Objective To investigate the influence of long-term blood purification on atherosclerosis(AS) in maintenance hemodialysis(MHD) patients by measuring serum levels of homocysteine(Hcy) and carotid artery intima-medial thickness(IMT).Methods Seventy-two MHD patients visiting our hospital from June 2006 to December 2007,according to their blood purification mode,were assigned to three groups: hemodialysis(HD,n=25),hemodialysis+hemodiafiltration(HD+HDF,n=30),hemodialysis+continuous renal replacement therapy(HD+CRRT,n=17).Another 25 healthy subjects served as controls.Serum Hcy was detected before and after one-year treatment and meanwhile color Doppler ultrasonography was performed to check IMT of carotid artery and atheromatous plaques.Results The serum levels of Hcy,IL-6,MCP-1,hs-CRP in MHD patients were significantly higher than those in healthy controls(P

4.Effects and mechanism of lycopene on the proliferation and apoptosis of human hepatoma cells

Objective To investigate the effects and mechanism of lycopene on the proliferation and apoptosis of human hepatoma cell HepG2.Methods The human HepG2 cells in logarithmic growth phase were treated with lycopene (5,10,20 μg/ml) and cisplatin (40 pg/ml).The cellular growth inhibition rate was calculated by methyl thiazolyl tetrazolium (MTT) after 48 h,and cell cycle and apoptosis rate were analyzed by flow cytometry.The expressions of Bax,Bcl-2 and cleaved Caspase-3 proteins were detected by Western blotting.Results After 48 h intervention,the cellular growth inhibition rate of human HepG2 cell in blank control group was 0,and in lycopene (5,10,20 μg/ml) groups and cisplatin group were (21.3 ±4.2)%,(40.5 ±7.6) %,(63.8 ± 9.1) %,(37.8 ± 5.9) %,with significant difference (F =37.905,P =0.000);and compared with blank control group respectively,the differences were statistically significant (t =208.124,P =0.000;t=394.637,P=0.000;t=628.592,P=0.000;t=257.168,P=0.000).The G0-G1 phase rates in lycopene (10,20 pg/ml) groups were (54.0 ± 2.9) % and (67.3 ± 3.6) %,compared with blank control group (37.9± 1.5)%,the differences were statistically significant (t =4.508,P =0.024;t =10.673,P =0.006).The G2-M phase rates in lycopene (10,20 μg/ml) groups were (8.5 ± 0.6)% and (4.7 ± 0.5)%,compared with blank control group (18.4 ± 0.8) %,the differences were statistically significant (t =9.975,P =0.008;t =13.864,P =0.003).The apoptosis index (AI) in lycopene (5,10,20 μg/ml) groups and cisplatin group were (19.5 ± 4.8) %,(43.0 ± 9.2) %,(67.6 ± 10.1) % and (36.9 ± 6.8) %,compared with blank control group [(3.6 ± 1.7)%],the differences were statistically significant (t =18.617,P =0.001;t =34.295,P =0.000;t =51.437,P =0.000;t =29.804,P =0.000).The expressions of Bcl-2,Bax and the ratio of Bax/Bcl-2 in lycopene (10,20 μg/ml) groups and cisplatin group were 0.42 ± 0.09,0.43 ±0.14,1.02 ±0.39;0.27 ±0.08,0.76 ±0.19,2.81 ±0.85;0.34 ±0.11,0.31 ±0.09,0.91 ±0.40,respectively.Compared with blank control group (0.59 ±0.17,0.18 ±0.06,0.31 ±0.12),the expressions of Bcl-2 were significantly down-regulated,and the differences were statistically significant (t =4.327,P =0.023;t =11.064,P =0.006;t =5.182,P =0.018),the expressions of Bax were significantly upregulated,and the differences were statistically significant (t =9.837,P =0.008;t =17.349,P =0.001;t =10.165,P =0.007),the ratios of Bax/Bcl-2 were significantly increased,and the differences were statistically significant (t=11.521,P=0.006;t=18.194,P=0.001;t=9.537,P=0.008).The expressions of cleaved Caspase-3 protein in lycopene (5,10,20 μg/ml) groups and cisplatin group were 0.25 ± 0.07,0.34 ±0.11,0.46 ± 0.18 and 0.17 ± 0.05,compared with blank control group (0.08 ± 0.03),the differenees were statistically significant (t =8.307,P =0.009;t =13.067,P =0.006;t =16.218,P =0.003;t =5.202,P =0.019).Conclusion Lycopene has inhibitive effect on the growth of human HepG2 cells perhaps through inhibiting proliferation and inducing apoptosis,which may be related to its effects of altering the cell cycle and the expressions of apoptosis-related genes and proteins.
5.The clinical analysis of IL-6,IL-15 in peritoneal fluid and serum in patients with endometriosis

Objective To investigate the relationship between the levels of intedeukin(IL)-6, IL-15 and endometriosis (EM). Method The levels of IL-6, IL-15 in the peritoneal fluid (PF) and serum of 74 patients with EM (EM group) and 46 patients without EM (control group) were measured by double-an-tibody ELISA. Results Higher levels of IL-6 in PF and serum were observed in EM group [(1017.81±361.98) ng/L,(455.47±161.52) ng/L]than those in control group [(284.63±70.50) ng/L,(149.37± 43.09) ng/L], and there was significant difference (P<0.01). The levels of IL-6 in PF and serum in EM group with stage Ⅲ-Ⅳ [(1253.44±189.63) ng/L, (556.50±93.34) ng/L]were significantly higher than those in patients with stage Ⅰ-Ⅱ [(582.81±107.75) ng/L, (268.96±63.48) ng/L](P < 0.01). There was positive relationship between the levels of IL-6 in PF and serum (r=0.950, P=0.01). The levels of IL-15 in PF in EM group [(333.45±63.94) ng/L]were significantly higher than those in control group[(203.85± 70.52) ng/L](P<0.01). No significant difference was found in the levels of serum IL-15 between EM group and control group (P>0.05). No significant difference was observed either in the levels of IL-15 in PF and serum between patients with stsge Ⅲ-Ⅳ and stage of EM Ⅰ-Ⅱ (P>0.05). Conclusions The increase of IL-6, IL-15 in PF may contribute to the development of EM. Serum IL-6 levels are of clinical diagnostic value in patients with EM.
6.Multiple risk factor analysis of recurrent respiratory tract infection after Mycoplasma pneumoniae pneumonia in children

Objective To investigate the occurrence trend and risk factors of recurrent respiratory tract infection during the ifrst year after Mycoplasma pneumoniae pneumonia (MPP) in children. Methods The clinical data of 133 children complete-ly recoved from MPP and one year follow-up after MPP were included in this study, MPP IgM IgG double antibody titer were measured in different time slots (3 month, 6 months, 9 months, 12 months) after discharge. Information on frequency of recurrent respiratory tract infections, respiratory tract infection site, and drug use within one year were collected. Possible factors affecting the occurrence of recurrent respiratory tract infection were analyzed by means of single factor and multi factor analyses. Results In 133 patients, the recurrent rate was 31.58%in the ifrst year;aged 3-6 years old (OR=2.29,95%CI:1.13~4.64), or continuous positive or negative to positive antibodies (OR=4.47,95%CI:1.47~13.65), or low CD4/CD8 (OR=10.26,95%CI:3.30~31.90), or low IgA (OR=1.90,95%CI:1.06~3.40) is independent risk factor of recurrent respiratory tract after MPP;immune enhancer therapy is an independent protective factor (OR=0.29,95%CI:0.11~0.78). Conclusions Immune function disorders in the ifrst year after MPP were independent risk factors of recurrent respiratory tract infection. MP antibody positive without clinical symptoms and sustained antibody positive can not prevent the recurrence of respiratory tract infection without use of antibiotics. Immune enhancer was advocate to adjust immune function and reduce the incidence of repeated respiratory tract infection.
7.Cognitive functions in first-episode schizophrenia and unaffected first-degree relatives of different patients

Objective To investigate cognitive functions in first-episode schizophrenia and unaffected first-degree relatives of different patients.Methods A total of 52 patients with first-episode schizophrenia (patient group),48 unaffected first-degree relatives of different patients (relatives group) and 50 healthy normal controls (control group) was recruited in the study.The attention,memory,and executive functions of all the subjects were assessed by a battery of neuropsychological tests,including cancellation test (CT),trail making test (TMT),digit span (DS),visual reproduction (VR),verbal fluency task (VFK),Wisconsin card sorting test (WCST) and HANOI tower.Results There were statistically significant difference among three groups in net scores of CT,TMTA,TMT-B,backward and total scores of DS,scores of VR,VFK,total errors and perseverate errors of WCST and all indicators of HANOI Tower (P <0.05 or P < 0.01).Post Hoc multiple comparisons showed that,compared with the control group,the patient group had worse scores in all tests above except TMT-A in the relatives group (P <0.05 orP <0.01).However,compared with the patient group,the relatives group had better scores in net scores of CT,TMT-A,TMT-B,scores of VR,VFK,total errors and perseverate errors of WCST (P < 0.05 or P < 0.01).Conclusions Patients with first-episode schizophrenia have global cognitive functions deficits.Unaffected first-degree relatives of different patients also have cognitive deficits to a certain extent.Our study indicates that cognitive functions deficits may be viewed as a biomarker for candidates reflecting genetic liability for schizophrenia.
8.Therapeutic Observation of Electroacupuncture plus Moxibustion for Infantile Cerebral Palsy

Objective To observe the clinical efficacy of electroacupuncture plus moxibustion in treating infantile cerebral palsy. Method Eighty eligible patients with infantile cerebral palsy were randomized into a treatment group of 42 cases and a control group of 38 cases. The control group was intervened by physiotherapy (PT), while the treatment group was given electroacupuncture at scalp acupoints, body acupuncture, and moxibustion along the Governor Vessel in addition to the treatment given to the control group. The two groups were treated once a day, 6 months in total. The comprehensive ability and the Gross Motor Function Measure (GMFM) were evaluated before and after the interventions in the two groups, and the clinical efficacies were compared.Result After 6-month treatment, the total effective rate was 92.9% in the treatment group versus 78.9% in the control group, and the total effective rate of the treatment group was superior to that of the control group (P<0.05). The motor function and activities of daily living were significantly improved in both groups after the interventions (P<0.05,P<0.01); after the treatment, there were significant differences in comparing the motor function and activities of daily living between the two groups (P<0.05). The GMFM scores of each domain were significantly changed after the treatment in both groups (P<0.05); after the treatment, there were significant differences in comparing the GMFM scores of each domain between the two groups (P<0.05).Conclusion Compared with being used alone, conventional PT can more significantly benefit the improvement of motor function of cerebral palsy patients when electroacupuncture and moxibustion were added.
9.Establishment and application of objective evaluation system for thyroid imaging reporting and data system classification in ultrasound

Objective To select the ultrasonic appearances which effect the benign and malignant of thyroid lesions,and to explore the establishment and applications of supersonic evaluation system for thyroid imaging reporting and data system (TI-RADS) classification.Methods The ultrasound images of 1 080 thyroid nodule cases were retrospectively analyzed by Logistic equation,and the ultrasonic appearances were screened to identify benign and malignant of thyroid lesions.All appearances were obtained weights.The TI-RADS classification analysis software was assigned,which was used to analyze 332 patients with thyroid diseases and to verify the diagnostic performance of the TI-RADS classification system.Results Five ultrasonic features were selected into the regression model,including aspect ratio (odds ratio [OR]=3.61),margins (OR=3.83),composition (OR=11.46),echogenicity (OR=14.12),microcalcification foci (OR =48.82).Using objective evaluation system for TI-RADS classification in ultrasound in diagnosing 332 cases,the area under the ROC curve was 0.91 (P<0.05).Conclusion Relatively the weight and formation of malignant indicators should be more concern than the number of ultrasonic malignant thyroid lesions indicators.The objective evaluation system for TI-RADS classification in ultrasound has relatively high diagnosis capability which established by the methods of Logistic equation.
10.Experimental studies for botulinum toxin type A to antagonist the VIP/PACAP expression on nasal mucosa in allergic rhinitis rat.

OBJECTIVE:
To explore the expression and significance of vasoactive intestinal peptide and Pituitary adenylate cyclase activiting polypeptide (VIP/PACAP) of nasal mucosa in rats with allergic rhinitis (AR), and the function of botulinum toxin-A(BTX-A) to inhibit the expression of VIP/PACAP in AR.
METHOD:
Thirty Sprague-Dawley rats were randomly divided into 3 groups, which were the AR group, the intervention group, and the control group. In the AR group, ovalbumin was used to sensitize healthy rats. In the intervention group, BTX-A was dripped into the nasal cavity of AR rats 7 times. In the control group, only physiological saline was used to drip into the nasal cavity of AR rats. Changes of the rats' behavior were observed. ELISA were used to detected the concentration variation of serum IFN-γ and IL-4. Histopathology and immunohistochemistry were employed to observe morphology in the rats' nasal mucosal and the expression of VIP/PACAP. Statistical analysis was also made.
RESULT:
(1)The typical symptoms marks of nasal scratching, sneezing, nasal blockage and rhinorrhea of AR group (7.5 ± 0.50) were higher than intervention group (1 ± 0.27) and control group (0.8 ± 0.31). (2) Comparing to intervention group and control group, the serm IFN-γ of the AR group obvious reduced (P < 0.05), the serm IL-4 of the AR group obvious rose (P < 0.01), and the serm Th1/Th2 (IFN-γ/IL-4) of the AR group obvious reduced (P < 0.01). (3) Comparing to intervention group and control group, the cilium loss, inflammatory cells infiltration, and inflammatory cells exudation of nasal mucosa in AR group were more obviously (P < 0.01), and the intervention group of the 3 indexes was obviously than control group. (4) The expression of VIP in the rats' nasal mucosa of the AR group (13.27 ± 2.74) were more intense than intervention group (5.21 ± 2.18) and control group (3.56 ± 5.30) (P < 0.01), and the expression of PACAP in the rats' nasal mucosa of the AR group (20.97 ± 2.14) were more intense than intervention group (6.33 ± 3.04) and control group (4.63 ± 1.25) (P < 0.01). (5) In all the 3 groups, there was positive correlation between expression of negative in VIP/PACAP and Thl/Th2 cell infiltration(r were respectively -0.340 and -0.223, P < 0.05).
CONCLUSION
The VIP/PACAP in the rats' nasal mucosa may play an important role in pathogenesis of AR, and BTX-A could improve the symptoms of AR through inhibition of the expression of VIP/ PACAP.
