1.Effect of CFSE labeling on proliferation, differentiation and secretion of rabbit bone marrow mesenchymal stem cells

Objective To explore the experimental conditions of labeling rabbit bone marrow mesenchymal stem cells (Rb-MSCs) with 5,6-carboxyfluorescein diacetic succinimidyl ester (CFSE) and to investigate the impact on the biological characteristics of Rb-MSCs in vitro.Methods Rb-MSCs were separated and purified by whole bone marrow adherent culture and then were identified by morphology and surface markers.Rb-MSCs were labeled with CFSE and the labeling effect was measured by flow cytometer.The proliferation capacity of labeled cells was detected with CCK-8.The differentiation capacity of labeled cells was investigated by being induced to osteoblasts and lipoblasts.The capacity of labeled cells to secret vascular endothelial growth factor (VEGF) was detected by VEGF ELISA kits.Results The primary Rb-MSCs adhered in 48 h,being fusiform and colony-like growth.Subculture cells became fibroblast-like cells in order with uniform configuration.Most (above 98%) cultured cells expressed the surface markers CD29 and CD44 except for CD45.Compared with other labeling conditions,10μmol/L final concentration of CFSE and 10 min was the best one with a 100% labeling rate and high fluorescence intensity.Compared with unlabeled cells,the ability of the labeled cells to proliferate and to secrete VEGF was not significantly decreased (P > 0.05).Moreover,the labeled cells had osteogenic and adipogenic differentiation capacity.Conclusions It was a simple and efficient method to label Rb-MSCs with CFSE,especially in a short-term.The capacity of cell proliferation,differentiation,and secretion were not affected.
2.Value of contrast-enhanced ultrasound in analysing testicular and epididymal mass lesions

Objective To investigate the differential value of contrast-enhanced ultrasonography (CEUS) and time-intensity curves(TIC) in diagnosing testicular and epididymal mass lesions.Methods CEUS via intravenous bolus injection of SonoVue and TIC were performed for quantitative analysis of testicular and epididymal mass lesions.Forty-one patients with 42 testicular and epididymal mass lesions and 26 normal testicles were examined with CUES,the perfusion progress were recorded dynamically,which findings were compared with surgery.Results Twenty-three (54.76%) malignant masses displayed enhanced pattern as evenly enhanced,fast-in and fast-out (8.7%),evenly enhanced,fast-in and slow-out (65.2 % ),unevenly enhanced,fast-in and slow-out (26.1% ).Ninteen (45.24 % ) benign masses revealed enhanced pattern as unevenly enhanced,fast-in and slow-out ( 10.5 %),evenly enhanced,slow-in and slowout ( 10.5 % ),unevenly enhanced,slow-in and slow-out ( 36.9 % ),without enhancement ( 42.1% ).There was statistical difference of peak intensity,time to peak and areas among malignant group,benign group and normal group ( P < 0.05).Conclusions CEUS combined with TIC could provide differential diagnostic value for testicular and epididymal mass lesions.
3.Survey of factors associated with deep venous thrombosis after artificial liver treatment in patients with liver failure

Objective To survey the factors associated with deep venous thrombosis after artificial liver treatment in patients with liver failure. Methods A retrospective survey was used in our hospital from January 2014 to December 2016 consecutive liver failure patients by artificial liver treatment, collected patient data, including general demographic information, medical history. Results Medical history data were collected from 189 compliance cases, including deep venous thrombosis group (11 cases),no deep venous thrombosis group(178 cases),and 5.82%(11/189)of deep vein thrombosis.There were significant differences in age(χ2=7.17, P=0.027), catheterization(χ2=4.99, P=0.025), number of successful venipuncture(χ2=10.856, P=0.004),artificial liver frequency(χ2=67.481, P<0.01), activity status(χ2=9.607, P=0.022), D-dimer(t=12.318, P<0.01), infection(χ2=17.231, P=0.001)and other factors in thrombosis group and thrombus group(P<0.05).Logistic regression showed that age(OR=1.643, P=0.01),activity status(OR=1.643,P=0.01),number of successful venipuncture(OR=6.049,P<0.01),D-dimer(OR=2.532, P=0.005)and infection(OR=2.463, P=0.008)were independent risk factors for thrombosis. Conclusions Deep vein thrombosis after artificial liver injury in liver failure is not uncommon, and the prevention of deep vein thrombosis after artificial liver surgery is strengthened, especially for elderly, absolute bed, venous puncture injury patients, elevated D-dimer and infected patients.
