1.Inducing Effects of Retinamide (RII) on Tumor Cells with Different Metastatic Ability

Newly synthesized retinoic acid derivative-retinamide (RII) was employed as differentiation inducing agent. Cultured tumor cells of epithelial origin were exposed to 10-5mol/L RII for 5 passages. These cells included mouse fore stomach carcinoma (MFC, lung metastatic rate 85%), human nasopharyngeal carcinoma (CNE-2Z, 56%), and clonal variants from CNE-2Z, CNE2L2 (100%) and CNE2L4 (13%) . The results of serial observation were as follows. The growth, clone-forming ability, motility, invasiveness of these tumor cells were obviously inhibted. Their adhesion to fibronectin and laminin was increased. Morphological ultrastructure changes, mainly of surface structure, were also observed. These changes suggested that RII made metastatic cells less aggressive and showed a tendency toward differentiation. After exposured to RII, different changes of oncogene and antioncogene expression of these tumor cells were detected. For example, RE caused expression of nm23in MFC cells, but down regulated (decreased) its expression in CNE2L2 cells and similar changes of rasH, fos, nm23, Rb. Expression were observed for CNE-2Z cells and its clonal variants CNE2L2 CNE2ZL2. RII down regulated both these oncogene and antioncogenes in both CNE-2Z and CNE2L2, but up regulated all of them in CNE2L4. The results indicated that oncogene and antioncogene may play different roles in different tumor cells, the same factor (RII) may lead to different changes of gene-expression in different metastatic tumor cells. So the function of oncogene and antioncogene may be of relativity and may be influenced by multifactors. Our data were mostly from in vitro experiments. It could not be deduced completely to a level as a whole in vivo.
2.Influence of medicative diet for food therapy on hepatic function of patients with gastric cancer and adjuvant chemotherapy after operation

Objective To approach the alleviating effect of medicative diet for food therapy on hepatic function of patients with gastric cancer and adjuvant chemotherapy after operation and supply references for effective treatment and nursing. Methods We divided 123 patients with gastric cancer after operation into the test group (65 cases) and the control group (58 cases).The control group adopted routine treatment project while the test group received medieative diet for food therapy based on routine treatment project. The changes in hepatic functions were compared between the two groups. Results The changes in hepatic functions of the test group were better than those of the control group (P<0.01 ). Conclusion Medicative diet for food therapy could significantly restore the seriously damaged hepatic function after chemotherapy.
3.Imaging features of Von Hippel-Lindau syndrome type Ⅰ

Objective To analyze the imaging characteristics of Von Hippel-Lindau Syndrome (VHL)typeⅠ.Methods 12 patients diag-nosed as VHL type Ⅰ were collected and all the patients had underwent 3.0T MRI and 64-slice spiral CT plain and enhanced scan. Imaging data was analyzed by two senior radiologists to summarize its imaging featueres.Results Multiple hemangioblastomas (HGBLs)in cerebellum,spinal cord or brainstem were seen in all the patients.Flow void effect with varied degree was seen in the lesions.Dilated feeding and drainage blood vessels around lesions was seen in 6 cases.Multiple kidney and pancreas cysts were seen in all the patients.There were 4 cases with syringomyelia,7 cases with ocular retinal angioma,5 cases with renal cell carcinoma,2 cases with ground-glass-like lesions and micronodulars in lung.Conclusion VHL type Ⅰ has complex imaging performance.Ima-ging examination is important for early detection of VHL disease.
4.Construction and efficacy identification of a lentiviral vector harboring RNAi based on gene NSBP1

Objective To construct and identify the efficacy of a lentiviral vector harboring RNAi sequence targe-ting NSBP1 gene. Methods Three siRNA targeting the NSBP1 mRNA were designed, the pGCSIL-GFP-NSBP1 lentivirus vectors were constructed and confirmed by DNA sequencing. A total of 293T cells were co-transfected with pGCSIL-GFP-NSBP1, pHelper1.0 and pHelper2.0 for the virus stocks produced, the titer of the virus was test-ed. After lentivirus transfecting into DU145 ceils, Western-blot and MTT methods were used to determine the ex-pression and biological activity of NSBP1 gene, the cells were transplanted into nude mice, then inhibitive effect was observed. Results PCR analysis and DNA sequencing demonstrated that the RNAi sequence targeting the hu-man NSBP1 gene was successfully inserted into the lentiviral vector. The titer of the recombinant]entiviral vector was 2 × 10~8TU/mL. NSBP1 protein expression level in transfected cells was significantly decreased and growth rate of cells transfected with lentivirus was decreased by MTT assay, the downregulation of NSBP1 reduced growth rate of transplantated tumor, whereas tumorgenicity was not influenced. Conclusion The construction of the]entiviral vector of NSBP1 has been successfully prepared and NSBP1 plays an important regulatory role in androgen-inde-pendent prostate cancer cell proliferation.
5.Research on Noninvasive Diagnosis for Coronary Heart Disease Based on Neural Network

Objective To extract characteristic parameters of ECG signals a new method of non-invasive diagnosis for coronary heart disease with artificial neural network. Methods ECG signals were digitized with A/D converter and filtered to eliminating the noise. Span of QRS interval, R-R interval,and voltage of S-T segment of filtered ECG were detected. These 3 characteristics were as the input parameters of the input layer. Samples were trained with an improved 3-layers back propagation(BP) artificial neural network, as trained samples. The non-trained samples were recognized with these BP neural networks. Results After 12 samples had been trained about 1500 times, the BP neural network could accurately distinguish samples of coronary heart disease from the trained samples and also recognize 20 non-trained samples, 19 to be correct except one. Conclusion It is showed that based on BP network and characteristic parameters of ECG, a new and promising method of non-invasive diagnosis for coronary heart disease has been found.
6.Color Doppler ultrasonic and clinical analysis of elderly degenerative valvular disease

By use of color Doppler ultrasonography (ATLHDI 5000,made in USA),a prospective study was made in 362 aged (greater than or equal to 60 years) patients (252 males and 110 females) for a period of 3 years.There were 176 patients (48.2%,130 males and 46 females) had degenerative vavular disease (DVD).The incidence increased with the aging of the population.There were no evident differences between males and females.Degenerative aortic valvular deformation was most frequently found (in 171 cases),followed by mitral (in 24 cases).The complications were hypertension,diabetes and coronary heart disease.It suggests that the DVD in the elderly mainly occurs in aortic valves,then in mitral value.Its incidence increases with aging.Complications such as hypertension,diabetes and coronary heart disease are important risk factors of DVD.
7.The relationship between early recurrence after radical resection and the expression of Cx43 in AFP positive and HBV related HCC tissues

Objective To study the expressions of Cx43,CD105 and VEGF in HBV related HCC tissues and the relationships between Cx43 expression and recurrence and prognosis after cancer radical resection in HCC patients stratified by serum AFP levels. Methods The expressions of Cx43,CD105,VEGF in 234 HBV related HCC tissues were examined by tissue microarray and two-step methods of PV-6000 of immunohistochemistry and the expressions of Cx43 in 20 frozen HCC specimens were examined by RT-PCR. Results Cx43 in HCC tissues was positive as examined by both immunohistochemistry and RTPCR methods.Positive Cx43 expression is correlated with lower early recurrence ( Log Rank P =0.001 ),longer disease free survival (Log Rank P =0.026 ) and overall survival( Log Rank P =0.000 ) as showed by the Kaplan-Meier analysis in patients with AFP < 400 μg/L. The expression of Cx43 is an independent prognostic factor.The positive expression of Cx43 related with lower positive expression of CD105 and VEGF (P =0.018,0.023 ),and correlated with histological differentiation (P =0.002),the number of focus (P =0.033 ),blood vessel tumor embolism ( P =0.029 ). Conclusions The expression of Cx43 is correlative with the expression of CD105 and VEGF,and is predictive of HCC early recurrence and poor prognosis after radical hepatectomy in HBV related HCC patients with serum AFP < 400 μg/L.
8.Clinical assessment of modified early warning score and SMART-COP on predicting mechanical ventilation in patients with severe influenza A H1N1

Objective To evaluate the predictive value of modified early warning score (MEWS) and SMART-COP score on mechanical ventilation in patients with severe influenza A H1N1. Methods Fifty cases diagnosed with severe influenza A H1N1 were retrospectively analyzed. The MEWS and SMART-COP score were calculated. The area under the receiver operating characteristic (ROC) curve (AUC) was evaluated using ROC curve. MEWS, SMART-COP score and AUC were analyzed by Z test. Results The AUCs of MEWS and SMART-COP score for predicting mechanical ventilation were 0. 923 and 0. 889, respectively, which were not significantly different (Z=0. 548, P =0. 584).Conclusion Both of MEWS and SMART-COP score are predictive factors of mechanical ventilation in the patients with severe influenza A H1N1.
9.Phenotype in 6 patients with mitochondrial DNA G13513A mutation

Objective To report 6 Chinese patients with mitochondrial encephalomyopathy caused by mitochondrial DNA(mtDNA)G13513A mutation and discuss the mitochondrial phenotype associated with this mutation based on the data of our patient series as well as the reports by others.Methods Direct sequencing of polymerase chain reaction(PCR)products or PCR-RFLP analysis Was performed to screen mtDNA G13513A mutation in 35 cases with mitoehondrial encephalomyopathy.who carried no mtDNA common mutations(1arge 8eale deletion,A3243G,T3271 C,A8344G,or T8993G/C).The clinical features,MRI changes were retrospectively collected and analyzed.Published studies of all patients with mtDNA G13513A mutation were also reviewed.Results Six patients were identified carrying mtDNA G13513A mutation.All patients presented stroke-like episodes with hemianopsia.hemiparesis or hemiparesthesia.Three adult patients presented clinical and radiological features of adult-onset mitochondrial myopathy,encephalopathy,lactic acidosis,and stroke-like episodes(MELAS),including stroke-like episodes,epilepsy,headache,short stature,sensorineural deafness,multifocal lesions on parietal,occipital and temporal lobes on cranial MRI scans.Three iuvenile.onset patients presented the clinical and brain MRI features of MELAS-Leigh syndrome(LS)overlap syndrome.In addition to the stroke-like episodes,they also showed brain stem lesions with dysarthria,ataxia,and ophthalmopJegia. Brain MRI revealed asymmetrical lesions in the cortex of the oecipital and temporal lobes,as well as symmetrical lesions in the bilateral basal ganglia and brainstem.Muslce biopsy showed ragged redfibem in 5 patients.The infant-onset LS or Leigh-like syndrome with mtDNA G135 13A was described in the English literature.Conclusions mtDNA G13513A mutation is a common pathogenic mutmion for mitochondrial encephalomyopathy,which can result in Leigh syndrome,MELAS-LS overlap syndrome and adult MELAS.The onset of various phenotypes is relatively age-dependent.
10.The effect of β-adrenoceptor blockers on G protein and heart function in rats with acute myocardial infarction

ObjectiveTo investigate the effect of β-adrenoceptor (β-AR) blockers on G protein and heart function changes in rats with acute myocardial infarction (AMI) MethodsWistar rats with AMI induced by left anterior descending coronary branch ligation were randomly divided into compared with sham operation group. Eight weeks after therapy, hemodynamics was assessed by inserting catheters and the level of G protein was detected by Western blot analysis. ResultsCompared with the sham operation group, systolic blood pressure(SBP), diastolic blood pressure (DBP), left ventricular systolic pressure(LVSP) and left ventrieular pressure maximal rate of rise and fall(±dp/dtmax) in AMI group were significantly decreased, while left ventricular end diastolic pressure (LVEDP) and Gs and Gi protein levels were significantly increased (all P<0.05). Compared with AMI group, LVSP and ± dp/dtmax were increased, but LVEDP and Gi protein level were significantly decreased in metoprolol and carvedilol group. LVEDP and Gi protein level were decreased in carvedilol group compared with metoprolol group. ConclusionsCarvedilol can effectively suppress the change of G protein and improve the heart function after AMI, and the effect is better than that of metoprolol. This may be related with its β2-AR blocking effect.
