Objective To obtain the specific antigens of the expressed major capsid proteins from Noroviruses with different sub-genotypes in Beijing area. Methods The full-length genes of the major capsid proteins (VP1)were obtained through the amplification of the VP1 encoding gene in the recombinant plasmids pBST-CR2987(G Ⅱ-3)and pBST-CR2932(GⅡ-4),which represented different Norovirus geno-types. The full-length genes were sub-cloned into the expression vector pET-30a(+),resulting in a recombinant plasmid, with which the BL21 competent cells were transformed, and the expression of the gene was induced by adding IPTG to the growth culture. The expression of the major capsid proteins were analyzed with Coomassie blue staining after SDS-PAGE, and assayed by Western blot with serum from human. Results (1)The major capsid protein genes of CR2987 and CR2932 were sub-cloned into expression vector pET-30a(+). The VP1 encoding genes were 1647 bp in length for CR2987 and 1623 bp for CR2932. The open reading frames(ORF)coded for 549 and 541 amino acids for these two proteins, respectively. (2)The expressed VP1s were present primarily as inclusion bodies, and the maximal amount of the expressed proteins occurred at 4-6 h after IPTG induction.(3)These VP1s could be recognized by specific immune serum against VP1 of Norovirus as well as His-tag antibody. Conclusion The VP1s of CR2987 and CR2932 are expressed in BL21 E.coli cells. The expressed VP1s could react with specific immune serum against VP1 of Norovirus, indicating that the expressed VP1s are of antigenicity.

Objective To investigate the relationship of the expression of phosphatidylinositol-3-kinase(PI3K)subunits,P85 and P110 to the pathogenesis of psoriasis. Methods Immunohistochemical staining for P85 and P110 was performed in the tissue specimens from patients with psoriasis(n=30),chronic dermatitis(n=20),seborrheic keratosis(n=20),squamous cell carcinoma(n=20),basal cell carcinoma(n=30)and normal human controls(n=10).The absorbance of immunostained tissue was quantified with image analysis system (Q550CW,Leica,Manheim,Germany).Statistical analysis was carried out by ANOVA,Results Among these groups,a significant difference was observed in the expression level of P110 in the epidermis(F=35.64,P<0.01),as well as in that of P85(F=59.98,P<0.01)and P110(F=323.23,P<0.01)in the lymphocytes infiltrating the lesion.Increased expression of P110 was found in the epidermis of psoriatic lesions compared with the lesions in the other disorders, whereas no significant difference was noticed among the other disorders. In the case of P85 and P110 expression in the lesion-infiltrating lymphocytes infiltrating the lesion, psoriasis and squamous cell carcinoma significantly differed from the other disorders, while no difference was observed between psoriasis and squamous cell carcinoma (P>0.05).Conclusions The high expression of P110 might be closely correlated to the hyperproliferation of keratinocytes;but filrther study is needed to clarify the relationship of increased expression of P85 and P110 to the activation and proliferation of lymphocytes in psoriatic lesions.

AIM: To study the mRNA and protein expression of Kang ai1 (KAI1) tumor suppressor gene and to determine the relationship between KAI1 and invasiveness and metastasis of cervical cancer. METHODS: The expression of KAI1 metastasis suppressor was detected by immunohistochemistry in paraffin slides and by real-time quantitative polymerase chain reaction (RT-PCR) in fresh tissue. The samples included 20 cases of normal cervical tissues, 20 cases of cervical intraepithelial neoplasia (CIN) and 40 cases of cervical carcinoma. The results of the gene expression combined with the pathological and clinical data were also analyzed. RESULTS: The expression of KAI1 protein and mRNA was related to the tissue differentiation of cervix. The positive rates of KAI1 expression were the highest in the normal cervical tissue, the middle in CIN and the lowest in cervical carcinoma with significant difference among three groups (P<0.01). The expression of KAI1 protein was not related with the grade of CIN (P>0.05). However, both mRNA and protein expression of KAI1 were related to the differentiation and the clinical stages of cervical cancer (P<0.01) and also related to the metastasis of the cancer. The positive rates between the non-lymphatic metastasis and lymphatic metastasis (P<0.05) were significant different. Cox regression and logistic regression showed that the tissue differentiation, clinical stages, lymphatic metastasis and expression of KAI1 were all related factors with recurrence and prognosis of cervical cancer. CONCLUSION: The down-regulation of KAI1 tumor suppressor gene at both mRNA and protein levels is related to the differentiation, clinical stages and metastasis of cervical cancer, indicating that the expression of KAI1 is a prognostic factor for cervical cancer.

Objective To investigate the role of phosphorylated extracellular signal-regulated kinase (p-ERK), mitogen-activated protein kinase (MAPK)/ERK kinase (MEK), and nuclear factor-κB (NF-κB) in the pathogenesis of psoriasis vulgaris. Methods Immunohistochemistry and Western blot were used to detect the expression of p-MEK, p-ERK and p-NF-KB in tissue samples from 30 patients with psoriasis vulgaris and 15 normal human controls. The average optical density of immunostaining and relative grey scale of immuno-bloting were calculated. Results The average optical density of immunostaining for p-MEK, p-ERK and p-NF-KB was 0.36 ± 0.03, 0.36 ± 0.04 and 0.26 ± 0.04, respectively in lesion samples of psoriasis, significantly higher than that in normal control tissue (0.22 ± 0.02, 0.18 ± 0.03 and 0.16 ± 0.03, all P < 0.01). A significant increase was also observed in the relative grey scale of p-MEK, p-ERK and p-NF-κB in psoriatic lesions compared with the normal controls (1.41 ± 0.14 vs 0.54 ± 0.10, 2.35 ± 0.34 vs 1.86 ± 0.12, 1.07 ± 0.15 vs 0.87 ± 0.08, all P < 0.01). Conclusions The expressions of p-MEK, p-ERK and p-NF-κB are enhanced in lesions of psoriasis vulgaris, and the abnormal activation of upstream and downstream molecules in the MAPK signaling pathways might be involved in the pathogenesis of psoriasis.
