Objective To study the role of anti-HLA-IgG antibodies levels in serum before renal transplantation.Methods Anti-HLA-IgG antibodies levels were detected by means of ELISA-LATM in 184 patients before renal transplantation from Jan. 1999 to Dec 2002. Acute rejection occurrence was compared between anti-HLA-IgG class-Ⅰantibody positive group and anti-HLA-IgG class-Ⅱantibody positive group, and between both class-Ⅰand class-Ⅱantibody positive groups and anti-HLA-IgG antibody negative group, respectively. Results In the 155, 7, 9 and 13 cases respectively negative for Anti-HLA-IgG antibodies, positive for anti-HLA-IgG class-Ⅰantibodies, anti-HLA-IgG class-Ⅱantibodies and both class-Ⅰand class-Ⅱantibodies, there were 18, 2, 4, 8 cases of acute rejection with the occurrence rate being 11.6?% , 28.57?% , 44.44?% , 61.53?% , respectively ( P

Objective To explore the value of transthoracic echocardiography(TTE) and 64-slice spiral computed tomography(CT) in the diagnosis of complex congenital heart disease(CHD). Methods Ninety-seven patients diagnosed as CHD by TTE underwent 64-slice spiral CT for cardiovascular examination. The results were compared with the results by cardiovascular angiography and from surgery. Results The total diagnosis accordance rated by TEE was 90. 2% and that by spiral CT was 92.5%. There was no difference in diagnosis accuracy rate between TTE and spiral CT. The diagnosis accuracy rate in intracardiae defomities by TTE was 99.2 %, higher than 87.50% by spiral CT. However, the diagnosis accuracy rate in extracardiac defomities and ventricular-arterial connections was 99. 0% by spiral CT, higher than 78. 6% by TTE. The combination of TTE and spiral CT can raise the diagnosis accuracy rate to 99. 1%. Conclusions TTE is of significant value in complex CHD diagnosis,especially in the diagnosis of intracardiac defomities. The combination of TTE and spiral CT can raise the diagnosis accuracy rate of various kinds of complex CHD.

Objective To evaluate the effeciency and safety of the implantation of phakic iris-fixated intraocular lens for high myopia.Methods 12 cases(18 eyes)with the mean spherieal equivalent refraction was(-15.12?5.23)diopters of myopia,following up 3~6 months after surgery(averaging 5.1 months)were enrolled.Visual acuity,intraocular pressure,contrast sensitivity,glare sensitivity,corneal endothelium and the stable of intraocular lens and the turbid level of lens were observed.Results A postoperation uncorrected visual acuity of(0.73?0.21)was observed in 18 eyes at 6 months.There was significant difference as compared with preoperation corrected visual acuity of(0.52?0.17)(P

Aim To investigate the effect and the possible mechanism of aminoguanidine (AG) on the lipopolysaccharide(LPS)-induced acute lung injury in rats. Methods Male SD rats were randomly divided into 3 groups: ① Control group; ② LPS group; ③ AG group. AG was administrated in AG group, saline was administrated in control group and LPS group. LPS group and AG group were further divided into 2 subgroups according to the duration of ALI:3 h+3 h group and 6 h+3 h group. In AG group and LPS group, LPS was administrated. Saline was administrated in control group. Expressions of NOS mRNA in the lung tissue were respectively measured with RT-PCR method;Apoptosis, the expression of Caspase-3 protein, Bcl-2 and bax were respectively detected with Flow Cytometry (FCM), Western blot analysis, and immunohistochemisty (IHC); the pathological changes of lung tissue were observed by light and electron microscope. Results Compared with that of the control group, the expression of iNOS mRNA was significantly increased; the eNOS mRNA was significantly decreased; apoptosis of pulmonary cells, the expression of Caspase-3 and Bax protein were significantly increased; the expression of Bcl-2 was decreased in alveolar and airway epithelial cells in LPS group respectively. Degree of ALI was gradually worsened after administration of LPS. In AG(3 h+3 h) group, the percentage of apoptotic cells, the expression of caspase-3 and Bax protein were decreased respectively, the expression of Bcl-2 and the ratio of Bcl-2 /Bax were increased and the lung damage was improved respectively compared with that of the LPS(3 h+3 h) group. Conclusion Relatively early administration of AG could ameliorate LPS-induced acute lung injury in rats. The possible mechanism is that AG could reduce the expression of iNOS mRNA,Caspase-3 and Bax protein,increase the expression of Bcl-2 protein, and regulate the balance between Bcl-2 and Bax protein.

Objective To explore the relationship between the stress during pregnancy and the NMDA-induced spasm. MethodsThe pregnant rats were divided into two groups: pregnant control group(PC group)and pregnatal stress group(PS group). The rats in PS group were forced to swim in cold water of 4℃ for five minutes. Pups of the two groups were injected NMDA 7mg/kg intraperitoneally on postnatal day 13 to induced infantile spasm-like attacks. Then the clinical behavior of pups of the two groups were observed and recorded. The ultrastructure of mitochondria was detected with scanning electron and appearance of neurons was observed with HE staining. NMDA receptor and its subuniforms (NR2A, NR2B) in hippocampus, cerebral cortex, hypothalamus and raphe nucleus of brainstem were estimated by immunohistochemistry. Results The average score of pups in PS group is higher than in PC group(7.84 ±1.01 vs 5.90 ± 1.12). There were swelling neurons and no cytonecrosis in hippocampuss of both two groups. The uhrastructure of mitochondria in hippocampuss and raphe nucleus of brainstem showed great changes in pups of PS group as membrane in mitochondria of pups in PS group. NR1, NR2A and NR2B distributed mainly on the membrane and also in cytoplasm and cellular nucleus. The NR1 expressions in hippocampus, cerebral cortex, hypothalamus and raphe nucleus in pups of PS group were elevated than those of PC group and the P value is 0.001, 0.009, 0.015, 0.001 respectively. The NR2B expressions in hippocampus, cerebral cortex hypothalamus and raphe nucleus in pups of PS group were elevated than those of PC group and the P value is 0.004, 0.002, 0.003, 0.002 separately. NR2A expressed more in hippocampus in pups of PS group than that in PC group,but there was no difference in cerebral cortex, hypothalamus and raphe nucleus between two groups and the P value is separately 0.006,0.067, 0.7, 0.5. Conclusions The mitochondria of neurons in rat pup were hurt by maternal severe stress daring pregnancy, which made the energy metabolism of neurons changed. Maternal stress can also elevate the expression of NR1, NR2A and NR2B in cerebral cortex, brainstem and hippocampus and increase the morbidity of infantile spasm-like attacks.

OBJECTIVE: To build information repository of the carrying rate of neonatal deafness gene in Shijiazhuang. METHOD: Blood samples were collected from the heel in 3-days neonates. Mutations of the deafness related genes were detected by the method of fluorescent PCR. Neonates received the detection of 6 mutation sites from 3 genes, including GJB2 (235delC, 299-300delAT), SLC26A4 (IVS7-2A> G, 2168A> G), mitochondrial DNA12S rRNA(1494C>T,1555A>G). RESULT: There were 384 neonates who carried mutations among 421 subjects and the carrying rate was 4.08%, 158 (1.68%) newborns carried heterozygous mutations and 1 (0.01%) case carried homogeneous mutation of GJB2 (235 delC), 55 (0.58%) neonates carried heterozygous mutations of GJB2 (299-300delAT); 133 (1.41%) neonates carried heterozygous mutations and 1 (0.01%) homogeneous of SLC26A4(IVS7-2A>G),19 (0.20%) newborns carried heterozygous mutations of SLC26A4 (2168A>G). The numbers of neonates who carried homogeneous and heterogeneous mutation of mitochondrial 12S rRNA gene were 14 and 3 with carring rates of 0.15% and 0.03%. Two newborns were found to carry more than one mutation. One carried 235delC, IVS7-2A>G and 1555A>G and another carried 235delC and IVS7-2A>G. CONCLUSION The main mutational patterns were 235delC from GJB2 gene and IVS7-2A>G from SLC26A4 gene in Shijiazhuang newborns. The carrying rate information repository of neonatal deafness gene has been built preliminarily.

OBJECTIVETo explore the effects of different acupuncture depths of Lianquan (CV 23) for dysphagia after stroke.

METHODSTwo hundred and fifty-one patients with dysphagia after stroke were randomly divided into a deep acupuncture group (group A, 85 cases) a shallow acupuncture group (group B, 83 cases) and a glossopharyngeum acupuncture group (group C, 83 cases). Based on the conventional treatment, the glossopharyngeum acupuncture which was pricking without needles retained at lingual surface, posterior pharyngeal wall of the affected side, Jinjin (EX-HN 12) and Yuye (EX-HN 13) was used in the three groups. After the glossopharyngeum acupuncture, Lianquan (CV 23) was acupunctured 60-70 mm in the group A and 30-40 mm in the group B. The needles were all retained for 30 min. The treatment was applied once a day for 30 days in the three groups. Water swallowing test evaluation scale was observed before and after treatment, and the efficacy was assessed in the three groups.

RESULTSThe total effective rate was 95.3% (81/85) in the group A, which was better than 85.5% (71/83) in the group B and 83.1% (69/83) in the group C (both P < 0.05). The water swallowing test scores were decreased apparently in the three groups (all P < 0.01), and scores in the group A were the most obvious decline (P < 0.01). The decrease of scores in the group A was more remarkable than those in the group B and the group C after two weeks of treatment (both P < 0.05).

CONCLUSIONAcupuncture at Lianquan (CV 23) can effectively improve the dysphagia after stroke. The acupuncture depth can affect efficacy and the effect of deep acupuncture is better.

To evaluate the value of left ventricular diastolic function in type 2 diabetes mellitus (DM) using dual-gate Doppler and relative factors, we included 50 non-obesity and hypertension-free DM patients into the controlled group in the study along with 48 age-and-gender-matched healthy volunteer subjects. The peak early diastolic velocity (E), peak later diastolic velocity (A), deceleration time (DT), anterior mitral annulus diastolic peak velocity (e') , isovolumic relaxation time (IVRT), E/A, E/e', Tei index and T(E-e), were measured with dual-gate Doppler. 20 subjects were randomly selected for repetitive analysis. Study showed statistical difference in E/A, DT, e', E/e', IVRT, Tei index and T(E-e), between the two groups (P < 0.05). Linear regression analysis showed positive correlation between T(E-e), and IVRT, course of DM patients and T(E-e), (Beta = 0.295, P = 0.020), and HbA1c control level and T(E-e), (Beta = 0.399, P = 0.010). Repeated analysis showed good reproducibility for both within and between groups. Dual-gate Doppler has clinical value in evaluating left ventricular diastolic function in type 2 diabetes mellitus patients. The course of type 2 diabetes mellitus patients and HbA(1C) control level were both closely related with left ventricular diastolic function.

Objective To investigate tissue distribution of Hylotelephin in Beagle dogs and excretion of Hylotelephin in rats. Methods A high performance liquid chromatography (HPLC) with UV detection was developed to study the concentrations of Hylotelephin in biological samples, taking anthracene as an internal standard, Benzoyl chloride as the pre-column derivatization of Hylotelephin and methanol-water as the mobile phase. Results After a single intravenous dose of 10.6 mg/kg Hylotelephin in beagle dogs, parent drug was widely distributed to virtually all tissues in 5 min and the concentration of Hylotelephin in most tissues at 90 min were lower than those at 5 min obviously. Hylotelephin was mainly distributed in kidney, liver and spleen, secondly in heart, lung and intestine. The parent drug concentration in kidney, liver and spleen was similar to that in blood at the same time point. After a single intravenous dose of 36 mg/kg Hylotelephin in rats, the excretion of the parent drug in urine, feces and bile amounted to 88.45%, 0.61% and 1.08% of the dose, respectively. The parent drug excretion amounted to 67% of the dose in 3 h. Conclusion Hylotelephin was distributed and eliminated in Beagle dogs rapidly. It was mostly distributed in kidney, liver, spleen and plasma. The parent drug excretion was 88% via urine.
