1.Effects of genistein on proliferation, differentiation, content of matrix calcium and mineralization in primary cultured rat osteoblasts

BACKGROUND: Osteoporosis has been one of the main reasons for old people' s in decline living standard and shortening of lifespan. The onset of osteoporosis is related to the loss of functional coupling in osteoblast and osteoclast. So observation of the regulative effect of medicines on osteoblast is a good method to evaluate their preventive and therapeutical efficacy in osteoporosis.OBJECTIVE: To investigate the effects of genistein on osteoblast culture in vitro.DESIGN: A randomined, controlled and single-blind study based on rat osteoblasts.SETTING: Department of laboratory in amunicipal hospital.MATERIALS: The experiment was completed in the Room of Isotope, Department of Nuclear Medicine, China-Japan Friendship Hospital of Beijing from February to December 2001. Ten SD rats of 24 hours old were provided from Beijing Weitong Lihua Experimental Animal Ltd. Co. (license:SCXK11-00-0008 ). The laboratory was SPF grade.METHODS: Skull osteoblasts of rats were selected to culture in vitro. Microculture tetrozolium, p-nitrophenyl phosphate, atomic absorptiometry and alizarin red were used to observe the effect of genistein on proliferation,activity of alkaline phosphatase(ALP), contents of matrix calcium and the number of mineral nodes of osteoblasts cultured in vitro.MAIN OUTCOME MEASURES: Morphological observation, determination of proliferation ratio, ALP stain, determination of matrix calcium accumulation and number of mineralized nodes with alizarin red stain.RESULTS: It was found that genistein stimulated the proliferation of osteoblasts, improved the ALP activity, and increased the contents of matrix calcium and the number of mineral nodes in cultured osteoblasts.CONCLUSION: Genistein has the effects on stimulating the proliferation,differentiation, maturation and mineralization of osteoblasts cultured in vitro.
3.Study on Mechanism of Effects of Total Flavone of Rhizoma Drynariae on Osteoblasts Cultured in Vitro

Objective: To study the mechanism of effect of total flavone of rhizoma drynariae in groups with different dosage on osteoblasts cultured in vitro. Methods: To culture osteoblasts with UMA-106-01 osteoblast strain and to observe the activity of ALP and the endosmosis of 3H-TdR. Results: The total flavone of rhizoma drynariae increased the activity of ALP in cells cultured with UMR-106-01 strain. The ALP activities correspondingly changed in the 24h, 48h and 72h, which related to dosage effect and time effect. Among them, the activity in the 48h is the most ideal one. And the amount of endosmosis of the total flavone of rhizoma drynariae to 3H-TdR increased more remarkably in the 48h than that in the 24h and it also related to the time effect. Conclusion: The total flavone of rhizoma drynariae can promote the differentiation and multiplication of osteoblasts.
4.Effect of dexmedetomidine on patients′ inflammation during the CPB and the effect on patients′ renal and hepatic function

Objective To observe the effect of dexmedetomidine on patients′ inflammation during CPB and protective effect on kidney and liver. Methods 60 cases undergoing cardiac valve replacement under CPB were randomly divided into NS group and Dex group. Blood samples were taken before induction , before ascending aorta blocked, end of CPB, 24, 48 and 72 hours after operation. The serum level of HMGB-1, TNF-α, IL-6, BUN, Cr and ALT are tested. Blood WBC and N% are also counted. Results WBC, N% and HMGB-1, TNF-α, IL-6, BUN, Cr in Dex group significantly decreased at time point T2 ~ T6 (P < 0.05) compared with NS group. But ALT in Dex group only decreased at time point T 2 and T5 compared with NS group (P < 0.05). Conclusion Dexmedetomidine can significantly decrease inflammatory factor during CPB and improve renal function after surgery.
5.Determination of Influenza Virus H5N1 and H7N9 Using MASA Technology.

To set up a new rapid method for the rapid determination of influenza virus H5N1 and H7N9 basing on the Multi-Analyte Suspension Array (MASA) technology. Sequence analysis and design of degenerate primers and specific probes were set in the comparison and analysis of H5, N1, H7 and N9 genes. In combination with MASA technology, these primers and probes were used for the determination of samples of H5N1 and H7N9 and other subtypes ( H1N1, PH1N1, H5N2, H3N2 and H9N2). We developed a rapid determination method. This method had high specificity and sensitivity that could detect H5N1 and H7N9 at one time, and could detect samples that containing 10 copies of H5N1 and H7N9. This determination method could be used for rapid determination of influenza virus H5N1 and H7N9 at one time.

6.Real-time PCR Detection Method for the Reston Subtype of the Ebola Virus.

We aimed to develop a real-time polymerase chain reaction (PCR) detection method for the Reston subtype of the Ebola virus. The NP gene of the Reston subtype of the Ebola virus was selected as the detection object. Sequences of different subtypes of Ebola viruses were aligned using Clustal W software. The most unique and conserved regions of the Reston subtype of the Ebola virus were recruited as candidate sequences for specific primers. Primer Express and Primer Premier 5. 0 software were used to filter the optimal pair of primers for detection. Real-time PCR was carried out using optimized parameters and positive DNA prepared by serial (tenfold) dilution of a recombinant plasmid and by plotting a standard curve. In addition, the reproducibility, accuracy, and specificity of the assay were tested. Results showed that the sensitivity of detection of the Reston subtype of the Ebola virus by real-time PCR could reached 10(2) copies/microL. The linear relationship (R2) reached 0.997, the slope of the standard curve was -0.3101, and amplification efficiency was 110.145%. A sharp and narrow melting peak appeared at 79.94 degrees C for all standards in different dilutions. In conclusion, a fast and sensitive real-time PCR detection system for the Reston subtype of the Ebola virus was developed. This system could be used as a supplementary diagnostic and monitoring approach for basic and clinical studies on the Reston subtype of the Ebola virus. The detection system does not require expensive technology or specialist operators.

7.Common and variant peak ratio in IR fingerprint of extraction from Radix Glycyrrhizae by dual index sequence analysis

AIM: To set up a new method to analyze IR fingerprint spectra, which was in line with the cha racteristic of traditional Chinese medicine. METHODS: In this paper. two new indexes, common peak ratio and variant peak ratio, were applied and its values were calculated by means of the sequential analysis, in which each sample's IR fingerprint spectra was set up and the common peak ratio sequences were arranged in order of size in comparison with other samples. RESULTS: The method could distinguish the water extract of Radix Glycyrrhizae of different varieties and different areas. CONCLUSION: The dual index sequential analysis enables us to distinct two or more herb's IR fingerprint.
8.A multi-center clinical trial of natural calcined bone repair material in repairing bone defect after tooth extraction

Objective:To study the efficacy and safety of natural calcined bone repair material(NCBM)in repairing bone defect af-ter tooth extraction.Methods:A randemized,double-blinded,parallel,positive control(Bio-Oss)and multi-center clinical trial was employed.Imaging examination was used as the main efficacy evaluation index,surgical wound healing,rejection reaction,bone me-tabolic changes,bone infection signs were the subordinate efficacy evaluation indexes,the incidence of adverse reactions was observed for safety evaluation.Results:280 cases were included,269 cases completed the trial.In NCBMand Bio-Oss group the effective rate of imaging examination was 93.08% and 93.70%(P >0.05)respectively.The wound healing time of the 2 groups was less than 7 days,no rejection reaction,bone metabolic change and bone infection sign were observed.The incidence of adverse events in NCBM and the Bio-Oss group was 0.72% and 2.14%(P >0.05)respectively.Conclusion:The efficacy and safety between natural cal-cined bone repair material is not inferior to Bio-Oss in repairing bone defect after tooth extraction.

Objective:To compare the outcomes of guide bone regeneration by calcined bovine bone and Bio-Oss graft material in alveolar ridge preservation after tooth extraction.Methods:280 patients were divided into two groups randomly.Each patient had single tooth extracted.The sockets were filled with calcined bovine bone in 140 patients and Bio-Oss graft in another 140 patients. After shaping,all the sockets were covered with Bio-Gide membrane.Buccal mucoperiosteal flap was released and sutured to close the alveolar sockets.The patients were regularly examined at the 1st,12th and 24th week after surgery.Physical examination and X-ray evaluation were applied to compare the outcomes of the two materials in alveolar ridge preservation.Results:No infection and re-jection occurred.The radiographic results showed the width and height of the alveolar bone were preserved well at the 12th and 24th weeks.No statistically difference was found in the two groups at the 1st and 24th weeks(P>0.05).Conclusion:The two graft ma-terials can effectively preserve alveolar bone after tooth extraction.
10.Risk factors for lymph node metastasis of early colorectal cancer and indices of endoscopic resection

Objective To analyse the risk factors for lymph node metastasis of early colorectal cancer and the therapeutic indication for endoscopy.Methods The clinical data of the 269 early colorectal cancer patients who underwent surgical treatments between January 2009 and December 2013 in the West China hospital of Sichuan University were analyzed retrospectively.Pathologic features were compared between different histological types and investigated by univariate and multivariate analysis of their possible risk factors for lymph node metastasis.Results Univariate analysis showed that tumor size (P =0.029),depth of tumor invasion (P =0.006),histological type (P =0.000) and lymphatic involvement (P =0.035) were correlated with lymph node metastasis.Multivariate analysis revealed that tumor size (OR =5.385,95% CI:1.156-25.075,P =0.032)and histological type (OR =5.145,95% CI:1.553-17.053,P =0.007) were independent risk factorsfor lymph node metastasis.Comprehensive analysis showed that lymph node metastasis could not be found in patients with tumor invading the mucous layer.However,lymph node metastasis occurred if the tumor invaded the submucous layer.The larger and less differentiated tumor was,the higher occurrence of lymph node matastasis would be (P < 0.05).Conclusion Endoscopic resection is recommended for those with early colorectal cancer localized in the mucous layer and without lymph node metastasis.It is recommended that patients with submucosal carcinomas undergo a preoperative endoscopic ultrasonography or pathological screening for the most appropriate surgical treatment,as the larger and the lower differentiated the tumor is,the higher chance the lymphatic metastasis is.
