1.PCR test of Helicobacter pylori and“Helicobacter macacae”infections in rhesus and cynomolgus monkey breading colonies

Objective To investigate the status of Helicobacter pylori and “Helicobacter macacae” infection in rhesus and cynomolgus monkeys in China.Methods With the use of 16S rRNA specific primers for Helicobacter spp and Helicobacter pylori ( HP ) from published literatures, and new 16S rRNA specific primers designed for “Helicobacter macacae” ( HM ) , we investigated the infection status of these two Helicobacter spps in both of 45 rhesus and 90 cynomolgus monkeys by qPCR or conventional PCR on stool samples.Results All three primer sets for 16S rRNA exhibited excellently sensitivity and specificity.Both the infection rates of HP and HM were 100% among 45 young adult rhesus monkeys.The infection rate of HP and HM in 90 young adult cynomolgus monkeys were 100% and 97.8%, respectively.Conclusions Helicobacter pylori and “Helicobacter macacae” are present in almost every artificially bred adult rhesus and cynomolgus individuals which may adversely affect the health of laboratory monkeys and the accuracy of related animal experiments.

3.The effects of IL-24 on the differentiation and function of osteoclasts

Objective:To study the effect of IL-24 on the differentiation and function of osteoclasts.Methods:Mature osteoclasts were isolated from long bones of neonate rats.Optimal multiplicity of infection(MOI)of AdCMV-EGFP was determined.Then osteo-clasts and RAW264.7 cells were transfected with AdCMV-EGFP and AdCMV-IL-24 respectively.The function of osteoblasts was studied by the observation of bone resorption lacunae,the differentiation of RAW264.7 cells was evaluated by the expression of osteo-clast related genes examined with real-time PCR.Results:Osteoclasts were TRAP positive with more than 2 neclei.MOI=400 was suitable for AdCMV-EGFP transfection.The resorption lacunae area in AdCMV-IL24 transfected cells was larger than that in AdCMV-EGFP transfected cells(P<0.05).Real-time PCR showed that under induced conditions,osteoclastic related genes NFATc1,CTSK and TRAP were changed in RAW264.7 cells transfected with AdCMV-IL-24.Conclusion:IL-24 may promote the differentiation and function of osteoclasts.
4.Quantification of left ventricular mechanics using vector-velocity imaging, a novel feature tracking algorithm, applied to echocardiography and cardiac magnetic resonance imaging.

BACKGROUNDTagged magnetic resonance imaging (MRI) is the non-invasive golden standard to measure myocardial deformity. Tissue Doppler Imaging can be used to assess myocardial deformity, however, it has the limitation of angle-dependence. Our study aimed to compare left ventricular torsion and strains measured by velocity-vector imaging (VVI) using echocardiography (echo-VVI) and MRI (MRI-VVI), and to validate them against harmonic phase tagged MRI (HARP MRI).
METHODSA total number of 34 subjects (14 normal and 20 patients) were evaluated. Apical and basal image of left ventricular short axis view were acquired for measurements of apical and basal rotation, circumferential and radial strain using both echo-VVI and MRI-VVI. An apical four-chamber view was obtained for measuring the distance between the apical and basal levels.
RESULTSThe correlations of segmental rotations, circumferential and radial strains were high between echo-VVI and HARP MRI, while the agreement of apical rotation was poor. Left ventricular torsion showed much better correlation and agreement between echo-VVI and HARP MRI than apical rotation: the coefficient was 0.97, P < 0.001. The correlation between MRI-VVI and HARP MRI in quantifying rotational parameters and strains was similar with echo-VVI and HARP MRI. Echo-VVI could discriminate normal and dysfunctional ventricles on either hypertensive or dilated cardiomyopathy.
CONCLUSIONThe data from this study show that (1) it is feasible to quantify left ventricular torsion and myocardial strain using echo-VVI and MRI-VVI in normal subjects, patients with left ventricular global systolic dysfunction and segment systolic dysfunction; (2) the agreement among all mechanical parameters derived from echo-VVI, MRI-VVI, and HARP MRI remained with clinically acceptable ranges.

5.Circular RNA circ_100367 as a diagnostic marker for thyroid cancer and its relationship with immune-related factors

Objective:To investigate the diagnostic value of circular RNA circ_100367 in thyroid cancer (THCA) and its relationship with immune-related factors.Methods:According to the data chip provided by the National Center for Biotechnology Information (NCBI) website, the differentially expressed circRNAs in THCA were analyzed, then circ_100367 was included in this study. The serum of 175 THCA patients and healthy people were collected, and the expression levels of circ_100367 and its linear transcript DCAF8 mRNA in serum samples were detected by qRT-PCR, and the correlation between circ_100367 and DCAF8 was calculated. The correlation between the expression of circ_100367 and the clinicopathological characteristicsof the patients, immune infiltration level and immunosuppressive factor PD-1 was analyzed.Results:Compared with serum of healthy people (1.00±0.37) , expression level of circ_100367 in serum of THCA patients was significantly increased (1.37±0.41) ( t=8.80, P<0.001) , and there was no significant difference in DCAF8 mRNA expression ( t=1.67, P=0.095) , but circ_100367 was positively correlated with DCAF8 mRNA expression ( r=0.17, P=0.028) . Analysis of expression and clinicopathological characteristics of circ_100367 showed that compared with patients in M0 group (1.26±0.40) , circ_100367 was overexpressed in M1 and Mx patients (1.43±0.40) ( t=2.63, P=0.009) ; compared with N0 patients (1.24±0.36) , circ_100367 was overexpressed in serum of N1 and Nx patients (1.45±0.42) ( t=3.48, P=0.001) ; compared with serum of patients with negative lymph node detection (1.28±0.36) , circ_100367 was overexpressed in serum of positive patients (1.42±0.43) ( t=2.14, P=0.034) ; compared with T1+T2 stage patients (1.30±0.37) , circ_100367 expression was overexpressed in serum ofT3+T4 patients (1.40±0.43) ( t=2.22, P=0.028) . Analysis of the expression and immune infiltration levels of circ_100367 found that highly expressed circ_100367 was associated with CD8+ T cells ( r=0.25, P=0.024) , macrophages ( r=0.22, P=0.038) , CD4+ T cells ( r=0.25, P=0.020) and B cell ( r=0.23, P=0.033) levels. The expression of circ_100367 was also positively correlated with the immunosuppressive factor PD-1 ( r=0.19, P=0.011) . Conclusion:circ_100367 can be used as a marker for the diagnosis of THCA and its expression is strongly correlated with immune-related factors.
6.Expression and significance of NT-proBNP,GFAP and HIF-1α in inflammatory injury in patients with cerebral hemorrhage

Objective To investigate N terminal pro B type natriuretic peptide (NT-proBNP),glial fibrillary acidic protein (GFAP)and hypoxia inducible factor-1 (hypoxia inducible factor-1α,HIF-1α) expression and there significance in inflammatory injury in patients with cerebral hemorrhage.Methods We selected 80 patients with acute intracerebral hemorrhage treated in our hospital from January 2018 to December 2019 as the observation group and 80 healthy people who underwent physical examination in our hospital in the same period as the control group.After routine treatment,NT-proBNP,GFAP and HIF-1α in patients serum and cerebrospinal fluid were detectedα、Tumor necrosis factor-α(TNF-α) interleukin-6 (IL-6),and correlation analysis was carried out.Results NT-proBNP,GFAP and HIF-1α in serum and cerebrospinal fluid of patients in the observation group、TNF-α and IL-6 levels in the control group were higher than those in the control group (P<0.05).NT-proBNP,GFAP and HIF-1α in serum and cerebrospinal fluid of the observation group after treatment,as well as TNF-α and IL-6 levels were lower than those before treatment (P<0.05).NT-proBNP,GFAP and HIF-1α in serum and cerebrospinal fluid increased with the aggravation of the disease、TNF-α and IL-6 levels were positively correlated with the degree of intracerebral hemorrhage (P<0.05).NT-proBNP,GFAP and HIF-1α in serum and cerebrospinal fluid were associated with inflammatory injury in intracerebral hemorrhage.There was a significant positive correlation with IL-6 level (P<0.05).NT-proBNP,GFAP and HIF-1α.It was closely related to inflammatory injury in patients with intracerebral hemorrhage and was an independent risk factor for intracerebral hemorrhage complicated with inflammatory injury (P<0.05).Conclusion The levels of NT-proBNP,GFAP and HIF-1α are closely related to inflammatory injury and prognosis in patients with cerebral hemorrhage.The detection of NT-proBNP,GFAP and HIF-1α levels has certain clinical value in judging the disease and prognosis.
7.Effect of Fangji Fulingtang on Macrophage Polarization and Oxidative Stress in Mouse Model of Myocardial Fibrosis

ObjectiveTo investigate the effects of Fangji Fulingtang on macrophage polarization and oxidative stress in the mouse model of myocardial fibrosis. MethodThe mouse model of myocardial fibrosis was established by subcutaneous injection of isoproterenol (ISO, 5 mg·kg-1·d-1). Fifty C57BL/6J mice were randomly assigned into control (0.9% NaCl), model (0.9% NaCl), low- and high-dose (3.315 g·kg-1·d-1 and 13.26 g·kg-1·d-1, respectively) Fangji Fulingtang (FFD-L and FFD-H, respectively), and metoprolol tartrate (Meto, 15 mg·kg-1·d-1) groups, with 10 mice each group. After 2 weeks of treatment, the heart appearance, cardiac weight index (CWI), heart weight (HW)/tibia length (TL) ratio, and myocardial histopathological alterations were observed. Meanwhile, the serum levels of creatine kinase-MB (CK-MB), transforming growth factor-β1 (TGF-β1), tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-6, IL-10, malondialdehyde (MDA), superoxide dismutase (SOD), and glutathione (GSH) were measured by enzyme-linked immunosorbent assay (ELISA). The expression levels of CD86 and CD206 were observed by immunohistochemical staining. ResultCompared with the model group, the FFD-L, FFD-H, and Meto groups showed improved heart appearance, decreased CWI and HW/TL ratio (P<0.01), lowered serum levels of CK-MB, TGF-β1, TNF-α, IL-1β, and IL-6 (P<0.05, P<0.01), and elevated IL-10 level (P<0.05). Furthermore, the three groups showed reduced infiltration of inflammatory cells, myocardial injury, collagen deposition, and myocardial fibrosis, decreased CD86, SOD, and GSH (P<0.01), and increased CD206 and MDA (P<0.01). ConclusionFangji Fulingtang can mitigate ISO-induced myocardial fibrosis by regulating macrophage polarization and oxidative stress.
8.Preliminary establishment of a novel localization method for sacral nerve foramen puncturing

【Objective】 To establish a novel localization method for sacral nerve foramen puncture by analyzing the characteristic of sacral nerve foramen trying to help improve the success rate of sacral foramen puncture. 【Methods】 Clinical data and sacrococcyx CT and three-dimensional reconstruction imaging data of 158 patients who received sacral nerve modulation (SNM) during Jan.2019 and Aug.2022 in our hospital were retrospectively analyzed.The distance between inferior margin of articulatio sacroiliaca and the internal edge of the 3rd neural foramen (D1), and the distance between the internal edge of the 3rd neural foramen and sacral midline (D2) were measured, and the ratio of D1 and D2 was calculated for precise intraoperative positioning. The measurement data characteristic and puncture results were analyzed. 【Results】 A total of 89 males and 69 females were included, with an average age of (49.0±16.9) years.The average D1, D2, and D1/D2 were (29.6±4.9) mm, (13.8±3.2) mm, and (2.2±0.6), respectively.Female patients had greater D1 \[(30.7±5.5) mm vs.(28.7±4.2) mm, P=0.010\] and D1/D2 \[(2.4±0.7) vs.(2.1±0.5), P=0.001\] than male patients.Compared with adults, the adolescents had smaller D1 \[(29.8±4.7) mm vs.(25.7±5.4) mm, P=0.006\].After precise intraoperative positioning using this positioning method, 158 patients were successfully punctured, 152 (96.20%) had excellent intraoperative neural response, 2 (1.27%) had good response, and 4 (2.53%) had average response. 【Conclusion】 The surface projection of sacral nerve was approximately at the middle-inner 1/3 of the inferior margin of articulatio sacroiliaca to sacral midline.Through measuring distance from inferior margin of articulatio sacroiliacato sacral midline via X-ray, sacral nerve situation could be performed in surface, which might be a secure way to accessorily situate electrode implantation site for sacral neuromodulation.
