1.Effects of Different Times of Needle Retention on Cognitive Function in Vascular Cognitive Impairment No Dementia

Objective To investigate the effects of different times of needle retention on cognitive function in patients with vascular cognitive impairment no dementia (VCIND). Methods One hundred and four VCIND patients were randomly allocated to group A of 35 cases, group B of 34 cases and group C of 35 cases. All the three groups received acupuncture plus medication. Of them, group A was given scalp acupuncture with 30 min retention of needles;group B, with 1 hr retention of needles;group C, with 10 hrs retention of needles. The Montreal Cognitive Assessment (MoCA) score and the ADL score were recorded in the three groups of patients before and after treatment. Results There were statistically significant pre-/post-treatment differences in the MoCA score and the ADL score in every group (P<0.05). There were statistically significant post-treatment differences in the MoCA score and the ADL score between group A or B and group C (P<0.05). Conclusions Acupuncture is an effective way to treat VCIND. It can improve cognitive function in the patients. Scalp acupuncture with 10 hrs retention of needles has a more marked effect.
2.Construction of hIAP-2 siRNA plasmid and its effects on breast cancer cell line MCF-7

Objective:To construct an expression plasmid of siRNA against gene hIAP-2 and to assess its effect on breast cancer cell line MCF-7.Methods: A hIAP-2 siRNA plasmid was constructed with mU6pro vector containing U6 promotor and its RNAi effect on MCF-7 cells was detected by RT-PCR and Western blot.The effects of the plasmid on MCF-7 proliferation were analyzed by MTT assay,on the cell cycle by flow cytometry,and on the cell morphology by Hoechst staining.The activity of caspase-3 was determined with its substrate Ac-DEVD-pNA and the expression of some related proteins was detected by Western blot.Results: The hIAP-2 siRNA plasmid knocked down hIAP-2 expression in MCF-7 cells specifically and obviously,inhibited tumor cell proliferation and arrested the MCF-7 cells at G_(1) phase.After hIAP-2 gene was silenced,MCF-7 cells became multinuclear and had macronucleus;procaspase-3 was activated and its expression was increased.The protein levels of I?B?(p37) and p21~(waf1) increased,of NF-?B(p65) decreased and Cyt C remained unchanged.Conclusion: hIAP-2 in MCF-7 cells plays a crucial role in cell proliferation and cell cycle progression.Mitotic cell death might be the main way of the cell death;G_(1) arrest after DNA damage might be associated with the p21~(waf1) increase.

4.The analysis CYBB gene mutation and prenatal diagnosis in X-linked chronic granulomatous disease

Objective To analyze the clinical feature of X-linked chronic granulomatous disease (X-CGD) and gene mutation of CYBB. Method The clinical data of X-CGD in one child and the results of CYBB gene detection in his family were reviewed. Results This boy had onset in the neonatal period and presented with recurrent severe pulmonary infection as his main manifestation. Results of nitroblue tetrazolium test (NBT) in both non-stimulation group and LPS stimulation group in the child were 0, and neutrophil oxidation index (NOI) was 1.15. Gene analysis showed a deletion mutation in exon 6 of CYBB gene in the child (579-582delATTA), which resulted in frameshift mutation started from coding sequence of 189—isoleucine (I) and stop codon occured in advance in the 212th amino acid (I189fsX212). Both the child's mother and grandmother were carriers of the mutated gene. The same deletion mutation was not found in the CYBB gene in the amniocyte from the mother's next child. Conclusion One case of X-CGD patient with CYBB gene mutation and his families were diagnosed by gene detection. Prenatal diagnosis can avoid the birth of children with X-CGD.
5.Application of systems biology method in the research of traditional Chinese medicine

Systems biology is a new science of the 21st century, which resembles traditional Chinese medicine (TCM) in many aspects such as study method and design. Adoption of systems biology approach would do much help for exploring the scientific connotation of TCM syndrome and the modernization of Chinese herbal medicine. Its technological platforms, such as genomics, proteomics and metabonomics, provide powerful tools for the study of the essence of TCM syndrome and the function of herbal compound recipe. Compared with other molecular biological methods, such as genomics and proteomics, metabonomics method is more direct and more concise, especially for providing an effective method for the study of TCM therapy in preventing and treating kidney diseases.
6.Changes of nitric oxide synthase in hippocampus of rats tolerant to and dependent on the anticonvulsant action of flurazepam

Aim To study the role of nitric oxide by measuring the expression and activity of nitric oxide synthase in hippocampus of rats tolerance and dependence to the anticonvulsant action of flurazepam.Method Animal models of toleran and dependence to flurazepam were established. Nitric oxide synthase in hippocampus was measured using Western blot and immunohistochemistry, and its activity was evaluated using colorimetry. Result Hippocampal nNOS increased in rats tolerance to flurazepam. In rats dependence to flurazepam, hippocampal nNOS increased but without statistical significance. Total NOS activity increased dramatically both in flurazepam tolerane and dependene rats.Conclusion NO may play a role in rats tolerance and dependence to the anticonvulsant action of flurazepam.
7.The Immediate Treatment of Acute Primary Angle Closure Glaucoma with High Blood Pressure

Objective To investigate the effects on intraocular pressure and blood pressure following anterior chamber paracentesis in acute primary angle closure glaucoma with high blood pressure.Methods 64 eyes of 64 glaucoma patients with high blood pressure were received anterior chamber paracentesis.the intraocular pressure and blood pressure were measured before and after paracentesis.Results The mean systolic blood pressure(SBP) and diastolic blood pressure(DBP) before paracentesis of 64 glaucoma were 195.10?15.24mmHg and 117.1?11.76mmHg respectively,after administration of paracentesis,the SBP and DBP were 138.82?20.86mmHg and 93.19?12.07mmHg respectively,the intraocular pressure also significantly decreased from 68.64?10.05mmHg to 10.66?4.41mmHg.There was statistical difference in SBP,DBP and intraocular pressure before and after administration of paracentesis.Conclusions Administration of paracentesis in acute primary angle closure glaucoma with high blood pressure can significantly reduce intraocular pressure and blood pressure simultaneously.
8.Improvement of Sitagliptin on Islet B-cell Function in Patients with Type 2 Diabetes:A Meta-analysis

OBJECTIVE:To evaluate the improvement effects of sitagliptin on islet B-cell function in type 2 diabetic patients systematically,and to provide evidenced-based reference. METHODS:Retrieved from PubMed,Cochrane library,EMBase, CJFD,Wanfang database,VIP and CBM,RCTs about sitagliptin alone or combined with routine plan(trial group)vs. placbo alone or combined with routine plan(control group)in the treatment of type 2 diabetes were collected. Two reviewers independent-ly screened studies according to exclusion and inclusion criteria,extracted data,and assessed the methodological quality according to Cochrane Manual 5.1.0. Meta-analysis was performed by using RevMan 5.3 software. RESULTS:A total of 5 RCTs were includ-ed,involving 1253 patients. The result of Meta-analysis showed that changes of islet B cell function index(Homa-B)[sitagliptin alone group:MD=9.21,95%CI(4.16,14.25),P<0.001;drug combination group:MD=7.24,95%CI(0.80,13.68),P=0.03] and changes of insulin resistance index(Homa-IR)[sitagliptin alone group:MD=-0.40,95%CI(-0.44,-0.36),P<0.001;drug combination group:MD=-0.35,95%CI(-0.63,-0.07),P=0.02] of trial group were significantly better than those of control group,with statistical significance. CONCLUSIONS:Sitagliptin shows certain therapeutic efficacy in improving islet B cell func-tion and insulin resistance.
9.The application of the theory of cognition, emotion, will and behavior in research integrity educational practice for the medical postgraduate students:based on experiences from United States

Objective The study analyzes the inadequacies of the research integrity educational practice for the medical postgraduate students in the present stage of China.Furthermore,the paper puts forward improvement suggestions and clears the direction for further efforts.Methods From the perspective of the educational law about cognition,emotion,will and behavior,the comparative analysis method is used,based on the rich experience in the United States.Results There are inadequacies of the research integrity educational practice for the medical postgraduate students in China,including that students lack knowledge learning motivation,and the pattern of education is simple.There are not enough emotional experience channels and the participation of students.Students lack honesty will training,and the intensity of supervision and punishment is not strong.The honesty recognition mechanism is weak,and cannot be used to guide practice.Conclusions In order to improve the level of the research integrity for the medical postgraduate students in China,we should optimize the system,follow the law of education,strengthen the transmission of knowledge,put the academic honesty education into normal work,enhance the emotional experience,build a participatory mechanism,highlight the positive guide,and increase the intensity of punishment.
10.Changing rule of epidermal stem cells in the survival of autologous full-thickness skin graft

BACKGROUND: Study on epidermal stem cells (ESCs) is satisfying. However, the amount and the distribution dynamic changing rule in each skin layer of ESCs during survival of skin graft need to be investigated.OBJECTIVE: To observe the changing rule of ESCs in the microenvironment during the survival of full-thickness skin graft.DESIGN: Prospective animal experiment based on controlled observation.SETTTNG: Department of Burn and Plastic Surgery, First Hospital Affiliated to Nanhua University.MATERIALS: Forty-two Wistar rats of clean grade and either gender, aged 2 to 3 months, weighing about 200 to 250 g,were provided by the Experimental Animal Center of South China University. After being raised for 1 week, the rats were divided into 7 groups according to observation time: post-operation 24 hours, 3 days, 5 days, 7 days, 14 days, 21 days and 30 days groups, with 6 rats in each group. Primary antibody: rabbit anti-rat integrinβ1 polyclonal antibody (Boster Biological Company, Wuhan); mouse anti-rat p63 monoclonal antibody (Santa cruz Company); Second antibody: goat anti-rabbit lgG (Zhongshan Biological Company, Beijing), goat anti-mouse lgG (Zhongshan Biological Company, Beijing);S-P immunohistochemical kit and DAB kit (Zhongshan Biological Company, Beijing).METHODS: This experiment was carried out in the Animal Laboratory of Nanhua University in May 2006. ①An area of (1.5×1.5) cm2 autologous full thickness skin graft model was made on the back of the all the rats. The color change of skin graft was observed at different time points. Meanwhile, whether hematocele and fluidify as well as binding to wound appeared under the skin graft were investigated. ②During the operation, a few skin graft was taken from each rat and immediately fixed with 100 g/L neutral formaldehyde solution and labeled, serving as blank control group. ③Skin graft packages of rats in the post-operation 24 hours, 3 days, 5 days, 7 days and 14 days were open at corresponding time points. Skin tissues were cut from the center of each wound and labeled; Skin graft packages of rats in post-operation 21 days and 30 days groups were open at postoperative 14 days. Rats in the two groups were raised till post-operation 21 days and 30 days, respectively, and skin tissue was cut from the center of skin graft. ④Pathological sections were made for haematoxylin and eosin (HE) staining and immunohistochemical staining.MAIN OUTCOME MEASURES: ①The amount of inflammatory cells, epithelization degree and the level of fibrocytes in the section. ② Integrin β1 and gene p 63 positive cell rate.RESULTS: Forty-two Wistar rats were involved in the final analysis. ①Gross observation of wound: At postoperative 1 to 2 days, the skin graft was pale and edematous; At postoperative 3 days, the color of skin graft did not turn better, but skin graft closely attached to the wound; At postoperative 7 days, the color of skin graft obviously turned better,epidermal color was close to normal, skin graft more closely attached to the wound, and some hair roots grew out of the wound; At postoperative 14 days, wound border and wound had healed completely; At postoperative 30 days, the color of skin graft became darker than normal, and the hair grew in the wound as in the normal skin. ②Observation results of HE stained pathological sections: Cellular edema and small quantity of inflammatory cell infiltration were found in the skin graft of the former 4 groups; Partial keratinocytes were necrotic and fell off at postoperative 5 days. ③Changing rule of distribution and amount of positive cells: Before operation, β1 integrin and gene p63 positive cells were mainly distributed in the epidermal basal membrane and eminence of hair follicle. Positive cells appeared in each epidermal layer from postoperative 24 hours and gradually increased. At postoperative 14 days and 21 days, many positive cells appeared in each epidermal layer. At postoperative 30 days, positive cells were distributed mainly in the basal layer of epidermis, and slightly in the other layers. Among the immunohistochemical sections, genic substance p63 positive cells were more thanβ1integrin positive cells at the same time point. p63 positive cells began to decrease from postoperative 24 hours,reached the lowest level at postoperative 3 days, then were gradually increased, reached or were close to the preoperative level at postoperative 7 days, reached the highest level at postoperative 21 days, subsequently were gradually decreased, and were still higher than preoperative level at postoperative 30 days.CONCLUSION: ①During the survival of full-thickness skin graft, ESCs are firstly decreased, then gradually increased, and even over the level in the normal skin. ②During the survival of full-thickness skin graft, firstly, ESCs arrange disorderly in the skin, and then they gradually gather in the epidermal basal membrane and eminence of hair follicle.
