1.Analysis of the Target Antigen Recognized By The Monoclonal Antibody (2H3)

The McAb 2H3 to different target cells was investigated by using an indirect ELISA meth-ods. The result showed that the 2H3 could bind to the ConA actiuated mouse T lymphoblastsand CTLL-2 cells (an IL-2 dependent T cell line)but not to the resting mouse splenocytes,lymph node cells and thymocytes. The binding of FITC-anti-IL-2R to the activated mouse spleencells and CTLL-2 cells was seen as tested by immunofluorescence analysis. This binding reactionwas blocked when the target cells were previously treated with 2H3, but the binding reaction ofFITC-anti-Ia McAb to these target cells couldn't be blocded by 2H3. Further analysis of the tar-get antigen by using Western-Blotting technigue showed the molecular weight of this antihenrecognized by 2H3 was of 50~60KD in reducing condition. According to these data, target anti-gen recognized by the McAb 2H3 may be the IL-2R molecule(P_(55) mole cule)on the cell surface.
2.An in vitro study of obturation efficiency and outcomes of vertical condensation of warm guttapercha using the four-handed technique

ObjectiveTo evaluate the therapeutic efficiency and outcomes of vertical condensation of warm gutta-percha obturation using the four-handed technique.MethodsA split-tooth model constructed with lateral grooves and depressions was used to compare vertical condensation of warm gutta-percha obturation with and without the four-handed technique.Respectively 10 times of obturation were done in the four-handed group and the independent operation group.The operation time and defect replication quality were recorded.Evaluation of defect replication quality was on an ordinal scale 0 to 2 grade based on how much each defect was replicated.The results were statistically analyzed.ResultsThere were signiticant differences in treatment time between the four-handed group and theindependent operation group,as well as in treatment outcomes between two groups.ConclusionsUse of the four-handed technique can improve therapeutic efficiency and outcomes of vertical condensation of warm gutta-percha obturation.
3.Role of hypoxia-induced factors in cancer development mediated by cancer stem cells

Hypoxia and hypoxia-induced factors (HIFs) are main regulators for tumor stem cells, metastasis-initiating cells and their differentiated progenies to adapt to the environment which lacks oxygen and nutrient in the process of cancer development. HIFs are up-regulated in many tumors, including leukemia, glioblastoma, melanoma, prostate cancer, breast cancer and pancreatic cancer, in where they are especially highly expressed in hypoxic regions. HIFs activation can induce expression of numerous stem cell related genes and multidrug resistance genes, which may play important roles in tumour and stem cell-mediated self-renewing, energy metabolism alternation, invasion, metastasis, angiogenesis and treatment resis?tance of neoplastic cells. Consequently, it will provide new clues for cancer therapy after investigating the role of HIFs in tar?geted regulation and metabolic pathway modulation in various stem cell-mediated tumor cells.
4.Observation on the influence of ademetionine for the proteometabolism and fibrosis indexes of patients with chronic hepatitis B

Objective To observe the influence of ademetionine for the proteometabolism and fibrosis indexes of patients with chronic hepatitis B.Methods110 patients with chronic hepatitis B in our hospital from January to December 2016 were randomly divided into control group(conventional treatment of hepatitis B group)55 cases and observation group(conventional treatment and ademetionine group)55 cases.The serum expression levels of proteometabolism and fibrosis indexes of two groups before the treatment and after the treatment were detected and compared.ResultsThe serum expression levels of proteometabolism and fibrosis indexes of two groups before the treatment were compared,there were no significant differences,while the serum expression levels of proteometabolism indexes of observation group after the treatment were all higher than those of control group,the serum expression levels of fibrosis indexes were all lower than those of control group,there were all significant differences(all P<0.05).ConclusionThe ademetionine can significantly improve the expression state of proteometabolism and fibrosis indexes of patients with chronic hepatitis B,so the application value for the patients with chronic hepatitis B is relatively higher.
5.Epileptic seizure in the elderly: a retrospective analysis of 72 cases

Objective To retrospectively study the clinical features in elderly patients with epilepsy.Methods Clinical data of 72 elderly patients with epilepsy aged over 60 years from outpatients and inpatients were studied.Results In the 72 cases,cerebrovascular disease associated with epilepsy occurred in 52 cases (72.2%),with partial seizures in 50 cases (69.4%),and with generalized seizures in 22 cases (30.6%).In 38 cases with epilepsy secondary to cerebral infarction,the cerebral cortex infarction occurred in 26 cases (68.4%).In 14 cases with epilepsy secondary to cerebral hemorrhage,cerebral cortex hemorrhage occurred in 11 cases (78.6%).In interictal electroencephalogram (EEG) of the 72 cases,8 cases had normal EEG (11.1%),64 cases had abnormal EEG (88.8%).31 cases (48.4%) presented with sharp wave,spikes wave or tip--slow composite wave,33 cases (51.6%) presented with low amplitude of slow wave activity,which were non-specific abnormalities.Conclusions The main cause of seizures in the elderly is cerebrovascular disease.Partial seizures is the main type of seizure.Most of seizures caused by cerebrovascular disease occur in the region near the cortex.
6.Expression of interleukin-37 in placenta and its relationship with the pathogenesis of severe preeclampsia

Objective To investigate the expression of interleukin-37 (IL-37) in placenta tissue and its relationship with the pathogenesis of severe preeclampsia. Methods All the patients were recruited in Qilu Hospital of Shangdong University from November 2012 to November 2013. Among them, thirty patients with severe preeclampsia were assigned to the preeclampsia group, and thirty-one healthy pregnant women were assigned to the control group. Immunohistochemistry of SP was used to detect the IL-37 protein expression in placenta tissue of the two groups. The expression level of IL-37 in placenta tissue of the two groups was detected by western blot. Besides, reverse transcription(RT)-PCR was used to detect the expression of IL-37 mRNA. The correlation between the expression of IL-37 mRNA and the delivery gestational age, body mass index(BMI)was analyzed. Results (1) IL-37 were detected in the placenta of both the preeclampsia group and the control group, and the expression site mainly located in the syncytiotrophoblast, with a small amount in cytotrophoblast.(2)The expression levels of IL-37 protein in the preeclampsia group and the control group were 0.59 ± 0.39 and 0.88 ± 0.22, respectively. The IL-37 mRNA levels in the preeclampsia group and the control group were 0.55 ± 0.17 and 1.11 ± 0.21, respectively. Both decreased significantly when compared to the control group (P<0.05). (3) Significant correlation between the expression of IL-37 mRNA and the delivery gestational weeks(r=0.209, P>0.05) was seen neither in the preeclampsia group nor in the control group(r=-0.053, P>0.05). In the severe preeclampsia group, the pregnant women′s BMI had no significant correlation with IL-37 mRNA expression of placenta tissue(r=0.102, P>0.05), neither did the control group(r=-0.115, P>0.05). Conclusion IL-37 expression is significantly lower in severe preeclampsia placenta tissue than that in the normal pregnant women, which may play a protective role in the course of severe preeclampsia.
7.Relationship between folic acid metabolism-related enzyme gene polymorphism and susceptibility of abnormal pregnancy

Objective To analyze the relationship between methylenetetrahydrofolate reductase (MTHFR)C677T,A1298C and methionine synthase reductase(MTRR)gene polymorphism with abnormal pregnancy history.Methods 549 normal women (control group)and 300 women with the abnormal pregnancy history(observation group)were taken as the subjects by adopting the case control research method.The oral mucosa epithelial cells were collected for extracting genomic DNA.The MTHFR and MTRR gene polymorphisms were detected by the gene sequencing method.Results The distribution frequency of MTHFR 677TT genotype in the abnormal pregnancy group was significantly increased compared with the control group (10.00% vs.3.46%,χ2 =15.25,P <0.01);the distribution frequency of MTHFR-1298CC genotype in the abnormal pregnancy group was significantly in-creased compared with the control group (11.00 vs.4.01%,χ2 =15.66,P <0.01);the distribution frequency of MTRR A66G gen-otype had no statistical difference between the two groups(χ2 =3.02,P =0.082).The interactive analysis of 2 genes indicated that simultaneous carrying the MTHFR A1298C mutation site and MTRR A66G mutation site increased the possibility of abnormal pregnancy occurrence (OR=1.52,P =0.011).Conclusion MTHFR C677T and A1298C have a certain correlation with female ab-normal pregnancy occurrence.
8.Expression of Gas6 in placenta and decidua tissues and its relationship with the pathogenesis of preeclampsia

Objective To investigate the expression of growth arrest-specific protein 6 (Gas6) in the placenta and decidua tissues and its relationship with the pathogenesis of preeclampsia. Methods All the patients were recruited in Qilu Hospital of Shangdong University from October 2013 to June 2014. Among them, thirty-two women with early-onset severe preeclampsia who received cesarean section were assigned to the preeclampsia group, and thirty healthy pregnant women who received cesarean section were defined as the control group. Blood glucose, blood lipids, platelet count, D-dimer levels and other clinical indicators of the two groups were detected. Immunohistochemistry of SP was conducted to identify the localization of Gas6 protein in the placenta and decidua tissues. And reverse transcription (RT)-PCR was performed for quantitative analysis of Gas6 RNA expression in placentas. The correlations between placental Gas6 mRNA levels with clinical indicators were analyzed. Results (1) The gestational age at delivery, blood pressure, serum albumin, platelet count and birth weight of fetuses showed statistically significant differences between the two groups(P<0.05). (2) The Gas6 protein expressed in the cytoplasm and nucleus of the syncytiotrophoblasts and decidual cells in the placenta and decidual tissues of the two groups. (3) The Gas6 mRNA expression elevated significantly in the placenta of preeclampsia group (0.60 ± 0.38) when compared to that of the control group (0.34±0.22;P<0.05). (4) The expression of Gas6 mRNA was positively related with body mass index, diastolic blood pressure, systolic blood pressure, free fatty acids and creatinine (P<0.05), while it was negatively associated with serum albumin(P<0.05). Conclusion The high expression of Gas6 in the placenta and decidua tissues may be related to the pathogenesis of severe preeclampsia.
9.Echocardiographic determination of the prevalence of primary cardiac valve myxomatous degeneration

Objective To determine the capability of echocardiography to identify primary cardiac valve myxomatous degeneration (PCVMD) compared to pathological findings and to determine the echo features of PCVMD. Methods Echocardiograms were retrospectively compared with pathological findings of 1080 patients who underwent surgery for moderate or severe valve regurgitation. PCVMD of the mitral,aortic and tricuspid valves was retrospectively identified, with a comparison of the echocardiography and pathology findings,to calculate the prevalence of PCVMD,and to summarize its echocardiography features.Results A total of 104 patients were diagnosed with PCVMD (prevalence of 9. 62%) with 117 lesions.Valvular regurgitation were confirmed by echocardiography in all patients( 100 % ). Valve morphology change including valve prolapsed, valve thickening and redundancy were confirmed by echocardiography in 101 lesions(86.3 % ), but suggestive diagnosis were done by echocardiography in only two patients. Conclusions In patients undergoing surgery for valvular regurgitation,a high prevalence of PCVMD was found. PCVMD had distinctive echocardiographic features,suggesting its preoperative diagnosis.
10.A Study on Quality of the Precise Powder Decoction Pieces of Medicinal Flowers Lonicerae japonicae Flos

This study aimed at evaluating the quality of the precise powder decoction pieces (PPDP) of L.japonicae Flos (LJF) compared with the traditional commercial slices with chemical fingerprint methods and DNA molecular identification technology.Different specifications of PPDP were prepared,their dry extract contents were in contrast with that of commercial slices.The three batches of commercial slices were collected,and the content uniformity,fingerprint and similarity evaluation before and after the mixing and pulverization were studied by HPLC-DAD and DNA sequence alignment.As a result,the paste rate of PPDP was slightly higher than that of the traditional commercial slices.The dissolution of chlorogenic acid of PPDP was higher than that of the traditional commercial slices.RSD of inter-assay dissolutions of chlorogenic acid of commercial slices was 11.93%,which was reduced to 8.29% after mixing and preparing into PPDP.The fingerprint showed that the slimilarity of the fringerprint of the mixed and powdered LJF was elevated with 7 common peaks.All the common peaks were increased at different levels.In conclusion,compared with traditional commercial slices of LJF,PPDP apparently improved the dissolution rate and the quality uniformity,indicating that the boiled powder of CRP obviously presented vantages in clinic.
