1.Clinical effects and influence on serum creatinine and blood urea nitrogen of Danggui Niantong pill in treatment of gouty nephropathy

Objective To analyse clinical effects and influence on serum creatinine and blood urea nitrogen of Danggui Niantong pill in treatment of gouty nephropathy. Methods 80 patients with gouty nephropathy were divided into two groups and each with 40 cases.The control group were treated with general therapy,and observation group was treated another with Danggui Niantong pill. Results After treatment,effective rate of observation group was 92.50%,higher than that of control group 72.50%(P<0.05).After treatment,renal function serum markers and urine related factors level of observation group were lower than control group(P<0.05).ESR,WBC and N level of observation group were lower than that of control group(P<0.05).The proportion of adverse reactions of observation group was 15.00% and control group was 12.50%.Conclusion Danggui Niantong pill can improve clinical effects of gouty nephropathy,reduce renal damage amd does not increase adverse reactions.
2.Development of Chromatographic Fingerprint of Rhizoma Alismatis Orientalitis by HPLC-ELSD

Objective A fingerprint of Rhizoma Alismatis Orientalitis' decoction pieces was established by HPLC-ELSD. Methods The gradient elution mode was applied in chromatographic separation by mobile phase of methanol and water at flow rate of 0.5 mL? min-1 in a Diamonsil TM C18 chromatographic column (250 mm ? 4.6 mm, 5 ? m), and recording time was 70 minute. The parameters of evaporative light scattering detector were set as follows: the temperature of the drift tube was 82.5 ℃ and gas flow rate was 1.9 L ? min-1.The data was analyzed with cluster analysis and similarity calculation software. Results The fingerprint showing 15 common peaks was obtained. The results of cluster analysis and similarity calculation showed there existed differences in fingerprints of 16 batchers of medicinal slices of Rhizoma Alismatis Orientalitis. Conclusion The method is effective and reproducible for the quality control of medicinal slices of Rhizoma Alismatis Orientalitis.
3.Effect of Yiqi Huoxue Tablets on Blood Coagulation and Fibrinolysis System of Cerebral Apoplexy Rats Induced by Artificial Cold Wave

Objective To study the effect of Yiqi Huoxue Tablets on blood coagulation and fibrinolysis system of rats with cerebral apoplexy induced by artificial cold wave. Methods SD rats were randomized into 4 groups. The apoplexy-prone renovascular hypertensive rat model was established by the bilateral renal arteries clamping method. From the 14th day after the modeling operation, the treatment group was given Yiqi Huoxue Tablets (1.89 g·kg-1·d-1), the positive control group was given aspirin (80 mg·kg-1·d-1), and the model group and the blank group were given the same volume of distilled water, once a day. After 112 days, the rats were put into the artificial cold wave box staying for 3 days to inducing apoplexy, and then thrombin-activatable fibrinolysis inhibitor (TAFI) and protein C ( PC) , thrombin-antithrombin complexes ( TAT) and plasmin-antiplasmin ( PAP) levels were measured. Results The plasma levels of TAFI, PC, TAT, and PAP in Yiqi Huoxue Tablets group were lower than the model group, the difference being statistically significant ( P<0.05) . Conclusion The preventive and therapeutic mechanism of Yiqi Huoxue Tablets for cerebral apoplexy is probably related with the regulation of the balance between blood coagulation and fibrinolytic system of apoplexy-prone renovascular hypertensive rats.
4.The study on changes of serum gastrin level and its influencing factors in carcinogenesis of gastric mucosa

Objective To explore the common factors' influence on serum gastrin level,and reveal the value of serum gastrin in the diagnosis of gastric precancerous lesions and gastric cancers.Methods Serum gastrin level of 3 906 serum samples was detected by ELISA.Results Serum gastrin level of patients over 60 years old was distinctively higher than that of other age groups.Serum gastrin level in antral diseases was higher than that in diseases of the body.HP positive group was higher than the negative one.The level progressingly increased from normal group to superficial gastritis and to gastric erosion or ulcer;and it decreased from gastric erosion or ulcer to atrophic gastritis,while it increased from atrophic gastritis to dysplasia,and it decreased from dysplasia to gastric cancer.Serum gastrin level was distinctively lower in gastric cancer than in other groups.Conclusion For people over 60 years old,their serum gastrin level has the tendence to increase.The location of gastric disease is an important factor affecting serum gastrin level.HP can increase the serum gastrin level.During gastric precancerous stage,the serum gastrin level may gradually increase with the diseases' progression,and decrease when gastric cancer is formed.Serum gastrin is a good marker to differentiate benign and malignant gastric diseases.
5.PCR fingerprinting discriminating between Candida albicans and Candida dubliniensis

Objective To develop a rapid PCR fingerprinting for discriminating between Candida albicans and Candida dubliniensis isolates. Methods Genomic DNA purified from the two species was amplified by single primer PCR. Oligonucleotide of the minisatellite-specific core sequence of the wild-type phage M13 (5′-GAGGGTGGCGGTTCT-3′) was used as primer and the amplified products were analyzed by gel electrophoresis and microfluidic DNA chip assays. Results Of 17 candida isolates, the PCR fingerprinting generated five strain-specific bands for C. albicans and C. dubliniensis respectively, allowing identification to species level between them. The other bands were minor different in their species. By microfluidic DNA chip, the DNA fragments in size of amplified products for the C. dubliniensis were 960,1177,1297,1495,1797 bp and for the C. albicans 653,1323,1531,2021,2875 bp. Conclusions C. albicans and C. dubliniensis have distinguishable pattern by PCR fingerprinting using the single primer. The microfluidic DNA chip is proposed here as a simple, rapid and highly reproducible tool, especially for the epidemiological investigation.
6.Clinical efficacy and perinatal outcome of nifedipine for severe preeclampsia: Meta-analysis

Objective To assess the efficacy,side effects and perinatal outcome of nifedipine compared with other antihypertensives for treating severe preeclampsia in pregnant women. Methods Randomized controlled trials (RCTs) that comparing nifedipine with other antihypertensives for severe preeclampsia were searched in PubMed,EMBase,Cochrane library,CNKI and VIP database etc( till January 2012).The quality of the included RCTs was evaluated,and Meta-analvsis was performed with Rev Man 5.1software.Results Nine trials were included,involving 386 women in the nifedipine group,and 378 women in other antihypertensives group.Compared with other antihypertensives,nifidepine was associated with greater effective control of blood pressure ( OR =2.65,95% CI:1.65 -4.25,P <0.01 ).There was no clear difference in the time needed to control blood pressure ( W.M D =- 3.64,95% CI:- 10.90 - 3.61,P =0.32 ).Nifedipine could prolong gestation better than other antihypertensives ( WMD =5.14,95% CI:3.29-6.99,P <0.01 ).There were no clear differences in maternal side effects headache (P =0.28),palpitation ( P =0.06 ),and nausea vomiting ( P =0.28 ).No noticeable difference was found between the two groups in the Apgar score at five minutes ( WMD =- 0.21,95% CI:- 0.32 - 0.91,P =0.72),neonatal respiratory distress syndrome ( OR =1.24,95% CI:0.57 - 2.67,P =0.59 ),or perinatal deaths ( OR =0.49,95% CI:0.22 - 1.11,P =0.09 ).Conclusion Nifedipine is associated with greater effective control of blood pressure and prolongation of gestation,with no additional neonatal respiratory distress syndrome or perinatal deaths,compared with other antihypertensives for women with severe preeclampsia.
7.To Predict Hypertention by Pregnancy in Counting Nuclear Red Blood Cells in Peripheral Blood

Objective To predict the Hypertention by pregnancy in counting nuclear red blood cells pe-ripheral blood. Methods Blood samples were obtained from 69 pregnant women with the gestational age from 12 to 18 weeks. Peripheral blood mononuclear cells were isolated by density gradient centrifugation and then analyzed the cells marked with PE-GPA/FITC-CD71 monoclonal antibodies on the flow cytometer. Correlations between them were analyzed after tracking of hypertention by pregnancy. Results Percentage of GPA+/CD71+ cells in healthy pregnant women was 0.89 ± 0.81%, while that in pregnant women with hypertention was 3.57 ± 3.48%. There was a significant difference between the two groups (P<0.05), and the percentage of GPA+/CD71+cells in the group of hypertention by pregnancy was much higher than that in the healthy group. Conclusions The increased nuclear red blood cells in maternal peripheral blood related to hypertention by pregnancy. The counting of nuclear red blood cells in maternal peripheral blood have potential value to predict hypertention by pregnancy.
8.The predictive value of coronary heart disease (CHD) risk factors in coronary atherosclerosis heart disease

Objective To investigate the predictive value of cardiovascular risk factors in coronary atherosclerotic heart disease.Methods A total of 400 patients who underwent coronary angiography (CAG) in our hospital was divided into multiple vascular lesion group,single vascular lesions group,and non-CAD group according to the result of CAG and their clinical data were analyzed retrospectively.The correlation between cardiovascular risk factors and coronary artery lesions was analyzed,and the independent risk factor of CAD was screened by multi-factorial logistic regression analysis.Results There was significant difference in total cholesterol (TC),triglycerides (TG),high density lipoprotein cholesterol (HDLC),fasting blood glucose (FBG),inflammatory cells,carotid artery plaque,brachial-ankle pulse wave velocity (baPWV),high-sensitivity C-reactive protein (Hs-CRP) between the CAD group and the non-CAD group (P < 0.05 or P < 0.01).The number of coronary artery lesion branch was increased significantly when risk factors,such as age,body mass index(BMI),hypertension,diabetes mellitas,smoking,carotid artery plaque,TG,TC,low density lipoprotein cholesterol (LDL-C),FBG,WBC,monocytes (M),neutrophils (N),neutrophils/lymphocytes (N/L),baPWV,and Hs-CRP.Other risk factors including TC,HDL-C,L were decreased with a statistically significant difference (P < 0.05).There was no significant relation among,and left ventricular ejection fraction (LVEF)%.The most significant risk factor was carotid artery plaque that was independently associated with coronary heart disease (b =1.264,P < 0.01),followed by smoking (b =1.204,P <0.01),HDL-C (b =1.104,P <0.01),TC (b =1.082,P <0.01) diabetes mellitus (b =0.956,P <0.01),baPWV increased (b =0.741,P <0.01),WBC (b=0.721,P <0.01),hypertension (b =0.602,P <0.01),the age (b =0.538,P <0.01),and HsCRP(b =0.421,P < 0.01).Conclusions The results suggest that the hypertension,hyperlipidemia,smoking,age,baPWV,inflammatory cells,Hs-CRP,and carotid artery plaque was a significant independent CHD risk factors.
9.Plasma-exosomal miR-18a-3p regulates CITED2 expression to affect cumulus cells apoptosis and cell cycle distribution in polycystic ovary syndrome

Objective:To investigate the effect of plasma exosome miR-18a-3p on of apoptosis and cell cycle cumulus cells (CCs) in polycystic ovary syndrome (PCOS) via targeting CITED2.Methods:qRT-PCR assay was used to detect the expression of miR-18a-3p in plasma, plasma-derived exosome and CCs of patients no matter with PCOS and non PCOS. After being transfected with miR-18a-3p mimic, the exosome was co-cultured with CCs, the cell cycle distribution and apoptosis of CCs were detected by flow cytometry assays. Gene ontology (GO) analysis was performed and CITED2 was included in follow up experiments. pcDNA3.1-CITED was transfected into CCs and then co-cultured with exosome to explore the co-effect of miR-18a-3p and CITED2 on the cell cycle and apoptosis of CCs.Results:The plasma-derived exosome isolated from CCs with PCOS were identified successfully, and the expression of miR-18a-3p was significantly decreased in plasma, exosome and CCs in PCOS, compared with that in non-PCOS (all P<0.05) . CITED2 could be regulated as a target of miR-18a-3p in CCs. Compared with NC group, overexpression of miR-18a-3p could significantly decrease proportion of CCs cells in G0/G1 phase and inhibit their apoptosis in PCOS patients (all P<0.05) . The effect of over-expressing miR-18a-3p could be partially reversed by up-regulating CITED2 (all P<0.05) . Conclusions:Plasma exosomal miR-18a-3p has the effect to induce the S phase of CCs cells, subsequently inhibit apoptosis, then restrain the progression of PCOS. Plasma exosomal miR-18a-3p is expected to play a paramount role in the target therapy of PCOS.
10.Association between Toll-like receptor gene polymorphism and susceptibility to non-human immunodeficiency virus-related cryptococcal meningitis in patients from Fujian Province

Objective:To explore the association between Toll-like receptor (TLR) gene polymorphism and susceptibility to non-human immunodeficiency virus (HIV)-related cryptococcal meningitis in patients from Fujian Province.Methods:A total of 101 non-HIV patients with cryptococcal meningitis and 270 healthy controls in Huashan Hospital, Fudan University and Cangshan Hospital District, The 900th Hospital of the Joint Logistics Support Force from Fujian Province were prospectively enrolled in this case-control study. Genomic DNA was extracted. Genotyping was performed by multiplex SNaPshot technology on eight TLR single nucleotide polymorphism (SNP) which were reported to be related to cryptococcal meningitis but still lacking validations. The differences of gene polymorphism distributions were compared between all patients and healthy controls, and between patients without predisposing factors and healthy controls. Data were analyzed by chi-square test or Fisher exact test.Results:Except for TLR1 rs5743563, the distributions of allele frequency in seven tested TLR SNPs (TLR1 rs5743604, TLR2 rs3804099, TLR4 rs1927907, TLR6 rs3796508, TLR6 rs5743794, TLR9 rs164637 and TLR9 rs352140) were in Hardy-Weinberg equilibrium. Comparisons between cases and controls found that TLR2 rs3804099 T/T genotype (52.5%(53/101) vs 40.4%(109/270), odds ratio ( OR)=1.63, χ2=4.378, P=0.036) and TLR6 rs5743794 G/G genotype (44.6%(45/101) vs 32.2%(87/270), OR=1.69, χ2=4.877, P=0.027) were correlated with high risks of cryptococcal meningitis, while TLR6 rs3796508 G/G genotype ((83.2%(84/101) vs 92.6%(250/270), OR=0.40, χ2=7.271, P=0.007) and TLR9 rs164637 C/C genotype (96.0%(97/101) vs 100.0%(270/270), Fisher exact test , P=0.005) were found to be protective factors. Seventy out of 101 patients had no predisposing factors. Comparison between patients without predisposing factors and healthy controls also found similar results. TLR6 rs5743794 G/G genotype (52.9%(37/70) vs 32.2%(87/270), OR=2.36, χ2=10.216, P=0.001) was risk factor, while TLR6 rs3796508 G/G genotype (81.4%(57/70) vs 92.6%(250/270), OR=0.35, χ2=7.906, P=0.005) and TLR9 rs164637 C/C genotype (97.1%(68/70) vs 100.0%(270/270), Fisher exact test, P=0.042) were protective factors. Conclusion:TLR gene polymorphism is significantly associated with non-HIV-related cryptococcal meningitis, which indicates that TLR might play an important role in the pathogenesis of cryptococcal meningitis.
