2.Correlation between preoperative bile duct dilatation evaluation and prognosis of hilar cholangiocarcinoma

Objective:To evaluate the correlation between the prognosis of patients with hilar cholangiocarcinoma and the degree of bile duct dilatation in MRCP .Methods:The clinical data of 89 patients with hilar cholangiocarcinoma undergoing radical operation at Tianjin Nankai Hospital from Jan 2009 to Dec 2013 were analyzed retrospectively.Results:Tumor size ( P=0.024), Bismuth-Corlette classification ( P=0.048) and tumor stage ( P=0.013) were related factors of biliary dilatation. Tumor differentiation ( P=0.002), R 0 resection ( P=0.002) and biliary dilatation ( P<0.001) were independent predictors of disease-free survival (DFS). Conclusion:The imaging evaluation of the degree of biliary dilatation has a certain predictive value for the prognosis of patients with hilar cholangio-carcinoma.
3.Application value of systematic prenatal ultrasound in prenatal screening of twin pregnancy

Objective To explore the application value of systematic prenatal ultrasound in prenatal screening of twin pregnancy. Methods Retrospectively analyzed 393 cases of twin pregnancy who underwent systematic prenatal ultrasound examination in our hospital from October 2012 to December 2017, and 27 cases of twin pregnancy malformation were included in the study. By comparing the data of prenatal ultrasound examination with follow-up data and pathological anatomic results, the detection rate and diagnostic coincidence rate of fetal malformation in prenatal screening of twin pregnancy by systematic prenatal ultrasound examination were analyzed. Results Among 27 cases of twin pregnancy malformation, 19 cases were fetal structural malformation and 8 cases were unique malformation of twin pregnancy. Nineteen cases of twin pregnancy fetal structural malformation occurred in one of the two fetuses in twin pregnancies, and all of which were single malformations. Eight cases of unique malformation of twin pregnancy were often involved in double fetuses. Seventeen cases of fetal structural abnormalities and 8 cases of unique malformation of twin pregnancy were diagnosed correctly by systematic prenatal ultrasound examination, and the diagnostic coincidence rate of prenatal ultrasound was 100% (25/25). Two cases of structural malformation were missed, and the detection rate of fetal malformation was 92.6% (25/27). Conclusion The systematic prenatal ultrasound in prenatal screening of twin pregnancy has high detection rate of malformation and diagnostic coincidence rate, which has important clinical application value.
4.Effects of rapamycin on expression of ten kinds autophagy-related miRNAs in RAW264.7 macrophages

To detect the influence of rapamycin on the expression of miR-30b,miR-200a and miR-17-5p etc in macrophages and provide the basis to study the regulation of miRNA in autophagy mechanism of macrophages .Methods: Small RNA was extracted at different times after stimulated with rapamycin in cultured RAW 264.7 cells.After using the stem-loop reverse transcription primers to reverse transcribed into cDNA ,the expression of miR-30b ,miR-30c,miR-106a,miR-214,miR-183,miR-200a, miR-376c,miR-17-5p, miR-142-3p, miR-377 was detected by Real-Time PCR.Results: After RAW264.7 cells was treated by rapamycin for 2,4,6 and 8 hours,the expression of miR-17-5p and miR-106 increased (More than 2.1 times,P<0.05) in 2,4 and 6 hours;miR-214 was up regulated in 2 and 8 hours (More than 2.4 times,P<0.05);miR-30b,miR-30c,miR-183,miR-200a,miR-376c and miR-142-3p was up regulated in 2,6 and 8 hours (2.4 times,P<0.05 );while miR-183 and miR-200a was down regulated at 4 hours(More than 2.1 times,P<0.05);miR-30b was significantly low expression in 8 hours (more than 50 times,P<0.05);miR-377 was up regulated at 4 hours (more than 2.5 times,P<0.05),but was significantly down regulated at 2 and 8 hours (More than 50 times,P<0.05) Conclusion: The expression of miR-200a,miR-30b,miR-377,miR-30c,miR-376c and miR-17-5p is significantly changed after rapamycin stimulated RAW264.7 macrophages,indicated the miRNA may plays an important role in autophagy through the regulation of autophagy-related genes.
5.A survey on the caries and allocation of oral medical resource in west area of Chongqing

Objective To discuss the allocation of oral medical resource in west area of Chongqing,and investigate the caries prevalence in these population.Methods Study samples was raised with the multistage stratified random cluster sampling method,then data was analyzed to compare the allocation of oral medical.Resource,awareness degree on oral health and caries prevalence in each age grade between urban and rural area.Results In west area of Chongqing,the allocation of oral medical resource and awareness degree on oral health were better in urban area than those in rural area.In each age grade,the caries prevalence is higher in rural area,when compared with that in urban area.In addition,the caries prevalence of 5 years old group and 12 years old group is the same between male and female in both urban area and rural area (P>0.05).Nevertheless,in 35-44 years old and 65-74 years old group,the caries prevalence was higher in female compared with male in both urban area and rural area (P<0.05).Conclusion In west area of chongqing,the rural allocation of oral medical resource is bad and needs improvement,the awareness of oral hygiene is weak among population of rural area.In west area of Chongqing,women in 35-44 years and 65-74 years old should pay more attention to caries prevention and treatment.
6.Research advances in the pathogenesis of drug-induced liver injury

In recent years, with an increasing number of drug types and unreasonable drug use, the incidence and mortality of drug-induced liver injury (DILI) have been increasing. The pathogenesis of DILI is complex, and may involve liver injury caused by the direct effect of drugs, immune-mediated liver injury, mitochondrial injury, and bile duct injury, etc. This article investigates the pathogenesis and its important role in the prevention and treatment of DILI, and reviews the research advances in the pathogenesis of DILI.
7.Effect of PNPLA3 I148M mutation on expression of TGF-β1 in rat hepatic stellate cells

ObjectiveTo investigate the mechanism of action of PNPLA3 I148M mutation in the development and progression of non-alcoholic fatty liver fibrosis. MethodsThe lentiviral vectors carrying the mutant or wild-type PNPLA3 I148M gene were constructed and transfected into rat hepatic stellate (HSC-T6) cells. Quantitative real-time PCR was applied to measure the mRNA expression of transforming growth factor β1 (TGF β1). The t-test was applied for statistical analysis. ResultsThe lentiviral vectors carrying the mutant or wild-type PNPLA3 I148M gene were successfully constructed and transfected into HSC-T6 cells, and a HSC-T6 cell line with stable expression of the mutant or wild-type PNPLA3 gene was established. Compared with the cell line carrying the wild-type gene, the cell line carrying the mutant gene showed significantly higher mRNA expression of TGF β1 (1.25±0.15 vs 0.48±0.07; t=11.826, P<0001). ConclusionPNPLA3 I148M mutation can increase the expression of TGF β1 in HSC-T6 cells, which provides a new cell model and new research ideas for investigating the role of PNPLA3 I148M mutation in non-alcoholic fatty liver fibrosis.
8.The effects of the compound lavender ointment on experimental guinea pigs scald、anti-inflammation and analgesia

Objective To study the main pharmacodynamics effect of compound lavender ointment on the scald, anti-inflammation and analgesia.Methods Sixty guinea pigs were divided into normal groups,model groups( superficialⅡ°scalding models) ,compound lavender ointment high,median,low dose groups,and jing wan hong ointment groups,then observation of the wounds and healing was made and recorded.Histopathological examination was made to examine the wounds,and ELISA to analyze serum TNF-α、IL-6、IL-8.Xylene ear swelling model and writhing test were used to observe the analgesic and anti-inflammatory effects of the compound lavender ointment.Results Compound lavender ointment can relieve the wounds swelling and exudation,which may be related to the decrease of proinflammatory cytokines(P <0.01), and promote the wound healing significantly( P <0.01) ,that the side-effects were not observed.High dose of compound lavender ointment can inhibit ear swelling and have certain analgesia with the writhing tests.Conclusion Compound lavender ointment have the promoting wound healing,anti-infectious and antalgic effects on the scald.
9.miR-17,miR-181,miR-106,miR-30 and miR-495 level differences in peripheral blood of patients with ankylosing spondylitis

Objective:In order to investigate the role of microRNA in the pathogenesis of ankylosing spondylitis patients,we detected the peripheral blood of patients with ankylosing spondylitis in miR-17,miR-181,miR-106,miR-30 and miR-495 expression, thereby to study the role of microRNA regulation of clinical and diagnostic value in the pathogenesis of ankylosing spondylitis provide new ideas.Methods:Collection of patients with ankylosing spondylitis and normal peripheral blood,peripheral blood mononuclear cells were isolated and extracted PBMC small RNA,using primers specific stem-loop reverse transcribed into cDNA,and build a mature miR-17,miR-181,miR-106,miR-30 and miR-495 T-carrier,standard curve,the use of stem-loop method by Real-time PCR technology to detect miR-17,miR-181,miR-106,miR-30 and miR-495 expression level.Results: In this study,92 cases of clinical samples were collected,of which 61 patients with AS,normal 31 cases.By Real-time PCR detection showed that compared with normal subjects,there was upregulation of miR-106 (P>0.05) and miR-30 (P>0.05) in the peripheral blood of patients with ankylosing spondylitis;down-regulation were miR-181 ( P<0.05 ) , miR-495 ( P<0.05 ) and miR-17 ( P>0.05 ) , in which the miR-181 and miR-495 was statistically significant.Conclusion:Compared with normal, there are differences in the peripheral blood of patients with ankylosing spondylitis expression of miR-495 and miR-181,which may be targeted to regulate TLR-4,HLA-B,DVL,GSK/3βgenes,this may be found in the pathogenesis of ankylosing spondylitis studies provide new ideas,to become a new clinical diagnosis markers.
10.Investigation into the urinary metabolomic profiling of spontaneously hypertensive rat (SHR) based on 1 H-NMR analysis

Objective To investigate the application of metabolomics in research of the metabolic profiling of spontaneously hypertensive rats ( SHR) and Wistar rats using NMR combined with PLS-DA.Methods SHRs were fed with normal diet for 1 week and then the urines of normal rats and SHR hypertensive model rats were collected at 1, 2, 3, and 4 weeks.The metabolic profiles were analyzed using 1H-NMR.Partial least squares discriminant analysis (PLS-DA) was used to determine the differential biomarkers .Results The systolic blood pressure (SBP) of SHRs were significantly higher than that of normal rats.Using PLS-DA and orthogonal partial least squares discriminant analysis (OPLS-DA), it was also possible to distinguish them in the principal components score plot .In the SHRs, the levels of some amino acids and some small molecular substances were higher than those in the control rats .Conclusions NMR combined with PLS-DA is a promising approach to provide the information on metabolic changes related to the pathophysiological processes in spontaneously hypertensive rats .
