1.Comparison between three sampling methods in the study of placental pathology

Objective:The pathological characteristics of tissue obtained from the placental bed biopsies (method A), placental basal plate sections (method B) and placental bed vacuum suction(method C) were evaluated to find a best sampling method to study the uterine spiral arteries. Methods:Histology obtained from 33 patients using method A and B and obtained from 51 patients using method C was analysised ,retrospectively. The differences of tissue and the spiral artery pick-up rate were compared. Results:Both the decidua and myometrium could be obtained by method A and C. 2.Effect and mechanism of nimesulide combined with cisplatin on proliferation and apoptosis of lung cancer

ObjectiveTo evaluate the effects of selective cyclooxygenase-2 inhibitor nimesulide lone and combined with cisplatin on tumor growth,and Ki67 and Caspase-3 expression in lung cancer xenografts in nude mice. Methods The mice were randomly divided into 4 groups:the control group,the nimesulide group,the cisplatin group and the nimesulide combined with cisplatin group. A549 cells were injected into BALB/c nude mice subcutaneously.On the 21st day after treatment tumor tissues were collected,and the xenografts growth were observed. The expression of Ki67 and Caspase-3 were detected by immunohistochemical method.ResultsNimesulide combined with cisplatin could significantly inhibited the xenografts growth compared with nimesulide or cisplatin.The tumor inhibition rate was 44.33% in the nimesulide group,53.61% in the cisplatin group and 80.41% in the nimesulide combined with cisplatin group (P < 0.05).Immunohistochemical analysis showed that the expression rates of Caspase-3 was significantly increased in the nimesulide combined with cisplatin group (67.43 ± 23.57 ) %,the cisplatin group (48.40 ± 20.37 ) %,and the nimesulide group (38.65 ± 15.37)%,compared with the control group (27.63 ± 13.03)% (P < 0.05).The expression rate of Caspase-3 was significantly increased in the nimesulide combined with cisplatin group compared with the nimesulide group or the cisplatin group (P < 0.05 ).The expression rate of Ki67 was significantly decreased in the nimesulide group combined with cisplatin group (24.34 ± 15.90)%,the cisplatin group (40.85 ± 22.47)% and the nimesulide group (53.33 ± 19.67)% compared with the control group ( 80.43 ± 16.88 ) % ( P < 0.05 ).The expression of Ki67 was significantly decreased in the nimesulide combined with cisplatin group compared with the nimesulide group or the cisplatin group ( P < 0.05 ).Conclusion Nimesulide can inhibit human lung cancer A549 cells xenografts in nude mice growth,Nimesulide enhanced the inhibitory effects of cisplatin.The mechanism may be related to inhibition of tumor cell Ki-67 expression,increased expression of Caspase-3,inhibition of tumor cell proliferation,and inducing tumor cell apoptosis.
3.Clinical observation of general anesthesia for caesarean section

Objective To investigate the safety and feasibility general anesthesia for cesarean section in term matemal.Methods Hospital in January-December 2014 admitted there are contraindications to spinal anesthesia or reject elective spinal anesthesia in term maternal 42 cases were general anesthesia.By intravenous injection of propofol 1.5-2 mg/kg and succinylcholine 1.5-2 mg/kg rapid induction,after video laryngoscopes with endotracheal intubation,immediate surgery to remove the fetus,neonates 1,5 min Apgar score.Results 42 cases of maternal disposable endotracheal intubation success rate of 100%,no regurgitation and aspiration;all fetuses removed within 10 min after induction of anesthesia,neonatal 1,5 min Apgar scores were 8 points or more,with no respiratory depression.Conclusion When patients have contraindications to spinal anesthesia or spinal anesthesia refused general anesthesia is a safe and viable alternative,as long as the rational use of anesthetics,general anesthesia for cesarean section and newborn is safe.

5.The effect observation of different doses of Dezocine used in postoperative analgesia of pediatric tonsillectomy and adenoidectomy

Objective To observe safety and effectiveness of Dezocine in postoperative analgesia of pediatric tonsillectomy and adenoidectomy by the continuous intravenous infusion of different doses,and to explore appropriate dosage of Dezocine for pediatric postoperative analgesia.Methods Elective tonsillectomy and adenoidectomy require-ments of postoperative analgesia in children 120 cases,they were randomly divided into A,B,C,D groups of 30 cases by using number table.After surgery patient -controlled intravenous analgesia(PCIA)two days,A,B,C groups were given Dezocine 0.2,0.3,0.4mg·kg -1 ·d -1 continuous intravenous infusion,D group received Sufentanil 1μg· kg -1 ·d -1 continuous intravenous infusion.Observed and recorded pain scores and sedation scores for postoperative 6h,12h,24h and 48h;recorded adverse reactions,pressing the number and effective pressing times of children within 48h after surgery.;recorded overall evaluation of postoperative analgesia of children and parents.Results A group at each time point pain score was significantly higher than B,C,D three groups,the difference was statistically significant (tAB =12.003,PAB =0.049;tAC =7.542,PAC =0.036;tAD =40.990,PAD =0.009);A,B,C three groups at each time point sedation score was significantly lower than D,the difference was statistically significant(tDA =3.894,PDA =0.030;tDB =5.020,PDB =0.039;tDC =36.103,PDC =0.016);D group of adverse reactions were significantly higher than A,B,C three groups,the difference was statistically significant(χ2DA =20.738,PDA =0.000,χ2DB =4.35,PDB =0.029,χ2DC =5.98,PDC =0.025);A group within 48h postoperative analgesia press times and the number of valid pressing were significantly higher than B,C,D three groups,the difference was statistically significant(tAB =3.003, PAB =0.036;tAC =5.720,PAC =0.031;tAD =37.157,PAD =0.015);B,C two groups of children and parents to evaluate postoperative analgesia overall satisfaction rates were 90.0%,93.3%,significantly higher than A 63.3%,D 60.0%,and the difference statistically significant (χ2AB =5.310,PAB =0.031;χ2DC =19.640,PDC =0.000). Conclusion Dezocine 0.3mg·kg -1 ·d -1 continuous intravenous infusion for pediatric tonsillectomy and adenoid-ectomy is safe and effective in postoperative analgesia and fewer adverse reactions,and should be promoted.
6.PULSE POLAROGRAPHIC DETERMINATION OF NITROGEN

A simple and rapid method was described for determining nitrogen content of a Kjeldahl digest by pulse polarography. The amount of sample needed was as small as 200mg. The digest was diluted and pipetted into a solution of formaldehyde in an acetate buffer. The hexamethylenetetramine produced by reaction between ammonia and formaldehyde was determined electrochemically at -0.9v.This method proved itself a better reproducibility. The recovery rate of nitrogen was 94.5% to 103.1%. Calibration curve of nitrogen in the range of 20 to 120 ?g/ml showed linear relation.Human milk and eleven different food-stuffs were analysed. There was no significant difference between the results by this method and Kjeldahl method.
7.Mesenchymal stromal cells for cerebral infarction

Although there are many drugs for the treatment'of cerebral infarction, no satisfactory efficacy has achieved. Mesenehymal stromal cells (MSCs) have multilineage differentiation potential and self-renewing capability, and can differentiate into neurons. The transplanted MSCs can improve neurological deficit after cerebral infarction, promote the recovery of neurological function, and thereby reduce the disability of cerebral infarction through the mechanisms such as releasing nutritional factors and growth nutritional factors, promoting angiogenesis. The use of MSCs for cell transplantation is likely to be an ideal means in the treatment of cerebral infarction.
8.Therapeutic Observation of Acupuncture-moxibustion for Postsurgical Gastroparesis and Effect on Gastrin and Motilin

Objective To observe the clinical efficacy of acupuncture-moxibustion in treating postsurgical gastroparesis syndrome (PGS) and the effect on gastrin (GAS) and motilin (MTL).Method Fifty PGS patients were randomized into a treatment group and a control group, 25 cases in each group. The control group was intervened by conventional medication, including effective gastrointestinal decompression, nutrition support, and psychotherapy; the treatment group was by acupuncture-moxibustion therapy in addition to the intervention given to the control group. The time of restoring gastric motility, the distention and fullness grading score, and serum GAS and MTL levels were observed, and the clinical efficacies were also compared.Result The total effective rate was 100.0% in the treatment group versus 92.0% in the control group, and the between-group difference was statistically insignificant (P>0.05). According to statistical analysis, there was a significant difference in comparing the time of restoring gastric motility between the two groups (P<0.01). The distention and fullness grading scores, GAS and MTL levels were significantly changed after intervention in both groups (P<0.01). The distention and fullness grading score, GAS and MTL levels of the treatment group were significantly different from that of the control group after intervention (P<0.01).Conclusion Acupuncture- moxibustion plus medication is an effective approach in treating PGS, and it can up-regulate the GAS and MTL levels.
9.The role of rdxA gene in Helicobacter pylori resistant to metronidazole

8 mg/L). The rdxA gene fragment (886 bp) of H. pylori was amplified by PCR and then sequenced. Finally polymorphism analysis was performed in the rdxA gene sequence from those strains resistant or susceptible to Mtz and standard strains H.pylori 26695. Results A total of 47 H.pylori strains were isolated from 51 patients positive for rapid urease test. Mtz resistance rate was 34% by E-test method and 31.9% by breakpoint susceptibility testing. Among them 15 strains were resistant to Mtz by the two tests. The rdxA genes of both 15 strains resistant to Mtz and 8 strains susceptible to Mtz were sequenced. Compared with that of strain H.pylori 26695, the sequence of rdxA gene nucleotide in 15 H.pyloristrains resistant to Mtz showed similarity ranging from 90% to 95% and being 92.5% on average, and the sequence of rdxA gene nucleotide in susceptible strains showed similarity ranging from 94% to 95% and being 94.5% on average. In most resistant strains, the rdxA genes occurred point mutation, single nucleotide insertion or deletion, however, the insertion or deletion of large nucleotide fragment were rare, and mutational site were variable. Conclusions The rdxA gene mutation in H.pylori strains resistant to Mtz may result from point mutations in several bases or insertional mutations or deletional mutations in one base, without insertion or deletion of large neuclectide fragment, and mutational position being variable.
10.Inhibitory effect of licoric extracts on activity of tyrosinase

AIM: To study the inhibitory effects of the licoric extracts on the activity of tyrosinase, which is constituted of monophenolase and diphenolase. METHODS: The 50%(V/V) alcohol is used as extractant. To investigate the effects of the licoric extracts on the monophenolase and diphenolase activity, the absorbance of the solution containing several concentrations of the licoric extracts was measured at 475 nm and 390 nm, using the L-tyrosine and pyrocatechin as the substrate, respectively. RESULTS: The licoric extracts has showed a strongly inhibition on the activity of tyrosinase, either monophenolase or diphenolase. The IC_ 50 is 1.94 and 2.94 mg?ml -1 , respectively. CONCLUSION: The licoric extracts has inhibitory effects on both monophenolase and diphenolase. The diphenolase activity test system using pyrocatechin as the substrate is a useful procedure to study the activity of tyrosinase.
