1.Literature analysis of semaglutide-induced adverse drug reaction

OBJECTIVE To explore the characteristics of adverse drug reaction (ADR) induced by semaglutide and provide a reference for clinically safe medication. METHODS Using search terms such as “semaglutide” and “adverse reactions” in both Chinese and English, the case reports about ADRs caused by semaglutide were searched and analyzed from PubMed, Web of Science, SpringerLink, CNKI, Chinese Medical Journal Full-text Database, Wanfang Medical Network and VMIS. RESULTS Overall 14 literature were included, involving 17 patients. Among 17 patients, 9 were female and 8 were male, with the age ranging from 25 to 80 years. Eight patients were given two or more drugs, and eight patients took 0.25 mg as the initial dose; the ADR occurred most frequently within 6 months (94.12%). Sixteen patients’ symptoms improved or recovered after drug withdrawal and symptomatic treatment. Eleven patients did not mention whether to continue to use semaglutide in the future. Nine patients underwent ADR correlation evaluation, and 1, 3, 1 and 4 cases were determined to be “definite”,“ probable”,“ possible”, and “doubtful” respectively. Semaglutide-induced ADRs involved multiple organs or systems, most of which were the digestive system (35.29%), followed by skin tissue (29.41%). Among them, acute gastric dilation, severe liver injury, calculous cholecystitis, bullous pemphigoid, eosinophilic fasciitis, acute kidney injury, acute interstitial nephritis, depression and acute hemolytic anemia were not mentioned in the instruction. CONCLUSIONS ADRs caused by semaglutide can occur in all ages, mainly within 6 months after medication, and mainly involve the digestive system, skin tissue, etc. Great attention should be paid to pharmaceutical care for those patients with liver and kidney dysfunction, neuropsychiatric diseases, glucose-6-phosphate dehydrogenase deficiency, etc. When ADR occurs, drug withdrawal and symptomatic treatment should be performed promptly to ensure patients’ medication safety.
2.Investigation and Analysis of Medication Safety among 24 Medical Institutions in Linyi City

OBJECTIVE: To investigate the current situation of medication safety in 24 public medical institutions(referred to as “hospital”) from Linyi city and the differences in medication safety between urban and rural areas. METHODS: ISMP self-assessment scale [including 10 key elements (Ⅰ-Ⅹ,such as “Ⅰ patient’s information” “Ⅱ drug information”), 20 key characteristics and 270 evaluation projects] developed by Institute of Safe Medication Practices was used to investigate 24 hospitals in Linyi city. The implementation rates of 10 key elements in urban and rural hospitals were analyzed statistically, and the top 10 evaluation projects were listed for the key elements with the lowest implementation rate. The key elements and top 10 evaluation projects with the largest difference in the implementation rate were compared between urban and rural hospitals. Radar maps were used for comparison and analysis intuitively. RESULTS: A total of 24 hospitals were surveyed, including 12 in urban and 12 in rural areas; there were significant differences in the implementation rate of 10 key factors among 24 hospitals; the elements with the highest implementation rate were “Ⅶ environmental factors, workflow and staffing pattern”(56.55%);the elements with the lowest implementation rate was “Ⅱ drug information” (26.77%). Among 33 evaluation projects of “Ⅱ drug information”, the implementation rate of No. 36 project (12.50%, related to opioids) was the lowest. Among the implementation rates of 10 key elements in 12 urban hospitals and 12 rural hospitals, the key elements with the greatest difference was “Ⅳ drug label, packaging and naming” (differing by 44.44%,59.72% vs. 15.28%); Among“Ⅸ patient education”evaluation project with the greatest gap, there was the greatest difference in No. 199 project (related to patients’ active participation in medication, 58.33% in urban, 4.17% in rural). CONCLUSIONS: The results of medication safety investigation in 24 hospitals from Linyi city show that all the item in each hospital needs to be improved expecially in the implementation of “Ⅱ drug information”. Rural hospitals should strengthen medication education for patients.
3.Breeding peanut variety Yuhua 7 by fast neutron irradiation and tissue culture.

Creating new germplasms and breeding new cultivars in peanut by radiation mutagenesis and tissue culture were conducted in this study, aiming to develop new breeding method of peanut. Mature seeds from Luhua 11, the most commonly grown peanut cultivar in Northern China, were treated by fast neutron irradiation. Then the embryo leaflets were separated from the irradiated seeds and inoculated on the media, and the regenerated seedlings were obtained through somatic embryogenesis pathway. The regenerated seedlings were grafted, acclimated and then transplanted into field and the selfed pods were harvested from 83 regenerated plants. The progenies were selected by the pedigree method, and 107 mutants were obtained from the progenies of the 83 regenerated plants. Different mutants showed obvious variation in many agronomic traits, including main stem height, branch number, pod shape and size, seed coat color, inner seed coat color, oil content and protein content etc. Yuhua 7, a new peanut variety with low oil content, early maturity and waterlogging tolerance was obtained. The yield of Yuhua 7 was over 14% higher than that of the mutagenic parent Luhua 11, and the oil content of kernels was 47.0%, lower than that of parent Luhua 11 with 52.1% oil content. Yuhua 7 had passed peanut variety regional multi-location trials in Liaoning Province in 2016 and its average yield was 13.8% higher than that of the control variety Baisha 1017. It had also passed national peanut variety registration, and the registration ID is "GPD peanut (2018) 370105". The results show that irradiation mutagenesis combined with tissue culture is an effective method for creating new germplasm and breeding new varieties of peanut.
