were higher significantly, and the aortic plaques area was (54.00?4.16)% in Ch group. Znpp-IX injection markedly decreased aortic HO-1 activity and CO production, the aortic plaques area was (61.13?3.50)%. Compared with Ch group, hemin injection increased markedly not only serum total bilirubin concentration, but also aortic HO activity and CO production, and the aortic plaques area was (17.88?3.01)%. Conclusion The HO-1/CO system may play a protective role against atherosclerosis. Enhancement of HO-1 activity is possibly the basic function of HO-1/CO system in prevention of atherosclerosis.
