Background and purpose:Hepatitis B virus X protein (HBx) and hypoxia inducible factor-1α(HIF-1α) play key roles in hepatocarcinogenesis and the development of hepatocellular carcinoma. Positive correlation on the expression of these 2 proteins in hepatocellular carcinoma tissues has been found, whereas the underlying mechanisms have not been fully elucidated. This study focused on the role of HBx in regulating HIF-1α and the underlying mechanisms in hepatocellular carcinoma cells. Methods:The expression plasmids were transfected into Huh7 cells with LipofectemineTM 2000. Western blot analysis was applied to detect the expressions of HIF-1αand HIF-1β protein. The transcriptional activity of HIF-1α was detected by the commercial analysis kits. The mRNA levels of HIF-1αand its target genes, including vascular endothelial growth factor (VEGF) and multi-drug resistance gene 1 (MDR1), were detected by quantitative real-time PCR (qRT-PCR). Immunoprecipitation analysis was applied to detect the interaction of HIF-1α, HBx and protein von Hippel-Lindau (pVHL). Results:Huh7 cells transfected with HBx plasmid led to sharp increase of HIF-1αprotein and transcriptional activity, as well as the mRNA of VEGF and MDR1 (P<0.05). However, the mRNA level of HIF-1αwas not obviously changed after HBx transfection (P>0.05). Meanwhile, HBx also signiifcantly impaired the function of pVHL in mediating the degradation of HIF-1αby ubiquitin hydrolase. This finding was further confirmed by the immunoprecipitation analysis, which showed that HBx could directly bind to pVHL, but not to HIF-1α. Conclusion:HBx may inhibit the inter-activation between pVHL and HIF-1αthrough directly binding to pVHL, and thus enhance the stability and transcriptional activity of HIF-1α.

Objective To evaluate the effects of lipid emulsion on parenteral nutrition associated liver dis ease (PNALD) in old tumor patients. Methods A retrospective study was performed with 402 patients in Renji Hospital from January 2003 to December 2008. Patients were retrieved according to the following criteria: (1)age ≥60 years; (2) confirmed diagnosis of tumor, had no evidence of metastasis, and tumor was completely resected before receiving parenteral nutrition; (3) liver and kidney function was in normal range before receiving parenteral nutrition; (4) parenteral nutrition days ≥7; and (5) parenteral nutrition was infused in "all in one" bag via central venous. Patients with history of viral hepatitis or died in parenteral nutrition episode were excluded. These 402 patients aged (71.7 ±6.8) years and the average parenteral nutrition time was (10. 2 ±5.9) (range, 7-61 )days. In 311 patients (77.4%), non-protein calorie was obtained from carbohydrate and lipid and 91 patients (22. 6% ) just obtained non-protein calorie from carbohydrate. Results The total prevalence of PNALD was 15.2% (61/402). The prevalence of PNALD was 8.8% (8/91) in patients receiving parenteral regiment without lipid and 17.0% (53/311) in patients receiving parenteral nutrition with lipid, and there was no significant difference in prevalence of PNALD between two groups (χ2 = 3.72, P = 0.07 ). Lipid type and amount showed no significant effects on PNALD ( P > 0.05 ). The fever days ( P < 0. 001 ), baseline level of alanine transaminase (P <0. 001 ) and γ-glutamyltransferase (P <0. 001 ) were risk factors for liver injury by logistic regression. Conclusion Lipid emulsion can be safely used in old tumor patients without affecting the occurrence of PNALD.

OBJECTIVE: Transplantation of stem cell, especially neural stem cell, has been proven effective in treating nervous system diseases in animal models, which provides new hopes for recovery of nerve functions in patients suffering from various nervous system diseases. Multiple studies have been conducted on the role of neural stem cells in repairing nerve functions, and several hypotheses have been proposed to explain how the stem cells or neural stem cells acts to recover nerve function.DATA SOURCES: We searched Medline for English articles from January 1997 to August 2003 with stem cell, neural stem cell, bone marrow stromal cell, stroke, ischemic injury, nervous system disease, and neurotrophic factor as the keywords. We also searched Wangfang database for Chinese articles with the same keywords from January 2003 to December 2004.STUDY SELECTION: The articles found in these two databases were primarily screened with the inclusion criteria as follow: the subjects should be animals or human; and the study should be the basic and/or clinical researches on(neural) stem cells.DATA EXTRACTION: Totally 72 English and Chinese articles were found in the two databases, among which 14 articles were closely associated with the present study and 10 were indirectly related. Eight articles were excluded for repetition in the contents. Finally 16 articles were included for analysis.DATA SYNTHESIS: The full text of the articles were reviewed and summarized. The results of these articles suggest that neural stem cells are valuable in treating cerebral vascular diseases, brain and spine injury, nerve degeneration diseases, and peripheral nerves diseases.CONCLUSION: Stem cells, especially neural stem cells, play active roles in treating nervous systeme diseases such as differentiation promotion, nerves nourishing, and nerve substitute.
