1.Clinical analysis of laparoscopy combined with deep hyperthermia in the treatment of pelvic absces

Objective To explore the clinical effect of laparoscopy combined with deep hyperthermia in the treatment of pelvic abscess.Methods Clinical data of 56 patients with pelvic abscess treated by laparoscopy combined with deep hyperthermia were retrospectively analyzed.Results 56 cases were completed in laparoscopic surgery,before surgery,after diagnostic accordance rate was 76.8%,average operation time was 51 minutes,postoperative average hospitalization was 5.1 days.After operation they were performed in vitro hyperthermia to increase the efficacy. Patients were followed up for 6 months,no recurrence.32 patients with fertility requirements of the fallopian tube,1 to 3 months after the operation of bilateral fallopian tube radiography,20 cases of patency,8 cases of tubal patency and poor.Conclusion Laparoscopy combined with deep hyperthermia in vitro is an ideal and safe treatment,which is conducive to protect the patients'reproductive function,especially for young patients with no pregnancy.
2.Exploration and Practice of Pharmaceutical Equipment & Workshop Design by Using the Task-driven Teaching

OBJECTIVE:To provide reference for the improvement of Pharmaceutical Equipment & Workshop Design teach-ing. METHODS:Task-driven teaching was adopted,CIDO engineering educational model was referred to,task was designed from the conception to run with task-based;theoretical and teaching exploration and practice was carried out by cross-learning of individ-uals and groups,applying of pilotscale experiment platform,developing course design and forming evaluation. RESULTS & CON-CLUSIONS:It changed the traditional teaching method that emphasized on class teaching to make students participate in multi-task learning,so that students can better grasp the equipment selection,understanding and application of workshop design in pharmaceu-tical process. The method gives full play to students' initiative and creativity and it is conducive to students’comprehensive ability for adapting the development of pharmaceutical industry.
3.Thinking and method of "Combinatorial Screening" based on complex systems principle for active components of Chinese materia medica compound

According to two theories,which are "traditional Chinese medicine researching is in the face of a complex opening gigantic system" and "Chinese materia medica compound(CMMC) is the natural combinatorial chemical libraries",the study on active fractions of CMMC has been facing to a complex system with a large number of data that is non-linear,multi-variable,and varieties related to each other.The thinking and method of "combinatorial screening" has been put forward based on these two theories,that is the principle "using the diverse structures of molecules to simulate the diverse organism" used in combinatorial chemistry and the data mining technique was taken into the study on complex system of CMMC.In order to create combinatorial libraries,the two methods were used to obtain the fractions of CMMC.Re-combination group of the CMMC was used,and various industrial separation techniques, such as resin,membrane,were used to replace the diversity of chemosynthesis routes to create "Building Block".Through data mining and knowledge discovery, the intrinsic correlation of CMMC "multi-components and multi-targets" between the chemical composition and the mechanism was carried out.The new technologies which aim at high-throughput screening of CMMC were explored,in order to provide the evidence for setting up an idea with "optimal separation" and optimization design about application system of refining technology.
5.Preparation and characterization of tetrandrine-loaded PLGA nanocomposite particles by premix membrane emulsification coupled with spray-drying method.

For effective inhalable dry-powder drug delivery, tetrandrine-PLGA (polylactic-co-glycolic acid) nanocomposite particles have been developed to overcome the disadvantages of nanoparticles and microparticles. The primary nanoparticles were prepared by using premix membrane emulsification method. To prepare second particles, they were spray dried. The final particles were characterized by scanning electron microscopy (SEM), dry laser particle size analysis, high performance liquid chromatography (HPLC), X-ray diffraction (XRD), differential scanning calorimetry (DSC), infrared analysis (IR) and confocal laser scanning microscope (CLSM). The average size of the primary particles was (337.5 ± 6.2) nm, while that second particles was (3.675 ± 0.16) μm which can be decomposed into primary nanoparticles in water. And the second particles were solid sphere-like with the drug dispersed as armorphous form in them. It is a reference for components delivery to lung in a new form.
6.Comparision of adsorption-desorption dynamics of huanglian jiedu decoction using different macroporous resins combinations.

OBJECTIVETo establish the techniques for the purification of Huanglian Jiedu decoction by macroporous resins combinations, and to lay the foundation of improvement of dosage forms in clinical application and realization of industrialized production about Huanglian Jiedu decoction.
METHODFour kinds of macroporous resins (X-5, S-8, HPD100, AB-8) were chosen and combined in six different forms. Adsorption and desorption experiments were carried out and contents of six index constituents, gardenoside, baicalin, palmatine, berberine, baicalein and wogonin, in different groups were compared before and after purification.
RESULTAdsorption rates of six index constituents in Huanglian Jiedu decoction of different combinations of macroporous resins groups were all nearly 100%, but desorption rates had remarkable differences.
CONCLUSIONThrough comprehensive comparison, macroporous resins combination 6 resulted in high contents of index suitable relatively. It is suitable for purification of Huanglian Jiedu decoction.

7.Preliminary study to investigate dynamic extract process of Huanglian Jiedu Tang and the mechanism of subsidence produce.

OBJECTIVETo study the phenomenon of subsidence emergence in the process of Huanglian Jiedu Tang decoction extraction, and the mechanism of subsidence emergence.
METHODUV was applied to determine the concentration of total alkaloids and total flavones; Simultaneous determination of berberine, jatrorrhizine, palmatine, baicalin and geniposide were carried out by HPLC; The solid holdup and precipitation rate were calculated by the formula. The relativity among these parameters was analyzed by the SPSS software program.
RESULTThe contains of berberine, jatrorrhizine, palmatine in total alkaloids showed a good correlation with total alkaloids. The correlation between baicalin and total flavones was lower than that between geniposide and total flavones. Compared to precipitation rate, solided hold up shows a larger relevance with index component
CONCLUSIONWith the change of time, the total alkaloids represented by berberine alkaloids and baicalin at a certain concentration can be regarded as the equilibrium point, or one generated by the critical point of precipitation, the reaction can generate "sediments"; because "precipitation objects" generated, reducing the concentration of the above-mentioned components, destroy the" balance". The relevant components of herbal medicine increased dissolution rate, at the same time might partly dissolved sediment, reaching a new equilibrium state.

8.Impact of patient-centered care services on the treatment compliance among patients with multidrug-resistant or rifampicin-resistant pulmonary tuberculosis

Objective To examine the effect of patient-centered care services on compliance to treatment among patients with multidrug-resistant (MDR) or rifampicin-resistant (RR) pulmonary tuberculosis (PTB), so as to provide the scientific evidence for promoting the widespread application of the appropriate nursing process of MDR/RR-PTB patients in the hospital. Methods The MDR/RR-PTB patients that were definitely diagnosed at the Sixth People’s Hospital of Nantong City during the period from January 2017 to October 2020 were enrolled. The patients with confirmed diagnosis of MDR/RR-PTB during the period January 2017 to December 2018 served as controls, who were given routine care in the hospital, and those with confirmed diagnosis of MDR/RR-PTB during the period January 2019 to October 2020 served as the care group, who were given patient-centered personalized care services, including one-to-one consultations, periodic group activities, informatization case management, and personal reminder for return visits. The proportion of inclusion into treatment, loss to follow-up, return visits and sputum examinations were compared between the care and control groups. Results A total of 104 MDR/RR-PTB patients were included, including 54 cases in the control group and 50 cases in the care group. There was no significant difference in gender and age distribution between the two groups (χ2 = 3.013, 1.336, P > 0.05). The proportion of inclusion into treatment was higher in the care group (100.00%, 50/50) than in the control group (87.04%, 47/54) (P = 0.013), and the proportion of loss to follow-up was lower in the care group (0, 0/43) than in the control group (19.05%, 8/42) (P = 0.002). In addition, the overall proportion of return visits was higher in the care group (93.09%, 377/405) than in the control group (83.56%, 371/444) (χ2 = 18.345, P < 0.001), and the proportion of sputum examinations was higher in the care group was (83.70%, 339/405) than in the control group (79.28%, 352/444) (χ2 = 2.737, P = 0.098). Conclusion Patient-centered care services facilitate the improvements in the proportion of inclusion into treatment and compliance to treatment and reduction in the proportion of loss to follow-up among MDR/RR-PTB patients, which deserves widespread applications.
9.Study on immunogenic cell death related proteins in nasopharyngeal carcinoma patients before and after radiotherapy

Objective:To explore the effect of clinical conventional fractionated dose radiation on the expression levels of immunogenic cell death (ICD) related proteins in patients with nasopharyngeal carcinoma (NPC).Methods:A total of 38 newly-treated NPC patients admitted to the Affiliated Cancer Hospital of Guizhou Medical University from November 2020 to December 2021 were enrolled, all of whom received induction chemotherapy and concurrent chemoradiotherapy, and another 20 healthy volunteers were selected as controls for a prospective study. The contents of ICD related proteins, namely calreticulin (CRT), high mobility group box 1 protein (HMGB-1) and heat shock protein 70 (HSP70) and the proportion of dendritic cell (DC) in the peripheral blood of patients were detected before treatment, after induction chemotherapy and after concurrent chemoradiotherapy, respectively. The correlation between the above indicators, general clinical data and short-term efficacy was analyzed by statistical methods such as t-test and analysis of variance (ANOVA). Results:The levels of HSP70 and HMGB-1 in peripheral blood of NPC patients before treatment were higher than those of healthy controls (both P<0.05). After concurrent chemoradiotherapy, the content of CRT was significantly higher than that before treatment ( P<0.05), whereas the difference before and after induction chemotherapy and the difference before and after concurrent chemoradiotherapy were not significantly correlated with the short-term efficacy of NPC patients. HSP70 level was significantly decreased after concurrent chemoradiotherapy ( P<0.001). There were no significant differences in the content of HMGB-1 after induction chemotherapy and concurrent chemoradiotherapy (both P>0.05). Conclusion:NPC patients receiving TPF regimen (docetaxel+cisplatin+fluorouracil) for induction chemotherapy and sequential cisplatin concurrent chemotherapy may induce ICD in NPC cells, and CRT has potential value in reflecting the clinical efficacy of NPC.
10.Establishment of fingerprint and determination of differential components of Sophora flavescens

OBJECTIVE To establish the fingerprint of Sophora flavescens, and to screen differential components and determine their contents. METHODS HPLC fingerprints of 12 batches of S. flavescens were established by using Similarity Evaluation System of Chromatographic Fingerprints of TCM (2012 edition); common peaks were identified and their similarities were evaluated. Chemical pattern recognition analysis [cluster analysis (CA),principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis(OPLS-DA)] were performed with SIMCA 14.1 and SPSS 23.0 software, and differential components which influenced the quality of S. flavescens were screen with variable importance in the projection(VIP)>1 as standard. Meanwhile, the contents of 4 kinds of differential components were determined by the same HPLC method. RESULTS There were 17 common peaks in the fingerprints of 12 batches of S. flavescens,and their similarities were all higher than 0.96. A total of 6 common peaks were identified, i.e. oxymatrine (peak 1), oxysophocarpine (peak 2), matrine (peak 10), trifolirhizin (peak 14), kurarinone (peak 16) and norkurarinone (peak 17). Results of CA, PCA and OPLS-DA showed that 12 batches of S. flavescens were divided into 3 categories according to producing area, i.e. S1-S7 (Shangzhou District of Shaanxi Province) were grouped into one category, S8-S10 (Yichuan County of Henan Province) into one category and S11-S12 (Chifeng City of Inner Mongolia) into one category. VIPs of matrine, norkurarinone, kurarinone and oxysophocarpine and the chemical components represented by peak 11 and 9 were all greater than 1. The contents of matrine, norkurarinone, kurarinone and oxysophocarpine in 12 batches of S. flavescens were 2.65-4.93, 1.54-3.44, 9.63-12.94 and 5.08-6.10 mg/g, respectively. CONCLUSIONS HPLC fingerprint of S. flavescens is established successfully in the study, and can be used to screen 6 differential components by combining with chemical pattern recognition analysis, which can provide reference for quality control of S. flavescens.
