1.Establishment and analysis of saliva two dimensional gel electrophoresis profiles of Type 2 diabetes patients

Objective:The purpose of this study was to establish two dimensional gel electrophoresis(2-DE) profiles of saliva of Type 2 diabetes patients,and to identify the differential proteomic expressions between normal persons and Type 2diabetes patients.Methods:Samples of saliva protein were separated by two dimensional electrophoresis with optimization.Differential proteomic expressions between the Type 2diabetes patients and the normal control persons were identified by two dimensional polyacrylamide gel electrophoresis,silver staining,image master 2-DE software analysis,peptide mass fingerprinting based on matrix assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF-MS),Bioworks and NCBI software database searching.Results:The two dimensional polyacrylamide gel electrophoresis profiles of saliva proteins were successfully established by 2-DE.twenty of the significant differential proteins were selected and identified by MALDI-TOF-MS.seven of them were finally identified.Conclusions:The 2-DE profiles of saliva proteins were established and the differential proteomic expressions were identified by proteome technique in our study.This can be an experimental basis for further research of the pathogenesis and treatment of Type 2 diabetes.
2.EFFECTS OF LIPOPOLYSACCHARIDE ON THE EXPRESSION OF SRC-SUPPRESSED C KINASE SUBSTRATE IN CULTURED ASTROCYTES

Objective To investigate the effects of lipopolysaccharide(LPS) on Src-suppressed C Kinase Substrate(SSeCKS) in cultured astrocytes. Methods Purified astrocytes were randomly divided into three groups:control group,singly stressed and multiple stressed groups.The expression of SSeCKS was detected by Real time RT-PCR and Western blotting,while immunocytochemistry was used to investigate the subcellular localization of it. Results Real time RT-PCR indicated that,after 12 hours incubation,both 100 ?g/L and 1mg/L LPS were able to elevate the levels of SSeCKS mRNA compared with control group,while 1mg/L LPS did not have a stronger effect than 100 ?g/L.Western blotting analysis showed 100 ?g/L LPS significantly increased the expression of SSeCKS.In time response experiments,the levels of SSeCKS expression enhanced three hours after the stimulation,peaked at the sixth hour,coincident with its rapid phosphorylation,and remained high until the 24th hour.Immunocytochemistry suggested a perinuclear translocation of SSeCKS,while the PKC inhibitor RO-31-8220 blocked it.Conclusion In cultured astrocytes,LPS can enhance the expression of SSeCKS,increase its phosphorylation level and change its subcellular localization.These effects are correlated with the PKC pathway,which indicates SSeCKS might participate in the signal transduction of inflammation in cultured astrocytes.
3.Contact analysis between artificial humeral head and glenoid fossa during humeral external rotation.

We developed a three-dimensional finite element model of the shoulder glenohumeral joint after shoulder arthroplasty including humerus shaft, scapular, scapular cartilage and eight muscles, while each of the muscles was simulated with 50 spring elements. To reduce the element number and improve the analytical precision, we used mixed tetrahedral and hexahedral elements in the model. We then used the model to calculate the biomechanics of the shoulder glenohumeral joint after hemiarthroplasty during humeral external rotation. Results showed that the maximum joint reaction force was 374.72 N and the maximum contact stress was 6. 573 MPa together with the contact areas at 40 degrees external rotation. These might be one of the reasons for prosthetic disarticulation, and would provide theoretical bases to prosthetic design.

4.Evaluation of uncertainty in determination of ethanol in human blood by auto HS-GC with internal standard curve method

ObjectiveTo evaluate the uncertainty for the determination of ethanol in human blood by auto-headspace gas chromatography (HS-GC) with internal standard curve method.Methods Each source of uncertainty, arising from the procedure of testing, was analyzed and conifrmed according to the guidelines of the uncertainty in measurement . After each uncertainty component was quantized, the combined standard uncertainty and the expanded uncertainty of the result were calculated.Results The relative uncertainty brought from the measurement repeatability, standard solution of the ethanol, the sample of blood, internal standard solution of the tert-butyl alcohol, the calibration cure, gas chromatography were 3.4%,0.71%,0.61%,0.41%,1.1% and 1.3% respectively; the relative expanded uncertainty of ethanol in blood was 3.9%.Conclusion The measurement uncertainty of the concentration of ethanol was came primarily from the measurement repeatability of sample, HS-GC and standard curve of ethanol.
5.Mechanism of the Reversal Effect of Acupuncture on the Fall of Blood Pressure Induced by Angiotensin-(1-7) in the Caudal Ventrolateral Medulla of Rats

Objective:To investigate the mechanism of the reversal effect of acupuncture on the fall of blood pressure induced by angiotensin-(1-7)[Ang-(1-7)] in the caudal ventrolateral medulla(CVLM) of rats. Methods: Ang-(1-7) and its selective receptor antagonist (D-Ala7), Ang-(1-7) and Ang-(779) were microinjected into the CVLM respectively to induce the change of blood pressure, during which, the concentrations of amino acid neurotransmitters were detected by means of microdialysis and high performance liquid chromatography(HPLC)combined with fluorescent detector, on the other hand, the effects of electro-acupuncture (EA) on the level of blood pressure and the concentrations of amino acid neurotransmitters were also observed. Results:Unilateral microinjection of Ang-(1-7) into the CVLM could decrease the blood pressure (BP), which was accompanied by an increase in the release of Glu and a decrease in the release of Tau in the same site; Whereas microinjection of Ang-779 into the CVLM could elevate BP, which was accompanied by a decrease in the release of Glu and an increase in the release of Tau; EA at acupoint Zusanli (ST 36) for 20 min could inhibit the fall of blood pressure induced by microinjection of Ang-(1-7) into the CVLM and the elevation of blood pressure induced by microinjection of Ang-779 into the CVLM, and in the meantime, it could inhibit the changes of the release of Glu and Tau induced by microinjection of Ang-(1-7) and Ang-779 into the CVLM. Conclusion: The reversal effect of EA on the fall or elevation of blood pressure induced by microinjection of Ang-(1-7) or Ang-779 into the CVLM might be related to the changes of Glu and Tau release.
6.Clinical investigation of docetaxel combined with nedaplatin in the treatment of recurrent epithelial ovarian cancer in elderly women

Objective To evaluate the efficacy and safety of concomitant docetaxal (DTX) and nedaplatin (NDP) therapy for recurrent epithelial ovarian cancer in elderly women.Methods Totally 42 elderly patients with histologically confirmed recurrent epithelial ovarian cancer received chemotherapy with DTX combined with NDP.DTX with the dose of 60-70 mg/m2 was administered intravenously on day 1,followed by NDP with the dose of 70-80 mg/m2 given intravenously on day 2.The treatment was repeated every 3 weeks.The efficacy was evaluated according to response evaluation criteria in solid tumors (RECIST) and serum CA125 level.Adverse reactions were assessed according to performance and standard criteria in toxicity of anticancer agents of WTO.Results 4 patients were switched to other chemotherapy strategies due to progression of disease after 2 courses of treament,and the other patients completed 4 to 6 courses.Among 42 patients,18 cases (42.9%)showed complete remission,10 cases (23.8%) showed partial remission,6 cases (14.3%) were in stable conditions,and 8 cases (19.0%) progressed to severe disease.The median progression-free survival (PFS) time was 7 months,and the efficacy rate (complete and partial remission) was 66.7%(28/42).The main adverse effects were alopecia,nausea and vomit,and leucopenia.Patients could get remission by receiving drug therapy.Conclusions Docetaxel plus nedaplatin therapy has a good therapeutic effect with low incidence of adverse reactions,high safety and well tolerance in elderly platinum-sensitive women and in those with platinum-resistant recurrent epithelial ovarian cancer.
7.The Clicinal Effect of Different Surgical Treatment for Laryngotracheal Stenosis

Objective To study the clinical effects of different surgical treatments for laryngotracheal steno-sis .Methods The clinical data of 56 patients with laryngotracheal stenosis from July 2006 to July 2014 were ana-lyzed .Of all 56 cases ,7 cases were children ,49 cases were aducts ;9 cases underwent CO2 laser resection ,5 cases underwent temperature controlled radiofrequency ablation and 5 cases received laryngeal mold under self -retaining laryngoscope ,6 cases received Keel placement after laryngofissure ,23 cases received T -shape tube implantation af-ter laryngoplasty or tracheoplasty and 8 cases underwent end-to -end tracheal anastomosis after cervical tracheoto-my .Results All the patients had been followed up for 6 months to 2 years postoperatively .Thirty nine cases of 56 were cured after single-operation ,and 10 of 56 were cured after repeated operations .No one suffered from laryn-gotracheal stenosis again .Five cases failed and survived with tracheal tube .2 cases lost to follow -up .No people die or had complications .Among all cases ,5 cases with laryngeal mold and 8 cases with tracheal anastomosis were cured after single-operation .Among 9 cases with lasere scar removal surgery ,5 cases were cured after an operation ,3 cases were cured after two operations ,1 case failed because of laryngeal cancer recurrence .Among 5 case with tem-perature controlled radiofrequency ablation ,3 cases were cured afrter an operation ,1 case was cured after two opera-tions ,1 case had no effect .Among 6 cases with keel place -ment ,4 cases were cured after one operation ,1 case was cured after two operations ,1 case had no effect .Among 23 cases with T -shape tube implantation ,14 cases were cured after an operstion ,5 cases were cured after two and more operations ,2 cases had no effects and 2 cases were lost .Conclusion In order to improve the therapeutic effects of laryngotracheal stenosis ,the treatment strategy should be personalized according to the degree of stenosis ,the position of stemtis ,and the impairment of laryngotra-cheal framework .
8.Ultrasound combined with tri-operators and the blood-oxygen functional image technology in diagnosis of breast cancer in young women

Objective To evaluate ultrasound combined with thri-operators and the blood-oxygen functional image (TOBOFI) technology in diagnosis of breast cancer in young women. Methods 221 breast tumor patients under 45 years old were selected to analyze their TOBOFI and ultrasonographic characteristics respectively. Based on pathological result, the two diagnostic methods were compared to determine the accurate rate. Results Of all the 221 patients, 82 (37.1%) were malignant and 139 benign. The sensitivity of TOBOFI and ultrasonography was 90.24% and 83.57% respectively, while the accurate rate of the two was 89.14% and 84 62% respectively. Accurate rate was improved to 93.90 % (77/82) if the two were combined. Conclusion TOBOFI has unique clinical value in diagnosing breast cancer in young women, especially in combination with ultrasound.

Heilongjiang Provincial Hospital has scored an initial success in dealing with medical disputes since the medical liability insurance and third-party mediation mechanism were introduced into the hospital in 2013.The paper identified problems found in the practice,and recommended the following:rationalizing the cost of insurance coverage,expanding scope of third-party mediation properly,enhancing professional authority in assessment of medical dispute cases,simplifying insurance compensation procedure,and consolidating the legal status of the medical dispute mediation institutions,for better resolution of such disputes.
